Studies on the Bioavailability of Berberine Preparations (베르베린 제제의 생체 이용율에 관한 연구)

  • In order to measure the bioavailability of Coptidis Rhizoma preparations, berberine was extracted by the mixture (1:1 v/v) of methanol and water from Coptidis Rhizoma and formulated into pills with some excipients. Disintegration time, dissolution rate, intestinal absorption rate, antibacterial effect, and serum concentration were measured. All pills were disintegrated about 60 minutes in gastric and intestinal juices and disintegration time did not change upon storage at $40^{\circ}C$ for 30 days. The pills manufactured using the extract which was extracted with the solvent containing dilute sulfuric acid showed faster dissolution and absorption from the small intestine than the other samples. It also showed not only the largest area under the serum concentration time curve and relative bioavailability, but also more potent antibacterial effect against gram-positive and gram-negative bacterias than the other samples. The correlation of the dissolution rate with absorption rate constant or $t_{max}$ showed a significant linear relationship.

  • This study explored the feasibility of applying Electrokinetic-Fenton process(EK-Fenton process) to remediation of contaminant sorbed on the soil possessed low-permeability. The addition of 0.01 N H$_2$SO$_4$ in the anode reservoir for the $H_2O$$_2$stabilization improved the stabilization of $H_2O$$_2$and the treatment effect of phenanthrene across the entire soil specimen. The use of $H_2O$$_2$and dilute acid as anode purging solution is a promising method treating of HOCs in low-permeability subsurface environments.

  • Fibrous adsorbents containing amidoxime group which make chelate complexes with uranyl ions are studied for the recovery of uranium from sea water. Acrylic fibers are used as base Polymer. The adsorption properties of uranium are carried out to examine pH effect, concen-tration dependence, adsorption rate, separation, and chelate complex. The results obtained are as follows; 1. Metal capacity of U (VI) ion is in the range of pH $2\~10.2$. Amidoxime group-containing fiber recover U (VI) ions existed in sea water or waste water in extremely small quantities. 3. Using amidoxime group-containing fiber Cu (II) and U (VI) are separated with each other in dilute nitric acid solution (pH 2.3). 4. U (VI) chelate complexes are conformed by tridendate ligands, which are coordinated with one nitrogen and two oxygens onto amidoxime group-containing fiber.

  • Novel amphiphilic, thermo-responsive polyaspartamides which showed both LCST (lower critical solution temperature), and sol-gel transition were prepared and characterized. The polyaspartamide derivatives were synthesized from polysuccinimide, the polycondensate of aspartic acid monomer, via successive nucleophilic ring-opening reaction by using dodecylamine and N-isopropylethylenediamine (NIPEDA). At the intermediate composition ranges, the dilute aqueous solution exhibited a thermally responsive phase separation due to the presence of LCST. The phase transition temperature was controllable by changing the content of pendent groups. In addition, a physical gelation, i.e. the sol-gel transition was observed from the concentrated solutions, which was elucidated by dynamic viscoelastic measurements. These novel injectable and thermo-responsive hydrogels have potential for various biomedical applications such as tissue engineering and current drug delivery system.

Effective test of lacquer in marine diesel engines

  • We perform an experiment on lacquer formation with simple test device. The anti-lacquer is one of important issues to increase durability, and to improve performance in the engines because the lacquer formation cause sticking of fuel injection pump, scuffing of cylinder liners, and increase of lubricant oil consumption in the marine diesel engines. We suggest this simple test in order to save enormous experimental cost in marine diesel engines, and in order to have ease in performing the various tests. The influences of the Base Number (BN) of lubricant oils and the sulfur content of fuel oils in the formation of lacquer are investigated. In order to investigate physical and chemical properties of lacquer, we perform a variety of tests such as, visual inspection, EDS. In addition, we investigate adhesion of lacquer by pull-off test quantitatively, and perform dissolution test with dilute sulfuric acid.

Fluorescence and Laser Light Scattering Studies of Modified Poly(ethylene-co-methylacrylate0 Ionomers on the Formation of Stable Colloidal Nanoparticles in Aqueous Solution

  • Fluorescence and dynamic light scattering measurements were applied to the study of formation and structure of aggregated colloidal particles in modified poly(ethylene-co-methylacrylate) ionomers in aqueous solution. Both 8-anillino-l-naphthalene-sulfonic acid (ANS) and pyrene were used as fluorescence probe to obtain the information on the structure of particle surface and inside, respectively. Three different ionomers used in this study started to aggregate at very dilute concentration, 3-8 x 10-6 g/mL. In this study, we demonstrate that the polyethylene ionomers can form stable nanoparticles. The hydrophobic core made of the polyethylene backbone chains is stabilized by the ionic groups on the particle surface. Such a formed stable nanoparticles have a relatively narrow size distribution with an average radius in the range of 27-48 nm, depending on the kind of ionic groups. Once the stable particles are formed, the particle size distributions were nearly constant. This study shows another way to prepare surfactant-free polyethylene nanoparticles.

Synthesis and Optical Properties of Poly[6'-(N-carbazolyl)hexyl-2-ethynylpyridinium bromide]

  • The polymerization of 2-ethynylpyridine by alkyl bromide carrying carbazole moiety, 6-(N-carbazolyl)hexyl bromide, relatively proceed well to give the corresponding poly[6'-(N-carbazolyl)hexyl-2-ethynylpyridinium bromide) in high yields under DMF reflux conditions without any initiator or catalyst. This polymerization was influenced upon the initial monomer concentration. The polymer yields and inherent viscosities of the resulting polymers were in the range of 34-85% and 0.11-0.21 dL/g, respectively. Instrumental analyses using NMR, IR, and UV-visible spectroscopies and elemental analysis indicated that the resulting polymer has a conjugated polymer backbone system carrying pyridine and n-hexyl carbazole moiety. The polymers were mostly brown powders and completely soluble in DMF, DMSO, nitrobenzene, and formic acid. The photoluminescence spectrum of the dilute polymer solution with the excitation at 383 nm exhibited two sharp peaks at 495 and 540 nm.

  • Microanalysis of organic sulfur compounds by means of combining thin layer chromatography and oxygen combustion flask method was attempted. The following procedure was found to be very simple and to give accurate results. The mixture of sulfa drugs was separated with T.L.C., and it was burned in a flask filled with oxygen, and the gas formed was absorbed in a dilute solution of sodium hydroxide. The solution was neuralized with hydrochloric acid and heated in a water bath. The sulfate ion formed was then treated with barium chromate solution and its absorbancy at 370 m.mu. was measured.

Evaluation of the degree of maturity of Chinese cabbage blades and midribs pretreated with dilute acetic acid solutions during Kimchi fermentation (초산 전처리 배추김치의 발효중 엽신 및 중륵별 김치숙성도 평가)

  • Effect of the pretreatment with acetic acid(0.01%, 0.05%, 0.1%) on the fermentation of Chinese cabbage Kimchi, and differences in the degree of maturity between Chinese cabbage blades and midribs were observed. To estimate the degree of maturity, correlation analysis was used with several variables such as pH, titratable acidity, volatile acidity, ascorbic acid content, resazurin test value and pH/acidity ratio. Based on the pH and titratable acidity, the degree of maturity of Chinese cabbage midribs fermented at 10$^{\circ}C$ until the optimum stage of fermentation increased more rapidly than those of the blades. The stage of maturity of Kimchi pretreated with 0.01% acetic acid were slightly prolonged, compared to those pretreated with acetic acid at the higher levels. The pH of Chinese cabbage midribs and blades pretreated with acetic acid was lower than those of control between 2nd and l0th day of fermentation. The volatile acidity of Chinese cabbage midribs and blades increased proportionally with the concentration of acetic acid. The ascorbic acid contents of Chinese cabbage midribs showed higher than those of Chinese cabbage blades. During the fermentation, the higher concentration of acetic acid was used, the less ascorbic acid content was remained. The pH/acidity ratio, volatile acidity, ascorbic acid content and resazurin test value of Chinese cabbage midribs and blades during Kimchi fermentation at 10$^{\circ}C$ showed good correlations with the pH and titratable acidity.

  • EHT and CNDO/2 calculations have been performed to determine conformations of acetamides and diacetamides, and of their protonated forms. Results show that: protonation is always favored on the cis position with respect to N due to greater attractive potential between $H^+$ and N; the trans-trans conformer of diacetamides gives the most preferred protonated form although the cis-trans conformer is the most stable one for the unprotonated diacetamides. Protonation on a carbonyl oxygen is predicted to increase both charge and orbital controlled $S_N$SN reactivities of the protonated carbonyl carbon due to increases in positive charge and AO coefficient of ${\pi}$-LUMO of the carbon atom. In the acid catalyzed hydrolysis of diacetamides therefore it appears highly probable that the rate determining attack by a water molecule occurs at the carbon of the protonated carbonyl group and the carbonyl carbon-nitrogen bond scission follows subsequently. This mechanism is consistent with that generally accepted for the hydrolysis of amides in dilute acid solution but disagrees with that proposed by Laureut et al., for acid hydrolysis of N-acetyl-lactams.
