  • A directed triple system of order ν, denoted by DTS(ν), is said to be $textsc{k}$-near rotational if it admits an automorphism consisting of exactly three fixed elements and $textsc{k}$ cycles of length (ν-3)/$textsc{k}$. In this paper, we obtain necessary and sufficient conditions for the existence of $textsc{k}$-near rotational DTS(ν)s for every positive integer $textsc{k}$.

  • An extended directed triple system of order v, denoted by EDTS(v), is a pair (V, (equation omitted)) where V is a v-set and (equation omitted) is a set of transitive triples of elements of V such that every ordered pair of elements of V is contained in exactly one member of (equation omitted). We obtain a necessary and sufficient condition for the existence of cyclic EDTS(v)s, and when k=1 or 2, we also obtain a necessary and sufficient condition for the existence of k-rotational EDTS(v)s.

  • Hydroxyproline (HYP) is a post-translational product of proline hydroxylation catalyzed by an enzyme prolyl 4-hydroxylase which plays a crucial role in the synthesis of all collagens, because the 4-hydroxyproline residues are essential for the folding of the newly synthesized collagen polypeptide chains into triple-heical molecules. Considering the role of collagen and its significance in many clinically important diseases such as liver cirrhosis, a great deal of attention has been directed toward the development of an assay at cell-based system.   • The CRISPR-Cas system is the adaptive immune system in bacteria and archaea against invading phages or foreign plasmids. In the type II CRISPR-Cas system, an endonuclease Cas9 cleaves DNA targets of phages as directed by guide RNA comprising crRNA and tracrRNA. To avoid targeting and destruction by Cas9, phages employ anti-CRISPR (Acr) proteins that act against host bacterial immunity by inactivating the CRISPR-Cas system. Here we report the backbone $^1H$, $^{15}N$, and $^{13}C$ resonance assignments of AcrIIA4 that inhibits endonuclease activity of type II-A Listeria monocytogenes Cas9 and also Streptococcus pyogenesis Cas9 using triple resonance nuclear magnetic resonance spectroscopy. The secondary structures of AcrIIA4 predicted by the backbone chemical shifts show an ${\alpha}{\beta}{\beta}{\beta}{\alpha}{\alpha}$ fold, which is used to determine the solution structure.

Single-Base Genome Editing in Corynebacterium glutamicum with the Help of Negative Selection by Target-Mismatched CRISPR/Cpf1

  • CRISPR/Cpf1 has emerged as a new CRISPR-based genome editing tool because, in comparison with CRIPSR/Cas9, it has a different T-rich PAM sequence to expand the target DNA sequence. Single-base editing in the microbial genome can be facilitated by oligonucleotide-directed mutagenesis (ODM) followed by negative selection with the CRISPR/Cpf1 system. However, single point mutations aided by Cpf1 negative selection have been rarely reported in Corynebacterium glutamicum. This study aimed to introduce an amber stop codon in crtEb encoding lycopene hydratase, through ODM and Cpf1-mediated negative selection; deficiency of this enzyme causes pink coloration due to lycopene accumulation in C. glutamicum. Consequently, on using double-, triple-, and quadruple-base-mutagenic oligonucleotides, 91.5-95.3% pink cells were obtained among the total live C. glutamicum cells. However, among the negatively selected live cells, 0.6% pink cells were obtained using single-base-mutagenic oligonucleotides, indicating that very few single-base mutations were introduced, possibly owing to mismatch tolerance. This led to the consideration of various target-mismatched crRNAs to prevent the death of single-base-edited cells. Consequently, we obtained 99.7% pink colonies after CRISPR/Cpf1-mediated negative selection using an appropriate single-mismatched crRNA. Furthermore, Sanger sequencing revealed that single-base mutations were successfully edited in the 99.7% of pink cells, while only two of nine among 0.6% of pink cells were correctly edited. The results indicate that the target-mismatched Cpf1 negative selection can assist in efficient and accurate single-base genome editing methods in C. glutamicum.

A Study Security Measures for Protection of VIP in the G20 Summit (G20 정상회의 시 주(主)행사장에서의 VIP 안전대책 방안에 관한 연구)

  • The threat factors available for occurrence given G20 Summit Meeting are expected leader terrorism, hostage terrorism, bomb terrorism, public facilities terrorism, and aircraft terrorism. As for the threat groups, which are expected in Korea, the North Korea, Islam extremist group, and the group such as NGO organization of being opposed to international meeting are regarded as having possibility of causing hazard. Thus, the purpose of this study is to suggest VIP Security-measure plans in the main site in preparation for G20 Summit Meeting. Accordingly, each country in the world is adopting 'the principle of Triple Ring' in common. Thus, it elicited a coping plan by 1st line(inner ring) 2nd line(middle ring) 3rd line(outer ring) based on this principle, and proposed even an opinion together that will need to be reflected in light of policy for the VIP security measures. In conclusion, as for the VIP Security-measure plans in the main site in preparation for G20 Summit Meeting, In the inner ring(safety sector), first, an intercepting measure needs to be devised for a spot of getting into and out of vehicles given the Straight Street. Second, the Walking Formation needs to be reinforced boldly in the exposed area. In the middle ring(security sector), first, the control plan needs to be devised by considering particularity of the main site. Second, there is necessity for adopting the efficient security badge operation plan that is included RFID function within security badge. In the outer ring(aid protective sector), first, there is necessity of preparing for several VIP terrorisms, of collecting information and intelligence, and of reinforcing the information collection system against terrorism under the cooperation with the overseas information agency. Second, the urgent measure training in time of emergency needs to be carried out toward security agent event manpower. Third, to maintain the certain pace in VIP motorcade, the efficient traffic control system needs to be operated. Finally, as for what will need to be reflected in light of policy for VIP security measures, first, there is necessity for allowing VIP residence to be efficiently dispersed to be distributed and controlled. Second, there is necessity for allowing impure element to misjudge or attack to be failed by utilizing diverse deception operations. Third, according to the reorganization in North Korea's Organization of the South Directed Operations, the powerful 'military-support measure' needs to be driven from this G20 Summit Meeting. For this, the necessity was proposed for further reinforcing the front back defense posture under the supervision of the Ministry of National Defense and for positively coping even with detecting and removing poison in preparation for CBR (chemical, biological, and radio-logical) terrorism.

The Revision of the Rules of the Workers' Party of Korea and the Organizational Changes of the 'Monolithic Guidance System of the Party Core': Focusing on Party-Government-Military Relations in Kim Jong Un Regime (조선노동당 제8차대회 당규약 개정과 '당중앙의 유일적 영도체계'의 조직적 변화: 김정은 정권의 당정군관계를 중심으로)

  • The Rules of the Workers' Party of Korea (WPK), revised at the eighth Party Congress in 2021, reflect the Kim Jong Un regime's changes in strategic lines and ideological justifications on North Korea's socialism and communism, and its recent stances against the external environment. Moreover, they contain critical changes in the party's organizational system encompassing the central and the provinces. This study explores the organizational changes of the "monolithic guidance system of the party core" stipulated by the new party rules in January 2021, based on the analysis of the entire nine revised rules of the WPK since 1945. In the 2021 Party Congress, the Kim Jong Un regime, which officially came to power after the fourth Party Conference in 2012, has institutionalized the monolithic guidance system centered on the party core, or the head of state, Kim Jong Un. The newly set leadership and execution system, which reorganized party, government, and military organizational structure and accompanied the relevant personnel changes, was derived from the attempts for reinstating the Kim Jong Un regime as a more normalized party-state structure before its 10th year in power in April 2022. The "monolithic leadership system of the party core" established a system of "organizational leadership" through the organization of the Central Committee, directed by the Party Head, or General Secretary. The institutionalization of the new system resulted from the ten-year development of the revival of the party-state structure, which compromised the status of the military and reconfirmed the party's control of the military. This study explains the new system from the perspectives of both institutionalization and top-down unity, shedding light on the new party-military-government relations of the Kim Jong Un regime. The analysis contributes to a better understanding and forecasting of the Kim Jong Un regime's governance, which currently strengthens the monolithic leadership system as a crisis management system in the face of the "triple hardships" of sanctions, Covid and disaster.
