Hepatoprotective effects of the aqueous extract from Taraxacum officinale (Dandelion) against Thioacetamide-induced hepatotoxicity in rats (Thioacetamide로 유발된 간 손상에 대한 Taraxacum officinale (Dandelion) 추출물의 효과)

  • The genus Taraxacum, known by the common name Dandelion, is a medicinal herb in the family Asteraceae. It has been traditionally used as a folk medicine for the treatment or prevention of various diseases due to its anti-inflammatory and anti-oxidative properties. In this study, we attempted to evaluate protective effects of Dandelion related with anti-oxidative activity to Thioacetamide (TAA)-induced liver damage. 36 rats were randomly assigned to six experimental groups : Control, Dandelion water extract (DWE), TAA, TAA&DWE 300, TAA&DWE 600, TAA&DWE 1,200 groups. Rats in DWE and TAA&DWE groups were pretreated with DWE (300, 600 or 1,200 mg/kg BW) by gavage for 7 days. All rats were treated intraperitoneally with TAA (200 mg/kg BW) or normal saline at 12 hours after last oral administration and sacrificed at 12 hours after last treatment. Levels of WBC and Neutrophil count were significantly decresed in TAA&DWE 1,200 group compared with that in TAA group (P<0.05). In TAA&DWE 600 and TAA&DWE 1,200 groups, serum AST, ALT, GGT levels were lower than TAA group (P<0.05). The serum TG level was significantly elevated in TAA&DWE groups compared with those in TAA group. Liver tissues from TAA group showed extensive histopathological changes, characterized by moderate or severe hepatocytes degeneration, inflammatory cell infiltration, and congestion. In the TAA&DWE group, The severity of histopathological lesions were decreased compared to those in the TAA group. The MDA concentration was significantly decreased and GSH content was significantly increased in the TAA&DWE 1,200 group compared to those in the TAA group. GR, CAT and GST activities in the TAA&DWE 1,200 group were significantly increased compared to those in the TAA group.

  • We investigated the physicochemical and antioxidant activity characteristics of Yakju with different ratios (0, 5, 10, and 15%) of dandelion (Taraxacum officinale) root powder in order to find out potential as a functional fermented rice alcohol. Yakju samples were measured to obtain total flavonoid content, total polyphenol content, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, and reducing power. Total flavonoid and polyphenol contents increased with increasing amounts of dandelion root. The DPPH radical scavenging activity increased with the addition of dandelion root. Also, the reducing power was higher in Yakju with dandelion root compared with controls such as gallic acid, butylated hydroxytoluene, and ${\alpha}$-tocopherol. It is concluded that the Yakju fermented with additional dandelion root is functionally superior to the rice alcohol without dandelion root.

  • This study was conducted to investigate the effects of dandelion leaf (Taraxacum officinale) extracts on hepatic antioxidative system in high cholesterol-fed rats. Four groups of rats were given high cholesterol diets containing 10 g cholesterol/kg and 2.5 g sodium cholate/kg for 6 weeks. The control group received a diet without dandelion leaf extract and the other three groups received dandelion leaf extracts, ie, water, ethyl acetate and ether extracts, respectively. There were no significant difference in cytochrome P-450 contents among four groups. Hepatic xanthine oxidase activity was significantly lower in water extract group than the other three groups. Superoxide dismutase activity was significantly lower in three dandelion leaf extract groups, but catalase activity was significantly higher in three dandelion leaf extract groups than control group. Glutathione peroxidase and glutathione S-transferase activities were significantly increased in water extract group than control group. Lipid peroxide content was decreased in water extract group than control group.

  • Dandelion(Taraxacum officinale), an oriental herbal medicine, has been shown to favorably affect choleretic, antioxidative and protection of gastric mucosa. The protective effects of common dandelion leaf and root extract were investigated by irradiation in Sprague-Dawley rats. Male SD rats, 6weeks, were orally injected with dandelion extract(100mg/kg) for 10days. Immediately after final injection, rats were whole body irradiated with 10Gy used irradiation facility(Elekta Linac, Sweden). At 24h-15days after irradiation, complete blood counted test and apoptosis in jejunal crypt cell. Stimulated recovery by the extract was observed in platelet but was not showed in the erythrocyte and leucocyte. The jejunal crypt cells were protected significantly(p<0.05) and the radiation-induced apoptosis was reduced(p<0.05). The survival rate were carried for 15days, the survival ratio was 15% and 85% for the control and experimental group. Observations intestinal inhibition of cell death, intestinal mucosa and tissue decreased systemic inflammation and vacuole. Base on these data, we propose that dandelion may be a useful radioprotector, especially since it is a relatively nontoxic natural product.

  • Dandelion (Taraxacum officinale) is a member of family Asteraceae that grows all over the Korea. Recently, dandelion was cultivated for medicinal crops because of its positive medicinal effects. However, dandelion is considered as a troublesome weed in grass lawns of golf course and orchards. This study was conducted to investigate the effect of plant growth regulators [gibberellins ($GA_3$); kinetin; salicylic acid (SA); ethephon)] with different concentration on seed germination control of dandelion. Seed germination rates were increased in all concentration of $GA_3$ and kinetin treatment compared to control. In the 0.5 mM of ethephon application, seed germination rate was more increased than that of control while seed germination rate was reduced in 1.0 and 1.5 mM of ethephon treatments. Seed germination rate was significantly decreased with different SA dilutions compared to control. The germination rate was more reduced when SA was applied in combination with $GA_3$ than only SA treatments.

The Chemical Composition of Taraxacum officinale Consumed in Korea (한국산 서양민들레(Taraxacum officinale)의 화학적 조성)

  • For centuries, dandelion(Taraxacum officinale) is widely used as a food and a medicinal herb. However, there is inadequate documented information on the chemical composition of T. officinale consumed in korea. This study was attempted to analyze proximate component, amino acid, free sugar, mineral and fatty acid composition in T. officinale to establish the value as an useful food material. Moisture, ash, crude protein and fat contents in leaf are 7.85, 11.35 21.6 and 5.12%, respectively. Moisture and ash contents of root were 7.73 and 4.82%. Crude protein and fat contents of root were 11.8 and 1.73%, respectively. The major free amino acid was asparagine, arginine, serine and proline etc. The total contents of free amino acid in leaf and root were 1476.98 and 2710.06 mg%, respectively. The major free sugar in T. officinale was glucose, fructose, sucrose and maltose. Sucrose was also detected in a large amount. The major fatty acid of T. officinale are palmitic acid, stearic acid, oleic acid, linoeic acid and linolenic acid. The fatty acid compositions of leaf and root were different each other. Linolenic acid(63.6%) was the major fatty acid of the leaf and linoleic acid(56.4%) was the major fatty acid of the root. Most of mineral contents were high in leaf and root. Especially K content was higher in leaf than root.

Quality Characteristics of Sulgidduk Containing Different Levels of Dandelion (Taraxacum officinale) Leaves and Roots Powder (민들레 잎과 뿌리 분말을 첨가한 설기떡의 품질 특성)

  • In order to develop a dandelion (Taraxacum officinale) with natural food applications, the effects of dandelion leaves and roots powder content on the physical, textural and sensory properties of sulggidduk were examined. Dandelion powders of leaves and roots at 0, 1, 2 and $3\%$ were added to the dandelion sulgidduk. In the dandelion roots powder in sulgidduk, the color intensity of L values and the values and moisture content decreased however, the sensory color intensity, dandelion flavor and overall acceptability increased with increasing the dandelion powder content. The sensory overall acceptability indicated the dandelion roots powder content at 3% had the highest overall and flavor scores. As the dandelion leaves content was increased, the moisture contents, lightness, adhesiveness and cohesiveness of sulggidduk decreased, while the sensory color intensity, sensory hardness, hardness of texture properties and dandelion flavor increased. The addition at $1\%$ of the dandelion leaves powder content with the dandelion sulggidduk and the addition at $3\%$ of the dandelion roots powder content with the dandelion sulggidduk showed the highest overall acceptability.

Biological Activity of Dandelion (Taraxacum officinale) extracts Fermented with Raw Sugar (당 첨가 민들레(Taraxacum officinale) 발효물의 생리활성)

  • Dandelions were fermented for 120 days at $15-20^{\circ}C$ in batches containing thirty and fifty percent raw sugar (FD30 and FD50). The total phenolic concentration of FD30 and FD50 were about $78.9{\pm}2.17$ and $59.35{\pm}2.56$ mg/g, respectively, both being higher than the 2$54.{\pm}1.49$ mg/g determined for DWE (dandelion water extract). The DPPH radical scavenging activity of FD30 and FD50 showed $IC_{50}$ values of $118.7{\pm}2.03$ and $123.40{\pm}2.15{\mu}g/mL$, respectively, and FD30 displayed the highest antioxidant activity. 2 mg/mL of FD30 and FD50 showed $8.8{\pm}1.72$ and $11.8{\pm}2.87{\mu}M$ production of NO, respectively, compared with $4.9{\pm}1.20{\mu}M$ of the dandelion extract. The protease, ${\alpha}$-amylase, and lipase activity of FD50 was the highest. The fibrinolytic activity of FD30 and FD50 were $0.56{\pm}0.28$ and $1.39{\pm}0.20$ unit/mg protein, respectively, which was substantially higher than the 0.28 unit/mg protein of DWE. In conclusion, the dandelions fermented by sugar showed improved antioxidant, anti-inflammatory and enzymatic activities.

Quality Characteristics of the Bread Added Dandelion Leaf Powder (민들레 잎분말 첨가에 따른 기능성 식빵의 품질특성)

  • Effects of adding of dandelion flour on the quality characteristics of bread were investigated. Dandelion flour was substituted at levels of 0, 0.5, 1.0, 2.0% to wheat flour for bread making, respectively. Quality characteristics of bread such as dough yield, dough microstructure, loaf volume, bread yield, crumb color, mechanical property and sensory evaluation were analyzed. Addition of dandelion flour to wheat flour increased dough yield, loaf volume and bread yield. And, addition of dandelion flour caused a decrease in the lightness and an increase in the redness. The results of texture evaluation revealed that hardness, chewiness, springiness of bread increased as the level of dandelion flour was increased. As the addition level of dandelion flour increased, flavor balance, bitterness, aftertaste, grassy odor of bread increased but overall acceptability, moistness decreased. In conclusion, bread with 0.5% leaf powder was the best quality in bread properties.

Genetic Diversity of the Original Plant for Taraxaci Herba, Taraxacum spp. by the Analysis of AFLP (AFLP 분석을 통한 포공영 기원식물 민들레의 유전 다양성 분석)

  • Collected germplasms of five representative dandelion species (Taraxacum ohwianum, T. platycarpum, T. platypecidum, T. officinale, and T. coreanum) were 104 lines from different habitates in Korea and China. Their genetic diversity was analyzed by genomic fingerprinting method using amplified fragment length polymorphism (AFLP). AFLP results of 6 primer combinations were revealed 1,176 total DNA fragments and 523 polymorphic bands with a 44.4% ratio of polymorphism. On the basis of similarity coefficient analysis by unweight pair group method with arithmetic averages (UPGMA), 104 dandelion germplasm lines were ranged from 0.64 to 0.99 and clustered distinct five group depending on the species. Furthermore, a principal coordinate analysis (PCA) by the application of multi-variate analysis indicated significantly greater differences among species than geographical origins.
