Effects of Dietary Lipid Sources and Levels on Lecithin : Cholesterol Acyltansferase Activity and Cholesterol Metabolism in Rats (식이지방의 종류와 수준이 흰쥐의 Lecithin : Cholesterol Acyltransferase 활성 및 콜레스테롤대사에 미치는 영향)

  • This study was carried out to investigate the effects of different sources and level of dietary lipid on lecithin : cholesterol acyltrasferase activity and cholesterol metabolism in male rats of Sprague-Dawley strain. The effects of different lipid sources was compared with sardine oil($\omega$3 EPA and DHA), beef tallow(SFA), perilla oil($\omega$3 linolenic acid) and corn oil($\omega$6 linoleic acid). Diets were formulated in such a way that 10%, 20% and 40% dietary energy were supplied with each of four experimental lipid sources. Control diet contained only non-lipid energy. A total number of 78 rats, equally divided into 13 groups, were fed the experimental diets for a period of 6 weeks. In vitro cultures were also carried out to study the cholesterol synthetic activity in the liver prepared from rats used in feeding trials. The concentration of plasma total cholesterol, HDL-cholesterol, LDL-cholesterol and HDL-C/T/C(total cholesterol) ratio were significantly (p<0.001) influenced by dietary lipid sources. Higher HDL-cholesterol and lower LDL-cholesterol concentration in plasma were obtained in rats fed $\omega$3 fatty acid supplemented diets(sardine oil and perilla oil group) compared to diets containing $\omega$6 and saturated fatty acid(corn oil and beef tallow group). In total cholesterol concentration of plasma, beef tallow group was significantly (p<0.001) higher than other lipid groups, and non-lipid group was significantly(p<0.05) higher than the lipid supplemented groups. The activity of lecithin : cholesterol acyltransferase(LCAT) in plasma was greatly(p<0.001) affected by dietary lipid sources and levels. In LCAT acivity of plasma, lipid supplemented groups were significantly(p<0.05) higher than non-lipid group, vegetable oil groups were significantly (p<0.001) higher than animal fat groups, and sardine oil group were significalylty (p<0.001) higher than beef tallow group. Also perilla oil group was significanlty (p<0.05) higher than corn oil group, and sardine oil group was significantly (p<0.05) higher than perilla oil group. Low lipid group, compared with medium or high lipid group, showed higher activity of LCAT in plasma. In cholesterol synthetic activity of liver tissues culture, sardine oil group($\omega$3 EPA and DHA) was significantly(p<0.001) higher than other lipid groups, non-lipid group was significantly(p<0.001) higher than the lipid supplemented groups, and amimal fat group were significantly(p<0.001) higher than vegetable oil groups, but the synthetic activity was not affected by dietary lipid levels.

Effects of Protein and Lipid Levels in Extruded Pellets on the Growth and Body Composition of the Olive Flounder Paralichthys olivaceus during the Summer and Whiter Seasons

  • Feeding trials were performed at two different water temperatures (summer and winter seasons) to identify suitable protein and lipid (energy) levels to be used in formulating extruded pellets for olive flounder. Experiments were conducted to determine the effects of protein and lipid levels in extruded pellets on the growth and body composition of the flounder in both feeding trials. Six experimental diets were prepared containing three protein levels (46%, 51%, and 56%) and two lipid levels (10% and 17%). In the first experiment, during the summer season ($22{\pm}2.2^{\circ}C$), a triplicate group of fish (initial weight, 114 g) were fed to satiation one of the six diets for 11 weeks. The highest weight gain was observed in fish fed the 56/17 (% protein/% lipid) diet, but this weight gain was not significantly different from that of fish fed the other diets, except for those fed the 46/10 diet. The feed efficiency and protein efficiency ratio of fish fed the 17% lipid diets were higher than those of fish fed the 10% lipid diets at each protein level. In the second experiment, during the winter season ($13{\pm}1.5^{\circ}C$), a triplicate group of fish (initial weight, 107 g) were fed to satiation one of the six diets for 9 weeks. Weight gain was not significantly different among all groups. The feed efficiency and protein efficiency ratio tended to increase with increasing dietary lipid level at each protein level. The whole-body crude lipid content of the of fish fed the 17% lipid diets was significantly higher than that of fish fed the 10% lipid diets at each protein level in both feeding trials. Based on the data obtained in this study, the inclusion of dietary protein at a level of 46% appears to be sufficient to support optimal growth, and increasing the dietary lipid level from 10% to 17% had no beneficial effects on the growth and feed utilization of olive flounder (110-300 g), except for fish fed a 56% protein diet in the summer season.

Correlation of Dietary and Serum Phospholipid Fatty Acid Composition and Serum Lipid Levels in Postmenopausal Women (폐경기 여성의 식이 지방산 및 혈청 인지질의 지방산 조성과 혈청 지질 농도의 관계)

  • The purpose of this study was to investigate the correlation of dietary and serum phospholipid fatty acids composition and serum lipid levels in postmenopausal women. Data about anthropometry, dietary intake, physical activities, serum lipid profile and serum phospholipid fatty acid composition were collected from eighty-five postmenopausal women. The subjects were classified as normocholesterolemia (NC), moderate hypercholesterolemia(MC) and phyperchollesterolemia(HC) according to their serum total cholesterol (TC) levels based on The Guideline for Korean Hyperlipidemia set/published by The Committee for Hyperlipidemia in Korea. The results were as follows. Total energy intake and the ratio of energy intake to energy expenditure were positively related, while dietary fiber intake was negatively related, with serum TC level. Dietary fat intake was positively related with serum LDL-C level. Dietary cholesterol, saturated fatty acid (SFA), monounsaturated fatty acid (MUFA) intake and the ratio of ingested saturated fat and cholesterol to calories (RISCC) were positively related, while the P/S ratio of dietary fat was negatively related, with serum TC and LDL-C levels. Serum phospholipid fatty acids composition was not significantly different among the three groups. SFA of serum phospholipid fatty acids was positively related, while P/S and M/S ratios of serum phospholipid fatiy acids were negatively correlated, with serum TC and LDL-C levels. We recommend that the dietary P/S ratio of postmenopausal women is increased by reducing their dietary SFA intake. With these changes in the diet, serum phospholipid fatty acid composition could be also changed, and serum lipids levels could be improved.

  • This study was conducted to examine the effects of mulberry fruit tea (MFT) on the levels of serum lipid profiles and serum cardiovascular disease markers in sixty middle-aged people(30 males and 30 females) who consumed MFT for 4 weeks. Anthropometric measurements, serum lipid profile levels, and serum cardiovascular disease markers were analyzed before and after consumption of MFT. After consumption of MFT, no significant differences were observed in anthropometric measurements, levels of serum aspartate transaminase (AST) and alanine transaminase (ALT) in males and females. Among serum lipid profiles, triglyceride and LDL-cholesterol were decreased significantly, whereas HDL-cholesterol was significantly increased.

Effect of Glutinous Barley Intake on Lipid Metabolism in Middle-Aged Rats Fed a High-Fat Diet

  • This study was designed to determine whether dietary glutinous barley (GB) affects lipid metabolism in middle-aged rats previously fed a high-fat diet. To induce obesity, 20 male 9-month-old Sprague Dawley rats were raised for 1 month on a diet containing 20%(w/w) lipid. The rats were allocated to 1 of 2 groups of 10 rats each and for the subsequent 2 months were fed an 8%(w/w) lipid diet containing well-milled rice (WMR) or GB powder. Rats fed the GB diet had significantly lower concentrations of plasma triglyceride, plasma total cholesterol, and liver cholesterol than rats fed the WMR diet. Fecal excretions of triglyceride and bile acids were significantly greater for the GB group than for the WMR group. In conclusion, dietary GB has positive effects on lipid metabolism: it decreases plasma cholesterol concentration by increasing fecal excretion of bile acids.

  • This study was conducted to investigate the synergic effect of dietary selenium and methionine levels on hepatic lipid metabolism in ethanol treated rats. Sprague-Dawley male rats were fed diets containing three levels of methionine(0,3 and 9g/kg diet) with or without selenium(0.45mg/kg diet). Ethanol was administered with 25%(v/v) ethanol orally at the same time once a day in ethanol group and isocalori sucrose was administered to the control group. The rate were sacrificed after 5 and 10 weeks of feeding period. Glutathione content was decreased by ethanol treatment and significantly increased in proportion to level of dietary methionine and was higher in selenium deficiency group than that of selenium admin-istration group. Lipid peroxide content was significantly increased in deficiency of both methionine and selenium(LMet-Se+EtOH) group. Total lipid triglyceride and cholesteol contents in liver were increas-ed and phospholipid content was decreased in ethanol treated group and ethanol treatment accelerated those increment and decrement in methionine deficiency(LMt) group and excessive methionine admin-istration(HMet)group.

Effects of the P/S Ratio of Dietary Lipids and Antioxidant Vitamin Supplements on the Level of Serum Lipids and Liver. Lipid Peroxidation in Rats Treated with DMBA

  • This study an analyzes the effects of the P/S ratio of dietary lipids and antioxidant vitamin supplements on serum lipids level and fatty acid profile, the degree of lipid peroxidation, and the antioxidant enzyme activities in the liver of rats treated with 7,12-dimethylbenz($\alpha$) anthracene(DMBA). P/S ratio of dietary lipids was made into 0.5, 1 and 2 by mixing palm oil, soybean oil, sesame oil and perilla oil at 10%(w/w) fat level and n-6/n-3 ratio was fixed to 4. Antioxidant vitamin of $\alpha$-tocopherol or $\beta$-carotene was supplemented in addition to vitamin mixture which was given at 1 % of the standard diet. female Sprague-Dawley strain rats, about 60 days old, were divided into three groups(LP : low P/S ratio(0.5), MP : medium P/S ratio (1.0), HP , high P/S ratio(2.0)) and each group was sub-divided into three groups(S ; standard, T ; tocopherol supplemented, C : carotene supplemented): Two weeks after feeding experimental diets, all groups were treated with a single dose of DMBA(2mg/100g BW) by gastric intubation and fed experimental diet for 9 week. The results were as follows ; 1) Serum total cholesterol(TC) level was not significantly influenced by diet but tended to be lower in HP groups compared to LP and MP groups. Triglyceride level was the highest in LP groups and the lowest in $\alpha$-tocopherol supplemented groups. 2) Thiobarbituric acid reactive substance(TBARS) level, representing lipid peroxidation in hepatic microsome, tended to be increased as the unsaturation of dietary lipids increases. $\alpha$-Tocopherol supplement significantly decreased TBARS level. 3) The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSHPx) in hepatic cytosol showed the tendency to be high with increasing P/S ratio of dietary lipids. SOD activity was not significantly influenced by antioxidant vitamin, but GSHPx activity was significantly increased in $\alpha$-tocopherol supplemented groups. In summary, high polyunsaturated fat diet was effective on reducing the serum level of total cholesterol and triglyceride, while it increased unsaturation and peroxidizability of serum fatty acid. With increasing P/S ratio of dietary lipids, lipid peroxidation was increased in the liver and antioxidant enzyme system was induced to inhibit lipid peroxidation against oxidative damage. $\alpha$-Tocopherol supplement was effective in lowering lipid peoxidation, but $\beta$-carotene supplement did not exhibit antioxidant effect. Effects of Packaging Methods on the Shelf Life of Selenium-Supplemented Chicken Meat during Refrigerated Storage

  • Effects of vacuum packaging and modified atmosphere packaging (MAP) on shelf life of selenium-supplemented chicken meat during refrigerated storage were examined. Body weight and feed efficiency were unaffected by dietary selenium level. Dietary treatments and packaging methods had no significant effects on level of microbial growth, lightness, and metmyoglobin content. MAP decreased purge loss (10.9-34.5%) and lipid oxidation (15.2-15.9%) more efficiently than vacuum packaging. Broiler chicks supplemented with ${\alpha}$-tocopherol or ${\alpha}$-tocopherol + selenium had similar TBA values. Dietary supplementation of 4 and 8 ppm selenium reduced lipid oxidation, and this effect was less significant in MAP breast meat.

Relationships of Potential Atherogenic Indices to Anthropometric Measurements, Dietary Intake and Dietary Behavior in Korean Obese Children

  • Epidemiological studies have indicated that a relationship between overweight and cardiovascular disease exists. To assess the relationship of unfavorable changes in serum lipid concentrations, apolipoprotein B, lipoprotein(a), blood sugar and blood pressure to anthropometric variables, nutrient intakes and dietary behavior,64 obese children were chosen as subjects and 24-hour dietary intake, dietary behavior as well as blood pressure, apolipoprotein B, lipoprotein(a), three kinds of cholesterol concentrations were measured in November 1976. More than half(57.8%) of the children had serum cholesterol concentrations over 175mg/d1, and 26.6% of them had over 200mg/d1 and this group could be classified as a high risk group. Abdominal obesity as measured with waist circumference and waist hip ratio was negatively co..elated to high density lipop개tein(HDL) cholesterol. Dietary behavior rather than nutrient intakes appeared to have associations with unfavorable lipid profiles. Total dietary behavior scores and fish and bean product consumption had strong correlations with potentially atherogenic lipoprotein concentrations. This study suggests that fir the early prevention of cardiovascular disease, waist hip ratio together with obese rate should be included in selective programs to identify risk group of children in Korea. furthermore, interventions in cardiovascular disease risk groups in obese children should emphasize maintenance of ideal body weight through reducing body fat and adopting desirable dietary behaviors such as increasing intake of fish or bean products. Effect of Dietary Fiber on the Serum Lipid Level and Bowel Function in Aged Rats (노화된 흰쥐에서 식이섬유질원이 첨가된 식이가 혈청지질과 장기능에 미치는 효과)

  • This study was performed to investigate the influence of dietary fibers on the serum lipid level and bowel function in aged rats. Fiber sources of experimental diets were made from mugwort(M), butterbur(B), apple(A), sea mustard(S) by drying and milling. Each of fiber sources was mixed into the diet at the level of 5, 15% of diet. Sprague-Dawley strain, 13 month old male rats were divided into 9 groups by randomized complete block design : C, M5, B5, A5, S5, M15, B15, A15, S15. The animals were fed ad libitum each of experimental diets for 4 weeks. Control group showed lower food intake compare to the other groups. There were no significant difference between 5% groups and 15% groups in food intake. Fecal weight, dry fecal weight and fecal water content of control group were significantly lower than those of experimental groups, and fecal water content was increased by increasing level of dietary fibber. Apple group showed the lowest values, sea mustard group showed the highest. The shorter transit time was observed in the group of higher intake of dietary fiber. At the same level of dietary fiber, transit time of sea mustard group was shorter than the other groups. With increasing age, serum triglycerides, total cholesterol were increased and HDL-cholesterol was decreased. Fiber fed groups showed lower serum TG, TC and higher HDL-c level compare to the control group. Absorption rates of calcium, magnesium and phosphorus was tend to be lower in the group fed dietary fiber sources than control group. Mucosa weight and maltase activity in the small intestine were decreased by increasing age. As intake of dietary fiber increaed, mucosa weight in the small intestine was not different but maltase activity was decreased.
