뼈 콜라겐의 탄소와 질소 안정동위원소에 기록된 6세기대 나주 영동리 고분군 피장자 집단의 식생활 양상 (Palaeodietary Reconstruction of 6th Century Naju Yeongdong-ri People Recorded in Stable Carbon and Nitrogen Isotope Analysis of Human Bone Collagen)

  • 뼈에서 추출된 탄소와 질소 안정동위원소 정보는 과거 사람들의 식생활, 영양상태, 생계경제와 고환경 등을 연구하는데 매우 중요한 자료이다. 본 연구에서는 6세기대 나주 영동리 고분군 제1호분 돌방무덤과 돌덧널무덤에서 출토된 옛사람 뼈 9개체에서 추출된 콜라겐의 안정동위원소 분석을 실시하였다. 탄소와 질소 안정동위원소 결과는 다음과 같다(${\delta}^{13}C=-19.5{\pm}0.7$‰, ${\delta}^{15}N=9.6{\pm}0.7$‰, (n=9)). 탄소 안정동위원소 분석 결과 나주 영동리 고분군 피장자 집단은 벼, 보리, 콩 등 $C_3$ 작물 위주의 섭취를 하였다고 추정되며, 질소 안정동위원소 값으로부터 동물성 단백질은 주로 육상 동물로부터 얻었다고 추정된다. 이 결과로부터 백제식 무덤 양식을 갖고 영산강유역 토착 세력의 매장 양식이 보이는 피장자 집단의 식생활 양상을 확인할 수 있었다.

이천 게걸무, 강화 순무, 조선무의 화학적 특성 및 효소활성 (Chemical Characteristics and Enzyme Activities of Icheon Ge-Geol Radish, Gangwha Turnip, and Korean Radish)

  • 본 연구에서는 이천 게걸무, 강화 순무 및 조선무의 품질 특성을 비교 분석하였다. 일반성분의 경우, 수분 함량은 이천 게걸무 87.78%, 강화 순무 92.73% 및 조선무 91.45%로 게걸무가 다른 무에 비하여 낮았다. 반면에 조단백, 조섬유 및 조회분 함량은 이천 게걸무가 강화 순무와 조선무에 비하여 약 1.7-1.8배, 1.2-1.6배 및 2.0-2.4배 높았다. 식이섬유함량은 이천 게걸무 11.32%, 강화 순무 13.26% 및 조선무 14.20%로 게걸무가 다른 무에 비하여 낮았다. 무기질 함량의 경우, 게걸무의 Mg, K, Ca 함량이 다른 무에 비하여 높았으며 P, Zn, Fe 함량은 다른 무와 유의적인 차이가 없었다. 가수분해 효소로서 소화와 관련된 ${\alpha}-amylase$활성의 측정 결과 이천 게걸무 9.97 unit, 강화 순무 14.87 unit 및 조선무 18.99 unit으로 나타났다. Protease 활성의 경우, 알칼리성은 이천 게걸무와 조선무가 각각 3.02 PU, 3.10 PU의 유사한 수준으로 순무에 비하여 높았고, 산성 protease 활성에서는 게걸무가 가장 높았다. 무의 매운맛 성분 및 기능성 관련 물질로 알려진 myrosinase 활성은 이천 게걸무가 36,990.81 unit으로 강화 순무나 조선무보다 높았다. 결론적으로 이천 게걸무는 강화 순무나 조선무에 비하여 조단백, 회분, 조섬유 및 무기질 중 Na, Mg, K, Ca의 함량이 높고 protease 활성과 myrosinase 활성이 높았다.

보리발효추출물로부터 분리한 다당의 대식세포 활성화 및 신호 전달 (Polysaccharide isolated from fermented barley extract activates macrophages via the MAPK and NF-κB pathways)

  • 보리는 높은 비율로 식이섬유를 함유하고 있어, 일반적으로 영양학적으로는 전곡 형태로의 섭취가 추천되고 있지만, 조직감과 소화율을 고려하여 발효물 형태의 소비가 바람직한 것으로 판단되고 있다. 본 연구는 효소당화 후, 효모 및 유산균 발효를 거쳐 조제한 보리발효물로부터 조다당 BF-CP를 분리하고, 대식세포에 대한 면역증강 효과 및 세포 내 신호전달을 규명하여 기능성 소재로의 이용방안을 모색하기 위해 계획되었다. BF-CP 획분의 일반화학적 특성을 분석한 결과 70.7% 글루코스 11.4% 자일로스 및 9.0% 아라비노스를 포함하여 91.1%의 중성당으로 이루어진 중성다당이었다. BF-CP는 RAW 264.7 대식세포주에서 농도의존적으로 IL-6, $TNF-{\alpha}$와 같은 사이토카인 및 NO의 생산능을 유도하는 등 높은 대식세포 활성능을 나타냈다. 또한 qPCR 분석을 통해, BF-CP 획분을 대식세포주에 처리하였을 때, 처리 농도에 비례하여 IL-6, $TNF-{\alpha}$ 및 iNOS의 mRNA 유전자 발현을 증가시킴을 확인할 수 있었다. 한편 Western blot을 활용한 신호전달 단백질 추적실험에서 BF-CP 획분을 대식세포주에 처리하였을 때, JNK, ERK 및 p38과 같은 MAPK 경로와 $NF-{\kappa}B$ 경로의 관련 단백질을 인산화시킴이 확인되었으며 그 활성은 BF-CP 농도에 의존적이었다. 이상의 결과로부터 보리발효물 유래 다당 BF-CP는 MAPK와 $NF-{\kappa}B$ 경로를 통해 대식세포를 활성화시키며, 이를 통하여 NO, IL-6 및 $TNF-{\alpha}$와 같은 면역활성화 관련 물질의 생산을 높은 비율로 유도시킨다는 것을 최종 확인할 수 있었다.

발효 쌀보리의 급여가 육성돈의 혈중 대사산물, 혈구조성 및 생산성에 미치는 영향 (Growth performance and, blood metabolites and leukocytes of growing pigs fed with fermented hulless barley)

  • 본 연구는 발효 쌀보리 급여가 돼지의 생산성, 혈구조성, 혈중 대사물질에 미치는 영향을 구명하고자 실시하였다. 공시동물은 총 45두의 3원 교잡 육성돈(요크셔${\times}$랜드레이스${\times}$듀록; 평균체중, $30.33{\pm}0.05kg$)을 이용하였고 사료 내 발효 쌀보리(Fermented hulless barley, FHB)는 0, 0.5, 1.0% 수준으로 첨가하였으며 처리구당 3반복, 반복당 5두씩 시험돈사에 임의로 배치하였다. 일당증체량은 0.5% FHB 처리구에서 0 및 1.0% FHB 처리구에 비해 증가하였으나 통계적 차이는 없었다. 일당사료섭취량 및 사료요구율은 처리구간 유의한 차이를 보이지 않았다. 총백혈구, 호중구, 림프구, 단핵구, 호산구 및 호염구를 비롯한 백혈구계 혈액세포 조성은 발효 쌀보리에 영향을 받지 않았다. 혈중 글루코오스는 대조구에서 0.5, 1.0% FHB 처리구보다 증가하였다. 알부민은 1.0% FHB 처리구에서 0.5% FHB 처리구에 비해 감소하였다. 그러나, 단백질, 지방, 에너지 및 광물질과 관련된 나머지 대사적 지표들은 발효 쌀보리 급여에 의한 영향이 관찰되지 않았다. 사양시험기간동안 시험동물에서 환축 또는 폐사가 관찰되지 않아 돼지의 건강관리측면에서는 효과를 나타낸 것으로 보인다. 그러므로 발효 쌀보리가 축산물 안전성에 미치는 영향을 구명하는 추가 연구를 통해 안전 축산물 생산 가능성을 제고하는 것이 필요할 것이다.

Lifestyle modifications in an adolescent dormi­tory: a clinical trial

  • Purpose: Childhood obesity is an increasing public health issue worldwide. We examined dietary patterns among adolescents in a dormitory school, identified obese adolescents and tried to intervene to improve food habits and physical activity. Methods: We conducted an experimental prospective longitudinal study based on 36 obese (body mass index $[BMI]{\geq}95th$ percentile) adolescents (aged 12-18 years) compared with controls (healthy children: normal age-appropriate BMI ($BMI{\leq}85th$ percentile). Six months' intervention included lifestyle-modification counseling (once a week by a clinical dietician), and an exercise regimen twice a week, 60 minutes each time, instructed by a professional pediatric trainer). Both groups underwent baseline measurements at the beginning of the study and 6 months later (arterial stiffness, blood pressure, pulse, weight and height, hemoglobin, creatinine, liver enzymes, highly sensitive C-reactive protein and complete lipid profile). Results: Twenty-one participants completed the study. Low compliance from participants, school staff and parents was observed (participation in planned meetings; 71%-83%). BMI significantly decreased from $32.46{\pm}3.93kg/m^2$ to $30.32{\pm}3.4kg/m^2$ (P=0.002) in the study group. Arterial stiffness was not significantly different between the 2 groups and did not change significantly after 6 months' intervention (P=0.494). No significant changes in CRP and lipid profile were observed after the intervention. Conclusion: Making lifestyle modifications among adolescents in a dormitory school is a complex task. Active intervention indeed ameliorates BMI parameters. However, in order to maximize the beneficial effects, a multidisciplinary well-trained team is needed, with emphasis on integrating parents and the school environment.

Milk Conjugated Linoleic Acid (CLA) Profile and Metabolic Responses of Dairy Cows Fed with High-temperature-micro-time (HTMT) Treated Diets Containing High Quantity Extruded Soybean (ESB)

  • A feeding trial was conducted to examine the effect of high-temperature-micro-time (HTMT) processing of diets containing extruded soybean (ESB) in high quantity on milk fat production, metabolic responses, and the formation of conjugated linoleic acid (CLA) and trans-vaccenic acid (TVA). Twenty-one multiparous Holstein cows in mid-lactation were blocked according to milk yield in the previous lactation. Cows within each block were randomly assigned to either normal concentrate or HTMT treated diets containing ESB (7.5% HTMT-ESB and 15% HTMT-ESB). It was hypothesized that the HTMT-ESB would affect the undegradable fatty acids in the rumen and, thus, would modify the fatty acid profile of milk fat. Both 7.5% and 15% HTMT-ESB did not affect milk yield, fat, protein, lactose and solid-not-fat (SNF), but the proportion of cis-9, trans-11 CLA in milk fat was significantly increased by these treatments. Content of TVA in milk fat was not affected by HTMT-ESB. The HTMT-ESB influenced the fatty acid profile in milk fat, but there was little difference between 7.5% and 15% of supplementation. HTMT-ESB feeding significantly decreased the concentration of plasma insulin and glucose, while plasma growth hormone (GH), triglyceride (TG), non-esterified fatty acid (NEFA) and HDLcholesterol were increased by 7.5% and 15% ESB-HTMT supplementation in comparison to the control group (p<0.05). However, no significant difference was observed in plasma LDL-cholesterol, insulin like growth factor (IGF)-1, T3, T4, and leptin concentrations among treatments (p>0.05). The present results showed that cis-9, trans-11 CLA production was increased by HTMT treatment of dietary ESB without reduction of milk fat, and the unchanged milk fat and yield was assumed to be associated with the constant level of thyroid hormones, leptin, and IGF-1.

Effect of production level and source of fat supplement on performance, nutrient digestibility and blood parameters of heat-stressed Holstein cows

  • The interactive effect of dietary fat supplementation and milk yield level on dairy cows performance under heat stress has not been thoroughly investigated. The purpose of this study was to evaluate the effect of production level, the source of fat supplements and their interaction on dairy cows performance under heat stress. In this study, 64 Holstein multiparous cows were divided into 2 groups and received one of two rations having either calcium salts of fatty acids (Ca-FA) or high-palmitic acid (PA) supplements (2.8% of DM; dry matter). After completing the experiment and based on maturity-equivalent milk, cows were divided into two groups of high-yielding (14,633 kg) and medium-yielding (11,616 kg). Average temperature humidity index (THI) was 71 during the trial period. Apparent digestibility of dry matter (p = 0.04), organic matter (p = 0.05), and neutral detergent fiber (NDF; p = 0.04) for cows fed Ca-FA were greater than cows fed PA. The milk fat content in high-producing cows was 0.3% greater than medium-producing cows (p = 0.03). The milk protein content in cows fed Ca-FA was greater than cows fed PA (p < 0.01). High-producing cows had greater serum cholesterol (p = 0.02) than medium-producing cows. The cows fed PA tended to have a greater BUN than cows fed Ca-FA (p = 0.06). Alanine aminotransferase and aspartate aminotransferase tended to be increased by PA, which indicates that cows in PA treatment may have experienced more adverse effect on the liver function than cows on Ca-FA. Therefore, under heat stress and in 90 d trial, milk production level does not affect the cows' response to PA or Ca-FA. Although cows fed Ca-FA received lower energy than those fed PA, they compensated for this shortage likely with increasing the digestibility and produced a similar amount of milk.

  • 본 연구는 열 스트레스 하에서 오리사료 내 대사에너지(ME) 수준이 오리의 간, 십이지장 융모, 미생물, 유전자 조절에 미치는 영향을 조사하였다. 총 240마리의 육용 오리 채리밸리(Anas platyrhynchos)를 4처리구로 완전임의배치 한 후 42일 동안 사육하였다. 처리구는 ME 2900 kcal/kg, ME 3000 kcal/kg, ME 3100 kcal/kg 및 ME 3200 kcal/kg로 구분하였다. 간 조직은 처리구 사이의 차이가 없었고, 십이지장 융모 및 창자샘 길이는 ME 3000과 비교할 때 2900은 10.58% 감소하였으나 3100, 3200과의 사이에 차이는 없었다. 맹장 Latobacillus는 ME 3000과 비교할 때 2900은 9.47% 감소하였으나 3100, 3200은 각각 2.52, 3.24% 증가하였다. Total aerobic bacteria, E. coli, Coliform bacteria는 ME 3000과 비교할 때 2900은 증가하였으나 3100, 3200은 차이가 나타나지 않았다. 간에서 HSP (heat shock proteins)-mRNA 중 HSP $90-{\alpha}$는 ME 3000과 비교할 때 2900은 48.60% 감소하였으며 3100, 3200은 차이가 없거나 증가하였다.

Relative Performance of Fattening Lambs on Raw and Processed Cottonseed Meal Incorporated Diets

  • In order to find the feasibility of feeding raw or processed cotton (Gossypium) seed meal (CSM), 30 male crossbred lambs were randomly assigned to 5 dietary treatments with 6 animals each. Lambs were fed each of the isonitrogenous and isocaloric concentrate mixtures containing 30% deoiled groundnut cake (DGNC) (reference diet), 40% raw, 45 minute cooked, 1% $Ca(OH)_2$ or iron (1 part free gossypol:0.3 parts iron) treated CSM (replacing about 50% nitrogen moiety of reference concentrate mixture). The concentrate mixture was fed to meet 80% of CP requirements (NRC, 1985) along with ad libitum maize (Zea mays) hay for 180 days. Incorporation of raw or processed CSM did not affect the palatability of diets as evidenced by higher (p<0.01) or comparable overall daily intakes of DM and protein (CP and Digestible CP) per kg $W^{0.75}$ by lambs in comparison to reference group. Intakes of DE and ME increased (p<0.01) due to inclusion $Ca(OH)_2$ treated CSM in diets, while the intakes were lower (p<0.01) on iron treated CSM based diet when compared with reference diet. The intakes of DM, CP, TDN, DE and ME by reference and experimental lambs were higher than the requirements stipulated by NRC (1985). The growth rate was highest (p<0.01) in lambs fed on diets with cooked CSM followed by those fed raw or $Ca(OH)_2$ treated CSM based diets. The growth of lambs fed on iron treated CSM diets was depressed (p<0.01) in comparison to that of other experimental lambs probably due to increased iron intake (889 mg/kg DM of diet) which was much higher than the toxic levels of 500 ppm (NRC, 1980). The lambs on various diets utilized DM with similar efficiency while the utilization of energy (TDN, DE and ME) was more efficient (p<0.01) when the diets contained raw or cooked CSM in comparison to that of reference diet. The lambs fed raw and $Ca(OH)_2$ treated CSM incorporated diets utilized DCP with higher (p<0.01) efficiency than the lambs fed reference diet. The feed cost per kg weight gain was lower (p<0.01) on raw, cooked and $Ca(OH)_2$ treated CSM based diets in comparison to reference diet. Cooking of CSM for 45 minutes further reduced the feed cost of weight gain.

Effect of Supplementing Microbial Phytase on Performance of Broiler Breeders Fed Low Non-phytate Phosphorus Diet

  • An experiment was conducted to study the production performance of broiler breeder females (25 to 40 weeks of age) fed either reference diet or low non-phytate phosphorus (NPP) diet with or without microbial phytase (500 FYT/kg) supplementation. A weighed (160 g/b/d) quantity of feed from each diet was offered daily to 40 replicates of one bird each housed in California type cage having individual feeders. Each cage was considered as a replicate. A continuous 16-h light per day was provided using incandescent bulbs. Body weight, egg production, egg weight, feed per egg mass, egg specific gravity, egg breaking strength, shell thickness, tibia ash and serum Ca and protein concentrations were not affected by reducing the NPP level from 0.30 to 0.18% in the broiler breeder diet. Supplementation of phytase (500 FYT/kg) enzyme to the diet containing 0.18% NPP had no added advantage on any of the above production parameters. The serum inorganic P was increased significantly (p<0.05) by either enhancing the NPP content from 0.18 to 0.30% or supplementing phytase @500 FYT/kg to the diet containing low P which were found comparable. Retention of Ca and P was positive on all the diets. P retention decreased significantly (p<0.05) with either increase in NPP content or phytase supplementation in the diet. Neither NPP nor phytase supplementation influenced bone mineralization in terms of tibia ash and strength. The hatchability was not influenced by either increasing the NPP content or supplementing the enzyme phytase. Similarly, the P concentration in the egg yolk and day old chick, day old and 14th day body weight and leg score was not altered by increasing the level of NPP or supplementing phytase enzyme. The mortality was within the normal limits in all the three dietary groups. Thus, it can be concluded that 0.18% NPP (288 mg NPP intake/b/d) in the broiler breeder' diet is adequate in sustaining the optimum performance from 25 to 40 wks of age. Enhancing the NPP content or supplementation of phytase (500 FYT/kg diet) to diet containing 0.18% NPP had no added advantage on performance.
