Cytotoxic Effects of Partially Purified Substances from Bacillus polyfermenticus SCD Supernatant toward a Variety of Tumor Cell tines

  • The cytotoxic effects of partially purified substances from Bacillus polylfermenticus SCD toward a variety tumor cell lines were studied. Cytotoxic activity was determined with regard to the A549 (human lung carcinoma), AGS (human stomach adenocarcinoma), DLD-1 (human colon adenocarcinoma), HEC-1-B (human uterus adenocarcinoma), SW-156 (human kidney carcinoma), and NIH/3T3 (murine normal fibroblast) cell lines using the MTT assay. Cytotoxic substances were partially purified through Diaion HP-20 columns and extracted with methanol or other organic solvents (n-hexane, chloroform, ethylacetate, and butanol). B. polyfermenticus SCD supernatant showed up to 60% inhibition of cell viability fer all five human cancer cell lines tested. When treated with 10 mg/mL of n-hexane, chloroform, ethylacetate, and butanol extract, HEC-1-B cells showed a 25,62,35, and 63% rate of inhibition respectively, and AGS cells showed a 72, 61, 44, and 67% rate of inhibition, respectively. At a concentration of 10 mg/mL, 100% methanol Diaion HP-20 extracts showed inhibition rates of 97.0% toward A-549 cells, 98.1% toward AGS cells, 81.6% toward DLD-1 cells, 83.5% toward HEC-1-B cells, and 92.7% toward SW-156 cells. These results indicate that partially purified fractions from B. polyfermenticus SCD have the potential to inhibit not only colon cancer cells, but also lung, stomach uterus, and kidney cancer cells. Further studies are needed to characterize the cytotoxic substances released in B. polyfermenticus SCD cultures.

Evaluation on Pollution Load Characteristics and Influence of Tributaries in the Hwangguji Stream (황구지천 유역의 오염부하 특성 및 지류 영향 평가)

  • This study investigated the pollution characteristics of the main pollution zone in the Hwangguji watershed and the influence of the tributary on the main stream. The characteristics of the main pollution zone, including, the water quality index (WQI), stream rating, load duration curve (LDC), delivery load density (DLD), and contribution of the tributary to the mainstream, were evaluated by time-series visual heatmap. The WQI of the mainstream of Hwangguji was lowered to the poor (IV) level from the inflow point of Suwon stream (SW) and the LDC excess rate in the T-P was higher than that of BOD5, especially for the wet season, suggesting that management of non-point source with T-P is preferred. The contribution (%) of the tributaries in the upstream section of Hwangguji watershed were BOD5 14.54%, TOC 15.67%, T-N 5.43%, and T-P 6.97%. In particular, the Suwon sewage treatment plant located in the mainstream showed a high contribution of BOD5 (64.40%) and T-P (53.54%), respectively, due to the high discharge rate (6.019 m3/sec). Meanwhile, Sammi and Gal stream have a large impact on the mainstream with high DLD and poor WQI. Thus, both streams were considered as pollution hot spots. These results provide useful basic data for preparing more effective water quality improvement and management plans in the watershed.

In Vitro Bactericidal and Anticancer Activity of New Metabolite, ARK42, Isolated from Aspergillus repens K42

  • A novel antibacterial metabolite, ARK42, was elated from a xerophilic fungal strain K42, and Identified as Aspergillus repens based on its morphological characteristics. The metabolite exhibited antibacterial activities towards Staphylococcus aureus, Bacillus cereus, and Pseudomonas aeruginosa, with MICs of 25, 12.5, and $3.125{\mu}g/ml$, respectively, and killed Pseudomonas aeruginosa with minimal bactericidal concentration (MBC) of $12.5{\mu}g/ml$. Furthermore, anticancer activities were demonstrated against human colon cancer DLD- 1 and lung cancer LXFL529 cells with an $IC_50$ of 10 and $1{\mu}g/ml$, respectively.

Anticancer and Anti-Inflammatory Activity of Probiotic Lactococcus lactis NK34

  • The anticancer and anti-inflammatory activities of probiotic Lactococcus lactis NK34 were demonstrated. Treatment of cancer cells such as SK-MES-1, DLD-1, HT-29, LoVo, AGS, and MCF-7 cells with 106 CFU/well of L. lactis NK34 resulted in strong inhibition of proliferation (>77% cytotoxicity, p < 0.05). The anti-inflammatory activity of L. lactis NK34 was also demonstrated in lipopolysaccharide-induced RAW 264.7 cells, where the production of nitric oxide and proinflammatory cytokines (tumor necrosis factor-α, interleukin-18, and cyclooxygenase-2) was reduced. These results suggest that L. lactis NK34 could be used as a probiotic microorganism to inhibit the proliferation of cancer cells and production of proinflammatory cytokines.

  • With respect to the effectiveness of types of knowledge on human diagnostic performance, the results of several experiments claimed that training with diagnostic rules (procedural knowledge) is more effective than training that provides theoretical knowledge (principle knowledge). However, we usually have the idea that understanding the principles of system dynamics is necessary for diagnosis in some situations. In this study, we pointed out some problems in the previous experiments that force to reinterpret their experimental conclusions. Accordingly, we conducted an experiment to reinvestigate the value of theoretical knowledge in two problem situations. A simulator system, which is named DLD, that is to diagnose an electronic device was created for this purpose. It is a context-free digital logic circuit which includes forty-one gates of three basic types. Our experiment investigated the marginal effects of theoretical knowledge over common diagnostic rules. The experimental results showed that the effectiveness of the instruction in theoretical knowledge is dependent on the complexity of diagnostic situations. This adds up an experimental evidence against the presumed ineffectiveness of theoretical knowledge and forward reasoning in fault diagnosis. Furthermore, the result suggests the source of the use of theoretical knowledge.

Cytotoxic Polyacetylenes from Aralia cordata (독활의 세포독성 폴리아세틸렌 성분)

  • An n-hexane extract of the roots of Aralia cordata Thunb. (Araliaceae) was found to show significant in vitro cytotoxic activity against P388D$_{1}$ lymphocytic leukemia cell in culture. Bioactivity-directed fractionation of this extract led to the isolation of four polyacetylenes, falcarindiol (1), dehydrofalcarindiol (2), falcarindiol-8-acetate (3) and dehydrofalcarindiol-8-acetate (4). Cytotoxicity of compounds 1 and 3 was found to be better than that of compounds 2 and 4 when these compounds were tested against eight in vitro tumor cell lines, namely, A549, HCT15, DLD1, MCF7, SKOV3, HL60, K562 and P388D$_{1}$. The fact that the cytotoxicity of compounds 1 and 3 against series of tumor cell lines was much stronger than that of compounds 2 and 4 suggested that the saturated carbon chain at the termial and the hydroxyl group at the C-3 are important for the activities. The requirement for the activity was further confirmed by synthesizing and assaying the acetate derivatives of compounds 1 and 2.

Analysis of Pollution Characteristics in the Mainstream and Its Tributaries of Gongneung Stream Using Water Quality Index and Pollution Load Data (통합수질지수 및 오염부하자료를 이용한 공릉천 유입지천과 본류의 오염특성 분석)

  • In this study, we identified the major pollution-zones of the mainstream and its tributaries of Gongneung stream and investigated their pollution sources based on water quality, flowrate and pollution-load data of the stream having the characteristics of the urban-rural complex to examine the effect of the tributaries on the water quality changes in the mainstream. The water quality and flowrate data were collected for 10 months (2018 ~ 2019) at 3 points of mainstream and ten tributaries. Water quality index (WQI), load duration curve (LDC), discharge load density (DLD)and delivery ratios for each tributary were obtained so as to investigate the pollution characteristics and some of the information visualized on GIS. The main pollution-zone in the Gongneung stream was in the middle and lower streams, and the tributaries that may affect the pollution of the area were JS, JY, SL and SM. JS and SL had low WQI levels (34.7/37.5) and DLD (kg/d/㎢) of BOD and T-P were relatively high in JY (99.2/6.00) and SL (60.0/2.07). BOD and T-P delivery ratios in JS were high (0.94/0.83), suggesting that JS had significant influence on the water quality of the main pollution-zone in the Gongneung stream. Meanwhile, SM having a high T-P delivery ratio (0.97) was found to be more affected by the non-point source due to the higher LDC excess rate (%) in the low flow compared to high flow. This study provides basic data on the water quality and pollution characteristics of the Gongneung stream, and the analysis results are expected to be used as examples for identifying the main pollution-zone and tributaries of stream and their pollution sources.

Antitumor Activity of Peptide Fraction from Traditional Korean Soy Sauce

  • Antitumor activities of a peptide fraction isolated from traditional Korean soy sauce (SSP) were investigated in vitro and in vivo using cancer cell lines and F9 teratocarcinoma-bearing BALB/c mice. SSP exerted a dose-dependent antiproliferative effect on P388D1 mouse lymphoma, F9 mouse teratocarcinoma, and DLD-l human colon cancer cells with $IC_{50}$ values of 11, 50, and $50\mug/ml$, respectively. Tumor growth in F9 teratocarcinoma-bearing BALB/c mice, orally administered with 80 and 200 mg/kg/day of SSPs, was inhibited 10.3% and 52.4%, respectively, and survival days increased by 11.9% and 22.1%, respectively, compared to the control group. The results of antitumor activities exerted by SSP in vitro and in vivo suggest the feasibility of using SSP as an antitumor agent.

  • We have developed and commercialize a time-of-flight - medium energy ion scattering spectrometry (TOF-MEIS) system (model MEIS-K120). MEIS-K120 adapted a large solid acceptance angle detector that results in high collection efficiency, minimized ion beam damage while maintaining a similar energy resolution. In addition, TOF analyzer regards neutrals same to ions which removes the ion neutralization problems in absolute quantitative analysis. A TOF-MEIS system achieves $7{\times}10^{-3}$ energy resolution by utilizing a pulsed ion beam with a pulse width 350 ps and a TOF delay-line-detector with a time resolution of about 85 ps. TOF-MEIS spectra were obtained using 100 keV $He^+$ ions with an ion beam diameter of $10{\mu}m$ with ion dose $1{\times}10^{16}$ in ordinary experimental condition. Among TOF-MEIS applications, we report the quantitative compositional profiling of 3~5 nm CdSe/ZnS QDs, As depth profile and substitutional As ratio of As implanted/annealed Si, Ionic Critical Dimension (CD) for FinFET, Direct Recoil (DR) analysis of hydrogen in diamond like carbon (DLC) and InxGayZnzOn on glass substrate.
