Effcts of Dangkiyeumja(當歸飮子) Water Extract of anti-allergic responses and on the Functions of Murine Immunocytes (當歸飮子 水抽出液이 抗ALLERGY 反應과 MOUSE의 免疫細胞機能에 미치는 影響)

  • This study were done to know the effects of Dangkiyeumja on the in vivo and in vitro immune responses of mice. The recipes of Dangkiyeumja used in this study enhanced such, cellular functions of immunocytes as phagocytic capacity of macrophages, rossett-eforming abilities of splenocytes and metabolic activities of lymphocytes, However, the same recipes decreased the formation of such reactive oxygen intermediates(ROI) as superoxide and hydrogenperoxide from the macrophages. The effects of the same recipes on the in vim immune responses was suppressive on the cellular immune response(CIR)measured by delayed-type hypersensitivity against dinitrofluorobenzene and mildly enhancing for the humoral immune response measured by antibody production against sheep red blood cells. The results of this study could be summarized as follow: 1. Administration of Dangkiyeumja enhanced the phagocytic activity of the murine macrophage. 2. Administration of Dangkiyeumja decreased the formation of ROI in the murine macrophage 3. Administration of Dangkiyeumja increased the number of the splenic rotte forming cells in the mouse. 4. Administration of DangKiyeumja did not effect the antibody production against sheep red blood cells. 5. Administration of Dangkiyeumja depressed the delayed-type hypersenitivity against dinitrofluoro benzene in the mouse. The result of this study suggest that Dangkiyeumja could ameliorate the hypersensitivity reactions by reducing the formation of ROI and decreasing the CIR without affecting the other functions of immunocytes.

Immunostimulating Activity by Protoplast Fusants between Ganoderma Iucidum and Lentinus edodes (영지와 표고의 융합체의 면역활성 증강작용)

  • On the inter-order protoplast fusants of Lentinus edodes and Ganoderma lucidum was the antitumor activity test performed and the fusant P22 was selected. The hot water extract of the cultured mycelia of P22 were purified by DEAE-cellulose chromatographya and the resulting purified fraction was designated as P22A. It was found to be a proteoglycan whose molecular weight was 47 kDa. When examined for immunopotentiation activity, P22A increased the number of colony forming unit in the bone marrow stem cells to 3-folds. It also potentiated the secretion of nitric oxide in activated macrophages to 2-folds. In humoral immune response, it increased the activities of the alkaline phosphatase in differentiated B cells to 1.6-folds and the number of plaque forming cells to 1.8-folds. In cellular immune response, it restored the depressed response of delayed type hypersensitivity in tumor bearing mice to normal level. These results suggest that P22A have potential to restore the decreased immune activity of the tumor bearing mice to normal level.

Immunosuppressive Activity of Wogonin from Scutellaria Radix on DNFB-induced Delayed Type Hypersensitivity in Mice (마우스에서 황금 유래 Wogonin의 Th1과 Th2 사이토가인 조절 효과)

  • In the present study, wogonin, a major flavone isolated from Scutellaria Radix were examined for its imunosuppressive activity on the 2,4-dinitro-fluorobenzene (DNFB)-induced delayed type hypersensitivity (DTH) in C3H/HeN mice. This compound inhibited selectively $TNF-\alpha$ and IL-4, but not IL-6, IL-10 and $IFN-\gamma$ on DNFB-induced Th1 and Th2 cytokines in a concentration-dependent manner. Interestingly, this compound increased the expression of heme oxygenase-1 (HO-1) in a concentration-dependent manner The above results reveal that wogonin possesses anti-inflammatory, humoral and cellular immunomodulatory, and stress reducing activities on the DNFB-induced DTH in mice and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

Effects of Dangkwiyughwangtang and Okbyoungpoongsangamibang on the immune response induced by Methotrexate in mice (Methotraxate로 유발된 생쥐의 면역반응(免疫反應)에 대한 당귀육황탕(當歸六黃場)과 옥병풍산가미방(玉屛風散加味方)의 효과(效果))

  • Objectives : In order to study the effect of Dangkwiyughwangtang and Okbyoungpoongsangamibang on the immune response induced by methotrexate in mice. Method : Delayed type of hypersensitivity, hemagglutinin titer, hemolysin titer, rosette forming cells, phagocytic activity for immune response, lymphocyte transformation, and productivity of Interleukin-2 were measured. Results : Body weight decreasing was significantly inhibited as compared with control group in both the Dangkwiyughwangtang and Okbyoungpoongsangamibang groups. Delayed type of hypersensitivity was significantly increased as compared with control group in both groups Hemagglutinin titer was significantly increased as compared with control group in both groups. Hemolysin titer was significantly increased as compared with control group in the Okbyoungpoongsangamibang group. Rosette forming cells were significantly increased as compared with control group in both groups. Phagocytic activity for immune response was slightly decreased in the Dangkwiyughwangtang group and slightly increased in the Okbyoungpoongsangamibang group insignificantly as compared with the control group. Lymphocyte transformation was significantly increased as compared with the control group in both the Dangkwiyughwangtang and Okbyoungpoongsangamibang groups. Productivity of Interleukin-2 was significantly increased as compared with the control group in both the Dangkwiyughwangtang and Okbyoungpoongsangamibang groups. Conclusion : Both the Dangkwiyughwangtang and Okbyoungpoongsangamibang groups enhance immunity in mice.

Study of the immunosuppressive activity of methanolic extract of Madhuca longifolia (Koenig)

  • The immunosuppressive activity of the Methanol extract of bark of Madhuca longifolia (Koenig) consisting of a mixture of saponins, flavonoids, tannins, steroids, phenol and glycosides was studied on the immune responses in mice. Methanol extract of Madhuca longifolia (MLL) was administered orally at doses of 50, 100 and 150 mg/kg/day to healthy mice divided into four groups consisting of six animals each. The assessment of immunomodulatory activity was carried out by testing the humoral (antibody titre) and cellular (foot pad swelling) immune responses to the antigenic challenge by sheep RBCs. Furthermore, the effect on hematological parameters as well as relative organ weight was determined. On oral administration MML showed a significant decrease delayed type hypersensitivity (DTH) response whereas the humoral response to sheep RBCs was unaffected. Thus MLL significantly suppressed the cellular immunity by decreasing the footpad thickness response to sheep RBCs in sensitized mice. With a dose of 100 and 150 mg/kg/day the DTH response was $7.66{\pm}2.75$ and $6.41{\pm}1.21$ respectively in comparison to corresponding value of $14.50{\pm}2.38$ for untreated control group. These differences in DTH response were statistically significant (P < 0.05). The study demonstrates that MLL shows preferential suppression of the components of cell-mediated immunity and shows no effect on the humoral immunity.

Effects of Swainsonine on the Cell-mediated Immune Responses of Lipopolysaccharide (리포포리사카라이드의 세포성 면역반응에 미치는 스와인소닌의 영향)

  • Effects of swainsonine (SW: 8${\alpha}$, ${\beta}$-indolizidine-1alpha, 2${\alpha}$, 8${\beta}$-triol from Locoweed) on the cellular and nonspecific immune responses of lipopolysaccharide (LPS) wer e studied in ICR mice. Mice were divided into 4 groups (10mice/group), and LPS was given to each mouse 1 hr after i.p. injection with 3.7mg/kg of SW by i.p. injection twice a week for 14 days at a dose of 2mg/kg. Immune responses of the delayed-type hypersensitivity response (DTH) to sheep red blood cells (s-RBC), phagocytic activity and natural killer (NK) cell activity were evaluated. LPS treatment didn`t affect NK cell activity, phagocytic activity, DTH to s-RBC compared with those in controls, and phagocytic activity of sareoma 180 tumor bearing mice. However, circulating leukocytes were significantly decreased. Combinaton of LPS and SW increased circulating leukocytes significantly compared vath that in LPS alone, and DTH to s-RBC, NK cell activity and phagocytic activities of normal and sarcoma tumor bearing mice were not affected. These findings indicate that SW didn`t affected the cellular immune responses suppressed by LPS but significantly increased circulating leukocytes.

  • The immunosuppressive effect of serum from mice heavily infected with BCG(BCG-serum) was evaluated. The delayed-type hypersensitivity reaction to sheep erythrocytes was depressed when BCG-serum was administered both systemically and locally at the same time of challenge. This study shows that serum of animal infected with mycobacteria contains a factor responsible for immunosuppression and factor may be important in understanding the mechanisms of allergy in mycobacteria] infections.

  • Experimental studies were done to research the clinical effects of Chihyosan and Chihyosangamibang on the Anti-allergic effect and pulmonary function of $O_3$ intoxicated Rats. Anti-allergic effect experiment consisted of vascular permeability responses to intradermal histamine and serotonin, 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, and delayed type hypersensitivity responses to Picryl Chloride and SRBC. Pulmonary function of $O_3$ intoxicated Rats experiment consisted of lung TBA value, water Contents of the lung, oxygen consumption time, and arterial blood $pCO_2,\;pO_2,\;HCO_3^-$, pH level. The results obtained as follows; 1. In the effects of Chihyosan and Chihyosangamibang on vascular permeability responses to intradermal histamine, both of chihyosan and Chihyosangamibang group revealed significant effect. 2. In the effects of Chihyosan and Chihyosangamibang on vascular permeability responses to intradermal serotonin, both of chihyosan and Chihyosangamibang group revealed significant effect. 3. In the 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against egg white albumin, Chihyosan groups revealed significant effect, but Chihyosangamibang groups revealed none significant effect. 4. In the delayed type hypersensitivity responses to Picryl Chloride, Chihyosan and Chihyosangamibang groups revealed none significant effect. 5. In the delayed type hypersensitivity responses to. SRBC, Chihyosan revealed none significant effect, but Chihyosankamibang revealed significant effect. 6. Both of Chihyosan and Chihyosangamibang groups revealed significant effect on decrease of the lung TBA value of lung. 7. Both of Chihyosan and Chihyosangamibang groups revealed significant effect on decrease of the water contents of right and left lung. 8. Both of Chihyosan and Chihyosangamibang groups revealed significant effect on decrease of oxygen consumption time. 9. In the decrease effect of arterial blood $pCO_2$ level, both of Chihyosan and Chihyosangamibang groups revealed none significant effect. 10. In the increase effect of arterial blood $pO_2$ level, both of Chihyosan and Chihyosangamibang groups revealed none significant effect. 1. In the decrease effect of arterial blood $HCO_3^-$ level, both of Chihyosan and Chihyosangamibang groups revealed significant effect. 12. In the increase of arterial blood pH level, Chihyosangamibang groups revealed none significant effect, but Chihyosan groups revealed significant effect. According to above stated results, both of Chihyosan and Chihyosangamibang are very usefully for treatment of cough, asthma, chronic obstructive pulmonary diseases and allergic pulmonary diseases.

The Effects of the Tumor Mass Size Inoculated in Immunologically Competent Balb/c Mice on Delayed-type Hypersensitivity Response (정상 면역 생쥐에 접종된 암세포주의 종괴 형성이 숙주의 지연성과민반응에 미치는 영향)

  • Background: Based on outstanding progresses in animal experiments, vaccines for some human tumors have been developed. However, clinical effects of these vaccines have been far below than expected. This discrepancy might come from differences between animal models and human patients with respect to immunocompetency. The immune status of mice after tumor inoculation has not been well studied, which make us cautious in interpreting and applying the results from mice to human. We evaluated cell-mediated immune responses in mice after tumor cell inoculation. Methods: Mice were inoculated with TA3Ha, CT26, or 4T1. Delayed-type hypersensitivity (DTH) responses were induced 2-4 weeks after inoculation using 2,4-dinitro-1-fluorobenzene as an antigen. The relationships between the severity of DTH responses and the duration of tumor inoculation or the size of tumor mass were analyzed. Results: In T A3Ha groups, DTH response was elevated 2 weeks after inoculation, but depressed after 4 weeks, compared to the control group. When analyzed based on the sizes of tumor masses elicited, DTH responses were inversely related to the mass size, especially in those greater than 10 mm in diameter. In CT26 groups, while the duration after inoculation did not affect the severity of DTH responses, those with large mass showed depressed responses regardless the duration of inoculation. 4T1 cells grew so slowly that the size of tumor mass was small even 4 weeks after inoculation, and this group showed much higher DTH responses compared to that of tumor-free group. Conclusion: At least in an experimental setting where tumor model was induced by inoculating tumor cell lines into immunologically competent mice, the host immune response was elevated in early stage, and then depressed in late stage when the mass grew over a critical size.

The Effect of Ethanol Administration on The Immune Response of Mice (에타놀투여가 마우스의 면역반응에 미치는 영향에 관한 실험적 연구)

  • The present study was undertaken in an effort to investigate the effects of alcohol on survival of mice and on their humoral and cellular immune responses, The immune responses examined were Arthus and delayed-type hyperrsnesitivity(DTH) reactions to sheep red blood cells(SRBC), contact hypersensitivity to dinitrofluorobenzend(DNFB), antibody response to thymus - dependent SRBC and to thymus -independent polyvinylpyroridone(PVP), and the recovery of Crytococcus neoformans from the liver, spleen, kidney and brain of experimentally infected mice. The administration of ethanol concentrations of 20% or less did not cause any change in survival rates as compared withs saline injected control group. In general, ethanol administration inhibited the Arthus and DTH reactions to SRBC, contact hypersensitivity to DNFB, and antibody response to both SRBC and PVP and it also decreased the resistance of mice to C. neoformans infection. Taken together, the present study stongly suggested that ethanol inhibits immune response and decrease the resistance of mice to C. neoformans infection.
