Highly Active Antiretroviral Therapy Alters Sperm Parameters and Testicular Antioxidant Status in Diet-Induced Obese Rats

  • The efficacy of highly active antiretroviral therapy (HAART) has led to an increase demand for therapeutic use, thereby necessitating investigation into drug toxicity. This study was designed to investigate the in vivo effects of HAART on sperm parameters and testicular oxidative stress in lean and obese rats. Wistar rats (males, n = 40, weighing 180~200 g) were assigned randomly into 4 groups and treated accordingly for 16 weeks as follows: Control (C): lean group fed with standard rat chow; Diet induced obesity (DIO): obese animals fed a high caloric diet; C + ART: lean animals treated with HAART; DIO + ART: obese animals treated with HAART. An antiretroviral drug combination of Tenofovir, Emtricitabine and Efavirenz at a dose of 17, 26 and 50 mg/kg/day was administered for the latter 6 weeks via jelly cube feeding. At the end of the experimental period, sperm analysis was performed on sperm collected from the caudal epididymis, while the testis was homogenized for antioxidant enzyme and lipid peroxidation assays. Results showed that HAART significantly decreased sperm motility (p < 0.05) in both lean and obese animals, and viability (p < 0.05) in the DIO group. Testicular glutathione, catalase and superoxide dismutase were significantly decreased (p < 0.05), while Thiobarbituric acid reactive substances (TBARS) levels were significantly increased (p < 0.05) when the DIO+ART group was compared to Control group. Thus, the decreased sperm qualities associated with HAART might be as a result of increased testicular oxidative stress prominent in obese animals.

Differences in Depression between Over weight and Normal Group who visited to Obesity Clinic for Diet Therapy (비만센터에 내원한 비만환자군과 정상군의 우울정도에 대한 비교고찰)

  • Objective : Obesity is related to psychological problems as well as physical problems. This study was designed to investigate the depression of over weight and normal group who visited to obesity clinic for diet therapy. Methods : 67 patients were selected(obesity group 42, normal group 25), they were treated medically in obesity clinic of Dong-Seo Oriental Hospital. We checked age, sex, education level, relative weight, BMI and Beck Depression Inventory(BDI) in two group. Results : The results were summarized as followed. 1. The ratio of Relative Weights are $137.45{\pm}12.77$ in over weight group, $108.48{\pm}9.11$ in normal group. 2. The ratio of BMI are $28.61{\pm}2.94$ in over weight group, $23.04{\pm}1.83$ in normal group. 3. The results of BDI score are $10.60{\pm}7.15$ in over weight group, $12.56{\pm}8.33$ in normal group. Conclusions : These results suggested that the patients who participated in diet therapy had mild depression and we should pay attention to psycological problem as well as the physical problem when we treat obesity.

A Suggestion to Improve Zinc Status of Type 2 Diabetic Women: Relationship among Zn, Protein and Phytate intake (제2형 당뇨병 여성의 아연영양관리 방안: 단백질 및 피틴산 섭취와의 관련성)

  • The purpose of this study was to analyze the relationships among zinc status, protein and phytate intake, and diabetic control indices of type 2 diabetic women. The mean age and the duration of diabetes were respectively 57.9±6.9 years old and 8.0±6.5 years. The mean daily energy intake of diabetic subjects was 1562 kcal. Both the zinc intake (6.2mg/day) and the zinc %RI (% of The Recommended Intake for zinc: 79.5%) of the diabetic participants were significantly lower than those of the control group (respectively p<0.01). As for the diabetic group, the higher the energy intake (kcal/day), the higher were the zinc intake (p<0.001) and %RI for zinc (p<0.001). Zinc intake was positively correlated with the protein (p<0.001), animal protein (p<0.001), and fat intake (p<0.001), but negatively correlated with the carbohydrate intake (p<0.001). Foods with high amount of phytate were the major source of zinc (p<0.01), but did not contribute to high zinc densities. The urinary zinc excretion was twice as high as in the diabetic group compared to the control group (p<0.001). In addition, the urinary zinc loss was positively correlated with the duration of diabetes (p<0.05), hyperglycemia (p<0.001) and insulin resistance (p<0.05). %RI for zinc was negatively correlated with the HbA1C (p<0.05). These results lead us to conclude that the appropriate intake of energy controlled by diet therapy could improve the total zinc intake and %RI for zinc in diabetic women. Also, normal blood glucose level controlled by diet therapy could improve the hyperzincuria. Dietetic practitioners should encourage consumption patterns that provide zinc-rich foods in the form of animal protein to improve the bioavailability as well as the total daily intake of zinc.

Medical-Nutrition-Therapy for Obese Patients with Type 2 Diabetes Mellitus Undergoing Metabolic Surgery (제2형 당뇨비만환자의 수술요법 시 임상영양치료 프로토콜 설정)

  • Currently, metabolic surgery (Laparoscopic Roux-en-Y gastric bypass, LRYGB) has an important role and should be recommended as an intervention in the management of obese patients with type 2 diabetes mellitus (T2DM). A successful outcome of surgery requires medical nutrition therapy. Therefore, we performed a retrospective study on 25 patients with T2DM who underwent LRYGB at Yeouido St. Mary's Hospital from October 2008 to May 2010. The patients were followed up for an average of 6 months after surgery (range: 2~19 months). Diabetes was resolved in 80% of the patients. Percentage of excess weight loss was (%EWL) was 56.2%. After surgery, fasting blood sugar (FBS) and HbA1c were significantly reduced (123 mg/dl, A1c 6.7%, P<0.001) and triglyceride was also significantly reduced to 107.6 mg/dl (P<0.05). As diets of the patients progressed from liquid to soft to regular diet, energy, carbohydrates, and fat intakes increased significantly (P<0.001). But protein intake did not change significantly. Nutrient intake of the patients after the surgery was significantly lower than the recommended diet for the non-surgery group. Patients experienced side-effects related to the diets after surgery, including hair-loss (76%), smelly gas (52%), vomiting (48%), etc. A significantly positive correlation was observed between vomiting and FBS (P<0.001). There was a significant relationship between side effects and the amount of nutrient intakes. Therefore, guide patients to a diet progression with treatment to minimize side effects, especially vomiting. And monitor their dietary life to be healthy and not to regain weight until remission of T2DM.

The Effects of Treadmill Exercise on Blood components, Antioxidant enzymes and Reactive Oxygen in Hyperlipidemic Rats (트레드밀 운동이 고지혈증 흰쥐의 혈액 성분과 항산화효소 및 활성산소에 미치는 영향)

  • PURPOSE: The purpose of this study was to investigate the effects of treadmill exercise on blood components and antioxidant system in hyperlipidemic rats. METHODS: Three weeks old male rats were randomly assigned into General diet(GD, n=10), High fat Diet(HD, n=10), and High fat diet+Treadmill exercise(HDT, n=10) groups. Treadmill exercise consisted of the treadmill running 5 times per week during 6 weeks(30 min/time for first 3 weeks and 60 min/time the other 3 weeks). RESULTS: The levels of triglyceride and total cholesterol were increased in HD group compared with GD group, and recovered to level of GD group by treadmill exercise(p<.05). Plasma glucose and insulin concentrations were increased in HD group compared with GD group, and recovered to level of GD group by treadmill exercise(p<.05). Glutathione(GSH) and glutathione reductase(GRD) concentrations were decreased in HD group compared with GD group, and these decreases returned to the level of GD group by treadmill exercise(p<.05). Xanthine oxidase(XO) and malondialdehyyde (MDA) concentrations were increased in HD group compared with GD group, and these increases retuned to the level of GD group by treadmill exercise(p<.05). CONCLUSION: This study showed that treadmill exercise application were effective treatment strategy on hyperlipidemia. Therefore, it could be considered as a treatment method in the patients with hyperlipidemia disease. Treadmill exercise, Hyperlipidemia, Blood components.

A Statistical Study on Sikryo-chanryo by Applying Database (데이터베이스를 이용한 식료찬요(食療纂要)의 통계적 연구)

  • This study was, based on traditional know-how indigenous to Korea, to systemize the knowledge on how to improve health by dining, and to make the best of it statistically. For this purpose, the knowledge in the Sikryo-chanryo(食療纂要), in Korean pronunciation and Siglyochan-yo in Chinese characters, which is an old text referring to diet therapy peculiar to Korea, was compiled into a database and analyzed statistically. Data processing was used as a 'Relational data model'. In addition, we have used nine data table to express diet therapy peculiar to Korea in the Siglyochan-yo. The software used for data construction was Microsoft Access 2014. As a result, the Sikryo-chanryo database, which can provide information on both disease treatment by food, medicines, and gourmet ingredients applicable to every kind of symptom, as well as the names of disease, was set up at in a PC interface. By employing the 'Relational data model', we can replace researching in the conventional method by employing the database.

Factors Associated with Attendance in a Nutrition Education Program for Hyperlipidemic Patients

  • This study was conducted to examine factors with attendance in hyperlipidemia nutrition eduction program among 101 hyperlipidemic outpatients (38 males 63 females) at Samsung Medical Center We employed the Health Belief Model (HBM) as the theoretical framework The individual nutrition education and counseling program was scheduled with 4- half hour session sin 2 to 4 weeks intervals. Upon initiation of the program a trained dietitian surveyed HBM constructs and psychosocial factors. The following were included perceived susceptibility to cardiovascular disease(CVD) perceived severity to CVD percieved benefits to diet modification perceived barriers to persistence in maintaining therapy and self efficicacy and social support from family Sociodemographic data health factors stress level nutrition knowledge, and 24-hour dietary recall behavior were also surveyed All these data was analyzed according to the number of nutrition sessions attended The subjects were 55.9$\pm$9.4 year old and 24.6$\pm$kg/㎡ Sociodemographic factors were not associated with the number of nutrition sessions attended HBM constructs and psychosocial factors were significantly associated with the number of nutrition sessions attended. According to Spearman correlation coefficients. From stepwis regression analyses using HBM constructs as independent variables perceive barriers to persistence in maintaining diet therapy (negative) proved to be the strongest predictors for the number of nutrition sessions attended (partial R$^2$= 72.3%) followed by perceived severity to CVD and self efficacy (model R$^2$=76.6% The findings indicate that HBM constructs and psychosocial factors were closely associated with patient attendance It suggests that information and guidance to minimize patients perceived barriers to diet therapy might help to improve patients to scheduled appointments in nutrition education programs.

  • This study aimed to verify the nutritional and curative effects of protein hydroysate and optimal ratio between protein and protein hydroysate as nitrogen source in rats with cysteamine-induced duodenal ulcer. Duodenal ulcer rat model was established by intraperitoneal injections of cysteamine. Sprague-Dawley, female rats weighing approximately 200g were intrapertionealy injected twice cysteamine(13mg/100g BW) at intervals of 3hours per day. This procedure was repeated 3 times at intervals of 3 days. Animals fed on 10% casein diet for injection periods. After last injection, 5 kinds of kiets (the ratio of casein and casein hydrolysate was 100 : 0(C100), 75 : 25(CH 25), 50 : 50(CH 50), 25 : 75(CH 75), 0 : 100(CH 100)) were given. The rate were sacrificed after feeding diet, 1, 3, 5 days. Ulcer index, hexosamine content of stomach and duodeum, gastric motility, trypsin activity, blood glutathione, plasma total protein, albumin, amino-N, urinalry urea nitrogen, creatinine, hydroxyproline and retention rate of nitrogen were analyzed for nutritional effects of diet treatments. There were no differences among diet groups in the view of the growth and diet treatments. There difference of ulcer curation by diet was appeared after 3 days. The ulcer indexes of C100 and CH 25 of 3, 5 days were significantly higher than those of CH 50, CH 75 and CH 100. This result was the same as hexosamine content of stomach, plasma protein, albumin concentration and nitrogen retention rate. The more casein hydrolysate diet had, the lower trypsin activity was. The more casein gydroysate diet had, the higher excretion of hydroxyproline was. These results show that protein hydrolysate can be applied in diet therapy for the patients with gastronitestinal ulcer. It suggests that it has curative effect of diet when nitogen sources include at least over than 50% of protein hydrolysate.

Clinical Case Studies on Hepatoma Patients with Dietary Therapy (식이요법에 의한 간암환자의 일부증상의 호전)

  • This study was performed to evaluate the effects of high supplementary diet of vitamins, minerals, and polysaccharides on enhancing immunity and preventing from cancer in patients with hepatocellular carcinoma(hepatoma). Dietary therapy for patients was performed with dried yeast, wheat germ. Gonoderma extract, and vegetable juice. The vegetable juice was made of fresh green edible vegetables. Ten patients of total 30 patients were considered as improved state in tumor sizes, AFP, or symptoms. Most of patients complained of weight loss, fatigue, indigestion, anorexia before dietary therapy, but most of symptoms disappeared after dietary therapy. There was no significant side effects because of dietary therapy except for occasional slight diarrhea.

  • The purpose of this study was investigation to examine the prevention diabetic mellitus and treatment effect on Gastrodia Elata Blume(GEB) dieted at prior and after induced diabetic application. Prior induced diabetic 3 weeks ago application GEB dieted. It is to analysis changes in body weight, blood glucose, SOD, CAT and histopathological findings. For the fingding significantly concentration diabetic rats were divided into 3 different experimental groups and each groups were induced diabetic. Experimental group Ⅰ (STZ-induced diabetic rats; n=10), experimental group Ⅱ (after induced DM and GEB dieted rats; n=10), experimental group Ⅲ (Prior GEB dieted thereafter DM induced; n=10). Prior and After GEB dieted application was that body weight, blood glucose were increase in experimental group Ⅱ, Ⅲ. Specially, group Ⅲ was significantly change than group Ⅱ at 1st weeks. and the level of CAT were significantly decrease in experimental group Ⅱ, Ⅲ than group Ⅰ. but SOD level was increase in experimental group Ⅱ, Ⅲ than group Ⅰ. In histological observation; group Ⅰ showed decrease in the intensity and incidence of vacuolations, cellular infiltration and hypertrophy of in liver and kidney. The Gomori's stain result, group Ⅰ showed disruption ${\beta}$-cell in pancrease.
