전신성 캔디다증에 대한 산사자 추출물과 Fluconazole의 병용요법에 의한 항진균 상승효과 (Synergistic Antifungal Activity against Disseminated Candidiasis by Combination Therapy of Crataegi Fructus Extract and Fluconazole)

  • In recent, there are increasing reports about pharmacological activities of Crataegi Fructus which has been used for many centuries as medicinal and food sources in East Asia. However, its antifungal efficacy needs to be investigated. Thus, in the current study, we determined synergistic antifungal activity of the Crataegi Fructus extract (CFE) when combined with fluconazole (FLC) against disseminated candidiasis due to Candida albicans. This fungus is one of the most problematic fungal pathogens. Data resulting from a microdilution susceptibility test showed that CFE had a dose-dependent antifungal activity. When the extract was combined with FLC, the activity was synergistic. For example, the antifungal activity by the combination of CFE at $20{\mu}g/ml$ plus FLC at $0.1{\mu}g/ml$ was 4 times more effective than antifungal activity by FLC alone at the same concentration (P<0.05). In the murine model of disseminated candidiasis, the combination therapy potentiated resistance of mice, resulting in 80% of C. albicans-infected animals surviving the entire period of 40 days observation, whereas mice given CFE alone or FLC alone all died with 17 and 23 days, respectively, although they survived longer than the untreated control animals (P<0.05). The CFE's antifungal activity seemed to be related to the blockage of hyphal production of C. albicans yeast cells. In summary, CFE has a synergistic antifungal activity, which can be produced by combining CFE with FLC. Thus, our data strongly indicate that CFE may be a potential candidate as an antifungal agent for combination therapy.

  • The growth in the number of single-member households is a critical issue worldwide, especially among the elderly. For those living alone, who may be unaware of their health status or routines that could improve their health, a continuous healthcare monitoring system could provide valuable feedback. Assessing the performance adequacy of activities of daily living (ADL) can serve as a measure of an individual's health status; previous research has focused on determining a person's daily activities and extracting the most frequently performed behavioral patterns using camera recordings or wearable sensing techniques. However, existing methods used to extract common patterns of an occupant's activities in the home fail to address the spatio-temporal dimensions of human activities simultaneously. Though multiple sequence alignment (MSA) offers some advantages - such as inherent containment of the spatio-temporal data in sequence format, and rapid identification of hidden patterns - MSA has rarely been used to extract in-home ADL routines. This research proposes a method to extract a household occupant's ADL routines from a cumulative spatio-temporal data log of occupancy collected using a non-intrusive method (i.e., a tomographic motion detection system). The findings from an occupant's 28-day spatio-temporal activity log demonstrate the capacity of the proposed approach to identify routine patterns of an occupant's daily activities and to reveal the order, duration, and frequency of routine activities. Routine ADL patterns identified from the proposed approach are expected to provide a basis for detecting/evaluating abrupt or gradual changes of an occupant's ADL patterns that result from a physical or mental disorder, and can offer valuable information for home automation applications by enabling the prediction of ADL patterns.

Optimization of the whole extract of Zarawand Mudaharaj (Aristolochia rotunda L.) root by Response Surface Methodology (RSM)

  • The chemical constitution of a drug has been accepted as an important basis for pharmacological action in Unani medicine. Various dosage forms have been developed on this concept, such as decoctions (Joshanda), infusions (Khesanda), extract (Rub / Usara), and syrup. Zarawand Mudaharaj (ZM.) / Aristolochia rotunda L. root was subjected to extraction process using Soxhlet's apparatus by using Response Surface Methodology (RSM) to design the number of random runs of the extracts with variation in the factors of temperature, the concentration of ethanol in water, time for extraction, for optimizing and maximizing the yield concentration. The data obtained, was analyzed with regression equation and ANOVA two-way summary to interpret the interaction of the factors for yield maximization. Minitab version 18 was used to design and analyze data. Validation of the optimum conditions for maximum yield of the whole extract of ZM. Root was carried out by re-run of the extract using the optimized conditions. The maximum yield percentage thus obtained using RSM was 20.87% whereas using these optimum conditions 21.35 % yield was obtained thereby validating the method. The association between the response functions and the process variables was identified by a three-factor recorded Box-Behnken design. In the present study RSM is used because itis a cheap and affordable method to optimize maximum yield percentage which may be reliably used by researchers. The study set in the surface conditions for ZM. root extraction by the Soxhlet apparatus for maximizing the yield percentage.

목표 구조물에 대한 점군데이터의 무손실 압축 기법에 관한 연구 (A Study on a Lossless Compression Scheme for Cloud Point Data of the Target Construction)

  • 본 논문에서는 지상라이다를 사용해 구조물을 측정한 점군데이터가 갖는 중복성을 피하고, 목표 구조물외에 불필요한 정보의 수를 감소시키도록 하는 점군데이터의 무손실 압축 기법을 제안한다. 제안된 방법을 적용하기 위해, 호프 변환을 이용하여 구조물과 지상라이다의 수평방향 사이의 각도를 찾아, 이를 점군데이터의 회전 변환에 적용하였다. 이로써 x축에 평행하도록 구성된 점군데이터에 대한 y좌표의 중복성은 기존의 데이터보다 많아지고, 따라서 압축률도 향상시킬 수 있다. 추가로, 불필요한 데이터를 찾아 정보량을 감소시키는 방법을 적용한다. 하나는 점군데이터를 데시메이션하는 것이고, 다른 하나는 목표 구조물이 갖는 y좌표의 범위를 찾아 목표로 하는 범위내 점군데이터만 추출하는 것이다. 제안한 방법은 실험을 통해 압축률이 향상되었음을 확인할 수 있다. 또한, 별도의 추가 정보 없이 점군데이터의 위치 정보만으로 데이터를 압축할 수 있고, 이 압축알고리듬으로 처리속도를 높일 수 있다.

  • 최근 다양한 정보검색 기법의 발달로 인해 사용자는 인터넷으로부터 유용한 데이터를 쉽게 획득할 수 있다. 이러한 데이터는 스트림 데이터와 유사하게 연속적으로 발생하기 때문에 인터넷에서 발생하는 대량의 데이터 중에서 사용자의 요구에 맞는 데이터만을 추출하기 위한 기법이 필요하다. 전통적인 데이터 추출 방법에서는 데이터를 먼저 데이터베이스에 저장한 후에 사용자의 요구를 질의로 변환하여 처리하였다. 그러나 이러한 방법은 연속적으로 발생하는 대용량의 데이터에 대하여 사용자 질의를 처리하는 경우에 비효율적인 문제가 있다. 이 논문에서는 인터넷 환경에서 대용량의 연속적인 데이터 스트림으로부터 사용자의 요구에 맞는 데이터를 추출하는 질의처리 기법을 제안한다. 먼저 인터넷에서 발생하는 데이터의 변화를 이벤트로 표현하고, 사용자의 요구사항을 프로파일로 정의한 이벤트-프로파일 모델을 제안한다. 그리고 이 모델을 기반으로 이벤트와 프로파일을 효율적으로 처리하기 위한 여과방법을 제안한다.

인삼 복용 전후 체표온도의 변화에 관한 임상적 연구 (A Clinical Study on the Effect of Ginseng to Control Superficial Body Temperature)

  • In Oriental medicine, Ginseng had been used for many diseases widely. It's main effects are suppling Qi and Promoting metastasis. The objective of this study is to examine the effect of Ginseng on control of body temperature. We conducted this study with 24 volunteers who had no problem in their health from November 2002 to January 2003. The body temperature were measured by DITI and thermometer before taking Ginseng and after 30minutes of the taking Ginseng extract 15g. We observed the difference of temperature among face chest and upper abdomen. All data were coded for computer analysis and significance were tested by Paired t-test and independent-t-test. According to our study, the axillary temperature show no signifiant change between two groups. The difference of temperature$({\Delta}T)$ between face and upper abdomen significantly increase after taking Ginseng extract(p<0.01). The difference of temperature$({\Delta}T)$ between face and chest significantly increase after taking Ginseng extract(p<0.05).

빌레나무 추출물의 항산화, α-글루코시다아제 억제 및 항미생물 활성 (Antioxidant, α-Glucosidase Inhibitory and Antimicrobial Activities of Extracts from Maesa japonica (Thunb.))

  • Maesa japonica (Thunb.) is an evergreen shrub belonged to the Myrsinaceae family, which was discovered in 2006 in South Korea. And, its biological functions have not been well studied. In this study, we determined the antioxidant activities, ${\alpha}$-glucosidase inhibitory effects and antimicrobial activities of methanol extract and the solvent fractions of M. japonica leaves and twigs. The highest antioxidant activity obtained by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging assay and reducing power assay was found in the ethylacetate fraction of twigs methanol extract, which contained the highest level of total phenolic compounds compared to the other fractions. In addition, ethylacetate fraction of twigs extract exhibited higher inhibitory activities against ${\alpha}$-glucosidase ($IC_{50}=0.8{\mu}g/m{\ell}$) compared to the $IC_{50}$ of the buthanol fraction ($IC_{50}=16{\mu}g/m{\ell}$) of leaves extract. It showed antimicrobial activities against Bacillus atrophaeus and Bacillus subtilis subsp. Spizizenii. Although the data is too limited, the current study is the first report on biological functions of M. japonica.

WHW$^{(R)}$의 랫드에서의 반복경구투여 독성에 관한 연구 (Thirteen-week repeated-dose oral toxicity study of the Modified Wenpitang-Hab-Wulingsan (WHW$^{(R)}$) in Sprague-Dawley rats)

  • Objectives : We investigated the repeated-dose toxicity of Wenpitang-Hab-Wulingsan(WHW), a Korean traditional medicine prescribed with twelve herbs, which has been used for the treatment of renal disease. Methods : WHW extract prepared by GLP company. WHW was supplemented by gavage at 0, 100, 500 and 1000 mg/kg/day for 13-week consecutive days. We recorded the clinical signs of toxicity, body weight, organ weights, hematology, gross and histological changes in target organs rats and clinical chemistry analysis for all rats. Results : WHW extract at all doses was shown no mortality or abnormal clinical signs in rats during at the observation period. Furthermore, there was no difference in body weight and food-take consumption, organ weight, gross pathological findings, and urine analysis among the groups of rats treated with different doses of WHW extract. The hematological analysis and clinical blood chemistry data were revealed no toxic effects from WHW-treated rats. Conclusions : The results suggest that WHW extract in rats is a wide margin of safety on a acute toxicity.

Protective actions of Rubus coreanus ethanol extract on collagenous extracellular matrix in ultraviolet-B irradiation-induced human dermal fibroblasts

  • Solar ultraviolet (UV) irradiation leads to distinct changes in the skin connective tissues by degradation of collagen, which is a major structural component in the extracellular matrix. UV irradiation induces the production of matrix metalloproteinases (MMP) capable of attacking native fibrillar collagen and responsible for inhibiting the construction of collagenous extracellular matrix. In this study, we attempted to investigate the protective actions of Rubus coreanus ethanol extract (RCE) on the MMP production and the consequent procollagen/collagen degradation in UV-B-irradiated human dermal fibroblasts. The analytical data showed that Rubus coreanus ethanol extract was mostly comprised of cyanidin 3-rutinoside. Pre-treatment of fibroblasts with this extract inhibited UV-B-induced production of MMP-1, MMP-8 and MMP-13 in dose-dependent manners. In addition, Western blot analysis and immunocytochemical staining assay revealed that RCE markedly augmented the cellular levels of procollagen/collagen declined in UV-B-exposed dermal fibroblasts. These results demonstrate that RCE blocks UV-B-induced increase of the collagen degradation by inhibiting MMP production. Thus, RCE may act as an agent inhibiting excessive dermal collagen degradation leading to the skin photoaging.

국내외산 녹차 추출액의 항균 작용 (Antibacterial Activities of Korean and Foreign Green Tea Extract)

  • This study was carried out to investigate antibacterial activities of the green tea extract according to storage temperature as death velocity constant. The survey data were collected from seven green teas (2 Korean, 2 Japanese, 2 Chinease and 1 Vietnamese) and experimental strains were used E. coli No. 11234, Ent. aerogenes No. 11783, Ps. fiu. orescens No. 11210, S. marscens No. 11808, St. mutans No. 11823 and V. parahaernolyticus No. 11965. The experimental results obtained were as follows: 1. Antibacterial activities of the green tea extracts according to the strains were strikingly difference. That is, those of the samples appeared to be conspicuous on [St. mutans No. 11823 (Range of death velocity constant a day after, at 20$\circ$C 4.85~5.22) and V. parahaemolyticus No. 11965 (4.70~6.00) while E. coli No. 11234 (0.59~-0.30) and Ent. aerogenes No. 11783 (0.24~-0.11)] were not so. 2. To storage temperature at 5$\circ$C than that at 20$\circ$C, antibacterial activities of the green tea extract on Ps. fluorescens No. 11210, S. marscens No. 11808, E. coli No. 11234, and Ent. aerogenes No. 11783 were sensitived but St. mutans No. 11823 and V. parahaemolyticus No. 11965 were not related to those. 3. Antibacterial activities to the strains were increased within two days.
