  • A composite material was produced as an artificial bone substitute which is gradually degrAded and replaced by the regenerated natural bones after implantation. To detect the effect of the material on the cell's activity, the composite specimens were placed in MEMs and incubated at $37^{\circ}C$ for one week. Human uterus cervical cancer origin HeLa 3 cells and mouse subcutaneous origin L929 cells were cul- tured in the specimen dissolved MEMs for 5 days to investigate cytotoxicity via cell growth rates. ${Na_2}^{51}CrO_4$ solution was added to the media, to label the HeLa 53 cells, and the released amount of $^{51}Cr$ was measured by a $\gamma$-counter. On the cell growth investigation, no significant cytotoxic phenomena were revealed in both HeLa S3 and L929 cell cultures. On the released 51CR from the incubated HeLa 53 cells, no significant cell degeneration was observed from the composite embedded MEMs.

In vitro Evaluation of Cytotoxic Activities of Essential Oil from Moringa oleifera Seeds on HeLa, HepG2, MCF-7, CACO-2 and L929 Cell Lines

  • Moringa oleifera Lam. (Moringaceae) is widely consumed in tropical and subtropical regions for their valuable nutritional and medicinal characteristics. Recently, extensive research has been conducted on leaf extracts of M. oleifera to evaluate their potential cytotoxic effects. However, with the exception of antimicrobial and antioxidant activities, little information is present on the cytotoxic activity of the essential oil obtained from M. oleifera seeds. Therefore, the present investigation was designed to investigate the potential cytotoxic activity of seed essential oil obtained from M. oleifera on HeLa, HepG2, MCF-7, CACO-2 and L929 cell lines. The different cell lines were subjected to increasing oil concentrations ranging from 0.15 to 1 mg/mL for 24h, and the cytotoxicity was assessed using MTT assay. All treated cell lines showed a significant reduction in cell viability in response to the increasing oil concentration. Moreover, the reduction depended on the cell line as well as the oil concentration applied. Additionally, HeLa cells were the most affected cells followed by HepG2, MCF-7, L929 and CACO-2, where the percentages of cell toxicity recorded were 76.1, 65.1, 59.5, 57.0 and 49.7%, respectively. Furthermore, the $IC_{50}$ values obtained for MCF-7, HeLa and HepG2 cells were 226.1, 422.8 and $751.9{\mu}g/mL$, respectively. Conclusively, the present investigation provides preliminary results which suggest that seed essential oil from M. oleifera has potent cytotoxic activities against cancer cell lines.

Cell differentiation and Anti-oxydative effect of Dioscoreae Rhizoma on HeLa Cell (산약(山藥)의 HeLa cell 분화에 미치는 영향과 항산화효과에 대한 연구)

  • Purpose: This study examined the Cell differentiation and the anti-oxidative effect of Dioscoreae Rhizoma on HeLa cells. We are interested in whether Dioscoreae Rhizoma is capable of causing apoptosis processes on HeLa cell, and whether cotreatment of NCS with Dioscoreae Rhizoma reduces cell viability. Methods: We used aqueous extract to treat HeLa cell with different concentrations treated with a water or a MeOH extract of Dioscoreae Rhizoma (0, x10, x20, x40, x80). The MTT (3, (4, 5-dimethyl-thiazol) 2, 5-diphenyl-tetraxolium bromide) reduction assay was employed to quantify the differences in cell activity and viability. Cells were stained with DAPI and visualized by fluorescent Microscope. The caspase-3, Bcl-2, PARP, p53 expression level was monitored using western-blotting techniques. The patterns of the changes in expression were scanned and analyzed. Results: The survival rate of cells treated with Dioscoreae Rhizoma extracts increased by 20% at specific concentration. The other side Dioscoreae Rhizoma extracts induced apoptotic features including chromatin condensation and fragmentation. And Dioscoreae Rhizoma extracts increased the expression of caspase-3, p53 and the cleavage of PARP protein. However, co-treatment with Dioscoreae Rhizoma with NCS attenuated the activations of p53 and PARP protein that were mediated by NCS treatment alone. This is indicated Dioscoreae batatas extracts attenuated cytotoxicity induced by oxidative agents including NCS. Conclusion: Our results suggest Dioscoreae Rhizoma extracts induce cell differentiation or apoptosis connected with concentration. Further elucidation of concentration of Dioscoreae Rhizoma awaits many other biochemical investigative studies.

  • TALP-32 is highly basic protein with a molecular weight of 32 kDa purified from human term placenta. Some basic proteins such as defensins and cecropins are known to induce cell death by increasing membrane permeability and some of them are under development as an anticancer drug especially targeting multi-drug resistant cancers. Therefore, we investigated cytotoxic effect and mechanism of TALP-32 When HeLa cell was incubated with TALP-32, cytotoxicity was increased in time and dose dependent manner. As time goes by, HeLa cells became round and plasma membrane was ruptured. Increase of plasma membrane permeability was determined with LDH release assay. Also in transmission electron microscopy, typical morphology of necrotic cell death, such as cell swelling and intracellular organelle disruption was observed, but DNA fragmentation and caspase activation was not. And necrotic cell death was determined with Annexin V/Pl staining. The cytotoxicity of TALP-32 was minimal and decreased or RBC and Hep3B respectively. These data suggests that TALP-32 induces necrosis on rapidly growing cells but not on slowly growing cells implicating the possibility of its development of anticancer peptide drug.

  • The combined effects of water-soluble fraction of Moschus (ME) and anti-tumor drug mitomycin C on the proliferation of human tumor cell-lines were estimated by MTT colorimetric assay. ME inhibited the proliferation of Hep G2, A540, HeLa, KHOS-NP and Balb/c 3T3 cells. Also, ME increased the cytotoxicity of mitomycin C on Hep G2, A549 and HeLa cells. In addition, ME enhanced the cell viability of murine splenocytes and human lymphocytes at the concentration of 100㎍/㎖. These results indicate that ME inhibits the proliferation of human tumor cells and increases the cytotoxicity of mitomycin C without cytoxicity on immune cells.

  • Cancer is one of the major health problems worldwide and its current treatments have a number of undesired adverse side effects. Natural compounds may reduce these. Currently, a few plant products are being used to treat cancer. In this study, goniothalamin, a natural occurring styryl-lactone extracted from Goniothalamus macrophyllus, was investigated for cytotoxic properties against cervical cancer (HeLa), breast carcinoma (MCF-7) and colon cancer (HT29) cells as well as normal mouse fibroblast (3T3) using MTT assay. Fluorescence microscopy showed that GTN is able to induce apoptosis in HeLa cells in a time dependent manner. Flow cytometry further revealed HeLa cells treated with GTN to be arrested in the S phase. Phosphatidyl serine properties present during apoptosis enable early detection of the apoptosis in the cells. Using annexin V/PI double staining it could be shown that GTN induces early apoptosis on HeLa cells after 24, 48 and 72 h. It could be concluded that goniothalamin showing a promising cytotoxicity effect against several cancer cell lines including cervical cancer cells (HeLa) with apoptosis as the mode of cell death induced on HeLa cells by Goniothalamin was.

  • The purpose of this study was to investigate effect of water extract of Carthami Flos on the proliferation of human cancer cell-lines. The effects of Carthami Flos on the proliferation of A431, HeLa, MOLT-4, K562 cells, Balb/c 3T3 cells, mouse thymocytes, splenocytes and human lymphocytes were estimated by MTT colorimetric assay. The results were as follows; 1. Carthami Flos did not effect A431, HeLa, MOLT-4, K562 cells. 2. The cytotoxicity of mitomycin C on K562 cells was increased by the combination of Carthami Flos. 3. Carthami Flos inhibited the proliferation of Balb/c 3T3 cells. 4. Carthami Flos stimulated the proliferation of thymocytes. 5. Carthami Flos stimulated the proliferation of splenocytes. 6. Carthami Flos stimulated the proliferation of human lymphocytes.

Curcumin-induced Growth Inhibitory Effects on HeLa Cells Altered by Antioxidant Modulators

  • Curcumin (diferuloyl methane), originated rhizomes of Curcuma longa L. has been suggested as an anti-inflammatory and anti-carcinogenic agent. In the present study, modulation of cytotoxic effects of curcumin on HeLa cells by different types of antioxidants was investigated. Cytotoxic effects of curcumin were significantly enhanced in the presence of superoxide dismutase (SOD) by decreasing $IC_{50}$ to 15.4 from $26.0\;{\mu}M$ after 24 hr incubation; the activity was not altered by catalase. The effect of curcumin was significantly less pronounced in the presence of 4 mM N-acetylcysteine (NAC). Low concentration (<1 mM) of NAC, however, increased the efficacy of curcumin. Cysteine and ${\beta}$-mercaptoethanol that have a thiol group, showed the similar biphasic patterns as NAC for modulating curcumin cytotoxicity, which was, however, constantly enhanced by ascorbic acid, a non-thiol antioxidant. In the presence of SOD, ascorbic acid, and 0.5 mM NAC, cellular levels of curcumin were significantly increased by 31-66%, whereas 4 mM NAC decreased the level. The present results indicate that thiol reducing agents showed a biphasic effect in modulating cytotoxicity of curcumin; it is likely that their thiol group is reactive with curcumin especially at high concentrations.

Increasing Effects of Apoptosis When Co-treated Scutellaria barbata D. Don. with Anti-cancer Drugs (반지련(半枝蓮)과 항암제 병용 투여에 의한 암세포 성장 저해에 관한 연구)

  • Purpose: This experiment was designed to find out increasing effects of S. barbata. co-treatment with anti-cancer drugs at cancer cell's growth inhibition effect. Methods: Divergent observational study of the S. barbata. co-treatment with Cisplatin treatment on HeLa cell. Cell viability using MTT assay, Cell Culture and Cytotoxicity Studies, Cell Cycle Analysis, Annexin V-FITC/PI assay, Cell morphological assessment, PARP cleavage using Western blotting analysis when HeLa cell were co-treated with Cisplatin and Scutellaria Barbata extracts. Results: When HeLa cell were co-treated with Cisplatin and Scutellaria Barbata extracts, we found out viability of HeLa cell, changing in the distribution of cell cycle, Annexin V-FITC staining, DAPI staining, PARP clavage protein assay by Western-blot. So Scutellaria Barbata extracts have increased apoptosis Conclusion: When co-treated Scutellaria Barbata extracts with anti-cancer drugs, the anti-cancer effects were increased. We still not sure which constituent apoptosis at cancer cells and activates anti-cancer effects suppressing, but we believe that it'll be revealed here after with following experiments.

Antitumor Activity of $\alpha$-Lactalbumin on the Tumor Cells ($\alpha$-Lactalbumin의 암세포 증식 저해효과에 관한 연구)

  • Bovine serum albumin(BSA), $\alpha$-lactalbumin($\alpha$-LA), $\beta$-lactoglobulin($\beta$-LG) and bovine immunoglobulin G (IgG) were investigated the cytotoxicity on tumor cell lines. $\alpha$-LA was showned a tendency of dose dependaent on cytotoxicity using WiDr. The growth of WiDr was inhibited 82% by 1mg/ml of $\alpha$-LA. However, IgG, BSA and $\beta$-LG were not shown the cytotoxicity on WiDr. When $\alpha$-LA was purified by using high pressure liquid chromatography(HPLC), the main component($\alpha$-LA) was eluted at 33.057 min and extremely small quantities eluted at 32.310 min. The cytotoxicity of main component (eluted at 33.057 min peak) was lower than commercial $\alpha$-LA. And the cytotoxic activity of hydrolyzed $\alpha$-LA and $\alpha$-LA treated with EDTA were lower than commercial $\alpha$-LA on tumor cells.
