Objectives: This study was designed to investigate the anti-obesity, anti-diabetic and anti-inflammatory effects of Platycodi radix on obese type 2 diabetes mouse model. Methods: Obese type 2 diabetes mouse model was induced by Surwit's high fat, high sucrose diet for 8 weeks. Models were divided into 4 groups of normal diet (ND, n=10), high fat and high sucrose diet (HFD, n=10), high fat and high sucrose diet with Platycodi radix (PR, n=10), and high fat and high sucrose diet with Metformin (Met, n=10). Body weights were measured every week. After 7 weeks fasting, blood sugar and oral glucose tolerance tests were conducted. After 8 weeks blood samples were taken from mouse hearts and analyzed biochemically. Lipid profile, fructosamine, leptin and weight of epididymal fat pad and liver were measured. Adipose tissue macrophage percentage was analyzed by fluorescence-activated cell sorting (FACS). Results: Compared with the HFD group, body weight, glucose level, fructosamine, weight of epididymal fat pad and adipose tissue macrophage percentage decreased in the PR group. Conclusions: These results suggest that Platycodi Radix has anti-obesity, anti-diabetic, and anti-inflammatory effects on obese type 2 diabetes mouse model.
Obesity is common disease resultly accumulated excess fat. In the model for obesity induced by high fat diet contains 30% fat, administration of ginseng extract inhibited increment of body weight, epididymal fat pads and enlargement of fat cell size. This was as the result of inhibition of lipogenesis in the liver and fat accumulation in the adipose tissues.

[Purpose] Blood glucose and insulin resistance were lower following hypoxic exposure in previous studies. However, the effect of hypoxia as therapy in obese model has not been unknown. [Methods] Six-week-old mice were randomly divided into chow diet (n=10) and high-fat diet (HFD) groups (n=20). The chow diet group received a non-purified commercial diet (65 % carbohydrate, 21 % protein, and 14 % fat) and water ad libitum. The HFD group was fed an HFD (Research Diet, #D12492; 60% kcal from fat, 5.24 kcal/g). Both groups consumed their respective diet for 7 weeks. Subsequently, HFD-induced mice (12-weeks-old) were randomly divided into two treatment groups : HFD-Normoxia (HFD; n=10) and HFD-Hypoxia (HYP; n=10, fraction of inspired=14.6%). After treatment for 4 weeks, serum glucose, insulin and oral glucose tolerance tests (OGTT) were performed. [Results] Homeostatic model assessment values for insulin resistance (HOMA-IR) of the HYP group tended to be lower than the HFD group. Regarding the OGTT, the area under the curve was 13% lower for the HYP group than the HFD group. [Conclusion] Insulin resistance tended to be lower and glucose uptake capacity was significantly augmented under hypoxia. From a clinical perspective, exposure to hypoxia may be a practical method of treating obesity.
Objectives : The purpose of this study was to investigate how Rhei Radix et Rhizoma affects on insulin resistance and adipose tissue inflammatory response in high fat diet induced obese C57BL/6 mice. Methods : Obesity was induced in C57BL/6 mice by high fat diet for 12 weeks. Models were divided into 3 groups (n=6) of normal diet, high fat diet (HFD), and high fat diet with Rhei Radix et Rhizoma and investigated for 12 weeks. We measured body weight, FBS and oral glucose tolerance test (OGTT), serum insulin, homeostatic model assessment-insulin resistance (HOMA-IR), weight of liver and epididymal fat pad. Inflammatory markers such as adipose tissue macrophage (ATM), tumor necrosis factor-${\alpha}$ and interlukin-10 and CD68 of epididymal adipocyte were determined to evaluate the effect of Rhei Radix et Rhizoma on adipose tissue inflammation. Results : Compared with the HFD group, we observed loss of body weight and epididymal fat pad weight, improvement of glucose level and HOMA-IR, reduction of ATM and gene expression of TNF-${\alpha}$, CD68 in the high fat diet with Rhei Radix et Rhizoma group. Conclusions : This study suggests that Rhei Radix et Rhizoma has effects on insulin resistance and adipose tissue inflammatory response in high fat diet induced obese mice.
This study was carried out to investigate the dietary effects of Monascus pilosus mycelial extract on obesity in high-fat with cholesterol-induced obese rat models. It was observed that M. pilosus mycelial extract contains $25.85{\pm}1.98mg%$ of total monacolin K without citrinin by highperformance liquid chromatography (HPLC). The rats were randomly divided into 2 groups; normal control and a high-fat with cholesterol diet group. The high-fat with cholesterol diet group was fed a 5L79 diet with an added 15% lard and 1% cholesterol supplemented diet for 3 weeks for induction of obesity. After induction, obesity was confirmed by checking obesity indexes, the animals were divided into 4 groups (n=5); first, the normal control (NC), and then taken from the obese model of rats, a high-fat with cholesterol diet obesity control group (HF), 0.5% M. pilosus mycelial extract supplemented high-fat with cholesterol diet group (MPMs), 2% conjugated linoleic acid supplemented high-fat with cholesterol diet group (CLA) for 7 weeks. Body weight gains, obesity indexes, and body fat contents in the experimental groups (MPMs and CLA) were decreased compared with HF group. Feed Efficiency Ratio (FER) in MPMs was significantly lower than that of HF without change of feed intake. These results suggested that the anti-obesity effects of the M. pilosus mycelial extracts (MPMs) could prevent obesity induced by high-fat with cholesterol diet possibly via inhibition of lipid absorption.
This study was conducted to investigate anti-oxidative and inflammatory inhibition effects of green tea and dietary fiber mixture on liver of high fat diet-induced obese rats. 21 male rats were divided into 3 dietary groups and control group (A), high fat diet-induced group (B), and high fat (HF) diet-induced + EQ diet-$S^{(R)}$ diet group (C). Immunoblotting and RT-PCR analysis revealed protein expression, and anti-oxidant analysis revealed MDA (malondialdehyde), GSH (glutathione), and free DPPH radical. As a results, Body weight and food consumption were not significantly different between groups. The levels of MDA and GSH were lower in HF + $EQS^{(R)}$ group than in HF group. Also, the $EQS^{(R)}$ demonstrated to be more effective than HF group for a DPPH radicals scavenging activities. In addition, protein and mRNA level of TNF-$\alpha$ in HF + $EQS^{(R)}$ group showed relatively more potent pro-inflammatory activity inhibition compared to HF group. These results suggest that green tea mixture (EQ diet-$S^{(R)}$) provide positive effects on anti-oxidative and inflammatory inhibition effects on obese animal model or obesity related diseases.
Objectives : Obesity is an important cause of diabetes, and lipotoxicity causes insulin resistance. In this study, we investigated the effects of Salvia miltiorrhiza on high fat diet-induced obese type 2 diabetic mouse models. Methods : Diabetes was induced in ICR male mouse (23~25 g) with Surwit's high fat, high sucrose diet. Mice were divided into 4 groups (n=10) of normal, control, Salvia miltiorrhiza, and metformin. After 8 weeks, body weight, OGTT, fructosamine, lipid profile, serum level of adiponectin and leptin, epididymal fat pad, liver weight and epididymal adipocyte size were measured. Results : Salvia miltiorrhiza significantly reduced oral glucose tolerance levels, fructosamine serum level, epididymal fat weight, and epididymal adipocyte size. Salvia miltiorrhiza also increased HDL-cholesterol, adiponectin and leptin serum levels. Conclusions : These results show that Salvia miltiorrhiza improves insulin resistance. Therefore we suggest that Salvia miltiorrhiza would be an effective treatment for obese type 2 diabetic patients.
This study was carried out to investigate the dietary effects of chitosan mixture (CM), an herbal medicine mixture with Sukjihwang (HS), and CM containing HS (CHS) on obesity in an induced obese model of rats fed high-fat only (HF), in which supplemented diets of 5% CM (HCM), 5% HS (HHS), or 2.5% CM-2.5% HS (HCH) was tested for 6 weeks. Body weight gains, obesity indexes, and body fat contents in the experimental groups (HCM, HHS, HCH) were decreased compared with HF group. The levels of serum triglyceride, total lipid, total cholesterol and LDL-cholesterol in the experimental groups were markedly decreased, however HDL-cholesterol levels in the experimental groups were slightly increased compared with HF group. In addition, although serum ALT and AST activity, and relative organ weights were lower than those of HF group, serum albumin contents were not significantly different in all experimental groups including the normal control group (NC). In conclusion, there are improved effects on obesity in the obese model of animals with all experimental diets supplementations, and the improvement degrees on obesity depend on the content and compositions of the herbal medicine mixture. Further study is needed on the anti-obesity mechanism of these diets.
BACKGROUND/OBJECTIVES: Seaweeds have been reported to have various health beneficial effects. In this study, we investigated the potential anti-obesity and anti-inflammatory effects of four types of domestic brown seaweeds in a high-fat diet-induced obese mouse model and bone marrow-derived macrophages (BMDM). MATERIALS/METHODS: Male C57BL/6N mice were fed low-fat diet (LFD), high-fat diet (HFD) or HFD containing Undaria Pinnatifida, HFD containing Laminaria Japonica (LJ), HFD containing Sargassum Fulvellum, or HFD containing Hizikia Fusiforme (HF) for 16 weeks. RESULTS: Brown seaweed supplementation did not affect long-term HFD-associated changes in body weight or adiposity, although mice fed HFD + LJ or HFD + HF gained slightly less body weight compared with those fed HFD at the beginning of feeding. Despite being obese, mice fed HFD + LJ appeared to show improved insulin sensitivity compared to mice fed HFD. Consistently, we observed significantly reduced blood glucose concentrations in mice fed HFD + LJ compared with those of mice fed HFD. Although no significant differences in adipocyte size were detected among the HFD-fed groups, consumption of seaweeds decreased formation of HFD-induced crown-like structures in gonadal adipose tissue as well as plasma inflammatory cytokines. BMDM from mice fed HFDs with seaweeds showed differential regulation of pro-inflammatory cytokines such as IL-$1{\beta}$ and IL-6 compared with BMDM from mice fed HFD by LPS stimulation. CONCLUSION: Although seaweed consumption did not prevent long-term HFD-induced obesity in C57BL/6N mice, it reduced insulin resistance (IR) and circulation of pro-inflammatory cytokines. Therefore, seaweeds may ameliorate systemic inflammation and IR in obesity partially due to inhibition of inflammatory signaling in adipose tissue cells as well as bone marrow-derived immune cells.

This study aimed to investigate the antidiabetic and antiobesity effects of Supungsunki-hwan partitioned prescriptions on obese type 2 diabetes mouse. Methods : Type 2 diabetes mellitus and obesity were induced by Surwit's high fat, high sucrose diet for 8 weeks. Mice were divided into 3 groups of ND (normal diet, n=10) HFD (high fat and high sucrose diet, n=10) and SPP (high fat and high sucrose diet with Supungsunki-hwan partitioned prescriptions, n=10) groups. Body weights were measured every week. After 7 weeks, fasting blood sugar and oral glucose tolerance tests were conducted. After 8 weeks, blood samples of all mice were taken from their heart and analyzed biochemically. At the same time, epididymal fat pad and liver weights were measured. Histological size of white adipocyte were measured as well. Results : Compared with a HFD group, body weight, fructosamine, epididymal fat pad weight and white adipocyte size decreased. High-density lipoprotein cholesterol levels increased in the SPP group. Conclusions : These results suggest that SPP has antidiabetic and antiobesity effects in high fat, high sucrose diet induced obese mice.
