Dithiothreitol Attenuates the Pathogenic Interaction Between Pseudomonas aeruginosa and Drosophila melanogaster

  • Infection of Drosophila melanogaster adults with Pseudomonas aeruginosa (PA14) can kill the flies within 48h. We found that the virulence of PA14 was significantly attenuated when treated with a reducing agent, dithiothreitol (DTT), prior to infection. Infection with DTT-treated PA14 elevated Metchnikowin expression at 22 h post-infection and the virulence of DTT-treated PA14 was not attenuated in Dif and Relish mutants. These results suggest that DTT pre-treatment of PA14 can aggravate certain virulence factors that may be required to paralyze fly immune responses, triggering Metchnikowin expression via Dif and Relish activations.

  • We have examined in vitro and in vivo radioprotective effects of a well-known thiol-containing compound, dithiothreitol (DTT). The treatment of both 0.5 and 1mM of DTT significantly increased clonogenic survival of ${\gamma}$-ray irradiated Chinese hamster (V79-4) cells. In order to investigate the possible radioprotective mechanism of DTT, we measured gamma-ray induced chromosome aberration by micronucleus assay. In the presence of 0.5mM or 1mM DTT, the frequencies of micronuclei were greatly reduced in all dose range examined (1.5-8 GY). Slightly higher reduction in micronucleus formation was observed in 1mM DTT-treated cells than in 0.5mM DTT-treated cells. In addition, incubation with both 0.5 and 1mM of DTT prior to gamma-ray irradiation reduced nucleosomal DNA fragmentation at about same extent, this result suggests that treatment of DTT at concentrations of 0.5 and 1mM reduced radiation-induced apoptosis. In vivo experiments, we also observed that DTT treatment reduced the incidence of apoptotic cells in mouse small intestine crypts. In irradiated control group 4.4${\pm}$0.5 apoptotic cells per crypt were observed. In DTT-administered and irradiated mice, only 2.1${\pm}$0.4 apoptotic cells per crypt was observed. In vitro and in vivo data obtained in this study showed that DTT reduced radiation-induced damages and it seems that the possible radioprotective mechanisms of action of DTT are prevention of chromosome aberration.

  • Redox signaling is one of way to regulate growth and death of cell in response to change of redox of proteins. To search whether translation is regulated by redox, we attempted in vitro translation assay under condition with or without DTT. Interestingly in vitro translation activity was increased up to 40% In the presence of dithiothreitol (DTT). Then we checked whether this positive effect by DTT was further accelerated by addition of thioredoxin (Trx). When a Trx purified from Saccharomyces cerevisiae was added to the in vitro translation extract, we observed a dose-dependent increase in translational activity. These results suggest the possibility of translation factors being redox-regulated via Trx in vivo.

  • Mononitrotoluenes were reduced to aminotoluenes using porphyrin as a catalyst in the presence of several types of reductants including hydrogen sulfide and 1, 4-dithiothreitol(DTT). Intermediates and final products of porphyrin-catalyzed reduction of mononitrotoluenes were identified and a pathway for the reduction of the nitro group to the corresponding amino group was proposed. The optimum pH for the reduction was determined. The catalytic activity of the porphyrin was confirmed by UV/VIS absorption spectra and basic kinetics of porphyrin-catalyzed reduction were studied. Of several types of reductants tested, DTT sodium hydrosulfite, and hydrogen sulfide were seen to give significant reduction of nitrobodies. When hydrogen sulfide was used as a reductant hydrogen sulfide and nitrotoluenes were removed simultaneously.

Effects of Magnetic Pole on Orientation of Bull Sperm Treated by Dithiothreitol or Heparin

  • Effect of the north (N) or south pole (S) of a magnet and dithiothreitol (DTT) or heparin sodium (H) on the orientation of bull sperm were studied. Sperm were collected from four Japanese Black bulls of Okinawa Prefectural Livestock Experimental Station were treated with various concentrations of DTT or H during the first and sixth days and were then exposed them to N or S with a magnetic field of 3,000 Gauss for 24 hours. Experimental results showed that both N and S significantly increased the perpendicular oriented percentage of bull sperm in most treatments not treated or treated with DTT or H. Anisotropy of magnetic field of bull sperm was the main cause which induced them to orient perpendicular to the magnetic direction. The results of the experiment also showed that, in most cases, the two polarities had the same effect on the orientation of bull sperm. However, in some cases, N exerted a stronger effect on the perpendicular percentage of bull sperm than S, such as with the 20 mM DTT treatment at the first and sixth days and the 100 units H treatment at the sixth day. These cases showed that the perpendicular oriented percentage of bull sperm at N was significantly higher than that of S. It was only the 100 units heparin treatment at S both at the first and sixth days that upward perpendicular orientation was higher than that at N. This might be due to the fact that protamin-DNA structure of a small number of bull sperm nuclei slightly changed because of heparin action.

  • A new simplified procedure for identifying human plasmin was developed using a DTT copolymerized agarose stacking gel (ASG) system. Agarose (1%) was used for the stacking gel because DTT inhibits the polymerization of acrylamide. Human plasmin showed the lowest activity at pH 9.0. There was a similar catalytically active pattern observed under acidic conditions (pH 3.0) to that observed under alkaline conditions (pH 10.0 or 11.0). Using the ASG system, the primary structure of the heavy chain could be established at pH 3.0. This protein was found to consist of three fragments, 45 kDa, 23 kDa, and 13 kDa. These results showed that the heavy chain has a similar structure to the autolysed plasmin (Wu et al., 1987b) but there is a different start amino acid sequence of the N-termini.

  • Artificial activation of oocytes is a prerequisite for the successful cloning by nuclear transfer. This study investigated the effect of the different combination of activation agents such as electric pulse (E), thimerosal (Thi) + dithiothreitol (DTT), 6-dimethylaminopurine (6-DMAP) or cycloheximide (CH) on the developmental ability of porcine embryos derived from parthenogenetic activation (PA). PA embryos activated with chemicals showed significantly higher developmental rate to the blastocyst stage compared to the embryos activated with E alone ($21.5{\sim}28.1%$ vs. 18.0%, respectively). Of chemicals, Thi + DTT supported higher development to the blastoryst stage (28.1%). There was no significant difference in 1 pronucleus (PN) formation rate $(59.9{\sim}64.7%)$, but 2PN formation rate was significantly higher in PA embryos with additional activation using chemicals $(7.2{\sim}9.7%)$. In conclusion, this study shows that chemical activation after electric pulse can increase the development of porcine PA embryos.

Protection of Mercury induced Acute Respiratory Injury by Inhaled Oxidizing Agent (수은에 의한 급성호흡손상시 산화물질의 억제효과)

  • Mercury vapor inhalation-induced acute respiratory failure(ARF) has been reported to be fatal. This study was designed to observe the possible mechanism of inhaled mercury vapor poisoning in the respiratory system. Sixty percent of rats(12/20) exposed to mercury vapor were dead within 72 hours of exposure whereas all the rats(20/20) exposed to mercury vapor combined with dithiothreitol(DTT) vapor survived. The histological observation showed that ARF was a direct cause of the death induced by mercury vapor inhalation, which was significantly circumvented by DTT vapor. Cyclic AMP mediated chloride secretion was inhibited by luminal side but not serosal side sulfhydryl blocking agents (Hf$^{2+}$ $\rho$-chloromercuribenzoic acid or $\rho$-chloromercuriphenyl sulfonic acid) in a dose-dependent manner in a primary cultured rat airway monolayer. The inhibitory component of cAMP induced chloride secretion was completely restored by luminal side DTT(0.5mM). these results suggest that the oxidized form(Hg$^{2+}$) of mercury vapor(Hg0) contribute to ARF and subsequent death. The finding is important as it can provide important information regarding emergency manipulation of ARF patients suffering from by mercury vapor poisoning.ing.

Reaction Mechanism of Purine Nucleoside Phosphorylase and Effects of Reactive Agents for SH Group on the Enzyme in Saccharomyces cerevisiae (Saccharomyces cerevisiae에서 얻은 Purine Nucleoside Phosphorylase의 반응기작과 효소에 대한 Sulfhydryl Reagent의 영향)

  • Kinetic analysis was done to elucidate the reaction mechanism of purine nucleoside phosphorylase (PNP) in Saccharomyces cerevisiae. The binary complexes of PNP${\cdot}$phosphate and PNP${\cdot}$ribose 1-phosphate were involved in the reaction mechanism. The initial velocity and product inhibition studies demonstrated were consistent with the predominant mechanism of the reaction being an ordered bi, bi reaction. The phosphate bound to the enzyme first, followed by nucleoside and base were the first product to leave, followed by ribose 1-phosphate. The kinetically suggested mechanism of PNP in S. cerevisiae was in agreement with the results of protection studies against the inactivation of the enzyme by sulfhydryl reagents, p-chloromercuribenzoate (PCMB) and 5,5'-dithiobisnitrobenzoate (DTNB). PNP was protected by ribose 1-phosphate and phosphate, but not by nucleoside or base, supporting the reaction order of ordered bi, bi mechanism. PCMB or DTNB-inactivated PNP was totally reactivated by dithiothreitol (DTT) and the activity was returned to the level of 77% by 2-mercaptoethanol, indicating that inactivation was reversible. The kinetic behavior of the PCMB-inactivated enzyme had been changed with higher $K_m$ value of inosine and lower $V_m$, and was restored by DTT. Inactivation of enzyme by DTNB showed similar pattern of K sub(m) value with that by PCMB, but had not changed the $V_m$ value, significantly. Negative cooperativity was not found with PCMB or DTNB treated PNP at high concentration of phosphate.

  • A technique for detection of Bacillus in soil and waste water treatment system was developed. Mixed col- tured solutions of 22 Bacillus strains were applied for selection of probe and pretreatment method for FISHAmong the three probes known as useful tool for FISH method,S-G-Bacill-0597-a-A-22 was best for detec-tion of Bacillus. Ninety five percent of DAPl count was observed with FISH method with S-G-Bacill-0597-a-A-22 probe. For increasing the permeability to Bacillus cell walls, pretreatment with lysozyme was betterthan that with lysozyme and SDS/DTT (dithiothreitol). Bacillus spore was not detected with FISH. So, Bacil-lus detection in ecosystem requires FISH with pretreatment of lysozyme and spore staining.
