In this study, the anti-allergic activity and mechanim of Salviae radix (SR) were investigated. The ethanol extract of SR showed significant inhibitory effect on degranulation from antigen-stimulated mast cells and it also inhibited the expression and secretion of TNF-${\alpha}$ and IL-4 in antigen-stimulated RBL-2H3 cells. In the mast cell-mediated local animal allergy model, it suppressed the passive cutaneous anaphylaxis in a dose-dependent manner. As its mechanism of action, SR inhibited the activating phosphorylation of Syk, a downstream signaling molecule of Src-family kinase, for the activation of mast cells. The results of the study indicate that the anti-allergic activity of SR is mediated by the inhibition of Src-family kinase in mast cells.
The aim of this study was to investigate the effects of Chungdaesan(CDS) by various administration routes on skin anaphylactic reaction. The most classic and popular skin reaction in vivo is passive cutaneous anaphylaxis(PCA) In this study, therefore, the author investigated the effect of CDS on PCA reaction activated by anti-dinitrophenyl immunoglobulin E antibody The results showed that CDS potently suppressed orally, topically, intraperitoneally, and intradermally administered. However, it did not show suppressive activity when intravenously administered. In addition CDS significantly inhibited anti-DNP IgE induced mast degranulation in mice skin. Moreover, CDS suppressed anaphylactic histamine release from mast cells induced by anti-dinitrophenyl immunoglobulin E antibody. These results indicate that CDS suppresses the PCA reaction by stabilization of mast cells in vivo and in vitro am] also suggest that the differential activity following administration routes may be caused by the difference of bioavailability.
The purpose of this research was to investigate effects of Bangpoongtongsungsan water extract(BTSE) on the immune reaction, anti-allergy action and anti-inflammatory action in BALB/c mice. The administration of BTSE (500mg/kg) enhanced the cell viability of thymocytes and the population of helper T cells in splenic T-lymphocytes. BTSE suppressed the production of nitric oxide, but enhanced the phagocytic activity in peritoneal macrophages. BTSE enhanced hemagglutination titer in mice. BTSE inhibited passive cutaneous anaphylaxis induced by egg albumin in rat, the lethal anaphylaxis induced by platelet activating factor and compound 48/80 in mice, and then inhibited the degranulation of peritoneal mast cells induced by compound 48/80. BTSE did not inhibit Arthus reaction, but inhibited the delayed type hypersensitivity induced by SRBC and contact dermatitis induced by DNFB. BTSE inhibited the acute hind paw edema induced by histamine after 30 minutes, the permeability of evans blue into peritoneal cavity induced by acetic acid and the writhing syndrome induced by acetic acid. These results suggest that BTSE has an immunopotentiative action, anti-allergy action and anti-inflammatory action via the inhibition of histamine release.
Neutrophils are professional phagocytes that provide defense against invading pathogens through phagocytosis, degranulation, generation of ROS, and the formation of neutrophil extracellular traps (NETs). Although long been considered as short-lived effector cells with limited biosynthetic activity, recent studies have revealed that neutrophils actively communicate with other immune cells. Neutrophils employ various types of soluble mediators, including granules, cytokines, and chemokines, for crosstalk with immune cells. Additionally, ROS and NETs, major arsenals of neutrophils, are utilized for intercellular communication. Furthermore, extracellular vesicles play a crucial role as mediators of neutrophil crosstalk. In this review, we highlight the extracellular mechanisms of neutrophils and their roles in crosstalk with other cells.
Eosinophils are terminally differentiated cytotoxic effector cells that have a role in parasitic infections and allergy by releasing their granule-derived cytotoxic proteins. However, an increasing number of recent observations indicate that eosinophils are not only associated with the pathogenesis of a wide range of diseases, but also contribute to the maintenance of homeostatic responses in previously underappreciated diverse tissues, such as the gastrointestinal (GI) tract and adipose tissue. In this review, we describe biological characteristics of eosinophils, as their developmental properties, permissive proliferation and survival, degranulation activity, and migration properties enable them to distribute to both homeostatic and inflamed tissues. We describe pathologic aspects of eosinophils with a role in asthma and in various GI diseases, including eosinophilic GI disorders, inflammatory bowel disease, and radiation-induced enteropathy. Finally, we discuss the beneficial role of eosinophils, which contribute to the resolution of pathogenic conditions and to the modulation of homeostatic biologic responses.

Neutrophils are innate immune cells that constitute the first line of defense against invading pathogens. Due to this characteristic, they are exposed to diverse immunological environments wherein sources for nutrients are often limited. Recent advances in the field of immunometabolism revealed that neutrophils utilize diverse metabolic pathways in response to immunological challenges. In particular, neutrophils adopt specific metabolic pathways for modulating their effector functions in contrast to other immune cells, which undergo metabolic reprogramming to ensure differentiation into distinct cell subtypes. Therefore, neutrophils utilize different metabolic pathways not only to fulfill their energy requirements, but also to support specialized effector functions, such as neutrophil extracellular trap formation, ROS generation, chemotaxis, and degranulation. In this review, we discuss the basic metabolic pathways used by neutrophils and how these metabolic alterations play a critical role in their effector functions.
Apoptosis signal-regulating kinase 1 (ASK1) is an upstream signaling molecule in oxidative stress-induced responses. Because oxidative stress is involved in asthma pathogenesis, ASK1 gene deficiency was investigated in animal models of allergic asthma. However, there is no study to investigate whether ASK1 inhibitors could be applied for asthma to date. Selonsertib, a potent and selective ASK1 inhibitor, was applied to BALB/c mice of an ovalbumin (OVA)-induced allergic asthma model. Selonsertib suppressed antigen-induced degranulation of RBL-2H3 mast cells in a concentration-dependent manner. The administration of selonsertib both before OVA sensitization and OVA challenge significantly reduced airway hyperresponsiveness, and suppressed eosinophil numbers and inflammatory cytokine levels in the bronchoalveolar lavage fluid. Histopathologic examination elucidated less inflammatory responses and reduced mucin-producing cells around the peribronchial regions of the lungs. Selonsertib also suppressed the IgE levels in serum and the protein levels of IL-13 in the bronchoalveolar lavage fluid. These results suggest that selonsertib may ameliorate allergic asthma by suppressing immune responses and be applicable to allergic asthma.
머위(Petasites japonicus) 잎에서 알레르기성 천식 억제 효능이 있는 bakkenolide B의 최적 추출공정을 반응표면 분석법을 이용하여 결정하였다. Box-Behnken design에 따라 설정한 추출온도(80, 100, $120^{\circ}C$), 추출시간(30, 45, 60 min) 및 추출용매의 pH (5, 7, 9)를 독립변수로 하고, 추출물의 특성 즉, bakkenolide B의 함량과 항알러지의 측정지표인 비만세포의 탈과립반응 억제효과를 종속변수로 하여 분석을 실시하였다. Bakkenolide B의 추출함량은 추출온도, 시간, pH의 유의적인 영향을 받았으며, 추출온도 $127.1^{\circ}C$, 시간 46.6분, 추출용매의 pH 7.7에서 최대치 121.6 ${\mu}g/g$이 예측되었다. 비만세포의 탈과립반응 억제효과의 회귀분석 결과는 유의성이 인정되지 않았고 낮은 상관계수를 보였다.
목 적 : 운동 중 과호흡으로 인해 기도 내에 열과 수분이 손실되어 기도 내의 삼투압이 높아지게 되고, 이로써 여러 세포들이 염증 매개체를 유리함으로써 운동 유발성 천식을 일으킨다고 추정하고 있다. 이에 본 연구에서는 알레르기 염증반응에 중요한 역할을 하는 호산구에 고삼투압으로 자극시킨 후 호산구의 활성화 여부를 알아보고자 하였다. 방 법 : 호산구 세포주인 EoL-1 세포에 각각의 삼투질 농도와 시간에 따른 superoxide anion의 생성을 측정하였고, EoL-1 세포에 15분 동안 고삼투압 자극(930 mOsm/kg $H_2O$)을 준 직후와 이후 등장성 배지에 옮겨 5시간 동안 배양 후 유세포 분석기를 이용하여 호산구 표면의 CD69와 ICAM-1의 발현을 측정하였고, 배양 상층액에서 ECP 농도를 측정하였다. 결 과 : EoL-1 세포에 15분간 고삼투압 자극을 준 직후 측정한 ECP 농도와 CD69 및 ICAM-1의 fluorescent intensity는 등장성 배지에 노출시켰을 경우와 비교하여 차이를 보이지 않았으며, 고삼투압 자극 후 등장성 배지에 5시간 동안 배양하였을 때에도 유의한 차이를 보이지 않았다. Superoxide anion의 생성에 있어서도 다양한 삼투질 농도에 따른 차이를 보이지 않았으며, 시간의 경과에도 차이를 보이지 않았다. 결 론 : 이번 연구에서는 운동 후 발생하는 기관지 수축의 조기 반응과 후기 반응과 관련하여 고삼투압 자극에 의한 호산구의 활성화를 관찰할 수 없었으나, 향후 연구에 있어 운동 유발성 천식과 관련한 호산구의 역할에 대한 많은 실험과 연구가 필요 하리라 사료된다.
Dendroaspis natriuretic peptide (DNP)는 최근에 green Mamba snake Dendroaspis angusticeps의 독(venom)에서 발견된 17개의 아미노산 disulfide링 구조를 포함하는 38개 아미노산 펩티드로 natriuretic peptide family와 그 구조가 비슷하다. Natriuretic peptide family는 사람과 흰쥐 비만세포로부터 히스타민을 유리시킨다고 알려져 있다. 그러나 DNP가 비만 세포로부터 히스타민을 유리 시키는 여부는 밝혀져 있지 않다. 본 연구는 DNP가 흰쥐 복강 비만세포로부터 히스타민을 유리시키는 여부와 함께 히스타민 유리기전을 구명하고자 실시되었다. 다양한 농도의 DNP를 흰쥐 복강 비만세포에 처리한 다음, 비만세포의 탈과립을 도립 광학현미경으로 관찰하였다. 히스타민 유리량은 방사효소법으로 측정하였고 세포내 칼슘 유입량과 cyclic GMP의 수준은 방사면역법으로 측정하였다. DNP는 흰쥐 복강 비만세포를 탈과립시켰고 농도가 증가함에 따라 비만세포로부터 히스타민 유리량이 증가되었다. 또한 DNP는 농도 증가에 비례하여 비만세포 안으로 세포밖의 칼슘을 유입시켰으며 세포안의 cyclic GMP의 수준을 증가시켰다. 이상의 결과로 미루어, DNP는 비만세포 안의 cyclic GMP와 칼슘의 농도를 증가시켜 비만세포로부터 히스타민을 유리시키는 것으로 생각된다.
