Development of pallet-scale modified atmosphere packaging for 'Tabor' tomatoes (토마토 'Tabor' 품종의 파렛트 단위 MAP 적용 연구)

  • This study was carried out to investigate the effect of modified-atmosphere packaging (MAP) on the quality change of "Tabor" tomatoes during long-term exportation periods. Hydroponics tomatoes were harvested at the turning stage, sorted, and box packed and then packaged in nylon film with a pallet. The packaged pallet was filled with a gas composition (5% $O_2$, 1% $CO_2$, and 94% $N_2$) and stored at $10^{\circ}C$ for three weeks. The quality changes in weight loss, firmness, color, acidity, soluble solids, and microorganism growth were measured every 7 day interval. During the initial storage, the pallet-scale MAP showed slightly higher weight loss and firmness changes when compared to the conventional pallet. The total color change (${\Delta}E$) during ripening was delayed 10% under MAP storage. Acidity, soluble solids and phenolic compound contents decreased with increases in storage time regardless of the storage method; however, the quality changes of tomatoes were delayed in the MAP pallet. Furthermore, the decay rate of the pallet-scale MAP stored for 14 days was less than that of the conventional pallet, and the number of microorganisms was approximately 30% lower in the pallet-scale MAP, showing a positive effect on marketability. These results suggested that the pallet-scale MAP of tomatoes could ensure higher quality and longer storage periods than conventional pallet storage.

  • Background: In the pathogenesis of acute lung injury induced by lipopolysaccharide(LPS), oxygen radiclls are known to be involved in one part. Superoxide dismutase(SOD) protects oxygen radical-induced tissue damage by dismutating superoxide to hydrogen peroxide. In eukaryotic cells, two forms of SOD exist intracellularly as a cytosolic, dimeric copper/zinc-containing SOD(CuZnSOD) and a mitochondrial, tetrameric manganese-containing SOD(MnSOD). But there has been little information about SOD gene expression and its regulation in pulmonary alveolar macrophages(PAMs). The objective of this study is to evaluate the SOD gene expression induced by LPS and its regulation in PAMs of rat. Method: In Sprague-Dawley rats, PAMs obtained by broncholaveolar lavage were purified by adherence to plastic plate. To study the effect of LPS on the SOD gene expression of PAMs, they were stimulated with different doses of LPS($0.01{\mu}g/ml{\sim}10{\mu}g/ml$) and for different intervals(0, 2, 4, 8, 24hrs). Also for evaluating the level of SOD gene regulation actinomycin D(AD) or cycloheximide(CHX) were added respectively. To assess whether LPS altered SOD mRNA stability, the rate of mRNA decay was determined in control group and LPS-treated group. Total cellular RNA extraction by guanidinium thiocyanate/phenolfchlorofonn method and Northern blot analysis by using a $^{32}P$-labelled rat MnSOD and CuZnSOD cDNAs were performed. Results: The expression of mRNA in MnSOD increased dose-dependently, but not in CuZnSOD. MnSOD mRNA expression peaked at 8 hours after LPS treatment. Upregulation of MnSOD mRNA expression induced by LPS was suppressed by adding AD or CHX respectively. MnSOD mRNA stability was not altered by LPS. Conclusion: These findings show that PAMs of rat could be an important source of SOD in response to LPS, and suggest that their MnSOD mRNA expression may be regulated transcriptionally and require de novo protein synthesis without affecting mRNA stability.

Population Phenology and an Early Season Adult Emergence model of Pumpkin Fruit Fly, Bactrocera depressa (Diptera: Tephritidae) (호박과실파리 발생생태 및 계절초기 성충우화시기 예찰 모형)

  • The pumpkin fruit fly, Bactrocera depressa (Tephritidae: Diptera), is one of the most important pests in Cucurbitaceae plants. This study was conducted to investigate the basic ecology of B. depressa, and to develop a forecasting model for predicting the time of adult emergence in early season. In green pumpkin producing farms, the oviposition punctures caused by the oviposition of B. depressa occurred first between mid- and late July, peaked in late August, and then decreased in mid-September followed by disappearance of the symptoms in late September, during which oviposition activity of B. depressa is considered active. In full-ripened pumpkin producing farms, damaged fruits abruptly increased from early Auguest, because the decay of pumpkins caused by larval development began from that time. B. depressa produced a mean oviposition puncture of 2.2 per fruit and total 28.8-29.8 eggs per fruit. Adult emergence from overwintering pupae, which was monitored using a ground emergence trap, was first observed between mid- and late May, and peaked during late May to early June. The development times from overwintering pupae to adult emergence decreased with increasing temperature: 59.0 days at $15^{\circ}C$, 39.3 days at $20^{\circ}C$, 25.8 days at$25^{\circ}C$ and 21.4 days at $30^{\circ}C$. The pupae did not develop to adult at $35^{\circ}C$. The lower developmental threshold temperature was calculated as $6.8^{\circ}C$ by linear regression. The thermal constant was 482.3 degree-days. The non-linear model of Gaussian equation well explained the relationship between the development rate and temperature. The Weibull function provided a good fit for the distribution of development times of overwintering pupae. The predicted date of 50% adult emergence by a degree-day model showed one day deviation from the observed actual date. Also, the output estimated by rate summation model, which was consisted of the developmental model and the Weibull function, well pursued the actual pattern of cumulative frequency curve of B. depressa adult emergence. Consequently, it is expected that the present results could be used to establish the management strategy of B. depressa.

An Analysis of Growth Conditions of old Trees in Yangdong Villages (양동마을의 노거수 생육실태 분석)

  • The purpose of this study is to provide the basic data by analyzing and analysing the Growth Conditions of old Trees in Yangdong village. This study investigated about the conducted on tree information, soil information, and tree health. The result are as follows. The trees information in Yangdong Village consists of Juniperus chinensis, Salix chaenomeloides, Salix pseudolasiogyne, Celtis sinensis, Zelkova serrata, Gleditsia japonica, and Gleditsia sinensis trees, The range of height was 4.0~17.0m, and the diameter was 0.51~1.34m, Juniperus chinensis trees of No.17 was most large. In the results of soil analysis, there showed that acidity was pH4.1~6.3, hardness of 5~48mm, organic matter content of 21.2~29.1g/kg, electrical conductivity(EC) of 0.34~1.76dS/m, available P2O5 of 79.8~451.6mg/kg, exchangeable K of 0.22~1.71cmol+/kg, exchangeable Ca of 4.98~7.44cmol+/kg, exchangeable Mg of 0.67~2.19cmol+/kg, exchangeable Na of 0.19~1.04cmol+/kg and cation exchange capacity(C.E.C) of 7.23~13.02cmol+/kg. As a result, the highest number of tree health levels is 8 of 11trees of Celtis sinensis, 2 of 7trees of Zelkova serrata, and 3 of Gleditsia sinensis, and 13 of 30 trees of health levels, The Older trees with high infection, spoil and hollowed part were the remaining trees except for the healthy part. Relatively, more than half of the number of targets is infected, decay, and the hollowed site, and it is necessary to perform surgery on the damaged area. In addition, preservation and protection measures should be implemented by supplying root nutrients for trees, controlling nutrients in the body to prevent secondary and tertiary damages that cause the infection site to metastasize to the health site, In order to continually monitor the trees, measures to improve the location environment and management of the trees should be sought.

Occurrences of Uranium and Radon-222 from Groundwaters in Various Geological Environment in the Hoengseong Area (횡성지역 다양한 지질환경에서 지하수 중 우라늄 및 라돈-222 산출특성)

  • Groundwaters in granite, gneiss, and two-mica granite formations, including faults, in the Hoengseong area are examined to determine the relationship between their uranium and radon-222 contents and rock types. The chemical compositions of 38 groundwater samples and four surface water samples collected in the study area were analyzed. Sixteen of the samples showing high uranium and radon-222 contents were repeatedly analyzed. Surface radioactivities were measured at 30 points. The uranium and radon-222 concentrations in the groundwater samples were in the ranges of 0.02-49.3 μg/L and 20-906 Bq/L, respectively. Four samples for uranium and 35 samples for radon had concentrations exceeding the alternative maximum contaminant level of the US EPA. The chemical compositions of groundwaters indicated Ca(Na)-HCO3 and Ca(Na)-NO3(HCO3+Cl) types. The pH values ranged from 5.71 to 8.66. High uranium and radon-222 contents in the groundwaters occurred mainly at the boundary between granite and gneiss, and in the granite area. The occurrence of uranium did not show any distinct relationship to that of radon-222. The radon-222, an inert gas, appeared to be dissolved in the groundwater of the aquifer after wide diffusion along rock fractures, having been derived from the decay of uranium in underground rocks. The results in this study indicate that groundwater of neutral or weakly alkaline pH, under oxidizing conditions and with a high bicarbonate content is favorable for the dissolution of uranium and uranium complexes such as uranyl or uranyl-carbonate.

Effect of Decay Rate Coefficients with Sulfur Denitrification due to Salt in Seafood Wastewater (수산물 가공폐수내 염분이 황 탈질 반응속도 상수에 미치는 영향)

  • This study examined the effects of the salinity of seafood wastewater on the sulfur denitrification process. An examination of the denitrification efficiency showed that the optimal EBCT was 1hr at an influent T-N concentration of 20mg/L or lower and 2-3hr at an T-N concentration of 30mg/L. An examination of the denitrification efficiency according to the nitrogen load showed that the legal effluent water quality criterion was satisfied when the influent load was maintained within $0.496kg/m^3/day$. On the other hand, the reactor volume increased when this was applied to the site. Therefore, the influent load should be within $0.372kg/m^3/day$ considering the denitrification and economic efficiency. At a load of $0.248{\sim}0.628kg/m^3{\cdot}day$, the k value was $0.0890{\sim}0.5032hr^{-1}$. The batch experimental results according to the $Cl^-$ concentration showed that at an influent nitrogen concentration of 30.0mg/L, the effect of the denitrification efficiency was not large below the salinity of $7,000mgCl^-/L$, but inhibition occurred above $9,000mgCl^-/L$. Calculations of the reaction rate constant according to the $Cl^-$ concentration showed that the reaction rate constant was $0.1049{\sim}0.2324hr^{-1}$ at a raw wastewater concentration of ${\sim}5,000mgCl^-/L$. In contrast, the k value was $0.1588hr^{-1}$ at $7,000mgCl^-/L$ and $0.1049hr^{-1}$ at $9,000mgCl^-/L$.

Changes in Pectin-degrading Enzymes activity during Storage of Satsuma Mandarin (온주밀감의 저장 중 성분과 펙틴분해효소의 변화)

  • Satsuma mandarin(Citrus unshiu Marc. var. miyagawa) was stored at $3^{\circ}C$ and 85% relative humidity, and then the changes of firmness, pectin- degrading enzymes activity and other physicochemical properties of citrus fruits during storage were investigated. Firmness of fruits with 2 m probe was decreased quickly from 1,176.8 g-force to 503.6 g-force, and moisture of peel and flesh were decreased from 75.3% to 74.9%, and from 91.8% to 90.7% during maturation, respectively. Decay ratio was increased to 18.75% after 90 days' storage, and after then it was increased rapidly. Weight loss was increased gradually to 24.5% during long-term storage. Firmness with 2 mm probe were decreased from 538.9 g-force to 336.9 g-force gradually during storage. Peel moisture was decreased from 75.8% to 72.6%, and flesh moisture was also decreased gradually from 90.3% to 88.3% during storage. Exopoly-galacturonase activity of peel and flesh were increased from 326.0 units/100 g to 534.9 units/100 g, and from 63.1 units/100 g to 81.0 units/100 g at 90 day's storage, respectively. After then, He enzyme activities were decreased from 394.0 units/100 g and 38.0 units/100 g, respectively. Pectinesterase activity of peel and flesh were increased from $14.4\;{\mu}mol$ to $38.8{\mu}mol$, and from $26.0{\mu}mol$ to $39.0{\mu}mol$ at 60 days' storage, respectively. After then, the enzyme activities were decreased to $6.0{\mu}mol$ and $8.2{\mu}mol$, respectively.

Cold Shock Response and Low Temperature Stable Transcript of DEAD-box RNA Helicase in Bacillus subtilis (DEAD-box RNA Helicase 유전자가 결핍된 Bacillus subtilis의 저온 충격 반응성과 저온 안정성 전사물)

  • We investigated the cold shock sensitivity of DEAD-box RNA helicase gene deleted strains of in Bacillus subtilis CU1065. To understand cold shock effects, cells were cultivated at $37^{\circ}C$ to log phase ($O.D_{600}$=0.5-0.6) and then temperature was shifted to $15^{\circ}C$. Cold shock slow down the growth rate of wild type and deleted strains of DEAD-box RNA helicase gene (ydbR, yfmL, yqfR, deaD). The growth rate of ydbR deleted strain is 5 times severely reduced compared to that of wild type strain (CU1065). But the growth rate of other three (yfmL, yqfR, deaD) deleted strains is nearly equal to the growth rate of wild type. Compared to $37^{\circ}C$, the amount of ydbR and yqfR mRNA transcripts are increased at the growth temperature of $15^{\circ}C$. On the other hands the mRNA transcripts of yfmL and deaD are not changed at both conditions of $37^{\circ}C$ and $15^{\circ}C$. Upon cold shock treatment ydbR mRNA transcript is clearly increased. After treatment of rifampicin (bacteria transcription inhibitor) the amount of ydbR mRNA was measured. Temperature shift from $37^{\circ}C$ to $15^{\circ}C$ and rifampicin treatment showed slowly decay of ydbR mRNA. But at $37^{\circ}C$ and rifampicin treatment ydbR mRNA is rapidly reduced. These results showed that cold shock induction of ydbR mRNA resulted from the stability of ydbR mRNA and not from the transcription induction of ydbR. In relation to these results, we found the cold box element of csp (cold shock protein gene) in 5' untranslated region of ydbR gene. Cold shock induction of ydbR is caused by the stability of ydbR mRNA like the stability of csp mRNA.

Estimation of Linkage Disequilibrium and Effective Population Size using Whole Genome Single Nucleotide Polymorphisms in Hanwoo (한우에서 전장의 유전체 정보를 활용한 연관불평형 및 유효집단크기 추정에 관한 연구)

  • This study was conducted to estimate the extent of linkage disequilibrium (LD) and effective population size using whole genomic single nucleotide polymorphisms (SNP) genotyped by DNA chip in Hanwoo. Using the blood samples of 35 young bulls born from 2005 to 2008 and their progenies (N=253) in a Hanwoo nucleus population collected from Hanwoo Improvement Center, 51,582 SNPs were genotyped using Bovine SNP50 chips. A total of 40,851 SNPs were used in this study after elimination of SNPs with a missing genotyping rate of over 10 percent and monomorphic SNPs (10,730 SNPs). The total autosomal genome length, measured as the sum of the longest syntenic pairs of SNPs by chromosome, was 2,541.6 Mb (Mega base pairs). The average distances of all adjacent pairs by each BTA ranged from 0.55 to 0.74 cM. Decay of LD showed an exponential trend with physical distance. The means of LD ($r^2$) among syntenic SNP pairs were 0.136 at a range of 0-0.1 Mb in physical distance and 0.06 at a range of 0.1-0.2 Mb. When these results were used for Luo's formula, about 2,000 phenotypic records were found to be required to achieve power > 0.9 to detect 5% QTL in the population of Hanwoo. As a result of estimating effective population size by generation in Hanwoo, the estimated effective population size for the current status was 84 heads and the estimate of effective population size for 50 generations of ancestors was 1,150 heads. The average decreasing rates of effective population size by generation were 9.0% at about five generations and 17.3% at the current generation. The main cause of the rapid decrease in effective population size was considered to be the intensive use of a few prominent sires since the application of artificial insemination technology in Korea. To increase and/or sustain the effective population size, the selection of various proven bulls and mating systems that consider genetic diversity are needed.

Performance Characteristics of MicroPET R4 Scanner for Small Animal Imaging (소동물 영상을 위한 MicroPET R4스캐너의 특성평가)

  • Purpose: Dedicated animal PET is useful equipment for the study of new PET tracer. recently, microPET R4 was installed in the Korea institute of radiology and medical science. In this study, we measured the characteristics of scanner. Materials and methods: Resolution was measured using a line source (F-18:65 ${\mu}Ci$, inner diameter: 0.5 mm). The line source was put in the axial direction and was moved from the center of field of view to outside with 1 mm interval. PET images were reconstructed using a filtered back-protection and ordered subset expectation maximization. line source (16.5 ${\mu}Ci$, 78 mm) was put on the tenter of axial direction to measure the sensitivity when the deadtime was under 1%. Images were acquired during 4 minutes respectively from center to 39 mm outward. Delayed count was subtracted from total count and then decay was corrected for the calculation of sensitivity. Noise equivalent count ratio and scatter fraction were calculated using cylindrical phantom. Results: Spatial resolution of reconstructed image using filtered back-projection was 1.86 mm(radial), 1.95 mm(tangential), 1.95 mm(axial) in the tenter of field of view, and 2.54 mm, 2.8 mm, 1.61 mm in 2 cm away from the center respectively. Sensitivity was 2.36% at the center of transaxial field of view. Scatter fraction was 20%. Maximal noise equivalent count ratio was 66.4 kcps at 242 kBq/mL. Small animal images were acquired for confirmation of performance. Conclusion: Performance characteristics of microPET R4 were similar with reported value. So this will be a useful tool for small animal imaging.
