대황 추출물의 항산화 활성 및 MMP-1 저해 활성 (Effect of Rheum undulatum Extract on Antioxidant Activity and Activity of Matrix Metalloproteinase-1 in Human Skin Fibroblasts)

  • 본 연구에서는 대황 추출물에 의한 항산화 효과, MMP-1 활성 저해효과 및 자외선이 조사된 사람 피부 섬유아세포에서 MMP-1 발현에 미치는 영향을 관찰하였다. 대황 추출물의 DPPH radicals과 초산소 음이온 라디칼 소거효과는 농도 의존적인 경향을 나타내었으며, DPPH와 초산소 음이온 라디칼을 각각 20 ${\mu}g$/ml에서 72%와 80% 정도 소거하여 우수한 항산화 효과를 나타내었다. 자외선이 조사된 사람 피부 섬유아세포에서 MMP-1의 발현 저해효과 측정에서 대황 추출물은 1 ${\mu}g$/ml에서 약 79.5%로 매우 우수한 합성 저해효과를 나타내었다. 또한 대황 추출물의 MMP-1 활성 저해효과는 20 ${\mu}g$/ml에서 약 70%의 우수한 활성 저해효과를 나타내었다. 이상의 결과를 종합하여 볼 때 대황 추출물은 항산화 효과 및 MMP-1 활성 저해효과와 자외선에 의한 MMP-1의 발현을 효과적으로 저해하는 것으로 보아 우수한 항노화 소재로써 이용될 수 있을 것으로 사료된다.

계혈등 추출물의 항산화와 사람 피부 섬유아세포에서의 Matrix Metalloproteinase-1 발현저해 효과 (Effects of the Spatholobi calulis extract on Antioxidation and Inhibition of Matrix Metalloproteinase in Human Skin Fibroblasts)

  • 본 연구에서는 계혈등 추출물에 의한 항산화 효과, in vitro MMP-1 효소 활성 저해 효과 및 사람섬유아세포에서 UVA에 의해 발현이 증가되는 MMP-1에 미치는 영향을 관찰하였다. 계혈등 추출물의 DPPH와 superoxide radical 소거효과는 처리농도가 증가함에 따라 농도 의존적으로 소거효과를 나타냈으며, $IC_{50}$은 각각 $45.81{\mu}g/ml$, $3.11{\mu}g/ml$로 DPPH와 superoxide radical을 소거하여 우수한 항산화 효과를 나타내었다. MMP-1 효소 활성 저해 효과도 $80{\mu}g/ml$에서 $97.33\%$를 저해하는 것으로 나타나 우수한 효과를 나타내었다. 사람섬유아세포에서 UVA에 의해 발현이 증가되는 MMP-1의 발현저해효과는 계혈등 추출물 $10{\mu}g/ml$에서 $74.66\%$로 단백질 수준에서 우수한 발현저해효과를 나타내었으며, mRNA수준에서도 계혈등 추출물은 모두 농도 의존적으로 발현 저해효과가 나타났다. 이상의 결과를 종합하여 볼 때 계혈등 추출물은 항산화 효과, MMP-1 효소 활성저해 효과와 UVA에 의한 MMP-1의 발현을 효과적으로 저해하는 것으로 보아 우수한 항노화 소재로써 이용될 수 있을 것으로 사료된다.

피부 노화 치료로서 저분자콜라겐펩타이드의 피부 항당화와 콜라겐 합성 효과 (The Effect of Low Molecule Collagen Peptide on Skin Anti-glycation and Collagen Synthesis as a Skin Aging Therapy)

  • 콜라겐 가수 분해물(collagen hydrolysate, CH)은 피부 진피 섬유아세포를 자극하여 콜라겐과 엘라스틴 같은 세포 외 기질 합성을 촉진함으로써 피부 노화 방지에 도움을 주는 것으로 알려져 있다. 최근 피부 노화를 일으키는 다양한 요인 중 최종당화산물(advanced glycation end products, AGEs)이 주목을 받고 있음에도 불구하고, 아직까지 CH가 AGE축적에 미치는 영향은 연구된 바 없다. CH는 피부 구조단백질의 생성을 촉진하여 AGE 축적에 영향을 줄 수 있으므로, 이를 확인하기 위해 트리펩타이드25%, Gly-Pro-Hyp 4%를 함유한 CH인 저분자콜라겐펩타이드(low molecule weight collagen peptide, LMCP)를 이용하여 임상시험을 수행하였다. 피부 조직 내 AGE 축적량을 평가하기 위해 AGE reader를 사용하여 피부자가형광(skin autofluorescence, SAF) 값을 측정하였다. 0.5%와 1.0% LMCP 용액을 8 주 동안 피험자의 전박에 도포한 결과, 시험부위의 SAF값이 대조부위에 비해 유의하게 감소하였다. 추가적으로, LMCP에 의한 피부섬유아세포의 콜라겐 합성 촉진을 평가하기 위해 CCD-986sk를 이용하여 in vitro test를 수행하였다. 그 결과, 800 ㎍/mL의 LMCP는 CCD-986sk의 human pro-collagen Iα1(COL1A1) 합성을 증가시켰다. 본 연구를 통해 LMCP도포가 콜라겐 합성을 촉진하여 항당화 효과에 도움을 줄 수 있다는 것을 확인하였으며, 이는 LMCP가 노화 방지 화장품 원료로써 잠재력이 있음을 시사한다.

섬유아세포에서 프로모터 다형성에 의한 Matrix Metalloproteinase-1의 발현에 관한 연구 (Study on the Expression of Matrix Metalloproteinase-1 by Promoter Polymorphism in Human Dermal Fibroblast)

  • 본 연구는 피부 섬유아세포에 자외선을 조사하거나 TNF-α를 처리하면 세포에 따라 MMP-1의 발현이 다르게 나타나는데, 이것이 MMP-1 프로모터의 다형성에 의해서 나타남을 밝히기 위해 수행되었다. 시판하는 23 종의 primary 섬유아세포에 대하여 MMP-1 프로모터의 -1607 부위의 유전형을 분석한 결과 6 개의 1G/1G 유전형, 10 개의 1G/2G 유전형, 7 개의 2G/2G 유전형을 가진 섬유아세포를 확인할 수 있었다. Hs68과 Detroit 551 세포주는 1G/2G 유전형을 가지는 것으로 확인되었다. 1G/1G 유전형은 TNF-α 처리에 의해 대조군에 비해 MMP-1이 2 배 높게 발현되었으며, 자외선에 의해서는 거의 발현되지 않았다. 1G/2G 유전형의 경우는 TNF-α 처리에 의해 MMP-1이 2.45 배 높게 발현되었으며, 자외선에 의해서는 1.4 배 MMP-1이 발현되었다. 2G/2G 유전형의 경우는 TNF-α 처리에 의해 MMP-1이 1.35 배 발현되었으며, 자외선에 의해서는 2.5 배로 높게 발현되었다. 즉 1G 유전형은 TNF-α에 의해, 2G 유전형은 자외선에 의해 발현이 유도되는 것으로 추정할 수 있으며, -1607 위치에 하나 더 삽입된 G에 의해서 Ets 전사인자가 결합할 수 있는 site가 만들어져서 MMP-1의 발현이 증가한다고 추정할 수 있으며, 피부 노화와 관련하여 섬유아세포에서는 이에 대한 연구가 전혀 진행되어 있지 않아서 향후 추가로 연구되어야 할 부분이다. 피부는 내인성 노화와 광노화의 영향을 동시에 받는 기관이므로, 피부 노화를 개선하기 위한 타겟으로 MMP-1의 발현을 분석할 경우에는 실험 조건에 적합한 유전형을 가지는 세포를 선택하여 연구를 진행하는 전략을 세워야 할 필요성이 대두된다.

자외선이 조사된 인간피부섬유아세포에 복분자 추출물이 미치는 영향 (Effects of Rubus coreanus Extracts on Ultraviolet-A Irradiated Cultured Human Skin Fibroblasts)

  • Sunlight, and in particular its UV component, is the major environmental trigger that underlies the major signs of human skin and skin cancer in general. Therefore, this study was carried out to investigate the UV protection effects of R. coreanus. R. coreanus was extracted by ultra high pressure extraction process at 500 MPa and $30^{\circ}C$ for 5 and 15 minutes. The cytotoxicity of the extracts extracted by ultra high pressure process on human dermal fibroblast cell CCD-986sk, human kidney normal cell HEK293, and human lung normal cell HEL299 was measured as 17.5%, 16.5% and 14.0%, respectively in adding $1.0\;mg/m{\ell}$ of the samples, which was much lower than that from conventional water extraction method at $100^{\circ}C$ as 23.2%, 22.5%, 21.2%. The secretion of $NO^-$ from macrophage showed $15.9\;{\mu}M$ on the R. coreanus extract from this process, which was higher than others. Prostaglandin $E_2$ ($PGE_2$) production from UV-induced human skin cells was also greatly decreased down to $510\;pg/m{\ell}$, compared to the control. From the results, we considered that the extracts from R. coreanus could be potent natural materials for skin anti-inflammation agent, and could be used as a potential anti-aging for the photo-damaged skin.

배양 혈청이 섬유아세포의 증식 및 교원질합성에 미치는 영향 (Effect of Serum Media on Fibroblast Proliferation and Collagen Synthesis)

  • Expanding cells ex-vivo is very important in tissue-engineering. Culture medium is usually supplemented with fetal bovine serum(FBS) in most of the experiments. However, cells grown in bovine serum media may posses the possibilities of disseminating bovine diseases and/or stimulating the patient's immune reactions. To overcome these problems, autologous or homologous serum should be used instead of the FBS. The purpose of this study is to compare cell proliferation and collagen synthesis depending on the kind of sera mixed on media and to provide a guideline on applying established experimental data to clinical cases. Human dermal fibroblasts were obtained from four patients. Five thousand cells per well in 96-well plates were incubated DMEM/F-12 Nutrient with varying serum mixture; 10% autologous serum, 10% homologous serum, and 10% FBS. Five days after incubation fibroblast proliferation and collagen production were determined by MTT assay and CICP enzyme immunoassay. The mean cell number were; $3.95{\times}10^4/well$, $2.97{\times}10^4/well$ and $2.30{\times}10^4/well$, respectively. The average amounts of collagen synthesized were; 238.13 ng/ml, 204.88 ng/ml, and 163.88 ng/ml in each. These results show that the use of human serum mixture may contribute to, not only preventing disseminated infection of bovine diseases. but also increase cell proliferation and collagen synthesis without simulating the patient's immune reactions.

당뇨족 궤양의 치료를 위한 신선 섬유아세포 동종이식 (Fresh Fibroblast Allograft as a Treatment for Diabetic Foot Ulcers)

  • Purpose: In order to overcome the limitations of the conventional cryopreserved fibroblast or keratinocyte allograft method used in the treatment of diabetic foot ulcers, we reported a pilot study in 2004 demonstrating promising results of a fresh fibroblast allograft method in eight patients. However, the number of cases was insufficient for full evaluation and the follow-up duration was not long enough to determine the efficacy and safety of the method. This encouraged us to conduct this follow-up study to fully evaluate the use of noncryopreserved fresh human fibroblast allografts in treating diabetic foot ulcers. Methods: Thirty-seven patients with diabetic foot ulcers were treated using fresh fibroblast allografts. Human dermal fibroblasts from healthy teenagers were cultured in DMEM/F-12 medium supplemented with 10% serum. The cultured cells were applied on the wounds immediately following debridement, with fibrin being used as a cell carrier. In eight weeks, percentages of complete healing, mean healing time, and patient satisfactions were assessed, with follow-up time ranging from 6 to 40 months. Results: Our study showed that 83.8% of the treated patients were complete healed. The time required for complete healing was $30.9{\pm}10.1$ days. Patient satisfaction scores for the experimental treatment were higher than those for the conventional method(mean scores of $8.1{\pm}1.1$ and $4.8{\pm}1.4$, respectively). No adverse events related to the study treatment occurred. Conclusion: The use of fresh human fibroblast allografts was found to be a safe and effective treatment for diabetic foot ulcers.

Polymeric nanoparticles as dual-imaging probes for cancer management

  • This article reports the development of biodegradable photoluminescent polymer (BPLP)-based nanoparticles (NPs) incorporating either magnetic nanoparticles (BPLP-MNPs) or gadopentate dimeglumine (BPLP-Gd NPs), for cancer diagnosis and treatment. The aim of the study is to compare these nanoparticles in terms of their surface properties, fluorescence intensities, MR imaging capabilities, and in vitro characteristics to choose the most promising dual-imaging nanoprobe. Results indicate that BPLP-MNPs and BPLP-Gd NPs had a size of $195{\pm}43nm$ and $161{\pm}55nm$, respectively and showed good stability in DI water and 10% serum for 5 days. BPLP-Gd NPs showed similar fluorescence as the original BPLP materials under UV light, whereas BPLP-MNPs showed comparatively less fluorescence. VSM and MRI confirmed that the NPs retained their magnetic properties following encapsulation within BPLP. Further, in vitro studies using HPV-7 immortalized prostate epithelial cells and human dermal fibroblasts (HDFs) showed > 70% cell viability up to $100{\mu}g/ml$ NP concentration. Dose-dependent uptake of both types of NPs by PC3 and LNCaP prostate cancer cells was also observed. Thus, our results indicate that BPLP-Gd NPs would be more appropriate for use as a dual-imaging probe as the contrast agent does not mask the fluorescence of the polymer. Future studies would involve in vivo imaging following administration of BPLP-Gd NPs for biomedical applications including cancer detection.

연육 제조과정에서 발생하는 어피 부산물의 활용을 위한 Cosmeceutical 활성 평가 (Evaluation of Cosmeceutical Properties of Fish Skin By-product Hydrolysates Collected During Surimi Manufacturing Process)

  • This study investigated the cosmetic effects of enzymatic hydrolytes of an aquatic by-product, fish skin. The skins of olive flounder Paralichthys olivaceus (PO) and Alaska pollock Gadus chalcogrammus (AP) were hydrolyzed using pepsin, Alcalase, and Protemax. Three enzymatic hydrolytes were obtained and the inhibitory effects of these hydrolytes on the aging-related enzymes tyrosinase, elastase, and collagenase were determined. The results indicated that the pepsin hydrolytes of PO and PA had stronger activities than the other hydrolytes. PO and PA also significantly reduced the intracellular reactive oxygen species levels in and improved the viability of H2O2-treated Vero cells; decreased nitric oxide production by and increased the cell viability of lipopolysaccharide-treated RAW 264.7 cells; and reduced intracellular reactive oxygen species levels and improved the viability of ultraviolet B irradiated HaCaT cells and human dermal fibroblasts. Furthermore, PO and PA remarkably reduced the intra- and extracellular melanin contents of alpha-melanocyte-stimulating hormone-stimulated B16F10 cells. These results demonstrate that PO and PA have potential for use in the cosmetic industry.

Effect of Oral Administration of Lactobacillus plantarum HY7714 on Epidermal Hydration in Ultraviolet B-Irradiated Hairless Mice

  • In this study, we evaluated the effect of Lactobacillus plantarum HY7714 on skin hydration in human dermal fibroblasts and in hairless mice. In Hs68 cells, L. plantarum HY7714 not only increased the serine palmitoyltransferase (SPT) mRNA level, but also decreased the ceramidase mRNA level. In order to confirm the hydrating effects of L. plantarum HY7714 in vivo, we orally administered vehicle or L. plantarum HY7714 at a dose of $1{\times}10^9CFU/day$ to hairless mice for 8 weeks. In hairless mice, L. plantarum HY7714 decreased UVB-induced epidermal thickness. In addition, we found that L. plantarum HY7714 administration suppressed the increase in transepidermal water loss and decrease in skin hydration, which reflects barrier function fluctuations following UV irradiation. In particular, L. plantarum HY7714 administration increased the ceramide level compared with that in the UVB group. In the experiment on SPT and ceramidase mRNA expressions, L. plantarum HY7714 administration improved the reduction in SPT mRNA levels and suppressed the increase in ceramidase mRNA levels caused by UVB in the hairless mice skins. Collectively, these results suggest that L. plantarum HY7714 can be a potential candidate for preserving skin hydration levels against UV irradiation.
