Synthesis and Biological Activities of the Alternating Copolymers Containing Cyclic Ether Rings along with Carboxyl or Hydroxyl Groups on Their Backbones

  • The alternating copolymers of dihydropyran(DP)-maleic anhydride(MA), dihydrofuran(DF)-MA and DF-vinylene carbonate(VC) were prepared by free radical copolymerization of DP or DF with MA or VC. The reactivity ratios for poly(DF-VC) were found to be less than unity (0.05, 0.04) and its alternating sequences were obtained by feeding an equimolar amount of the comonomers at the onset of copolymerizations. The copolymers were hydrolyzed to give poly(TP-CE), poly(TF-CE) and poly(TF-HE), whose cytotoxicities against normal and tumor cells (3LL, B16) were measured in vitro.

Scientific Analysis of Formulation Theory of Chungpesagan-tang; In vitro Cytotoxicity of Cisplatin Combined with Chungpesagan-tang

  • In vitro cytotoxic activities of cisplatin combined with Chungpesagan-tang or puerarin, which were treated with or without human intestinal bacteria, were measured. When cisplatin was combined with Chungpesagan-tang and its ingredient treated without intestinal bacteria, they did not affect the in vitro cytotoxicity of cisplatin against tumor cell lines. However, when cisplatin was combined with intestinal bacteria-treated Chungpesagan-tang and its ingredients, the cytotoxicities against SNU C4, L1210, A549 and P388 tumor cell lines were synergistically increased. Puerarin, which was isolated from Puerariae Radix, did not show in vitro cytotoxicity. However, its metabolite, daidzein, showed potent cytotoxicity against tumor cell lines and was synergistic by the combined usage of cisplatin. These results suggest that natural glycosides are not only prodrugs which can be transformed to active compounds by intestinal microflora, but the combined usage of cisplatin with natural components, such as daidzein, and herbal medicinal polyprescriptions, such as Chungpesagan-tang, may be a new method for prevention and minimization of the toxicity of cisplatin.

소청용탕 및 가미치효산이 평흡기 배장세포로부터의 뮤신 분비에 미치는 영향 (Effects of Socheongryong-tang and Kamichihyo-san on Mucin Secretion from Airway Goblet)

  • In the present study, the author intended to investigate whether two oriental medical prescriptions named socheongryong-tang(SCRT) and Kamichihyo-san(KCHS) significantly affect mucin release from cultured hamster tracheal surface epithelial(HTSE) cells. Confluent HTSE cells were metabolically radiolabeled with ³H-glucosamine for 24 hrs and chased for 30 min in the presence of SCRT or KCHS to assess the effect of each agent on ³H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Also, the effects of SCRT and KCHS on contractility of isolated tracheal smooth muscle were investigated. The results were as follows: (1) SCRT significantly inhibited mucin release from cultured HTSE cells, without cytotoxicity; (2) KCHS significantly increased mucin release without cytotoxicity; (3) SCRT and KCHS did not affect contractility of isolated tracheal smooth muscle. We suggest that the effects of SCRT and its components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have the possible inhibitory effects on mucin release from the viewpoint of management of hypersecretion of airway mucus.

Effects of Bambusae concretio Silicea on Suppression of Collagenolysis and Bone Resorption in Mouse Calvarial Osteoblasts

  • Objective : We studied the effect of Bambusae concretio Silicea (BCS) on bone metabolism. Methods : At first, we treated PTH, 1,25(OH)₂D₃, mononuclear cell conditioned medium (MCM) and IL-1 to osteoblast cells derived from mouse calvarial bone explants in vitro, and then investigated the activities of collagenolysis and bone resorption factors. Results : BCS extracts have no cytotoxicities in concentrations of 1-150 ㎍/ml. BCS had protective activity against PTH (5 units/ml), MCM (5%, v/v), 1,25(OH)₂D₃ (20 ng/ml), IL-1α(2 ng/ml) and IL-1β, (1 ng/ml)-induced collagenolysis in the mouse calvarial cells. And, pretreatment of BCS for 1 hr significantly reduced the collagenolysis. Furthermore, it was much more expressed at 16 hrs after BCS (50 ㎍/ml)-pretreatment. And, BCS significantly protected against enhanced collagenolysis induced by IL-1α and IL-1β. Conclusion : BCS extracts inhibited the bone resorption in mouse calvarial bone cell;, thus BCS could be used clinically for bone diseases.

  • Phytochemical investigation on dragon's blood of Dracaena cambodiana led to the discovery of two new flavonoid derivatives, cambodianin G (1) and cambodianin H (2). Their structures were elucidated on the basis of detailed spectroscopic analysis, including 1D and 2D NMR techniques and chemical methods. The two compounds were observed to exhibit antibacterial activities against Staphylococcus aureus, and compound 1 showed cytotoxicities against K562 and SGC-7901 cell lines.

Comparison of cytotoxicities and wound healing effects of diquafosol tetrasodium and hyaluronic acid on human corneal epithelial cells

  • This study aimed to compare the cellular toxicities of three clinically used dry eye treatments; 3% diquafosol tetrasodium and hyaluronic acid at 0.3 and 0.18%. A methyl thiazolyltetrazoiun (MTT)-based calorimetric assay was used to assess cellular proliferation and a lactate dehydrogenase (LDH) leakage assay to assess cytotoxicity, using Human corneal epithelial cells (HCECs) exposed to 3% diquafosol tetrasodium, 0.3% hyaluronic acid (HA), or 0.18% HA or 1, 6 or 24 h. Cellular morphology was evaluated by inverted phase-contrast light microscopy and electron microscopy, and wound widths were measured 24 h after confluent HCECs were scratched. Diquafosol had a significant, time-dependent, inhibitory effect on HCEC proliferation and cytotoxicity. HCECs treated with diquafosol detached more from the bottoms of dishes and damaged cells showed degenerative changes, such as, reduced numbers of microvilli, vacuole formation, and chromatin of the nuclear remnant condensed along the nuclear periphery. All significantly stimulated reepithelialization of HCECs scratched, which were less observed in diquafosol. Therefore, epithelial toxicity should be considered after long-term usage of diquafosol and in overdose cases, especially in dry eye patients with pre-existing punctated epithelial erosion.

Mucin 분비에 미치는 대청룡탕(大靑龍湯) 및 <석실비록(石室秘錄)> 급치법방(急治法方)에 대한 연구 (The Effect of Daecheongryong-tang and prescription A on airway mucin secretion)

  • Objectives : This study was done with intend to investigate whether two oriental medical prescriptions, daecheongryong-tang (DCRT) and prescription A (P-A) significantly affect mucin release from cultured hamster tracheal surface epithelial (HTSE) cells. Methods : Confluent HTSE cells were metabolically radiolabeled with $^3H$-glucosamine for 24 hrs and chased for 30 min in the presence of DCRT or P-A to assess the effect of each agent on $^3H$-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase (LDH) release. Also, the effects of DCRT and P-A on contractility of isolated tracheal smooth muscle were investigated. Results were as follows : 1. DCRT significantly inhibited mucin release from cultured HTSE cells, with significant cytotoxicity. 2. P-A significantly increased mucin release from cultured HTSE cells, with significant cytotoxicity. 3. DCRT inhibited Ach-induced contraction of isolated tracheal smooth muscle. 4. P-A also inhibited Ach-induced contraction of isolated tracheal smooth muscle. Conclusion: Results suggest that DCRT and P-A have regulating effects on mucin secretion from goblet cells. Further investigation is needed, because of the value in finding novel agents to this purpose, and these oriental medical prescriptions have potential for this role.

소 흉선 추출물의 암세포 증식 및 림프구에 미치는 영향 (Antitumor and Immunological Effects of Bovine Thymic Extract)

  • Thymic extract showed antitumor effect to sarcoma mice with higher dose$(200{\mu}g/mouse/day$ i.p., 4weeks) but not with low dose$(5{\mu}g/mouse/day$ i.p., 6 weeks). Direct cytotoxicities were exhibited against sarcoma 180, L1210 and MOLT-4 by MTT assay. The spleen weight of mice were increased but the number of circulating lymphocytes were not increased after long-term(2 weeks) administration of thymic extract. Evaluating the mitogenesis by MTT assay. $\%$ absorbance of human lymphocytes was not increased by thymic extract. Cell cycle statistics of S phase and $G_2/M$ phase was not increased in the presence of that by PI staining. The formation of rosette was induced, irrespectively of exposure time short-term(l hour) and long-term(2 weeks). The population of mouse blood T-cell to bind Lyt2-antimonoclonal antibody and to $L_2T_4$ were increased after administration of thymic extract$(2-200{\mu}g/mouse/day)$. From the above results, it is suggested that thymic extract exerts antitumor activity by stimulating T cells to differeniate in vivo but not in vitro.

수영의 항돌연변이 활성 및 세포독성 (Antimutagenic Activity and Cytotoxicity of the Whole Plant of Rumex acetosa)

  • The extract and fractions of Rumex acetosa (Polygonaceae), a Korean medicinal plant, were examined for their cytotoxicities against five cultured human tumor cell lines, i.e. A549 (non-small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCY15 (colon), using the SRB (sulforhodamine-B) method in vitro and antimutagenic activities by Ames test with Salmonella typhimurium TA98 and TA100 and SOS chromotest with E. coli PQ37. Among the tested samples, the methylene chloride fraction strongly inhibited the proliferation of each examined tumor cell line with $IC_{50}$ values ranged from 13.2 to $18.7\;{\mu}g/ml$ and showed potent antimutagenic activities with 96.1% and 96.7% at the concentration of 1 mg/plate against the mutagens, NPD and sodium azide, respectively. Its antigenotoxic activity was also very effective at the final concentration of $10\;{\mu}g/reaction$ tube against the mutagens, MNNG and NQO by SOS chromotest.

Also, the effects of CGGH and GRJS on contractility of isolated tracheal smooth muscle were investigated. Results : (1) CGGH and GRJS significantly increased mucin release from cultured HTSE cells, without cytotoxicity : (2) CGGH and GRJS did not affect contractility of isolated tracheal smooth muscle. Conclusions : These results suggest that the effects of CGGH and GRJS should be further investigated, and that it would be gainful to invesigate, from among oriental medical prescriptions, what novel agents have these mild expectorant effects on mucin secretion from airway goblet cells.
