식물체 및 퇴비 성분 분석을 위한 마이크로웨이브 분해법과 기존 습식 분해법의 비교 (Comparison of Microwave with Conventional Wet-Digestion Methods for the Element Analysis of Plant and Compost)

  • 마이크로파 시료 분해법이 식물체와 퇴비 중의 K, Ca, Mg, Cu, Zn, Mn등을 분석하는데 적합한가를 검정하기 위하여 기존의 습식분해법과 비교하였다. 분석시간은 기존의 습식분해법이 4~6시간 걸리던 것에 비하여 마이크로파 분해법에서는 1시간 이하로 단축할 수 있었으며, 두 분석법간의 각 성분별 분석값의 상관계수($R^2$)는 각각 K 0.98, Ca 0.97, Mg 0.91, Zn 0.94, Mn 0.99, Cu 0.99를 나타내었다. 칼륨과 칼슘의 경우 기존 습식분해법의 분석값이 더 높은 경향을 나타내었고 구리, 망간, 아연의 경우는 마이크로파 분해법의 분석값이 더 높은 경향을 나타내었으나 마그네슘의 경우 일정한 경향이 없었다. 그러나 칼슘의 경우를 제외한 그 외 성분들의 편기도(biases)는 10% 이내였다.

Pressure Cycling Technology-assisted Protein Digestion for Efficient Proteomic Analysis

  • In typical proteomic analysis, trypsin digestion is one of the most time-consuming steps. Conventional proteomic sample preparation methods use an overnight trypsin digestion method. In this study, we compared high-pressure cycling technology (PCT) during enzyme digestion for proteome analysis to the conventional method. We examined the effect of PCT on enzyme activity at temperatures of 25, 37, and $50^{\circ}C$. Although a fast digestion (1 h) was used for the standard protein mixture analysis, the PCT-assisted method with urea showed better results for protein sequence coverage and the number of peptides identified compared with the conventional method. There was no significant difference between temperatures for PCT-assisted digestion; however, we selected PCT-assisted digestion with urea at $25^{\circ}C$ as an optimized method for fast enzyme digestion, based on peptide carbamylation at these conditions. The optimized method was used for stem cell proteome analysis. We identified 233, 264 and 137 proteins using the conventional method with urea at $37^{\circ}C$ for 16h, the PCT-assisted digestion with urea at $25^{\circ}C$ for 1 h, and the non-PCT-assisted digestion with urea at $25^{\circ}C$ for 1 h, respectively. A comparison of these results suggests that PCT enhanced the enzyme digestion by permitting better access to cleavage sites on the proteins.

새로운 전처리 방법과 ICP-AES에 의한 폴리우레탄 중의 중금속 분석 (Determination of Heavy Metal in Polyurethane by New Digestion Method and ICP-AES)

  • A simple and convenient digestion method was developed for the determination of heavy metal in polyurethane sample using only nitric acid in open system without heating. This digestion method had prevented loss of volatile elements of Hg and As and reduced the pretreatment time for the sample digestion. The detection limit is lowered over 10 times than that of microwave digestion method. Recoveries in polyurethane material were 108% for As, 96 ~ 99% for Hg, 102 ~ 103% for Cr, 98 ~ 102% for Pb, 86 ~ 90% for Cd, and 90 ~ 93% for Cu.

Development of Continuous Flow Microwave Digestion Procedures for Analysis of Trace Metal in Water Using Ion Chromatography

  • A simple and rapid sample pretreatment process necessary for determination of metal oxides in water was proposed. Samples were injected into the continuous-flow tube installed inside the microwave oven and the treated samples were cooled before entered to the Ion Chromatography (IC) or Inductively Coupled Plasma (ICP). By coupling this microwave digestion system with IC or ICP, a fully automatic analytical procedures may be easily established. In this study, two different types of digestion methods were considered; the open tubing method (OTM) and the restraint tubing method (RTM). The RTM was proved to be 3 times faster in digestion period and 10 times higher in detection range than the OTM. Validation of proposed sample digestion system was carried out by using an ICP. The results showed that both of continuous-flow methods, the OTM and the RTM were comparable in accuracies with the conventional batch-type vessel digestion method.

비육용 곡물사료의 가공방법과 증체효율 (Grain Processing on Feed Efficiency for Beef Production)

  • The studies had been conducted to evaluate the grain processing effects for ruminants on starch digestion, body weight gain and feed efficiency since 1970. This research deals with experimental results on chemical structure, gelatinization, microbial starch digestion in rumen, intestinal starch digestion in rumen, roles of protozoa, intestinal starch digestion of bypass starch, limits to starch digestion in small intestine. The grain processing has different effects on digestion, weight gain and feed efficiency when different grain sources and contents is used, and the quality and quantity of roughage is different. The economical and efficient method of grain processing should be selected considering weight gain and feed efficiency enhancement than digestibility.

국내 토양오염 공정시험방법중 중금속 관련 오염평가의 문제점과 개선책 (The controversial points and a remedy on evaluation of heavy metal contamination in standard method for examination of soil in Korea.)

  • Heavy metals are extracted from stream sediments, roadside soils and sediments and soils and tailings from mining area using partial extraction, acid digestion and HF-digestion. Compared to amounts of heavy metals extracted using partial extraction, those extracted using acid digestion are higher by 2.0∼220.9 times in Cu, 2.4∼2806.1 times in Pb, 1.3∼121 times in Cd, 14.1∼1300885 times in Fe, 1.2∼271.5 times in Mn, 1.3∼372.5 times in Zn, 2.2∼1734.5 times in Cr. Although partial extraction, which extracts less amounts of heavy metals from soil compared to acid digestion, is used in domestic standard method for examination of soil, domestic soil standard for heavy metals in non-agricultural and industrial areas is higher than soil standard in foreign countries which use acid digestion, For improvement of the domestic standard method for assessment of soil, it is suggested to lower the domestic soil standard for heavy metals or to change pretreatment method for extracting heavy metals from partial extraction to acid digestion with modifying the soil standard.

음식물류폐기물의 성상별 온도변화에 따른 혐기성소화 효율 비교 연구 (Comparison of Anaerobic Digestion Efficiency with Different Temperature of Food Wastes)

  • A comparative study on the anaerobic digestion efficiency according to the temperature change was conducted considering the characteristics of domestic food wastes with high water content of about 80 % or more. The substrate was tested for anaerobic digestion efficiency in two substrates, a liquid component separated naturally from food waste and food waste itself. In the anaerobic digestion experiments, the digestion efficiency was the highest at $55^{\circ}C$ (thermophilic temperature). However, the digestion efficiency at $45^{\circ}C$(middle high temperature) was lower than that at $35^{\circ}C$(mesophilic temperature). The comparison of general food wastes anaerobic digestion requiring 30 days of hydraulic retention time to the liquid component indicated a stable digestion efficiency even after 15 days of hydraulic retention time.In the experiments conducted on food waste, the digestion efficiency at $55^{\circ}C$ was higher than that at $35^{\circ}C$. When the food waste, especially the liquid component originating from food waste, is treated by anaerobic digestion method, the mesophilic temperature and thermophilic temperature conditions are more favorable in the digestion efficiency than the middle high temperature ($45^{\circ}C$). However, when applying thermophilic or mesophilic temperature anaerobic digestion process operation in the field, the amount of energy input should be considered.

실리콘 웨이퍼 중의 금속 불순물 분석을 위한 시료 전처리 (Sample Pretreatment for the Determination of Metal Impurities in Silicon Wafer)

  • 실리콘 웨이퍼 중의 금속 불순물을 유도결합플라스마-질량분석기(ICP-MS)로 분석하기 위한 시료의 전처리 방법으로 마이크로파 분해법과 산분해법을 비교하였다 $HNO_3$-HF 혼합용액을 실리콘 웨이퍼 시료에 가하고 마이크로파 분해법과 산분해법으로 용해시킨 후, 용액의 실리콘 매트릭스는 증발 용기 안에서 Si-F의 형태로 증발시켜 분석하였다. 실제 시료에 spike한 Ni, Cr 및 Fe의 회수율은 두 전처리 방법에서 95∼106%였다. Cu의 경우는 산분해법이 회수율이 더 좋았고, Zn의 경우는 마이크로파 분해법으로 전처리 한 경우의 회수율이 더 좋았다. Spin coater로 Fe를 오염시킨 실리콘 웨이퍼를 산분해법과 마이크로파 분해법으로 전처리하여 ICP-MS로 분석한 결과, Fe의 농도는 위의 두 전처리 방법에서 거의 차이가 없었다.

이플루오린화 암모늄 시료분해 및 ICP/MS에 의한 암석 및 퇴적물 중 게르마늄 분석 (Analysis of germanium in rock and sediment by ICP/MS after ammonium bifluoride(NH4HF2) digestion)

  • 본 연구에서는 암석과 퇴적물 시료를 대상으로 이플루오린화 암모늄을 이용한 시료 분해법을 적용하여 open vessel digestion 후 유도결합 플라즈마 질량분석법(Inductively coupled plasma mass spectrometry, ICP/MS)으로 게르마늄을 분석하였다. 미국 지질조사소 표준물질 중 QLO-1, SDO-1을 사용하였고, 실제 시료로 GeoPT용 퇴적물, 현무암, 점토를 사용하였다. 휘발하여 손실되기 쉬운 것으로 알려진 게르마늄을 이플루오린화 암모늄($NH_4HF_2$) 시료 분해법을 사용함으로서 게르마늄 휘발 억제와 동시에 암석 및 퇴적물을 open vessel digestion하에서 간단하고 빠르게 분해 가능하였으며, 또한 이플루오린화 암모늄 분해법에 과산화수소를 함께 사용해도 게르마늄의 회수율에 영향을 끼치지 않았다. ICP/MS에 의한 게르마늄 분석 결과, MDL (method detection limit)은 0.015 ${\mu}g/g$, 게르마늄 회수율은 106~128% 이었다.

회화방법에 따른 폐 내 석면섬유의 투과전자현미경 분석 (Analysis of Asbestos Fiber in Lungs by Transmission Electron Microscopy According to the Digestion Method)

  • Objectives: This study was designed to establish an accurate analytical method for asbestos in a biological sample for determining occupational asbestos-related diseases and relief of the health effects of environmental asbestos. Methods: Biological samples were obtained from lungs of rats following intratracheal instillation of asbestos(Chrysotile, anthophyllite) and were prepared according to digestion method(Wet digestion, high temperature ashing, low temperature ashing). The samples were then analyzed for asbestos fibers using a transmission electron microscope equipped with an energy dispersive X-ray spectrometer. Results: Low temperature plasma ashing removed more of the organic components and reduced fiber loss compared to the wet digestion method, making specimens so prepared more suitable for transmission electron microsocpy. Conclusions: The low temperature ashing technique is the most accurate method for analyzing asbestos in biological samples.
