Evaluation on Anti-fungal Activity and Synergy Effects of Essential Oil and Their Constituents from Abies holophylla (전나무 정유의 항진균 효과와 유효성분의 시너지효과 평가)

  • This study was to investigate the antifungal activity of A. holophylla essential oil against dermatophytes, such as Epidermophyton floccosum, Trichophyton mentagrophytes and Trichophyton rubrum, and to determine the potential effective compound as dermatitis treatment. To evaluate the potential antifungal activities of A. holophylla essential oil and its fractions, paper disc diffusion and agar dilution method tested with morphological observation. Also, their major constituents were analyzed by GC/MS. To determine synergic effects of active ingredient from A. holophylla essential oil were carried out by checkerboard microtiter plate testing. The morphological changes of the dermatophytes exposed to active fraction G4 were observed by electron microscopes. As the results, the highest activities were identified in the fraction containing ${\alpha}$-bisabolol. A mixture of ${\alpha}$-bisabolol and bornyl acetate showed the synergy effects, expressing high potential effects. Also, morphological observation using electron microscopes showed a dramatic changes of cell membrane of E. floccosum and T. rubrum exposed to fraction G4 containing ${\alpha}$-bisabolol. In conclusion, A. holophylla essential oil and its constituents were expected to be used as antifungal agent or raw material for dermatitis therapy.

Comparison of Antifungal Activites of Monoterpenes and Sesquiterpenes in Essential Oil from Chamaecyparis obtusa against Dermatophytes (피부사상균에 대한 편백정유의 Mono- 및 Sesquiterpene 항진균 활성 비교)

  • This study was to compare the antifungal activitiy of monoterpenes with that of sesquiterpenes, which were major constituents of antifungal active essential oil of Chamaecyparis obtusa, to investigate which constituents had more effective against dermatophytes, and to evaluate the synergistic effect of combination of the antifungal active constituents. The antifungal activities of seven fractions (A, B, C, D, E, F, and G) from C obtusa essential oil by column chromatography were tested against Trichophyton mentagrophytes. Fraction D, E, F, and G were more active than fraction A, B, and C, and their major compounds were mono- and sesquiterpenes analyzed by GC/MS. Borneol, linalool, and ${\alpha}$-terpineol were selected as monoterpenes, and ${\alpha}$-cedrol, nerolidol, and ${\beta}$-eudesmol as sesquiterpenes, respectively. The antifungal activities of the constituents were respectively examined against Trichophyton mentagrophytes (KCTC6077), Microsporum canis (KCTC6591), and Microsporum gypseum. The constituents of sesquiterpenes were more active than those of monoterpenes. By comparing single and combined sesquiterpenes with fraction F containing the higher ratio of sesquiterpenes, combined sesquiterpenes were the most active. This result indicated that there was the synergistic antifungal effect against dermatophytes when sesquiterpenes were combined together.

Evaluation on Anti-Dermatophyte Effect of Larix (kaempferi) Essential Oil on the Morphological Changes of Eermatophyte Fungal Hyphae (피부사상균 균사의 형태학적 변화를 통한 일본잎갈나무 정유의 항진균 활성 효과 구명)

  • This study was to investigate the antifungal activity of Larix kaempferi essential oil against dermatophytes, Epidermophyton floccosum, Trichophyton mentagrophytes and Trichophyton rubrum. The active components of L. kaempferi against dermatophytes were determined (characterized by GC-MS), and the morphological changes of the dermatophytes exposed to the L. kaempferi essential oil were observed by electron microscope. Main component of L. kaempferi essential oil was (-)-bornyl acetate. In antifungal activity tests, MIC of L. kaempferi crude oil was 125 ppm on every fungi and 100% (agar dilution method) at more than 500 ppm. By using SEM and TEM, the fungal morphology of E. floccosum exposed to the L. kaempferi essential oil was different from that of normal hyphal morphology. Hyphae exposed to the L. kaempferi essential oil was damaged with distorted and collapsed surfaces. In addition, there were destruction and disorganization of organelles in cytoplasm and collapse of cell membrane. Active antifungal components from L. kaempferi essential oil were identified as terpene alcohol compounds like (-)-${\tau}$-muurolol, (+)-terpinen-4-ol, ${\alpha}$-terpineol, and ${\alpha}$-cadinol.

Antifungal and Antioxidant Activities of Volatile Organic Compounds Generated During the Drying Process of Chamaecyparis obtuse (편백나무 건조 중 발생하는 휘발성 유기화합물의 항진균 및 항산화 활성)

  • In this study, we evaluated antifungal and antioxidant activities of condensed liquid of volatile organic compounds generated during the drying process of Chamaecyparis obtuse. Condensed liquid of volatile organic compounds were extracted with hexane and ethyl acetate, respectively. The extracts were used for antifungal activity at range from 100 to 500 mg/L Antifungal activity of extracts increased as the extracts concentration increased, the activity against tested dermatophytes was high at 500 mg/L. Among extract solvents, antifungal activity was higher at hexane extract than the ethyl acetate. Especially, the antifungal activity against Trichophyton rubrum was higher than other tested dermatophytes. Antioxidant activity was 90% above 25 mg/L regardless of the extractive solvents, while the hexane extract showed high antioxidant activity below 25 mg/L. Major compounds of hexane extract were torreyol, alpha-cadinol, and tau-cadinol. However, major compounds of ethyl acetate extract were alpha-amorphene, alpha-cadinol, and gamma-cadinene.

  • The antifungal activity of methanol extracts against dermatophytes was the highest at root-bark methanol extract, and the highest inhibitory effect was revealed in petroleum ether fraction of root-bark methanol extract. Compound I and compound II with significant antifungal activity were isolated from the fractions by silica gel column chromatography. As a result of the instrumental analyses, compound I and compound II were already known alkaloids. Compound I was identified as 4-methoxyfuro[2,3-6]quinoline (dictamnine ; $C_{12}H_9NO_2$) and compound II was identified as 4-methoxy-lmethyl-2(1H)-quinolinone ($C_{11}H_{11}NO_2$). The MIC of compound I against T. mentagrophytes and T. rubrum was $40{\mu}g/m\ell$ and the MIC of compound II against the same fungi was $800{\mu}g/m\ell$.

In vitro Evaluation of Antidermatophytic Activity of Egyptian Bee Propolis in Combination with Plant Essential Oils in Sheep Hoof Plate: An Experimental Model

  • Bee propolis ethanolic extract with some plant essential oils was investigated for its antidermatophytic properties. The tested plant essential oils included jasmine, clove, lemon, Arabian jasmine, mint, rosa, olive and basil. The antidermatophytic activity has been compared to Naftifine-HCl and Clotrimazole used for dermatophyte treatment. Experimental model has been tested using sheep hoof plate for the in vitro tests to stimulate human nails. Mint, clove and basil with 4 mg/ml of bee propolis have a comparable efficacy to those of Naftifine-HCl and Clotrimazole. There is a great necessity for new effective low price and safe antidermatophyte agents to avoid recurrent infection. Propolis synergistic could be of great importance with essential oils of plants in dermatophyte therapy.

  • The present study is of ultra-fine structures of Trichophyton rubrum, Trichophyton mentagrophytes, Microsporum canis and Epidermophyton floccosum by means of electron microscopy and reveals the following. 1. In contrast to the bacteria, the normal fungus contains nuclear membrane, mitochondria, endoplasmic reticulun, distinct cell wall and cell membrane and secretory granules as observed in the higher plants and animals. 2. Thickening of the cell wall, inapparent cell wall, inapparent cell membrane with the appearance of electron thin area(ETA) and increase of inclusions were observed in the L-1 treated groups. 3. Thickening of cell wall and increase of ETA were more apparent in the Epidermophyton floccosum than the other groups. 4. Increase of electron thin area was thought to be associated with autolysis.

  • Microsporum gypseum is a geophilic fungus abundant in soil throughout the world. M. gypseum has been identified for the first time in Korea at 1976. The purpose of this study was to investigate epidemiologic aspects of M. gypseum. We performed the epidemiologic study an 198 patients with M. gypseum infections in from January 1976 to December 2005. From 100,012 patients, dermatophytes were isolated as follows; Trichophyton rubrum was isolated from 86,553 (86.5%), T. interdigitale 3,991 (4.0%), Epidermophyton floccosum 1.021 (0.1%), T. tonsurans 141 (0.1%). T. mentagrophytes 3.930 (3.9%), T. verrucosum 220 (0.2%), Microsporum canis 3.879 (3.9%), and M. gypseum 198 (0.2%). Number of patients with M. gypseum by site and age were as follows; most patients infected with tinea corporis were young people under 10 years. M. gypseum were most frequently isolated in summer and autumn.

  • The antifungal activity of salicylaldehyde-o-phenylenediimine, a derivative of Schiff base, was studied in vitro against various species of fungi, especially, dermatophytes. The antifungal tests were performed according to the tube-dilution method, and all subcultures were incubated at room temperature for 14 days. Salicylaldehyde-o-phenylenediimine was effective against fungi such as Trichophyton mentagrophytes, Trichophyton rubrum, Microsporum canis and Fonsecaea compacta. Trichophyton mentagrophytes was inhibited at $20{\mu}g$ per ml, Trichophyton rubrum at $40{\mu}g$ per ml, Microsporum canis at $6{\mu}g$ per ml, and Fonsecaea compacta at $2{\mu}g$ per ml.

  • This study was carried out to investigate the anti-fungal effect of Pyrethri Flos Boiled Solution (PFBS) from Chrysanthemum Cinerariaefolium Visiani on Microsporum canis, Microsporum gypseum and Trichophyton Mentagrophytes with 0.5 ml, 1ml, 2 ml. In comparative drugs, on Sabouraud's glucose agar medium to 2 ml respectively and PFBS on the rabbit skin lesions, in vivo experiments, were observed. The results obtained were as follows : PFBS slightly suppresed the growth of the fungi in 0.5 ml and markedly it in 1 ml and 2 ml. In comparative drugs, benzylbenzoate represented similar anti-fungal action in 2 ml compared with PFBS. While, salicylic acid and boric acid represented less suppression of growth of the fungi than PFBS in 0.5 ml. The effects of PFBS on the rabbit skin lesions inoculation with dermatophytes was almost normally recorved about Microsporum gypseum, Trichophyton mentagrophytes except Microsporum canis after 8 days.
