Research on the Delayed Family Formation Phenomenon among the Never-married Professionals in Korea: A Qualitative Study with Focus Groups (전문직 미혼 남녀의 만혼현상에 대한 연구)

  • This study uses qualitative data to examine the dynamics and barriers of the delayed family formation phenomenon among the never-married professionals in Korea. The data was drawn from focus groups of 14 never-married professionals aged 28 through 40. The investigation depicts several dimensions of the circumstances of never-married professionals that have not garnered much attention in the literature. This research provides initial evidence of critical influences within the socio-cultural contexts of the participants' dating behaviors as well as the willingness to many. Two different realities, based on gender, were discovered: both male and female voluntarily delayed marriage to pursue their career, education and success. As both reached their career goals, however, male participants experienced a rise in their value as a marriage prospect whereas females experienced decreased value as a marriage prospect. Hence, neo-traditional familism was the dominant value among the participants, but it also functioned as a major barrier to family formation because in the present value system, female professionals, must still struggle with the dual burdens of work and family management.

An effect of the financial resources on the willingness to marriage formation among the never married in their 30s (30대 미혼남녀의 결혼의향에 대한 경제자원의 효과)

  • The purpose of this study is to identify the factors that influence the willingness to marriage initiation(WMI) among the never married in their 30s. In particular, we examine the effect of financial resources on the WMI. A data set from the National Survey of Marriage and Childbirth (2009 Wave) were analyzed. We find that the effect of financial resources on the WMI are different by gender. Home ownership is a statistically significant factor in the logistic regression model for men, but not in the model for women. Women with higher earnings are more likely to be single and unemployed men are more likely to be single than employed men. In the Korean societal contexts, these findings support the bi-standard of financial resources between male and female in the marriage market.

  • Cleidocranial dysplasia (CCD) is an autosomal-dominant disease characterized by the delayed closure of cranial sutures, defects in clavicle formation, supernumerary teeth, and delayed tooth eruption. Defects in the Runt-related transcription factor 2 (RUNX2), a master regulator of bone formation, have been identified in CCD patients. The aim of this study was to identify the molecular genetic causes in a CCD family with delayed tooth eruption. The 23-year-old female proband and her mother underwent clinical and radiographic examinations, and all coding exons of the RUNX2 were sequenced. Mutational analysis revealed a single nucleotide deletion mutation (NM_001024630.4 : c.357delC) in exon 3 in the proband and her mother. The single C deletion would result in a frameshift in translation and introduce a premature stop codon [p.(Asn120Thrfs*24)]. This would result in the impaired function of RUNX2 protein, which may be the cause of delayed eruption of permanent teeth in the family.

The Impact of Educational Attainment on First Marriage Formation: Marriage Delayed or Marriage Forgone? (교육이 초혼 형성에 미치는 영향: 결혼 연기 혹은 독신?)

  • Using the Korean labor and Income Panel Study, this study examines the impact of educational attainment on first marriage formation in Korea. In particular, this study examines whether higher education is associated primarily with delayed marriage or with a higher likelihood of never marrying. The results show that the trend toward later and less marriage is occurring at all levels of educational attainment in Korea. The data also indicated that educational attainment showed differential effects on the risk of first marriage formation for Korean men and women. For those born before 1970, both highly educated men and women delayed marriage but caught up by marrying at higher rates at later ages. However, for those born after 1970, highly educated women were increasingly more likely to show the trend toward later and less marriage, while highly educated men were more likely to delay marriage but caught up by marrying at higher rates at later ages. Overall the evidence in this study is consistent with the argument that gender divisions make it difficult for women to balance work and family in Korea.

Functions of PUF Family RNA-Binding Proteins in Aspergillus nidulans

  • RNA-binding proteins are involved in RNA metabolism and posttranscriptional regulation of various fundamental biological processes. The PUF family of RNA-binding proteins is highly conserved in eukaryotes, and its members regulate gene expression, mitochondrial biogenesis, and RNA processing. However, their biological functions in Aspergillus species remain mostly unknown in filamentous fungi. Here we have characterized the puf genes in the model organism Aspergillus nidulans. We generated deletion mutant strains for the five putative puf genes present in the A. nidulans genome and investigated their developmental phenotypes. Deletion of pufA or pufE affected fungal growth and asexual development. pufA mutants exhibited decreased production of asexual spores and reduced mRNA expression of genes regulating asexual development. The pufE deletion reduced colony growth, increased formation of asexual spores, and delayed production of sexual fruiting bodies. In addition, the absence of pufE reduced both sterigmatocystin production and the mRNA levels of genes in the sterigmatocystin cluster. Finally, pufE deletion mutants showed reduced trehalose production and lower resistance to thermal stress. Overall, these results demonstrate that PufA and PufE play roles in the development and sterigmatocystin metabolism in A. nidulans.

Extracellular S100A4 negatively regulates osteoblast function by activating the NF-κB pathway

  • Patients with inflammatory bone disease or cancer exhibit an increased risk of fractures and delayed bone healing. The S100A4 protein is a member of the calcium-binding S100 protein family, which is abundantly expressed in inflammatory diseases and cancers. We investigated the effects of extracellular S100A4 on osteoblasts, which are cells responsible for bone formation. Treating primary calvarial osteoblasts with recombinant S100A4 resulted in matrix mineralization reductions. The expression of osteoblast marker genes including osteocalcin and osterix was also suppressed. Interestingly, S100A4 stimulated the nuclear factor-kappaB (NF-${\kappa}B$) signaling pathway in osteoblasts. More importantly, the ex vivo organ culture of mouse calvariae with recombinant S100A4 decreased the expression levels of osteocalcin, supporting the results of our in vitro experiments. This suggests that extracellular S100A4 is important for the regulation of bone formation by activating the NF-${\kappa}B$ signaling pathway in osteoblasts.

Physio-Ecological Studies on Stevia(Stevia rebaudiana Bertoni) (스테비아(Stevia rebaudiana Bertoni)에 관한 생리 생태적 연구)

  • Stevia (Stevia rebaudiana Bertoni) is a perennial herb widely distributed in the mountainous area of Paraguay. It belongs to the family Compositae and contains 6 to 12 percent stevioside in the leaves. Stevioside is a glucoside having similar sweetening character to surgar and the degree of sweetness is approximately 300 times of sugar. Since Korea does not produce any sugar crops, and the synthetic sweetenings are potentially hazardous for health, it is rather urgent to develop an economical new sweetener. Consequently, the current experiments are conducted to establish cultural practices of stevia, a new sweetening herbs, introduced into Korea in 1973 and the results are summarized as followings: 1. Days from transplanting of cuttings to the flower bud formation of 6 stevia lines were similar among daylengths of 8, 10 and 12 hours, but it was much greater at daylengths of 14 or 24 hour and varietal differences were noticable. All lines were photosensitive, but a line, 77013, was the most sensitive and 77067 and Suweon 2 were less sensitive to daylength. 2. Critical daylength of all lines seemed to be approximately 12 hours. Growth of plants was severely retarded at daylengths less than 12 hours. 3. Cutting were responded to short daylength before rooting. Number of days from transplanting to flower bud formation of 40-day old cuttings in the nursery bed was 20 days and it was delayed as duration of nursery were shorter. 4. Number of days from emergence to flower bud formation was shortest at short day treatment from 20 days after emergence. It was became longer as initiation of short day treatment was earlier or later than 20 days. 5. Plant height, number of branches, and top dry weight of stevia were reduced as cutting date was delayed from March 20 to May 20. The highest yield of dry leaf was obtained at nursery duration of 40-50 days in march 20 cutting, 30-40 days in April 20 cutting, and 30 days in May 20 cutting. 6. An asymptotic relationship was observed between plant population and leaf dry weight. Yield of dry leaf increased rapidly as plant population increased from 5,000 to 10,000 plants/10a with a reduced increasing rate from 10,000 to 20,000 plants/l0a, and levelled off at the plant population higher than 20,000 plants/l0a. 7. Stevia was adaptable in Suweon, Chengju, Mokpo and Jeju and drought was one of the main factors reducing yield of dry leaf. Yield of dry leaf was reduced significantly (approximately 30%) at June 20 transplanting compared to optimum transplanting. 8. Yield of dry leaf was higher in a vinyl house compared to unprotected control at long daylength or natural daylength except at short day treatment at March 20. Higher temperature ill a vinyl house does not have benefital effects at April 20 transplanting. 9. The highest content of stevioside was noted at the upper leaves of the plant but the lowest was measured at the plant parts of 20cm above ground. Leaf dry weight and stevioside yield was mainly contributed by the plant parts of 60 to 120cm above ground but the varietal differences were also significant. 10. Delayed harvest by the time of flower bud formation increased leaf dry weight remarkably. However, there were insignificant changes of yield as harvests were made at any time after flower bud formation. Content of stevioside was highest at the time of flower bud formation and earlier or later harvest than this time was low in its content. The optimum harvesting time determined by leaf dry weight and stevioside content was the periods from flower bud formation to right before flowering that would be the period from September 10 to September 15 in Suweon area. 11. Stevioside and rebaudioside content in the leaves of Stevia varieties were ranged from 5.4% to 14.3% and 1.5% to 8.3% respectively. However, no definit relationships between stevioside and rebaudioside were observed in these particular experiments.

A Small GTPase RHO2 Plays an Important Role in Pre-infection Development in the Rice Blast Pathogen Magnaporthe oryzae

  • The rice blast pathogen Magnaporthe oryzae is a global threat to rice production. Here we characterized RHO2 gene (MGG_02457) that belongs to the Rho GTPase family, using a deletion mutant. This mutant ${\Delta}Morho2$ exhibited no defects in conidiation and germination but developed only 6% of appressoria in response to a hydrophobic surface when compared to the wild-type progenitor. This result indicates that MoRHO2 plays a role in appressorium development. Furthermore, exogenous cAMP treatment on the mutant led to appressoria that exhibited abnormal morphology on both hydrophobic and hydrophilic surfaces. These outcomes suggested the involvement of MoRHO2 in cAMP-mediated appressorium development. ${\Delta}Morho2$ mutation also delayed the development of appressorium-like structures (ALS) at hyphal tips on hydrophobic surface, which were also abnormally shaped. These results suggested that MoRHO2 is involved in morphological development of appressoria and ALS from conidia and hyphae, respectively. As expected, ${\Delta}Morho2$ mutant was defective in plant penetration, but was still able to cause lesions, albeit at a reduced rate on wounded plants. These results implied that MoRHO2 plays a role in M. oryzae virulence as well.

EXPRESSION PATTERN OF RUNX2 IN MURINE TOOTH DEVELOPMENT (Mouse의 치아 발육시 Runx2의 발현 양상)

  • Runx2 is a transcription factor in homologous with Drosophila runt gene and it is essential for bone formation during embryogenesis and a critical gene for osteoblast differentiation and osteoblast function. Runx2-haploinsufficency causes cleidocranial dysplasia (CCD). CCD is an autosomal-dominant inherited disorder characterized by hypoplastic clevicle and delayed ossification in fontanelles and wormian bones. Dental defects are possibly shown to CCD patients : multiple supernumerary teeth, irregular and compressed permanent tooth crowns, hypoplastic and hypomineralized defects in enamel and dentin, an excess of epithelial root remnants, the absence of cellular cementum, and abnormally shaped roots. In addition, delayed eruption of the secondary dentition is a constant finding. The aim of this study is to evaluate the role of Runx2 in the tooth development and eruption through analyzing the expression pattern of Runx2 by in situ hybridization during crown (late bell stage) and root formation of tooth, using postnatal day 1, 4, 7, 14 and 21 mice mandibular molar teeth. mRNA of Runx2-full length is expressed in dental follicle and surrounding tissue at postnatal day1 and 4. At postnatal day 7, it is expressed in ameloblasts of occlusal surface of enamel and bone area surrounding the tooth. In comparison with previous stage, at postnatal day 14, it is expressed in ameloblasts of proximal surface of enamel. At postnatal day 21 it's expression is observed only in bone area. mRNA of Runx2-typeII is not expressed. At postnatal day 1 and 7. At postnatal day 14 and 21, it's expression is observed in the bone area. In this study, we suggest that Runx2 have a relation of ameloblasts differentiation and an important role to tooth eruption made by dental follicle during intraosseous eruption stage. Also we can confirm that Runx2 has a role to bone formation.

Germination of Yam Bean Seeds as Affected by Temperature and Its Productivity with Different Seeding Dates (얌빈의 온도별 발아특성과 파종시기에 따른 생산성 비교)

  • Yam bean (Pachyrhizus erosus) is a subtropical plant belonging to the Fabaceae family, and is a tuberous vegetable used as various food material with a crisp and juicy taste. This study was conducted to seek optimum sowing time of yam bean in Korea. For this, we surveyed germination properties by the different temperatures and compared the accumulation temperature (AT) and dry matter production (DMP) on growth stages of yam bean by the different sowing times. Two types of varieties cultivated mainly in Korea, Thailand local variety (TLV) and Cheongunmanma cultivar (CGMM), was used. The germination rate of yam bean was 86.0~94.0% at above $18^{\circ}C$, and germination days was longer at lower temperature. The times for flowering and tuber formation of CGMM were later than those of TLV, and the AT required for flowering, tuber formation and hypertrophy of CGMM were higher than those of TLV by $293^{\circ}C$, $280^{\circ}C$ and $108^{\circ}C$, respectively. Also, DMP of shoot and tuber in CGMM were greater than those in TLV. In sowing at April 25, tuber formation was slower than sowing after that time, and harvest index (HI) was relatively low due to delayed formation and hypertrophy of tuber. In sowing after June 9, DMP of shoot was relatively greater in early growth, but tuber was not sufficient to enlarge due to lack of growth days by cold and frost in late October. In sowing May 10 and 25, DMP of tuber and HI were the highest, because the change of day length and temperature gave an advantage to vegetable growth and tuber development. All above suggest that it was suitable to sow seeds on May for increment of tuber productivity in Korea.
