Antibacterial Activity of Vinegars on Streptococcus mutans Caused Dental Caries (충치 유발균 Streptococcus mutans에 대한 식초의 항균효과)

  • The antibacterial activity for S. mutans by using 5 kinds of vinegar was researched. As a result it was the highest at persimmon vinegar and then grape, apple, high calcium brown rice, brown rice vinegar in order. The antibacterial effect was shown at 1.5% of persimmon vinegar concentration and the growth was fully repressed at 4% of concentration. As an observation result with a transmission electron microscope(TEM), the growth was obstructed by destroying the cell wall and the cytoplasmic membrane. As a research result of the total phenol contents for each vinegar, the persimmon vinegar was the highest as 237.49 mg% and the antibacterial activity was increased when the total phenol contents were high.

Antimicrobial Effect of Coptidis rhizome Extract against Mutans Streptococci and Periodontopathogens

  • The purpose of the study was to investigate the antimicrobial activity of the methanol extract of Coptidis rhizome against the type strains of cariogenic bacteria, Streptococcus mutans and Streptococcus sobrinus, and the periodontopathogens, Porphyromonas gingivalis, Prevotella intermedia, Treponema denticola and Aggregatibacter actinomycetemcomitans. The antimicrobial activities of the crude extract and the methanol extract fractions of Coptidis rhizome separated by silica gel chromatography were evaluated by determining the minimal bactericidal concentration (MBC) values, using the microdilution method. The cell viability test of the extracts of Coptidis rhizome on the KB cells was also studied by methyl thiazolyl tetrazolium (MTT) assay. Our results showed that the 11th fraction (F11) of the methanol extract had the greatest antimicrobial activity against the tested bacteria, with no associated cytotoxicity on the KB cells, upto a concentration of $50{\mu}g/ml$. These results suggest that the silica gel chromatography fraction F11 of the methanol extract of Coptidis rhizome, could be useful in the development of oral hygiene products as an antimicrobial agent for the prevention of dental caries and periodontal diseases.

Antibacterial Activity of Hydrogen-rich Water Against Oral Bacteria

  • There are estimated to be about 700 species of bacteria in the oral cavity. Based on epidemiological investigations, some of these strains have been proposed as the pathogens responsible for oral diseases such as dental caries, gingivitis and periodontitis. Since electrolyzed hydrogen-rich water has been shown to have beneficial effects on human immunity, its use has increased. In our study, the antibacterial activity of hydrogen-rich water for oralagainst bacteria associated with oral disease was evaluated. The bacterial strains Streptococcus mutans, Fusobacterium nucleatum, Porphyromonas gingivalis and Tannerella forsythia were cultured in specific growth medium. S. mutans, F. nucleatum and P. gingivalis were soaked to thein both hydrogen water and tap water for 30 sec and then inoculated onto mitis-salivarius agar and brain heart infusion agar including supplemented withvitamin K and hemin, respectively. The numbers of bacterial colonies were then measured after cultivation for 48 hours. In the case of T. forsythia, which does not grow well on agar plates, inoculations into modified new oral spirochete (NOS) broth were performed and growth curve analysis was undertaken every day with a spectrophotometer. Hydrogen water showed antibacterial activity against all four bacterial strains in comparison with tap-water. We conclude from this that hydrogen water may have a positive impact on oral hygiene by helping to remove cariogenic bacteria and periodontopathogens.

FAILURE OF ODONTOGENESIS AFTER CHEMO-RADIATION THERAPY FOR RHABDOMYOSARCOMA (횡문근육종의 항암제-방사선치료 후 치아발육장애)

  • This report details a case of 8-year-old girl showing failure of odontogenesis after chemo-radiation therapy for the rhabdomyosarcoma at the age of 4. The observed results were as follows : 1. Past history revealed that she had received for a total radiation dose of 4430cGy, 29 fractions in 6 weeks and chemotherapy with vincristine, actinomycin D and cytoxan, followed as maintenance phase for 2 years. 2. The patient was symptom -free and appointed for the treatment of multiple dental caries. 3. Oral examination showed hypoplastic enamel on whole erupted permanent teeth and showed retarded eruption. 4. Conventional radiograms showed failure of root development including abrupt cessation of root formation and root agenesis, and microdontia, missing teeth, irregular enamel, dislocation of the impacted teeth. Additional finding showed good healing bone pattern on the left mandibular ramus and angle area. 5. Cephalometric analysis revealed failure of bite raising due to incomplete eruption of all the first molars and made it possible to suspect entrapped mandibular growth and then Class II tendency growth. 6. There was correlation between the time of chemo-radiation therapy and the damage of the teeth.

Dental Caries Extraction using YCbCr Color Model and ART2 Algorithm (YCbCr 색상모델과 ART2 알고리즘을 이용한 충치 추출)

  • 본 논문에서는 충치 환자의 진단을 위해 구강 영상에서 충치를 추출하는 방법을 제안한다. 먼저 구강은 붉은색을 띄고 치아는 흰색을 띈다는 특징이 있기 때문에, 구강 영상을 YCbCr 컬러모델로 변환한다. YCbCr 컬러모델에 임계치를 설정하여 붉은 영역을 검출해내고, 검출된 붉은 영역에 대해 이진화하여 치아 영역을 추출한다. 그 후, 모폴로지 기법을 이용하여 잡음 제거 및 치아의 빈 공간을 채운다. 치아 영역 추출 시 영상에 따라 치아 사이를 잇는 모서리 부분이 손실된 경우가 발생할 수 있기 때문에 치아 사이의 손실된 부분을 연결 한다. 치아 영역에 ART2 알고리즘을 적용하여 클러스터링하고 충치 후보 영역을 추출한다. 충치 후보 영역에 8방향 윤곽선 추적 기법을 적용하여 충치를 분석 및 추출한다. 실험 결과 81%의 추출 성공률을 보였고 다양한 형태의 충치를 효과적으로 추출할 수 있는 것을 확인하였다.

  • Autotransplantation is the surgical repositioning of an autogenous erupted or unerupted tooth from one site to another in the same individual. This treatment is indicated in traumatic tooth loss, teeth with severe caries, congenitally missing teeth, teeth with bad prognosis and in case of developmental anomalies of teeth. The following 2 cases describe the potential to utilize autotransplantation to replace hopeless teeth with sound wisdom teeth.

Chitosan-induced biomodification on demineralized dentin to improve the adhesive interface

  • Objectives: Metalloproteinase-inhibiting agents, such as chitosan, can prevent collagen degradation in demineralized dental substrates, thereby improving the adhesive interface. This study evaluated the bond strength (BS) and chemical and morphological characterization of the adhesive interface after applying chitosan solution to demineralized dentin. Materials and Methods: The 80 third molars were selected. Forty teeth underwent caries induction using the pH cycling method. The teeth were divided according to the treatment: distilled water (control) and 2.5% chitosan solution. The surfaces were restored using adhesive and composite resins. Half of the specimens in each group were aged, and the other half underwent immediate analyses. The teeth were sectioned and underwent the microtensile bond strength test (µTBS), and chemical and morphological analyses using energy-dispersive spectroscopy and scanning electron microscopy, respectively. Data analysis was performed using 3-way analysis of variance. Results: For µTBS, sound dentin was superior to demineralized dentin (p < 0.001), chitosan-treated specimens had higher bond strength than the untreated ones (p < 0.001), and those that underwent immediate analysis had higher values than the aged specimens (p = 0.019). No significant differences were observed in the chemical or morphological compositions. Conclusions: Chitosan treatment improved bond strength both immediately and after aging, even in demineralized dentin.

Enhancing the Antibacterial Effect of Erythrosine-Mediated Photodynamic Therapy with Ethylenediamine Tetraacetic Acid

  • This study evaluated the additive impact of ethylenediamine tetraacetic acid (EDTA) on erythrosine-mediated photodynamic therapy (PDT) against Streptococcus mutans (S. mutans) biofilm by measuring colony-forming units and applying confocal laser scanning microscopy. Fifty-six bovine incisors, free from dental caries or structural defects, were utilized in this study. Dentin specimens were created by cutting with a low-speed diamond disk under a continuous flow of water, resulting in dimensions of 6.0 mm × 3.0 mm × 2.0 mm. The specimens were categorized into 4 groups: Control, EDTA, PDT, and EDTA + PDT. S. mutans ATCC 25175 was employed to establish biofilm on the dentin specimens. A 17% EDTA solution was applied for 1 min. For PDT, erythrosine served as the photosensitizer. Finally, a light-emitting diode source (385 - 515 nm) was employed in this study. The PDT group exhibited a significantly lower bacterial count than both the control and EDTA groups (p < 0.001). The EDTA + PDT group demonstrated a significantly reduced bacterial count compared to the other 3 groups (p < 0.001). This study demonstrated that EDTA enhances the antimicrobial efficacy of PDT on S. mutans biofilm. Even at a low concentration of photosensitizer, the combination of EDTA and PDT yields a significant antibacterial effect.

Affecting Factors Related to Oral Health Conditions of Workers (근로자의 상실치아 실태와 관련요인)

  • This study was conducted to investigate the health behavior and the oral health status of workers engaged in a manufacturing business, and to analyze the related factors that affect their number if teeth loss. References for the study involved the materials of 526 individuals whose occupational categories are of a manufacturing business, amongst the survey participants in seven city or gun (county) area including Gyeongsan-si, Gyeongju-si, Mungyeong-si, Chilgok-gun, Yecheon-gun and Sangju-gun for the Community Health Survey (CHS) conducted in 2008. The CHS 2008 was conducted for three months from September to November 2008, and the survey details utilized in the study included the number of loss of teeth and teeth-brushing frequency, which was a part of CHS questionnaires and also additionally included for the study. The analysis with respect to the number of teeth loss on sociodemographic factors, was found to be significantly related to the age, educational status, average monthly income and occupation (p<.01). In the $x^2$-test in terms of the oral health status and the number of teeth loss, it was found that the teeth-brushing after lunch (p<.05) and an experience in an oral examination were statistically significant factors (p<.01). According to the multiple-regression analysis with the number of teeth loss being a dependent variable, the age, educational status, marital status and subjective oral health status were the statistically significant (p<.01). In conclusion, it is important to provide education on the teeth-brushing subjected to workers engaged in a manufacturing business with many dental caries and low educational status, and to recommend a regular scaling as well as to establish policy for creating conditions upon an oral health check-up and a tooth scaling and thus allow the maintenance of an oral health.

Antimicrobial Activities and Adherence Inhibition on Streptococcus mutans by Ethyl Acetate Extract from Caesalpinia sappan L. (소목(Caesalpinia sappan L.)의 에틸아세테이트 분획물이 Streptococcus mutans에 대한 항균활성 및 부착 억제)

  • Somok, the heart wood of Caesalpinia sappan is used in traditional Chinese medicine. Adherence of S. mutans to the tooth surface can result in the formation of a dental plaque. This study was performed to investigate the antibacterial activity and bacterial adhesion of ethyl acetate extract from C. sappan against S. mutans ATCC 25175. The bacteria were cultured in brain heart infusion(BHI) broth, and then incubated under 5% $CO_2$ at $37^{\circ}C$ for 18~24 hours. The antimicrobial activity of the ethyl acetate extract of C. sappan was then examined using the paper disc methods and MIC. In addition, bacterial adherence to hydroxyapatite was also examined. The ethyl acetate extract was shown to produce inhibitory effects and had MIC values of 125 mg/ml against S. mutans ATCC 25175. The ethyl acetate extract inhibited adhesion of S. mutans to saliva coated-hydroxyapatite beads(S-HA). At 24 hr, the ethyl acetate extract significantly reduced the adherence of S. mutans to S-HA beads relative to the control. The isolated active substance was identified as brazilin($C_{16}H_{14}O_5$) by $^1H-NMR$ and $^{13}C-NMR$. Thus, the application of C. sappan can be considered a useful and practical method for the prevention of dental caries.
