  • This study has been performed to investigate the potential effects of by-product discarded after probiotics production (BPPP) on growth performance, immune gene expression, innate-immunity status, and disease resistance of carp, Cyprinus carpio. For 3 weeks, carp were fed four diets containing different levels of BPPP at 0, 0.1, 0.2 and 0.5% per kg of normal diet. Every 7 days of feeding, immune-related gene expression, serum lysozyme activity and ACH50 were analyzed. Growth rates and challenge test with E. tarda were conducted after 3 weeks of BPPP feeding. Both lysozyme activity and ACH50 were significantly (p<0.05) increased in all BPPP supplemented groups compared to the control at every 7 day for 3 weeks of feeding trial. The gene expression of pro-inflammatory cytokines, IL-1β and TNF-α was significantly (p<0.05) up-regulated until 21 days of feeding in all groups except for 0.2% group on day 7 post feeding. The anti-inflammatory cytokine IL-10 gene expression was only significantly (p<0.05) increased in 0.1% group on day 7 and decreased (p<0.05) on day 14 in all BPPP supplemented groups. On day 21, the IL-10 gene expression was augmented (p<0.05) in all groups. SOD gene expression was significantly (p<0.05) increased compared to the control on day 14 and 21 post feeding, whereas no significant difference was observed on day 7. In challenging test, 0.2%, 0.1%, 0.5% and control group showed 80%, 70%, 60% and 40% of survival rate, respectively. Feed conversion rate was only improved in 0.5% group. In conclusion, the present study indicates that dietary BPPP suplementation improved growth performance, innate immune response and bactericidal activity in carp.

Effects of Dietary Supplementation of Fermented Rice Bran (FRB) or Fermented Broken Rice (FBR) on Laying Performance, Egg Quality, Blood Parameter, and Cholesterol in Egg Yolk of Hy-Line Brown Laying Hens

  • This experiment was aimed at investigating the effects of dietary supplementation with fermented rice bran (FRB) or fermented broken rice (FBR) on egg-laying performance, egg quality, blood parameters, and cholesterol level in egg yolk of Hy-Line Brown egg-laying hens. Altogether, 144 Hy-Line Brown egg-laying hens (32-week-old) were randomly allocated to one of 4 dietary treatment groups, with 4 replicates per treatment. Of them, 3 treatments diets were prepared by supplementing the basal diet with 0.1% probiotics (PRO), 1% fermented rice bran (FRB), or 1% fermented broken rice (FBR) at the expense of corn. Hen-day egg production was higher (P<0.05) in PRO and FRB treatment groups than in the basal treatment groups. However, feed intake, egg weight, egg mass, and feed conversion ratio did not differ among the treatment groups. Additionally, supplementation with FRB or FBR did not affect eggshell strength, eggshell thickness, egg yolk color, and Haugh unit during the feeding trial. There was no significant difference in leukocyte count. Total cholesterol level was lower (P<0.05) in the FRB treatment group than in the basal treatment groups. Asparate aminotransferase, alanine transferase, glucose, and albumin levels were unaffected by dietary supplementation with FRB or FBR. Egg yolk cholesterol level was lower (P<0.05) in the FRB and FBR treatment groups than in the basal treatment groups. In conclusion, dietary supplementation with FRB or FBR improved egg-laying performance, and reduced the levels of total serum cholesterol and cholesterol in egg yolk of Hy-Line Brown egg-laying hens.

Evaluation of Multi-microbial Probiotics Produced by Submerged Liquid and Solid Substrate Fermentation Methods in Broilers

  • Two experiments were conducted to evaluate multi-microbe submerged liquid (SLF) and solid substrate (SSF) fermented probiotic products in broilers. The SLF and SSF probiotics were comprised of Lactobacillus acidophilus ($1.1{\times}10^9$ and $4{\times}10^8$ cfu/g), Bacillus subtilis ($1.1{\times}10^9$ and $4{\times}10^9$ cfu/g), Saccharomyces cerevisiae ($1.5{\times}10^7$ and $1.0{\times}10^4$ cfu/g) and Aspergillus oryzae ($2.6{\times}10^7$ and $4.3{\times}10^7$ cfu/g), respectively. In Exp. 1, 640 day-old Ross chicks were allotted to 4 treatments, each comprising 4 replicates (40 chicks/replicate). The basal diet was prepared without any antimicrobials (negative control, NC), and 20 mg/kg avilamycin (positive control, PC), 0.3% SLF and 0.3% SSF probiotics were added to the basal diets as treatments. Birds fed PC and SSF diets showed improved (p<0.001) overall weight gain and F/G than birds fed NC and SLF diets; whereas, birds fed SLF diet had better weight gain and F/G than birds fed NC diet. Retention of CP was higher (p<0.05) in birds fed the SSF diet than birds fed PC, SLF and NC diets. Birds fed the SLF diet tended to have higher (p<0.10) cecal total anaerobic bacteria than birds fed PC and NC diets; whereas, lesser cecal coliforms were noticed in birds fed PC, SLF and SSF diets than birds fed the NC diet. In Exp. 2, 640 day-old Ross chicks were randomly allotted to 4 treatments in a $2{\times}2$ factorial arrangement. Each treatment had 4 replicates (40 chicks/replicate). Two different multi-microbe probiotic products (0.3% SLF or SSF) each with two different antibiotics (10 mg/kg colistin, or 20 mg/kg avilamycin) were used as dietary treatments. Birds fed the SSF diet had greater weight gain (p<0.001), better F/G (p<0.05), greater retention of energy (p<0.001) and protein (p<0.05), and lesser cecal Clostridium (d 35) than birds fed SLF diet. Birds fed the colistin-supplemented diet had less (p<0.01) cecal coliforms when compared with birds fed the avilamycin diet. Additionally, birds fed the avilamycin diet had greater energy retention (p<0.05) than birds fed the colistin diet. Thus, the results of this study suggest the multi-microbe probiotic product prepared by a solid substrate fermentation method to be superior to the probiotic product prepared by submerged liquid fermentation; moreover, feeding of probiotics with different antibiotics did not elicit any interaction effect between probiotic and antibiotic.

  • 세계적으로 축산 폐수로 인한 환경오염 문제는 매우 심각하게 거론되어져 왔으며 특히 네덜란드, 덴마크, 영국 등과 같은 축산 선진 국가에서의 연구는 매우 심도 있게 이루어져 왔다. 우리 나라의 경우도 최근 들어 축산업이 대규모 전업화 혹은 기업화 되면서 가축이 배설하는 분뇨가 심각한 수질 및 토양 오염원이 됨에 따라, 정부는 이들 축산 폐수를 억제하기 위한 일환으로 환경 시행법을 법률화하고 있다. 또한 농림수산부는 시설 개선과 축산 폐수의 처리를 위한 시설 지원에 많은 노력을 기울이고 있다. 특히 환경청이나 환경 관련 단체의 활동이 강화되면서 일반 국민의 수질오염에 대한 관심이 고조되고 있는 가운데 이에 부응하는 축산인들의 자체 노력이 가세하여 축산 폐기물로부터 발생하는 수질오염에 대한 경각심은 초미의 관심사로 대두되고 있다. 동물의 품종의 개량과 함께 생산성을 저해하는 각종 질병의 예방, 치료 약제, 성장 촉진 및 사료 효율을 개선할 수 있는 물질이 사료첨가제로 개발되어 사용되었다. 동물의 성장 능력을 향상시키고 사료 효율을 개선하기 위하여 사용되는 물질로서는 항생제(Thrasher, 1968), 효소제(권 등, 2003), 호르몬제(Chung, 1990) 등이 있으며, 최근에는 축산악취저감 및 사료 효율개선 등을 위한 생균제에 관한 연구(박 등, 2001; 길 등, 2004)가 진행되고 있다. 하지만, 양돈 산업에 있어 생산성 향상뿐만 아니라 환경 친화적인 축산을 위하여 양돈을 하면서 발생하는 오염 물질을 감소시키는 것이 가장 시급한 문제로 대두됨에 따라 질소의 배설량을 줄이기 위해서는 저단백질 사료를 급여하거나 단백질 소화율을 개선시킬 수 있는 소화효소제나 미생물제제, 효모제, 생균제 등을 이용하여 배설량 및 질소의 배출량을 감소시킨다는 연구들이 있었으며(민 등, 1992; Park 등, 1994), Sutton 등(1999)과 Shriver 등(2003)은 사료내 단백질 수준을 낮추고, 합성아미노산을 급여함으로써 질소 배설량을 감소시켰다고 보고하였다. 따라서 본 연구에서 친환경적인 사료 개발은 사료 효율 및 영양소 소화율의 개선, 분뇨내 질소 배출량 및 악취 발생량을 감소시킬 수 있는 저공해 사료 개발에 대하여 연구하고자 하였다. 저공해 사료 개발에 대한 시험에서는 육성돈의 사료에서 대사에너지 함량을 낮추고, α-1, 6-galactosidase와 β-1,4-mannanase를 포함한 복합 효소제를 첨가하여 성장 능력 및 소화율에 미치는 영향을 알아보고자 시험을 실시하였다. 사료내 낮은 CP의 함량과 복합 생균제를 첨가 급여 성장능력, 질소배설량 및 악취발생량에 미치는 영향을 조사하기 위하여 실시하였다.

Growth Performance and Post-Weaning Diarrhea in Piglets Fed a Diet Supplemented with Probiotic Complexes

  • Weaning stress can affect the growth performance and intestinal health of piglets. Dietary alternatives to antibiotics, such as dietary probiotics, especially those containing multiple microbial species, are a preventive strategy for effectively controlling post-weaning diarrhea. In this study, we investigated forty-eight crossbred piglets in three treatment groups for 21 days: the control and experimental groups were supplemented with Enterococcus faecium DSM 7134, Bacillus subtilis AS1.836 plus Saccharomyces cerevisiae ATCC 28338 (EBS) or Lactobacillus paracasei L9 CGMCC No. 9800 (EBL). On day 21, weaned piglets supplemented with two kinds of probiotic complexes showed increased growth performance and significantly reduced post-weaning diarrhea (p < 0.05). The EBS treatment increased acetic acid and propionic acid in the feces (p < 0.05), and the EBL treatment increased fecal acetic acid, propionic acid, butyrate and valerate (p < 0.05). Moreover, the fecal microbiota of the piglets changed markedly in EBL treatment. The addition of EBS and EBL may have similar effects on the prevention of diarrhea by improving the intestinal morphology and regulating the microbiota during the weaning period.

Innovative Dietary Intervention Answers to Baby Colic

  • Purpose: The purpose of this paper is to evaluate the efficacy of a lactose- reduced synbiotic partial whey hydrolysate in formula fed infants presenting with colic and the impact of this dietary intervention in mean crying time and quality of life. Methods: Forty infants with infantile colic were treated during one month with parental reassurance and the intervention formula (partial whey hydrolysate, reduced lactose, Bifidobacterium lactis BB12 and galacto-oligosaccharides) and were compared to a control group of 20 infants with infantile colic treated with parental reassurance and a standard infant formula. Parents completed a quality of life (QoL) questionnaire assessing the burden of infantile colic. Wilcoxon test, t-test and Mann-Whitney test were used to compare QoL scores before and after intervention as well as between the intervention and control group. Results: At inclusion, duration of crying did not differ between both groups. Crying duration decreased with 2.7 hours (from 3.2 to 0.5 hours) in the intervention group while duration of crying decreased only with 1.2 hours in the control group (p<0.001). Stool composition became looser in the intervention group, but defecation frequency did not change. The median scores of the QoL questionnaire improved significantly in the intervention group for all parameters. In the control group, parameters improved significantly also but not for the parent-child and social interaction. The score changes were significantly greater in the intervention than in the control group. Conclusion: The intervention formula (partial whey hydrolysate, synbiotic, reduced lactose) significantly reduced the duration of crying and improved QoL of the parents and infants.

RAW 264.7 대식세포에서 유산균으로 발효한 다시마와 톳의 항염증 효과 (Anti-inflammatory Effects of Fermented Laminaria japonica and Hizikia fusiforme Water Extracts with Probiotics in LPS-stimulated RAW264.7 Macrophage Cell Line)

  • This study was conducted to investigate alterations of seaweed composition upon Lactobacillus rhamnosus GG (LGG) fermentation as well as potential anti-inflammatory effects and mechanism (s) of water extracts and fermented water extracts of Laminaria japonica (LJ) and Hizikia fusiforme (HF) in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. Total polyphenol, total sugar, and reducing sugar contents were measured in LJ and HF water extracts before and after fermentation by LGG. Alterations of inflammatory cytokine levels in cell culture media were measured by ELISA, and levels of phosphorylation of c-jun NH2-terminalkinase (JNK) and extra cellular signal regulated kinase (ERK) were examined by Western blot analysis. LGG fermentation of LJ and HF altered total polyphenol and sugar contents in water extracts of LJ and HF. LPS-induced production of pro-inflammatory cytokines such as IL-6 and $TNF-{\alpha}$ was significantly reduced by HF-f compared to control in RAW264.7 cells. Consistent with reduction of anti-inflammatory cytokine, interleukin (IL)-6, and tumor necrosis factor $(TNF)-{\alpha}$ levels by HF-f, HF-f also significantly reduced phosphorylation of ERK and JNK in LPS-stimulated RAW264.7 cells. In addition, LJ-f and HF also significantly reduced phosphorylation of JNK and ERK induced by LPS in RAW264.7 cells. Overall, our result suggests that HF-f among the four tested seaweed extracts is the most potent anti-inflammatory agent, and its mechanism of action is partially mediated by reduction of JNK and ERK phosphorylation as well as IL-6 and $TNF-{\alpha}$ production in LPS-stimulated RAW264.7 cells.

광주지역 중년여성의 코로나바이러스 감염증 19 시대 식생활 및 라이프 스타일 변화 연구 (A Study on the Dietary and Lifestyle Changes of Middle-Aged Women in the Gwangju Area in the COVID-19 Era)

  • Objectives: This study aimed to investigate the changes in the eating habits and lifestyle of middle-aged women in Gwangju during the COVID-19 pandemic. Methods: A total of 428 middle-aged women aged between 40 and 60 participated in a survey relating to general information, food and lifestyle, health functional food, and menopausal symptoms. The correlation between the variables was analyzed. Results: In the positive habits, the intake of nutritional supplements for immunity enhancement increased the most, followed by the use of media to learn healthy eating tips, and diets including healthy food. Negative habits increased in the order of frequency of taking delivery orders, levels of stress or anxiety, and time spent sitting or watching movies. In the case of recommended foods, the intake increased the most in the order of eggs, fruits, vegetables, milk/dairy products, and seaweed. Non-recommended foods increased in the order of meat, bread, rice, and noodles. The awareness of health functional foods was in the increasing order of interest, knowledge, consumption experience, and purchase amount. The type of health functional food intake was in the increasing order of probiotics, multivitamin and mineral supplements, vitamin C, collagen, and omega-3. Menopausal symptoms were in the increasing order of bone and joint pain, poor sleep quality, emotional ups and downs, loneliness, and feeling of emptiness. In the correlation of major variables, positive habits showed a significant positive correlation with recommended food intake and the recognition of health functional foods. Negative habits showed a significant positive correlation with non-recommended food intake and a significant positive correlation with menopausal symptoms. Recommended food intake showed a significant positive correlation with health functional food recognition and intake and menopausal symptoms. Conclusions: This study suggests that it is necessary to establish social measures to reduce the negative effects of the COVID-19 pandemic on middle-aged women and to ensure effective self-management through a healthy lifestyle since the pandemic has a long-term impact.

Effects of Bacillus SW1-1 coated diets on innate immunity and disease resistance of olive flounder Paralichthys olivaceus against Edwardsiella tarda infection

  • Bacillus SW1-1 is a probiotic isolated from shrimp intestines. We investigated the effects of Bacillus SW1-1 coated diets on the growth, feed utilization, innate immunity, hematological parameters and resistance to Edwardsiella tarda in olive flounder (Paralichthys olivaceus). A commercial diet was used as the control (AP0) and two other diets were prepared by coating 0.25% (AP25) or 0.50% (AP50) probiotic powder which contains 1.0 × 107 CFU/g Bacillus SW1-1. Four replicate groups of olive flounder (153 ± 2 g) were fed one of the diets for 12 weeks. Growth performance and feed utilization of the fish were not significantly affected by the dietary Bacillus SW1-1. After the challenge with E. tarda, AP50 group showed significantly higher survival than AP0 and AP25 groups. Innate immunity and anti-oxidant capacity of the fish were not significantly affected after the feeding trial. However, after the E. tarda challenge, the innate immune parameters (immunoglobulin, lysozyme and anti-protease) were significantly improved in fish fed AP25 and AP50 diets compared to those in fish fed AP0 diet. After the challenge test, significantly lower glucose level was observed in AP50 group compared to AP0 group. These results indicate that dietary supplementation of Bacillus SW1-1 could increase the disease resistance of olive flounder against E. tarda infection. The optimum coating levels of Bacillus SW1-1 needs to be further elucidated.

Dietary encapsulated Bifidobacterium animalis and Agave fructans improve growth performance, health parameters, and immune response in broiler chickens

  • Objective: The present study was conducted to evaluate the effects of dietary supplementation with Bifidobacterium animalis, Agave fructans, and symbiotic of both encapsulated on growth performance, feed efficiency, blood parameters, and immune status in broiler chickens, and to compare these with diets including antibiotic growth promoters and without additives. Methods: A comparative experimental study was carried out with 135 male Ross 308 broiler chickens. Each trial was divided into 5 equal groups. Control group (CON) received a standard diet without growth promoter; GPA, a standard diet with colistin sulfate and zinc bacitracin (0.25 g/kg of feed); PRE, a standard diet with 1% Agave fructans; PRO, a standard diet with Bifidobacterium animalis (11.14±0.70 log CFU/g); SYM, a standard diet with B. animalis and Agave fructans. Results: A significant decrease in food consumption was found for the GPA, PRE, and SYM, compared to the CON group. The results show a better feed conversion index in PRE and GPA with respect to the CON group with the highest conversion index. Interestingly, the weight of the gastrointestinal tract shows a statistically significant difference between GPA and PRE groups. Moreover, the length of the gastrointestinal tract of the GPA group was less than the PRE group. In the total leukocyte count, there was a statistically significant increase in the GPA group compared to the CON, PRE, and PRO groups, and the heterophiles-lymphocytes index was lower in PRO. Regarding the cytokines, interleukin 10 (IL-10) decreased in PRO compared to CON and PRE, while IL-1β increased in the SYM group. Conclusion: Alternative treatments were shown to achieve similar productive results as growth-promoting antibiotics and showed improvement over diet without additives; however, they have immunomodulatory properties and improved the development of the gastrointestinal tract compared to the treatment of growth-promoting antibiotics.
