Distribution of Fatty Acids in Newly Developed Tissues of Soybean Seedlings

  • The objective of this study was to determine the fatty acid composition of newly developed tissues of germinated soybean seeds. Five soybean accessions with varied fatty acid composition were allowed to germinate in sand under greenhouse conditions. Seedlings were picked up after 4, 6, 8, 10 and 12 days of germination and freeze dried. The fatty acid composition of the newly developed tissues was analyzed by gas chromatography. Significant variation in fatty acid composition was observed between accessions, days of germination, and variety ${\times}$ day of germination in whole and the cotyledons. In the case of newly developed five tissues, significant variation in fatty acid composition were observed between days of germination except oleic acid for root, hypocotyl and epicotyl stem and except stearic acid for hypocotyl and unifoliate leaves while all the parameters were significantly different for accession. Significant interactions of accession and days of germination were observed for palmitic, linoleic and linolenic acid in all tissues; only for oleic acid in hypocotyl, epicotyl and unifoliate leaves; and only for stearic acid in root, hypocotyl, epicotyl and unifoliate leaves. During germination, the fatty acid composition of newly developed tissues changed dramatically but whole seedlings and cotyledons changed slightly. These tissues contained five major fatty acids as found in original seeds, but compositions were totally different from that of the seed: higher in palmitic, stearic and linolenic acid and lower in oleic and linoleic acid. New tissues conserved their fatty acid compositions regardless of genotypic variation in the original seeds.

Effect of Seed Priming Treatment on the germination of Sesame

  • This experiment was conducted to find out optimum priming treatment conditions to the sesame seed as a preliminary study for enhancing sesame germination properties. Effective priming agents and concentrations for sesame seed were $K_3PO_4$, 200 mM and PEG6000, -1.0 MPa respectively. Optimum priming temperature and duration were $15^{\circ}C$, 4 days in view of germination speed and germination percentage. PEG6000 with -1.0 MPa was selected as an efficient priming treatment condition at $15^{\circ}C$, 4 days. This study suggested that priming treatment to sesame seed would be an effective technique enhancing sesame seed germination and shortening time to the $T_{50}$ at the field condition, but the efficiency of priming treatments to the sesame seed would be strongly dependent on individual or integrated conditions of priming agent, concentration, temperature and duration etc.

미생물에 의한 자귀나무·참싸리·비수리 종자의 발아촉진 (The Increase of Seeds Germination in Albizzia julibrissin, Lespedeza cyrtobotrya and Lespedeza cuneata by Microbial Treatment)

  • Herbs and plants widely used for the ecological restoration were selected for germination rate analysis under treatment of microorganisms to determine ideal treatment conditions and medium for enhanced germination rate. Albizzia julibrissin, when submerged in a nutrient medium or distilled water, presented a decrease in germination period rather than increase in germination rate. When treated with microorganism culture solution (JM-2) for 24 hours, 90% germination was achieved in two days, which is sufficient evidence to conclude that such treatment accelerates the germination of Albizzia julibrissin. Germination period decreased for Lespedeza cyrtobotrya samples submerged in microorganism solution for 15 and 48 hours, however, increases in germination rates were not observed. Sample treated in the solution for 24 hours had increased germination rate and enhanced germination period. Microorganism solution treatment had a negative effect on germination for Lespedeza cuneata, unlike Lespedeza cyrtobotrya and Albizzia julibrissin. Microorganism treated seeds of Lepsedeza cuneata had a lower germination rate than that of the control with no treatment. However, submerging treatments in a nutrient medium or distilled water for 24 to 48 hours were proven effective with higher germination rates than control sample with no treatment. Physicochemical and Functional Properties of Germinated Glycine max Merr Soybeans

  • To investigate the applicability as the functional food materials of germinated Glycine max Merr soybeans, its biochemical characteristics and its abilities to inhibit platelet aggregation and hydrolyze alcohol were examined. With the progression of germination time, crude protein content gradually increased, and on the 5th day of germination it was 30.19%. However, crude fat content tended to decrease, and on the 5th day of germination it was 14.30%. Total amino acid content was highest on the 3rd day of germination at 80,875 mg%. The free amino acid content doubled from day 0 of germination (1,273.35 mg%) until the 5th day of germination (2,742.99 mg%). Fatty acid analysis revealed that linoleic acid was highest among all the samples, ranging from $53.55{\sim}56.00%$. Linolenic acid content slightly increased as the germination period was prolonged. The ability to inhibit platelet aggregation increased according to the germination period and then decreased again on the 5th day of germination; it was somewhat higher in the ethanol fraction. In measuring ADH, we found that the activity of the ethanol fraction increased with increasing days of germination. In the case of the water fraction, the activity decreased as germination was prolonged, and the ADH activity of the water fraction was higher than that of the ethanol fraction. Based on the above results, we deemed that the Glycine max Merr soybeans germinated for $2{\sim}3$ days were most pertinent for use as functional food materials.

  • 금낭화의 실생번식방법을 확립하기 위하여 $GA_3(100mg{\cdot}L^{-1})$와 냉습저장($3^{\circ}C$) 그리고 배병의 제거가 금낭화의 발아에 미치는 영향을 조사하였다. $GA_3$를 처리하지 않은 종자는 90일간의 냉습저장에도 발아를 하지 않았다. 그러나 $GA_3$처리는 냉습저장에 관계없이 금낭화의 발아를 촉진하였다. 배병을 제거한 경우 $GA_3$처리에 의한 발아촉진효과가 감소하는 경향을 나타내었다. $GA_3$처리 후 냉습저장에 의해서 또한 냉습저장 기간이 길어질수록 발아일수는 단축되었으며 발아속도는 촉진되었다. 이상의 결과로부터 $GA_3$처리는 금낭화의 발아촉진에 효과적이며 냉습저장처리는 $GA_3$의 효과에 상승적인 작용을 하는 것을 알 수 있었다.

Germination Characteristics of Some Red Rice Accessions

  • Nine accessions of red rices collected from different regions were tested for germination characteristics in relation to seed maturity and several storage conditions after harvest. No red rice seeds germinated at five days after anthesis (DAA). Wanjuaengmi (long-grain red rice) seeds at 10DAA germinated up to 30% of total samples, producing only abnormal seedlings, whereas 10D-AA-seeds of normal cultivar showed only 3.3% germinability. Some red rice seeds absorbed more water than cultivated varieties, and showed less decrease in germination rate than cultivated varieties when seeds were exposed at -1, -5 or -1$0^{\circ}C$ for up to 60 hours after soaking. Red rice accessions maintained more than 95% germination when stored. indoor for 120 days. Two short-grain red rices showed about 50% germination when overwintered in the field, while other long-grain red rices and cultivated rices germinated less than 10%. Germination of seeds overwintered in clay loam soil was lower than that in loam soil, and seeds on surface germinated less than those in 1~7cm depth.

  • 우리나라 중서부 천결지에서 많이 자생하는 바다 새를 사료작물화 할 수 있는지를 구명하기 위하여 채종후 저장조건, 광, 옹도, 당수깊이, 염농도 및 복토깊이가 바다새의 종자 발아에 미치는 영향을 조사하였던 바, 그 결과를 요약하면 다음과 같다. 1. 바다새 종자의 발아특성은 광의 유무 간에는 유의성이 없었으며, 광의 유무 보다는 채종후 저장 조건이 유의성이 컸고, 광과 저장조건 상호간에도 유의성이 없었다. 2. 바다새의 발아율, 평균발아일수, 발아세 및 발아계수에 대한 발아온도 효과는 환온조건 보다는 변온조건에서 양호하였고, 환온조건에서는 $35^{\circ}C$에서 환온조건에서는 $35/25^{\circ}C$에서 가장 우수하게 나타났다. 3. 담수 깊이별 바다새의 발아율, 평균발아일수, 발아세 및 발아계수는 처리 수준간 통계적 유의성이 커서 담수 깊이가 Ocm인 구에서 제일 좋았으며, 담수 깊이가 0.5cm에서 1cm로 담수 깊이가 깊어 질수록 발아율이 현저히 떨어졌고 3cm 이상 담수서는 전혀 발아가 되지 않았다. 4. 고농도형 바다새의 발아율, 평균발아일수, 발아세 및 발아계수는 처리 수준간 유의성이 커서 고농도 0.2%일 때가 발아가 제일 좋았고 고농도가 높아질수록 발아율이 떨어졌으나 고농도 1%에서도 30% 이상의 발아율을 보여 내염성이 높음을 알 수 있었다. 5. 바다새 종자의 복사 수준별 발아율, 평균발아일수, 발아세 및 발아계수는 처리 수준간 유의성이 커서 복사 깊이가 0.2cm일 때가 제일 좋았고 복사 깊이가 깊어질수록 발아특성이 떨어져 복토 깊이가 1cm 이상에서는 거의 발아가 되지 않았다.5회 예취구가 37.06%로 유의하게 낮았고 3회와 4glh 예취구에서는 각각 40.27과 40.53%를 나타내어 유의성이 인정되지 않았다. 6. 건물 생산효율은 3회와 5회 예취구의 30kg N/ha/cut 수준에서 18.9kg와 15.7kg을 나타내어 가장 높았고, 4회 예취구에서는 90kg N/ha/cut 수준에서 16.3kg으로 가장 높았다. 전 질소량은 예취번초별로 3회 예취구에서는 90kg N/ha/cut수준에서 0.52kg, 4회와 5회 예취구에서는 30kg N/ha/cut수준에에서 각각 0.51kg과 0.43kg의 높은 전 질소량을 나타내었다. 7. 경제적 액상구비 시용수준의 범위는 3회 예취구에서 107.2~151.0kg N/ha/cut, 4회 예취구에서 359.1~375.7kg N/ha그리고 5회 예취구에서 160.3~236.9kg N/ha이었으며, 이 때 얻어지는 경제적 한계수량은 각각 9.6~10.0ton/ha, 12.4~12.6 ton/ha 및 9.0~9.7 ton/ha의 범위를 나타내었다. 8. 액상구비의 시용수준에 따른 액상구비의 한계시용수준은 3회 예취구에서 420.0kg/ha, 4회 예취구에서 440.6kg/ha, 그리고 5회 예취구에서 666.3kg/ha로 추정되었다.례, 오버레이 용접기술 및 용접재료의 개발현황 둥을 중심으로 살펴봄으로서 아직 국내에서는 널리 알려지지 않은 본 기 술의 활용을 넓이고자 한다. within minimum time from beginning of the shutdown.및 12.36%, $101{\sim}200$일의 경우 12.78% 및 12.44%, 201일 이상의 경우 13.

가축 분 퇴비화과정의 온도에 따른 잡초종자 발아억제 효과 (The Effect of Temperature of Compost on the Germination Inhibition of Weed Seeds during Livestock Manure Composting)

  • 본 연구는 퇴비단 내에서 소리쟁이 종자와 피 종자의 발아력 억제효과를 분석하기 위하여 염소 분 퇴비, 젖소 분 퇴비 그리고 한우 분 퇴비를 대상으로 하여 실험을 수행하였다. 소리쟁이 종자와 피 종자를 거즈에 싸서 퇴비단 내에 묻어놓고 1.5일, 3일, 10일, 20일 경과 후 종자를 회수한 뒤 발아력을 측정하였다. 실험 결과 소리쟁이 종자와 피 종자의 발아율은 가축 분의 종류와 퇴비화 방법에 관계없이 퇴비단의 온도와 직접적인 상관관계를 가지는 것으로 나타났다. 온도가 $60{\sim}70^{\circ}C$인 조건의 퇴비단에서 1.5일 동안 머무른 소리쟁이 종자와 피 종자는 발아력을 완전히 상실하였다. 반면에 퇴비단 최고 온도가 $51^{\circ}C$ 이하인 퇴비단에서는 소리쟁이 종자와 피 종자가 퇴비단 내에서 20일간 머무른 후에도 10 % 정도의 발아력을 유지하였다. 이상의 결과를 종합해보면, 소리쟁이 종자와 피 종자의 발아율을 억제하기 위해서는 가축 분 퇴비화단계에서 최소 $55^{\circ}C$ 이상의 온도를 3일 이상 유지하는 것이 좋다. 또한 퇴비단 온도가 $60{\sim}70^{\circ}C$ 정도 상승한다면 약 1.5일 정도 체류하는 것으로도 소리쟁이 종자와 피 종자의 발아력을 억제할 수 있을 것이다. 퇴비화 방법을 기준으로 보면, 호기적 퇴비화 방법이 퇴비단의 온도상승기간이 더 짧았다. 따라서 호기적 퇴비화방법을 적용하는 것이 소리쟁이 종자와 피 종자의 발아력을 억제하는데 더 효율적일 것이다.

  • This study was carried out to investigate the chemical change of rutin and fatty acids of buckwheat during germination. The content of rutin in buckwheat before germination was 31.5mg% and that after 7 days was increased to 1, 660.3mg%. The content of stearic acid was increased slightly after 3 day during germination but those of other fatty acids were decreased gradually.

Effects of PEG Priming Treatment on Germination and Seedling Growth of Onion Seed(Allium cepa L.)

  • These experiments were conducted to evaluate the variability of seed germination and seedling growth with different levels of polyethylene glycol(PEG 6,000) solution in onion seed. Average germination percentage of seed primed in PEG solution with 1.00 and 0.75 MPa was higher than control, and that of seed primed in 1.50MPa was lower than unprimed control. Germination percentage(GP) of seed primed for 5 days was highest, and as the primed days become long, the GP was decreased. The GP of airation seed during the primed was higher than that of unairation seed, about 5% , respectively. The GP of washed seed after primed was higher than that of unwashed seed, but that of redried seed after primed was lower than that of the others. The highest GP cultivar was Chunjoogoohyung and the lowest GP cultivar was Seouldego in unredried seed after primed, but Chunjoojoonggo was highest and Jungpoonwhang was the lowest cultivar in redried seed after primed. As the PEG concentration increased, the seedling length(SL) was shortened, and seed primed for 15 days was longer than other treatments. The SL of primed seed was similar to GP. The SL of washed seed after primed was longer than that of others, but that of redried seed after primed was shortest among the others. The SL of Chunjoojoonggo and Nongwoodego was longest and Seouldego was shortest among the cultivars in unredried seed after primed, but that of Chunjoogoohyung and Chunjoojoonggo was longest and Seouldego was the shortest cultivars in redried seed after primed.
