본 연구는 치과 병의원 진료실 주변환경과 치과종사자의 휴대전화에서 분리된 coagulase-negative staphylococci (CNS)의 분리율과 항균제 내성양상 및 분자 역학적 특성을 분석하고자 하였다. 2014년 12월부터 2015년 1월까지 부산지역 10개의 치과 병의원에서 총 154개의 샘플을 수집하여 MALDI-TOF분석법을 통해 동정하였다. 항균제 감수성검사는 디스크 확산법을 시행하였고 mupA, mecA 유전자 보유현황 및 SCCmec type은 PCR과 염기 서열분석에 의해 결정하였다. 154개의 샘플 중 10개(6.5%)에서 CNS 균주(Staphylococcus epidermidis 5주, Staphylococcus capitis 2주, Staphylococcus warneri 2주, Staphylococcus haemolyticus 1주) 가 분리되었다. 항균제 감수성검사에서 penicillin 10주, mupirocin 6주, gentamicin 5주, tetracycline 3주 및 cefoxitin과 erythromycin 2주가 내성이었고 clindamycin, ciprofloxacin, teicoplanin, trimethoprim-sulfamethoxazole에 내성인 세균은 없었다. 2개의 CNS균주(S. warneri, S. haemlyticus)에서 mecA 유전자가 검출되었고 1개의 CNS균주(S. epidermidis)에서 mupA가 확인되었다. Methicillin 내성 CNS균주 가운데 1주는 SCCmec I형이었고 1균주는 SCCmec의 유전형이 구분되지 않았다. 본 연구를 통하여 다약제 내성을 보이는 CNS균주가 더 이상 우리나라 치과병의원 환경에서 드물지 않음을 알 수 있었다.
새로 개발된 조기 진단 장비들로는 laser fluorescence device(LFD), 초음파 진단 시스템, confocal laser scanning microscopy(CLSM), quantitative light-induced fluorescence(QLF) 시스템 그리고 digital imaging fiber-optic trans-illumination(DIFOTI) 시스템 등이 있다. 본 연구는 임상에서 사용되고 있는 DIFOTI시스템과 LFD를 이용하여 유치 교환 시기에 있는 환아 21명, 25개의 유치를 대상으로 각 치아당 $1{\sim}3$점을 선정하여 구강내에서 초기 우식법랑질에 대한 DIFOTI 이미지 촬영 결과와 LFD 계측값을 3회 측정하고 그 평균을 대표값으로 한 결과를 CLSM과 비교하여 진단 능력을 평가하였다. 실험실 연구에서는 인공우식 용액을 이용하여 수거된 40개의 유치를 협설면에 $2{\times}3mm$ 크기의 창을 형성하고 4, 8, 12, 16일간 탈회시키면서 그 변화를 DIFOTI 시스템과 LFD를 이용하여 측정하고 이를 CLSM과 비교 분석하여 다음과 같은 결론을 얻었다. 1. 구강내에서 촬영된 DIFOTI의 민감도는 88.2%이었고, 특이도는 76.9%이었다. 2. 구강내에서 측정된 LDD의 민감도는 76.5%이었고, 특이도는 69.2%이었다. 3. 실험실 연구에서 유치 법랑질의 탈회 기간에 따른 DIFOTI 광투과율의 회귀 분석한 결과, 탈회 시간에 따라 광투과율은 감소하였다(r=-0.96, p<0.05). 4. 실험실에서 탈회 기간에 따른 LFD의 측정값의 회귀 분석 결과 통계적 유의성을 보이지 않았다(p>0.05). 5. DIFOTI 이미지의 광투과율과 CLMS의 병소 깊이에 대한 상관 계수는 -0.688이었으나(p<0.05), LFD의 측정값은 유의성을 보이지 않았다.
Objectives: The purpose of this study was to investigate the involvement of TRPA1 in the cinnamaldehyde-induced pulpal blood flow (PBF) change in the feline dental pulp. Materials and Methods: Mandibles of eight cats were immobilized and PBF was monitored with a laser Doppler flowmetry at the mandibular canine tooth. To evaluate the effect of cinnamaldehyde on PBF, cinnamaldehyde was injected into the pulp through the lingual artery at a constant rate for 60 seconds. As a control, a mixture of 70% ethanol and 30% dimethyl sulfoxide (DMSO, vehicle) was used. To evaluate the involvement of transient receptor potential ankyrin 1 (TRPA1) in PBF change, AP18, a specific TRPA1 antagonist, was applied into the pulp through the Class V dentinal cavity followed by cinnamaldehyde-administration 3 minutes later. The paired variables of experimental data were statistically analyzed using paired t-test. A p value of less than 0.05 was considered as statistically significant. Results: Administration of cinnamaldehyde (0.5 mg/kg, intra-arterial [i.a.]) induced significant increases in PBF (p < 0.05). While administration of a TRPA1 antagonist, AP18 (2.5 - 3.0 mM, into the dentinal cavity [i.c.]) caused insignificant change of PBF (p > 0.05), administration of cinnamaldehyde (0.5 mg/kg, i.a.) following the application of AP18 (2.5 - 3.0 mM, i.c.) resulted in an attenuation of PBF increase from the control level (p < 0.05). As a result, a TRPA1 antagonist, AP18 effectively inhibited the vasodilative effect of cinnamaldehyde (p < 0.05). Conclusions: The result of the present study provided a functional evidence that TRPA1 is involved in the mechanism of cinnamaldehyde-induced vasodilation in the feline dental pulp.
Korean Panax ginseng C. A. Meyer (P. ginseng) is a well-known and one of the most important tonic herbs used in traditional Korean medicine. The pharmacological effects of P. ginseng have been reported by many researchers. Nevertheless, little is known between the mechanism of action and the active compounds. In this study, we performed a comprehensive proteomic analysis and protein categorization in order to understand the physiological characteristics of the major components in the adventitious roots of P. ginseng. Whole proteins extracted from the cultured adventitious roots of P. ginseng were separated by two-dimensional polyacrylamide gel electrophoresis (2-DE). Among the 1000 spots which were detected by silver staining, 113 spots were labeled and identified by matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF MS). Our results showed that 40 proteins were identified among the 113 spots, with a hit ratio of 35.3%. A number of proteins identified on the 2-DE gels (30%; 16 spots) were involved in energy metabolism. These proteomic data will be helpful to better understand the physiological and pharmacological effects of P. ginseng.
Purpose: The aim of this study was to investigate the combined effects of physical and chemical surface factors on in vivo bone responses by comparing chemically modified hydrophilic sandblasted, large-grit, acid-etched (modSLA) and anodically oxidized hydrophobic implant surfaces. Methods: Five modSLA implants and five anodized implants were inserted into the tibiae of five New Zealand white rabbits (one implant for each tibia). The characteristics of each surface were determined using field emission scanning electron microscopy, energy dispersive spectroscopy, and confocal laser scanning microscopy before the installation. The experimental animals were sacrificed after 1 week of healing and histologic slides were prepared from the implant-tibial bone blocks removed from the animals. Histomorphometric analyses were performed on the light microscopic images, and bone-to-implant contact (BIC) and bone area (BA) ratios were measured. Nonparametric comparison tests were applied to find any significant differences (P<0.05) between the modSLA and anodized surfaces. Results: The roughness of the anodized surface was $1.22{\pm}0.17{\mu}m$ in Sa, which was within the optimal range of $1.0-2.0{\mu}m$ for a bone response. The modSLA surface was significantly rougher at $2.53{\pm}0.07{\mu}m$ in Sa. However, the modSLA implant had significantly higher BIC than the anodized implant (P=0.02). Furthermore, BA ratios did not significantly differ between the two implants, although the anodized implant had a higher mean value of BA (P>0.05). Conclusions: Within the limitations of this study, the hydrophilicity of the modSLA surface may have a stronger effect on in vivo bone healing than optimal surface roughness and surface chemistry of the anodized surface.
Purpose: It is to compare and evaluate the change of the wear rate and phase variation of the Zirconia before and after the sintering after the grinding by a high speed equipment manufactured for the Zirconia. Methods: The specimen of the sintered Zirconia was manufactured as size of $15mm{\times}15mm{\times}2mm$. The grinding has been applied to each of all pieces of each test groups for a minute fit for each condition at same speed of 50,000 rpm by a diamond bur at high speed handpiece with injection of the air and water. For the observation of the surface before and after the sintering of the each test piece, the cross section of it was observed as 100 magnification by a scanning electron microscope after it was coated by PT, and the diffraction analysis was performed by XDR to compare the crystal phase of the Zirconia. The average surface roughness value of all specimens were evaluated. The wear test was performed at room temperature by applying a load of 1kg for 120,000 cycles for the chewing period 6 months. Wear was analyzed for the enamel cusps by measurement of the vertical substance loss with a laser scanner. Conclusion: The phase variation from the tetragonal phase to the monoclinic phase was confirmed in the test group of the pre-sintered Zirconia after the grinding, and the value of the surface roughness and the wear rate was increased in experimental group.
Cyclosporin A (CsA) has been used clinically as an immunosuppressive drug to prevent organ transplant rejection and in basic research as a mitochondrial permeability blocker. It has been reported that CsA has a protective role in severed neurons and a neurotrophic effect in neuronal cells. However, the molecular mechanisms underlying the stimulation of neuronal cell proliferation by CsA have not yet been elucidated. In our current study, we investigated CsA responsive proteins in PC12 cells using a systematic proteomic approach. The viability of these cells following CsA treatment increased in a dose- and time-dependent manner. Proteins in the CsA-treated PC12 cells were profiled by two-dimensional gel electrophoresis (2-DE) and identified by matrix-assisted laser desorption ionization time-of flight (MALDI-TOF) and electrospray ionization quadupole time-of-flight mass spectrometries (EIQ-TOFMS). This differential expression analysis showed significant changes for 10 proteins (6 up-regulated and 4 down-regulated) upon CsA treatment that were related to cell proliferation, metabolism and the stress response. These proteomics data further our understanding of the proliferation mechanisms of PC12 cells exposed to CsA and demonstrate that our methodology has potential to further elucidate the mechanisms and pathways involved.
Background: In this study, we investigated the potential use of keratin for oral tissue regeneration. Keratin is well-known for its effectiveness in skin regeneration by promoting keratinization and enhancing the elasticity and activity of fibroblasts. Because of its structural stability, high storability, biocompatibility, and safety in humans, existing research has predominantly focused on its role in skin wound healing. Herein, we propose using keratin proteins as biocompatible materials for dental applications. Methods: To assess the suitability of alpha-keratin protein as a substrate for cell culture, keratin was extracted from human hair via PEGylation. Viabilities of primary human gingival fibroblasts (HGFs) and human oral keratinocytes (HOKs) were assessed. Fluorescence immunostaining and migration assays were conducted using a fluorescence microscope and confocal laser scanning microscope. Wound healing and migration assays were performed using automated software to analyze the experimental readout and gap closure rate. Results: We confirmed the extraction of alpha-keratin and formation of the PEG-g-keratin complex. Treatment of HGFs with keratin protein at a concentration of 5 mg/ml promoted proliferation and maintained cell viability in the test group compared to the control group. HOKs treated with 5 mg/ml keratin exhibited a slight decrease in cell proliferation and activity after 48 hours compared to the untreated group, followed by an increase after 72 hours. Wound healing and migration assays revealed rapid closure of the area covered by HOKs over time following keratin treatment. Additionally, HOKs exhibited changes in cell morphology and increased the expression of the mesenchymal marker vimentin. Conclusion: Our study demonstrated the potential of hair keratin for soft tissue regeneration, with potential future applications in clinical settings for wound healing.
In case of skeletal Class III malocclusion with underdeveloped maxilla, the extraoral orthopedic force for the stimulation of maxillary growth or anterior reposition of the maxilla has been used clinically for the improvement of facial skeletal relationship. The purpose of this investigation was to examine the initial reaction of maxillofacial complex to the maxillary protraction by using extraoral orthopedic force. The dried human skull was used and this investigation was done by means of double exposure holographic interferometry. The protraction forces placed on the canine or the first molar were parallel, $10^{\circ}$ downward, $20^{\circ}$ downward to the occlusal plane. Fringe pattern of each protraction condition was compared and analized. The results were as follows: 1. Each maxillofacial bone displaced saparately. 2. More displacement was shown at the area of the teeth and the alveolar bone. 3. A counterclockwise rotation of the maxilla wa decreased by downward protraction and especially 20 degree downward protraction from the canine showed least rotation. 4. On the zygomatic arch, outward bend was observed and this effect was decreased by downward protraction. 5. On the zygomatic bone, the counter clockwise rotation was increased by the downward protraction. 6. When maxillary expansion was applied at the same time, outward and upward displacement with counterclockwise rotation was observed on the maxilla. 7. The lateral pterygoid plate of sphenoid bone was affected by maxillary protraction.
본 연구에서는 세라믹 브라켓 제거에 Er:YAG 레이저 조사가 도움이 되는지 알아보고, 브라켓 제거에 적합한 레이저 조사 방법을 연구하였으며, 또 이렇게 적용된 레이저가 치수와 법랑질에 손상을 주는지도 알아보았다. 총 190개의 치아, 단결정 세라믹 브라켓(MISO), 다결정 세라믹 브라켓(Transcend series 6000)과 KEY Laser3를 사용하였다. 실험군은 세라믹 브라켓의 종류(단결정, 다결정)와 레이저의 에너지(140, 300, 450, 600 mJ)에 따라 분류하였으며, 레이저를 브라켓당 두 곳에 1펄스씩 조사하고, 전단 강도를 측정하였다. 대조군은 레이저를 조사하지 않는 군으로 하였다. 레이저 조사에 의한 열 효과는 브라켓 하방 법랑질과 치수강에서 측정하였으며, 전단 강도 측정 후 치면에 남아있는 접착제의 양을 접착제 잔류 지수(adhesive remnant index)를 이용하여 평가하였다. 레이저 조사로 인한 접착제의 파괴 양상과 법랑질 표면 변화를 주사전자현미경으로 관찰하였다. 모든 세라믹 브라켓군에서 레이저 에너지가 증가할수록 전단 강도는 유의하게 감소하였다. 또한, 브라켓 하방 법랑질에서 최대 온도 변화는 평균 $3.78^{\circ}C$ 상승에 그쳤으며, 치수강에서 최대 온도 변화는 평균 $0.9^{\circ}C$ 상승에 그쳤다. 주사전자현미경을 이용한 법랑질과 접착제 단면 관찰에서 접착제 표면이 레이저에 의해 붕괴되어 분화구 모양의 구덩이로 관찰되었으며, 일부 시편에서 약 $10\;-\;30{\mu}m$의 법랑질 손상이 발견되었다. Transbond XT로 부착된 단결정 도재 브라켓(MISO)의 디본딩에 Er:YAG 레이저를 이용할 경우, 300 - 450 mJ의 레이저 에너지를, 그리고 다결정 도재 브라켓(Transcend series 6000)의 경우는 450 mJ 정도의 에너지를 사용하는 것이 효과적이고 안전할 것으로 생각된다.
