Controlled Release of Isonicontinic Acid Hydrazide from the Membrane-Coated Tablet

  • Membrane-coated tablet of isonicotinic acid hydrazide (INAH) which releases INAH at the zero-order kinetics was deveoped. It consisted of a soluble tablet core surrounded by a porous membrane which controls the diffusion rate. Tablet cores were prepared by compressing granules of INAH and polyvinyl chloride (PVC) dissolved in methyl ethyl ketone in which micronized sucrose were suspended. Diffusion rate of INAH from the tablet through the membrane was constant until the loaded INAH in the core was almost released. The rate was independent of pH of the dissolution medium. Water-soluble sucrose particles behaved as a poreproducing material in the water-insoluble PVC film coat. The pH independency of the rate was probably due to the high solubility of INAH in the water of wide pH range. The diffusion rate of INAH could be controlled by chnaging the composition of the membrane or the coat weight. This membrane-coated INAH tablet seemed to be a powerful candidate for the controlled release drug delivery system (DDS) of INAH or other highly watersoluble drugs.

Nb 석출 거동을 고려한 저탄소강의 상변태 모델 (A model for Phase Transformation of Microalloyed Low Carbon Steel Combined with Nb Precipitation Kinetics)

  • The dissolution and precipitation of Nb, which has been known as strong carbide-forming element, play a key role in controlling phase transformation kinetics of microalloyed steels. In this study, we analyzed both numerically and experimentally the precipitation behavior of Nb-microalloyed steel and its effect on the austenite decomposition during cooling. Nb precipitation in austenite matrix could be predicted by the thermo-kinetic software MatCalc, in which interfacial energy between precipitate and matrix is calculated. The simulated precipitation kinetics fairly well agrees with the experimental observations by TEM. Austenite decomposition, which is strongly affected by Nb precipitation during cooling, was measured by dilatometry and was modeled on the basis of a Johnson-Mehl-Avrami-Kolmorgorov(JMAK) equation. It was confirmed that the dissolved Nb delays the austenite decomposition, whereas, the precipitated Nb accelerates phase transformation during the austenite decomposition.

  • The interrelation into nucleation and thermodynamic parameters of solutions has been established by plotting of various dependencies: the enthalpy of dissolution, solubility product and super-solubility on ionic salt radii and also the extent of deviation from an ideal Debye -Huckel model of electrolyte solution on solubility product. The possible methods of perfect crystal growth from aqueous solution have been found a priori by separating of known set of pair values of solubility and super-solubility into no less than six-nine characteristic and distinctive sub-sets.

가바펜틴을 함유한 위체류성 정제의 제조 및 용출 평가 (Preparation and Dissolution Characteristics of A Gastro-Retentive Tablet System Containing Gabapentin)

  • The objective of this investigation was to develop a gastro-retentive(GR) dosage form of gabapentin and was to evaluate of its dissolution characteristics. GR tablet consists of expandable core tablet matrix and semi-permeable membrane coating. Poloxamer 407 and sodium bicarbonate were used to prepare the core matrix. Polyvinly acetate dispersion (Kollicoat $SR30D^{(R)}$) and polyvinyl alcohol-polyethylene glycol copolymer ((Kollicoat $IR^{(R)}$)) were employed to form the semi-permeable membrane. The GR tablets significantly expanded up to fivefold in simulated gastrointestinal fluids with no apparent damage of the coating membrane over 12 hours. Also, the swelling rate was controllable with the amount of sodium bicarbonate. The drug release was observed to be substantially sustained based on coating level. The release rate of gabapentin from the GR tablet was gradually slowed down as the coasting amount was increased. The gabapentin GR tablet with 8% coating level showed a pseudo-zero order release kinetics over 12 hours. These results suggest that this swellable GR tablet system having semi-permeable membrane coating can be applicable for hydrophilic drug substances like gabapentin.

Butolphanol tartrate 함유 구강점막 점착성 [P(AA-co-PEGMM)] 공중합체 필름의 평가 (Evaluation of buccal mucoadhesive [P(AA-co-PEGMM)] copolymer films containing butorphanol tartrate)

  • The mucoadhesive characteristics of [P(AA-co-PEGMM)] films by estimating the glass transition temperature $(T_g)$, analyzing surface energy and studying FT-IR was previously reported. In this study, the possibility of buccal mucoadhesive dosage form of [P(AA-co-PEGMM)] films by mucoadhesive force measurements and dissolution tests were also investigated. Mucoadhesiveness of [P(AA-co-PEGMM)] films was compared with cr-PAA and cr-PEGMM films crosslinked with 3% ethyleneglycol dimethacrylate (EGDMA). The buccal mucoadhesive force of [P(AA-co-PEGMM)] films increased with increasing content of PEGMM. [P{AA-co-PEGMM (18 mole%)}] films showed a significantly greater mucoadhesiveness than cr-PAA and cr-PEGMM films. The mucoadhesive force measured in normal saline (pH 5.0) was higher than that measured in phosphate buffer (pH 6.8) because of the pH dependence of hydrogels with carboxyl ions within the PAA. Moreover, the mucoadhesive force of [P{AA-co-PEGMM (18 mole%)}] films was at maximum after 2 hr attachment of buccal mocosa and it was maintained over $1\;N/cm^2$ for up to 10 hr. In dissolution studies, the release of butorphanol tartrate from [P(AA-co-PEGMM)] films increased with increasing PEGMM content, and films prepared with 18 mole% PEGMM gave almost zero order release kinetics.

V-Nb Microalloyed 강의 오스테나이트역에서 복합 탄질화물의 재용해 및 석출 거동 (Dissolution and Precipitation behaviors of Complex Carbonitrides in Austenite of a V-Nb Microalloyed Steel)

  • Dissolution and precipitation behaviors of complex carbonitrides in austenite of a V-Nb microalloyed steel were quantitatively examined through electrical resistivity measurement and transmission electron microscopy. The electrical resistivity increased with solution treatment temperature up to $1240^{\circ}C$ and then was saturated at $225n{\Omega}m$ for a holding time of 10 min. The electrical resistivity method was also used to quantitatively measure the isothermal precipitation kinetics of the complex carbonitrides in austenite. Nb-rich precipitates were observed in austenite at the early stages of precipitation, but Nb was replaced by V up to the equilibrium amount within the precipitates with further holding time. The time-temperature-precipitation diagram showed a C-type curve; nose temperature and its incubation time were $900^{\circ}C$ and 100 s, respectively.

카르베딜롤을 함유하는 경구제어 방출형 제제의 제조 및 용출특성 (Preparation and Dissolution Properties of Oral Controlled Release Formulation Containing Carvedilol)

  • 고혈압 치료제인 카르베딜롤을 모델약물로 하여 새로운 서방성 방출 제어형 매트릭스 정제를 제조하기 위하여 소수성 서방성 부형제인 Compritol 888 ATO와 친수성 고분자인 hydroxypropyl methyl cellulose (HPMC) 또는 polyethylene oxide (PEO)를 이용하여 방출 제어형 매트릭스 정제를 제조하였다. 카르베딜롤 방출 제어형 매트릭스 정제의 제조 시 Compritol 888 ATO의 비율과 친수성 고분자의 종류 및 비율, hot melt coating coagglutination (HMCC) rocess의 적용 유무에 따른 카르베딜롤의 방출 양상을 위하여 용출시험기를 사용하여 pH 1.2의 인공위액과 pH 6.8의 인공장액에서 24시간 동안 $37^{\circ}C$, 50 rpm으로 용출시험을 실시하였다. 그 결과, HMCC process를 적용한 모든 제제가 약물의 방출 제어에 매우 효과적인 것을 확인하였다. 또한 소수성 서방성 부형제인 Compritol 888 ATO의 비율에 따라 약물의 방출 양상 및 시간이 기존 일반정제에 비하여 약 95%의 용출률을 나타내었으며 24시간까지 지연됨을 확인할 수 있었다.

광미-물 상호반응에서 반응시간이 중금속 용출에 미치는 영향 (The Effects of Kinetics on the Leaching Behavior of Heavy Metals in Tailings-Water Interaction)

  • 이번 연구는 광미시료를 대상으로 반응시간(1, 2, 4, 7, 14, 21 및 30일)과 반응용액의 pH(1, 3 및 5)를 달리하여 실내 용출실험을 실시하였다. pH 5와 pH 3의 반응용액과의 용출실험에서는 반응 2일 후 최종 pH가 4.6-6.1 혹은 2.8-3.5로 안정화되었으며, 반응시간이 지남에 따라 점진적으로 낮아졌다. 황산염 농도와 산도는 7일 이후 증가하는 것이 관찰되었는데, 이는 황화광물의 산화작용때문인 것으로 판단된다 pH 5와 pH 3에서 용출된 Pb 함량은 반응기 간(1-30일) 동안 뚜렷한 변화가 보이지 않은 반면에 Zn, Cd 및 Cu는 시간이 지남에 따라 용출함량이 증가하는 경향을 보였다. 반응용액 pH 1에서의 용출특성은 Zn 형태(Zn, Cd, 및 Cu)와 Pb 형태(Pb)로 구분되었다. Zn 형태는 시간에 따라 용출함량이 증가되었고, 이는 지속적인 용해작용에 의한 것이다. Pb 형태는 시간에 따라 용출농도가 감소하였으며, 이는 황산염의 용출함량이 높아짐에 따라 용해된 Pb가 앵글레사이트(anglesit)로 침전됨을 지시한다. 높은 황산염 농도는 Fe, Zn, 및 Cd의 높은 용출함량과 밀접한 관련이 있다. 이들 용출실험 결과 Zn과 Cd의 용출과 산도는 광산주변 환경의 지표수와 지하수 수질에 악영향을 주는 요인이 될 수 있다. 용출반응에서 kinetics 문제는 광미가 빗물과 반응하여 유출수에서의 중금속 농도를 증가시킬 수 있는 중요한 요인이 된다.

HF-HCl 혼합 용액에서 $EAGLE^{2000TM}$ LCD 유리의 식각에 관한 연구 (A Study on Etching of $EAGLE^{2000TM}$ LCD Glass by HF-HCl Mixed Solutions)

  • 본 연구는 2.5MHF-xMHCl$(x:0\sim8)$ 혼합 용액에서 $EAGLE^{2000TM}$ LCD 유리의 식각에 관한 것이다. 유리의 용해공정에서 율속단계는 HF를 함유한 식각액과 유리와의 반응이었다. 그 증거로는 유리의 두께는 시간에 따라 직선적으로 감소하였고, 식각속도는 교반정도에 무관하게 일정하였으며, 활성화 에너지가 $35\sim45$ kJ/mol 범위였다는 점이다. 순수한 HF만 사용했을 때 보다 HCl이 첨가되면 식각속도는 증가하였고. 그 속도는 HCl 첨가량에 비례하여 증가하였다. 또한 HCl 첨가시 식각 후의 표면은 식각 전과 유사하였다. 이는 HCl 첨가에 의해 $H_{3}O^{+}$ 이온의 농도가 증가하여 각종 불화물의 용해도를 증가시키고, $H_{3]O^{+]$ 이온의 흡착은 Si-O 결합의 결합력을 감소시키기 때문에 일어난 현상으로 사료된다.

이온교환수지를 이용한 새로운 오메프라졸 복합체 개발 (Development of New Omeprazole-lon Exchange Resin Complex)

  • Omeprazole(OMZ)-cholestyramine(CHL) and various OMZ-Dowex resin complexes were prepared by reaction between OMZ and activated resins in 0.1N NaOH solution. And their physical properties were tested by means of infrared(IR), differential scaning caloimeter(DSC), X-ray diffraction. Chemical stability of OMZ-CHL was increased markedly compared with OMZ and the decomposition of OMZ-CHL followed the pseudo first-order kinetics and the rate constants were $2.743{\times}10^{-4}/day$ at $20^{\circ}C$, $7.83{\times}10^{-3}day^{-1}$ under 80% RH and $1.68{\times}10^{-2}day^{-1}$ under UV radiation, respectively. On the other hand, the rate constants of OMZ were $2.996{\times}10^{-4}day^{-1}$ at $20^{\circ}C$, $1.17{\times}10^{-2}day^{-1}$ under 85% RH, and $4.07{\times}10^{-2}day^{-1}$ under UV radiation, respectively. The rates of dissolution of OMZ-CHL bulk and OMZ-CHL tablet were 100% and more than 85% in 15 minutes, respectively, which were increased than OMZ base and OMZ-tablet. In the acute toxicological test, the value of oral $LD_{50}$(mouse) was 4.608 g/kg. OMZ-CHL was pelletized using lactose, polyethyle neglycol(PEG), D-sorbitol, Avicel PH 101, sodium laurylsulfate and polyvinylpyrrolidone(PVP) K-30, and enteric coated with HPMCP, Myvacet, acetone, ethanol and cetanol, of which dissolution rate was found to be more than 85% in 10 minutes. From the above results, it was found that OMZ-CHL is a useful means for development of new oral dosage forms of OMZ.
