Effect of Genistein on the Onset of Puberty in Female Rats (암컷 흰쥐의 사춘기 개시에 미치는 Genistein의 효과)

  • There is growing concern that dietary soy intake is associated with protection of breast cancer. However, questions persist on the potential adverse effects of the main soy constituent genistein(GS) on female reproductive physiology. In this study, we examined whether prepubertal exposure to GS affected on the onset of puberty and the associated reproductive parameters such as hormone receptor expressions in female rats. GS(100mg/kg/day) was administrated daily from postnatal day 25(PND 25) to the day when the first vaginal opening(VO) was observed, and the animals were sacrificed on the day after VO occurred. Gross anatomy and tissue weight were compared to test the GS's effect on the cell proliferation. Furthermore, histological studies were performed to assess the structural alterations in tissues. Specific radioimmunoassay(RIA) were carried out to measure serum LH levels. To determine the transcriptional changes in progesterone receptors(PR), total RNAs were extracted from ovary and uterus and were applied to semi-quantitative reverse transcription polymerase chain reaction(RT-PCR). As a results, advanced VO was shown in the GS group(PND $31.2{\pm}0.6$) compared to the vehicle group (PND $35.3{\pm}0.7$). GS treatment significantly increased wet weight of ovaries and uteri compared to the vehicle group. Increased serum LH levels were also shown in the GS group. Graafian follicles and corpora lutea(CL) were observed only in the ovaries from GS treated animals. Similarly, hypertrophy of luminal and glandular uterine epithelium were found only in the GS group. Collectively, these effects were probably due to the estrogenic effects of GS. In the semi-quantitative RT-PCR studies, the transcriptional activities of PR in both ovary and uterus from GS-treated group were significantly higher than those from the vehicle group. The present studies demonstrated that acute exposure to GS, at levels comparable to the ranges of human exposure, during the critical period of prepubertal stage activates the reproductive system resulting precocious puberty in immature female rats.

Effects of Single or Mixed Feeding of Lactobacillus and Yeast on Performance, Nutrient Digestibility, Intestinal Microflora, and Fecal NH$_{3}$ Gas Emission in Laying Hens (유산균과 효모의 단독 및 혼합 급여가 산란계의 생산성, 영양소 소화율, 장내미생물 변화 및 분의 암모니아 발생량에 미치는 영향)

  • Total 240 of ISA Brown layers were employed in this experiment to study the effects of single or mixed feeding of Lactobacillus and yeast on the performance and intestinal microflora of laying hens. They were randomly allocated to six dietary treatments; None(Control), Pichia farinosa(PF), Lactobacillus crispatus avihen1 (LCH), Lactobacillus vaginalis avihen1(LVH), LCH+PF, and LVH+PF. Viable microflora were added to meet 3${\times}$10$\^$6/ cfu PF and 10$\^$7/ cfu Lacrobacillus per g of feed. There were four replicates per treatment, and 10 birds per replicates. Laying performance was recorded for 10 weeks, followed by a metabolism trial during which nutrient utilization, pattern of intestinal microflora and fecal NH$\sub$3/ emission were examined. Egg production and daily egg mass of birds fed either single or mixed microorganisms were significantly higher than those of the control(P<0.05). Egg weight and feed intake were not statistically different among all treatments. However, feed conversion ratio tended to improve by the supplementation of microbes. Digestibility of crude protein, ether extract and crude ash tended to improve in Lactobacillus treatments, however, there were not statistically different. With regards to the number of intestinal microbes, number of anaerobes were increased in microbes feeding group. Eggshell quality of PF layers was significantly poorer than those of the other treatments. No consistent trend was found in Haugh Unit among all treatments. Fecal NH$\sub$3/ gas emission was significantly lower in LVH, LVH+PF and LCH+PF than the other treatments(P<0.05). From the result of this experiment, it could be concluded that single or mixed feeding of Lactobacillus and yeast improves the laying performance and decreases the fecal ammonia gas emission. No synergic effect was found when both microbes were mixed and fed to the layers.

A Study on Quality Characteristics of Pleurospermum kamtschaticum as a Functional Food Resource (누룩치의 기능성 식품재료화를 위한 품질특성에 관한 연구)

  • The aim of this study was to investigate Quality characteristics of Pleurospermum kamtschaticum to increase the value of functional food resources. To examine Quality characteristics of Pleurospermum kamtschaticum, various factors such as color, texture, fiber, minerals, tannin, crude proteins, crude lipids and sensory quality, were determined using physicochemical methods. The contents of dietary fiber were 0.57% and 0.54% in each 100 g of leaf and stem parts (freeze drying base). In mineral content, potassium was the highest value in both of leaf and stem parts (freeze drying base). The contents of tannin were 108.1 $\mu$g/mL and 20.9 $\mu$g/mL in leaf and stem parts (fresh base) respectively. Blanching stems showed significant difference in hardness from freeze-drying leaves. The antioxidative activity in ethanol extracts of Pleurospermum kamtschaticum was investigated using peroxide value and free radical scavenging activity. Free radical scavenging activity in ethanol extracts of Pleurospermum kamtschaticum was 43.5% in a 100 $\mu$g/mL level, and the antioxidative index was 1.09 in a 500 $\mu$g/mL level. For functional food using Pleurospermum kamtschaticum the contents of crude proteins and lipids were about 9% and 26%, respectively. The results from sensory evaluation of each treatment older age showed a better acceptability.

  • Medium-chain fatty acids (MCFA) are composed of 8-12 carbon atoms, and are found in coconut, cuphea, and palm kernel oil. MCFA were introduced into clinical nutrition in the 1950s for dietary treatment of malabsorption syndromes because of their rapid absorption and solubility. Recently, MCFA have been applied to Gastrointestinal Permeation Enhancement Technology (GIPET), which is one of the most important parts in drug delivery system in therapeutics. Therefore, to accumulate the MCFA in seed oil of rapeseed, much effort has been conducted by classical or molecular breeding. Laurate can be successfully accumulated up to 60 mol% in the seed oil of rapeseed by the expression of bay thioesterase (Uc FatB1) alone or crossed with a line over-expressing the coconut lysophosphatidic acid acyltransferase (LPAAT) under the control of a napin seed-storage protein promoter. Also, caprylate and caprate were obtained 7 mol% and 29 mol%, respectively, from plants over-expressing of the medium-chain specific thioesterase (Ch FatB2) alone or together with the chain-length-specific condensing enzyme (Ch KASIV). Despite the success of some research in utilizing parallel classical and molecular breeding to produce MCFA, commercially available seed oils have for the most part, not been realized. Recent research in the field of developing MCFA-enriched transgenic plants has established that there is no single rate-limiting step in the production of the target fatty acids. The purpose of this article is to review some of the recent progress in understanding the mechanism and regulation of MCFA production in seed oil of rapeseed.

Effect of Feeding Lactobacillus reuteri to Laying Hens on Laying Performance, Availability and Intestinal Microflora (Lactobacillus reuteri의 급여가 산란계의 생산성, 영양소 이용율 및 장내 미생물 변화에 미치는 영향)

  • A feeding trial was conducted to investigate the influence of feeding Lactobacillus reuteri culture (LR) on productive performance, intestinal microflora and availability in laying hens. Four hundred and eighty, Isa-Brown layers, 49 weeks of age, were fed diets supplemented with LR at the level of 0 (control), 0.1, 0.2, and $0.4\%$ of the diets for eight weeks. Egg production and egg weight were measured daily. Feed intake was weighed every two weeks. Egg quality was measured three times at the start, mid-term, and end of the experiment. Intestinal microflora were examined for Lactobacillus spp., E. coli and Salmonella at the end of the experiment. Overall egg production was the highest in $0.2\%$ LR (P<0.05), but that of $0.1\%$ or $0.4\%$ LR treatments did not significantly differ from that of control. Egg weight was significantly higher in LR feeding group than the control (P<0.05). Daily egg mass was significantly higher in $0.2\%$ and $0.4\%$ LR treatments compared to the control and $0.1\%$ LR (P<0.05). The number of jumbo and extra large eggs were increased in LR supplemented groups, especially in $0.1\%$ LR. Feed intake of layers fed LR supplemented diets tended to be lower than the control. However, feed conversion ratio significantly improved in LR supplemented groups (P<0.05). Availability of dry matter and crude protein improved significantly in $0.4\%$ LR treatment (P<0.05). But, those of ether extract and crude ash were not significantly different among treatments. Eggshell breaking strength and eggshell thickness were not significantly influenced by LR supplementation, and Haugh unit and yolk index were also similar to the control. Total number of Lactobacillus spp. in ileum and cecum fed LR supplemented diets were significantly higher than those of the control (P<0.05). There were no significant differences in intestinal E. coli and Salmonella in all treatments. Therefore, it is concluded that dietary supplementation of Lactobacillus reuteri culture can improve the laying performance, feed efficiency and intestinal Lactobacillus.

Effect of Feeding Multiple Probiotics on Performance and Fecal Noxious Gas Emission in Broiler Chicks (혼합 생균제의 급여가 육계의 생산성 및 계분의 유해가스 발생에 미치는 영향)

  • A study was conducted to examine the effect of dietary supplementation of multiple probiotics (EM) on growth performance, blood cholesterol, intestinal micro flora, and fecal gas emission in broiler chicks. A total of 450 one day old male broiler chicks (Ross $\times$ Ross) were divided into six treatments with five replications in each treatment for five weeks. Treatments were factorially designed with two levels of diet containing probiotics (DW; 0, $0.2\%$) and three levels of drinking water containing probiotics (DW; 0, 0.01, $0.1\%$). Basal diets contained $21.5\%$ CP and 3,100 kcal/kg ME for starting and $19\%$ CP and 3,100 kcal/kg ME for finishing period. Weight gain, feed intake, and feed conversions of birds fed with probiotics were not significantly different between Ds. Total cholesterol and triglyceride levels were significantly lower (P<0.05) in birds fed with DW $0.01\%$ or $0.1\%$ compared with no probiotics group, but there was no significant difference between D treatments. The number of E. coli, Salmonella and Lactobacillus in the ileum and cecum of the birds fed multiple probiotics were not significantly different from those of no probiotic groups. There were no significant differences in the $CO_2$ gas emissions of fecal between birds fed with Ds or among birds fed with DW. However, $NH_3$ gas emissions of DW $0.1\%$ were significantly lower (P<0.05) than DW $0\%$. In the results of this study, supplementation of probiotics tended to decrease the serum cholesterol and triglyceride compared to those of control groups and reduction of fecal $NH_3$ gas emission.

Effect of Feeding Red Ginseng Marc on Vital Reaction in Laying Hens under Stress Task (홍삼 부산물이 스트레스에 대한 산란계 생체반응에 미치는 영향)

  • This study was conducted to determine the possible use of Red Ginseng marc as stress inhibiter in thermal stress (temperature humidity index 86) and lipopolysaccharide (LPS) - exposed laying hens by investigating their effects on laying performance, blood biochemical parameters, immunoglobulin concentration and serum superoxide dismutase (SOD) like ability. A total of forty-five 52-wk-old laying hens (ISA Brown) were divided into 3 treatment groups with 5 replicates of 3 birds in each group. NC (negative control, no immune substances), PC (positive control, ${\beta}$-glucan 25 ppm) and RGM (Red Ginseng Marc 3%) were added in feed with respective substance. Egg production in RGM was significantly increased in comparison with NC groups for 8 weeks (P<0.05). On blood biochemical parameters, effects of ambient temperature is definite by showing significant difference in aspartate aminotransferase and others (P<0.05), but RGM both before and after thermal stimulation have no significant difference in comparison with other groups. And for 3 weeks after thermal stimulation, laying performance was also not significantly different among treatments. Immunoglobulin M content and SOD like activities after challenge with LPS were higher in the RGM and PC than NC (P<0.05). In conclusion, although ineffective as inhibiter in thermal stress, dietary supplementation of Red Ginseng marc improved SOD like activity and immune system by regulating immunoglobulin content in laying hens. These findings have laid the foundation for future studies of immunomodulation in laying hens fed Red Ginseng Marc and of evaluation of heat stress inhibitor.

Effects of Dietary Levels of Corn Distiller's Dried Grains with Solubles and Phytase on Performance and Nutrient Utilization of Broilers (옥수수 주정박의 수준과 Phytase의 첨가 유무가 육계의 생산성과 영양소 이용률에 미치는 영향)

  • This work was conducted to investigate the effects of the addition level of Corn distiller's dried grains with solubles (CDDGS) and phytase in broiler diets on the broiler performance and the ileal amino acid digestibility. Three eighty four Ross broiler (one-day-old) were assigned randomly to 8 treatments groups (12 birds/pen, 4 pens/treatment) as $4{\times}2$ complex factors of CDDGS 4 treatments (0, 5, 10 and 15%) and phytase 2 treatments (0, 500 FTU/kg), and fed experimental diets for 6 weeks (starter, 0~2 wk; grower, 2~4 wk; finisher, 4~6 wk). After the feeding trial was finished, 12 broilers (6 weeks) with similar weights were selected from all treatments to investigate the digestibility of nutrients and amino acids for the first week. The experimental diets contained similar ME and CP of all treatments. Body weight gains and feed intakes were high at phytase treatments (P>0.05), but there was no difference in feed intake of the chicks. The amino acid digestibilities of diets were improved for iso-leucine, lysine, threonine, valine, aspartic acid, proline, serine and tyrosine (P<0.05). Finally, this work showed that growth performance and amino acid digestibility were improved by the addition of CDDGS and phytase in broiler diets.

Effects of Applying Cattle Slurry and Mixed Sowing with Legumes on Productivity, Feed Values and Organic Stock Carrying Capacity of Whole Crop Barley and Rye (액상우분뇨 시용과 콩과작물의 혼파가 청보리와 호밀의 생산성, 사료가치 및 단위면적당 유기가축 사육능력에 미치는 영향)

  • This study was conducted to determine effects of applying cattle slurry and mixed sowing with legumes such as hairy vetch or forage pea on productivity, feed values and organic stock carrying capacity of whole crop barley and rye as winter forage crops, and to obtain organic forages together with higher soil fertility. Experimental plots consisted of 7 treatments, which were non-fertilizer, chemical fertilizer (containing phosphate and potassium: P+K), chemical fertilizer (containing nitrogen, phosphate and potassium: N+P+K), organic fertilizer, cattle slurry, cattle slurry application (mixture with hairy vetch), and cattle slurry application (mixture with forage pea) plots. Each treatment was triplicates, and seven treatments were allocated in a completely randomized block design. For whole crop barley or its mixture crops, annual dry matter (DM), crude protein (CP), and total digestible nutrients (TDN) yields of N+P+K plots were significantly (P<0.05) higher than other plots except for cattle slurry plots. The CP content of barley or its mixture crops was significantly higher tor N+P+K plot (9.8%) and mixture plots with legumes (8.6~9.7%) than those of other treatments. As 450 kg Hanwoo heifers were fed diets included 70% whole crop barley or 70% mixture crops with legumes, mixture plots are capable of raising average 1.7 to 1.8 heads/ha a year. For rye or its mixture crops, annual DM, CP, and TDN yields represented 6.9~7.1, 0.5~0.6 and 4.3~4.4 ton/ha, respectively. The N+P+K plot contained 10.8% CP, which was higher (P<0.05) than all other treatments. In case of 450 kg Hanwoo heifers fed diets included 70% rye or 70% mixture crops with legumes, mixture plots can rear average 1.9 heads/ha a year. When it was considered based on crop yields and organic stock carrying capacity, applying cattle slurry to whole crop barley or rye had the comparable yields and feed values to chemical fertilizer application. Moreover, whole crop barley and rye within cattle slurry plots had a greater combination with hairy vetch and forage pea, respectively, and their mixture crops with legumes had higher crude protein and TDN yields within cattle slurry plots. In conclusion, it would be expected that mixed sowing with legumes in the application of cattle slurry to grass crops could be substituted for imported organic grains as dietary protein sources in feeding organic livestock.

Complementary and Alternative Medical Therapies in Children and Adolescent with Chronic Disease : Utilizations and Patterns (만성 질환을 가진 소아, 청소년에게 시행되는 보완대체요법의 실태 연구)

  • Purpose : This study describes how often complementary and alternative medical(CAM) therapies, what patterns of CAM therapies utilization, what types of CAM therapies and why CAM therapies are used in children and adolescent with chronic disease. Methods : We interviewed and filled out questionnaires with parents of patients suffering from asthma, atopic dermatitis, congenital myopathy, epilepsy and so on from Mar 2004 to Aug 2004. Results : Two hundred fifty two parents answered questionnaires; 160 cases(63.5 percent) had used CAM therapies. Utilization of CAM therapies according to each chronic disease was as follows : congenital myopathy(100 percent), atopic dermatitis(91.1 percent), asthma(72.5 percent), chronic diarrhea (50.0 percent), epilepsy(17.1 percent) etc. Types of CAM therapy were as follows : dietary supplement (31.1 percent), herb medicine(23.4 percent), massage(14.2 percent), acupuncture(4.6 percent) etc. Sixty four(40.0 percent) case underwent more than two types of CAM therapies simultaneously. One hundred thirty three(83.1 percent) case were asked through non-medical prescription. Ninety nine(61.9 percent) case were recognized positive effect of CAM therapies. Parents' and childrens' ages were influential factors in CAM therapies utilization : the older the parents and the younger the children, the higher the utilization. Conclusion : Although patients receive treatment with western medicine, many of them not only undergo CAM therapies, especially more than two types, but also recognize the positive effects of CAM therapies. As a result, we suggest that it is easy to use of CAM therapies, and the positive effects of CAM therapies are prominent, domestically. Therefore, medical approach and study about the effectiveness and side effects of CAM therapies is necessary to avoid the imprudent use of such therapies.
