Involvement of Heme Oxygenase-1 in Orexin-A-induced Angiogenesis in Vascular Endothelial Cells

  • The cytoprotective enzyme heme oxygenase-1 (HO-1) influences endothelial cell survival, proliferation, inflammatory response, and angiogenesis in response to various angiogenic stimuli. In this study, we investigate the involvement of HO-1 in the angiogenic activity of orexin-A. We showed that orexin-A stimulates expression and activity of HO-1 in human umbilical vein endothelial cells (HUVECs). Furthermore, we showed that inhibition of HO-1 by tin (Sn) protoporphryin-IX (SnPP) reduced orexin- A-induced angiogenesis in vivo and ex vivo. Orexin-A-stimulated endothelial tube formation and chemotactic activity were also blocked in SnPP-treated vascular endothelial cells. Orexin-A treatment increased the expression of nuclear factor erythroid-derived 2 related factor 2 (Nrf2), and antioxidant response element (ARE) luciferase activity, leading to induction of HO-1. Collectively, these findings indicate that HO-1 plays a role as an important mediator of orexin-A-induced angiogenesis, and provide new possibilities for therapeutic approaches in pathophysiological conditions associated with angiogenesis.

Potential in vitro Protective Effect of Quercetin, Catechin, Caffeic Acid and Phytic Acid against Ethanol-Induced Oxidative Stress in SK-Hep-1 Cells

  • Phytochemicals have been known to exhibit potent antioxidant activity. This study examined cytoprotective effects of phytochemicals including quercetin, catechin, caffeic acid, and phytic acid against oxidative damage in SK-Hep-1 cells induced by the oxidative and non-oxidative metabolism of ethanol. Exposure of the cells to excess ethanol resulted in a significant increase in cytotoxicity, reactive oxygen species (ROS) production, lipid hydroperoxide (LPO), and antioxidant enzyme activity. Excess ethanol also caused a reduction in mitochondrial membrane potential (MMP) and the quantity of reduced glutathione (GSH). Co-treatment of cells with ethanol and quercetin, catechin, caffeic acid and phytic acid significantly inhibited oxidative ethanol metabolism-induced cytotoxicity by blocking ROS production. When the cells were treated with ethanol after pretreatment of 4-methylpyrazole (4-MP), increased cytotoxicity, ROS production, antioxidant enzyme activity, and loss of MMP were observed. The addition of quercetin, catechin, caffeic acid and phytic acid to these cells showed suppression of non-oxidative ethanol metabolism-induced cytotoxicity, similar to oxidative ethanol metabolism. These results suggest that quercetin, catechin, caffeic acid and phytic acid have protective effects against ethanol metabolism-induced oxidative insult in SK-Hep-1 cells by blocking ROS production and elevating antioxidant potentials.

퀴노아의 품질특성 및 생리활성에 대한 로스팅 온도의 영향 (Effects of Roasting Temperature on Quality Characteristics and Biological Activity of Quinoa)

  • The present study aimed to evaluate the effects of roasting temperature on the quality characteristics and biological activity of quinoa. Quinoa was roasted at 160, 200, and 220℃ for 20 min. The lightness (L*) of quinoa decreased, however, the redness (a*) increased as the roasting temperature increased. The yellowness (b*) was the highest at 160℃ and decreased at 200 and 220℃. The highest contents of total polyphenol, flavonoid, and quercetin were observed at 220℃, the highest roasting temperature. The highest radical scavenging activities of 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (73.65%) and α,α-diphenyl-β-picrylhydrazyl free radicals (47.82%) were found in roasted quinoa at 220℃. The α-glucosidase activity was inhibited by 62.13% at this temperature. The roasted quinoa at 220℃ also showed a significant cytoprotective effect against oxidative stress in HepG2 cells. These results could be useful in the development of food products using quinoa.

울금에 의한 혈관내피세포 보호 효과에 대한 연구 (Cytoprotective Effects of Radix Curcumae Aromaticae in Human Umbilical Vein Endothelial Cells)

  • In order to validate the use of Radix Curcumae Aromaticae as an anti-inflammatory drug in the traditional Korean medicine, I have investigated the effect of water-soluble extract of Radix Curcumae Aromaticae (ECA) on the expression of inducible heme oxygenase-1 (HO-1), which ha.s anti-inflammatory and cytoprotective effects stimulates, in human umbilical vein endothelial cells (HUVECs) stimulated with a high dose of pro-inflammatory tumor necrosis factor-alpha (TNF-α). The extract protected dose-dependently HUVECs against TNF-α-induced apoptosis, as measured qualitatively by a nuclear staining method using the fluoresoence DAPI and quantitatively by a flow cytometry using fluoresce-enhanced Annexin V antibody, and significantly Increased HO-1 expression, as determined by Western blotting analysis using anti-HO-1 antibody. Biockage of HO-1 activity by a pharmacological inhibitor reversed cytoprotection afforded by the extract, and treatment with carbon monoxide, one of HO-1 metabolites, resulted in cytoprotection comparable to the extract. These results suggest that ECA may have therapeutic potential in the control of endothelial disorders caused by inflammatory cytokines.

제주 연안 갈조류 추출물의 신경세포 보호효과 (Cytoprotective Effects of Phaeophyta Extracts from the Coast of Jeju Island in HT-22 Mouse Neuronal Cells)

  • 항암, 항염증 및 비만억제 등의 생리활성을 지닌 해조류는 최근 건강기능 식품, 기능성 화장품 그리고 의약품 산업 분야에서 미용과 건강식품 소재로 각광받고 있다. 본 연구에서는 10종의 갈조류 메탄올 추출물을 이용하여 1,1-diphenyl-2-picrylhydrazyl(DPPH) 라디칼 소거능과 아밀로이드 베타 단백질($A{\beta}$)의 신경독성에 대한 HT-22 신경세포 보호효과를 조사함으로써 천연물로부터 안전하고 새로운 신경세포 보호소재를 개발해내고자 한다. DPPH 라디칼 소거능의 경우 미역쇠를 포함한 8종의 갈조류에서는 비교적 낮은 활성산소 소거능을 보인 반면, 감태와 패에서 강력한 활성산소 소거능이 나왔다. $A{\beta}$의 신경독성에 대해 10종의 갈조류 추출물이 갖는 HT-22 신경세포 보호효과를 검증하기 위해 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) 분석과 APP, BACE1, iNOS 단백질의 발현양상 및 ERK1/2, p38, JNK1/2 단백질의 활성화 양상을 분석했다. MTT 분석 결과, $A{\beta}$의 신경독성으로부터 미역쇠가 $25{\mu}g/mL$의 농도에서 가장 효과적으로 세포를 보호하고 있는 것으로 나타났고, 알쏭이모자반, 불레기말, 바위수염, 짝잎모자반도 세포 보호효과가 있는 것으로 나타났다. 세포 보호효과가 있는 것으로 밝혀진 5종의 갈조류를 가지고 수행한 단백질 발현분석 결과, 미역쇠는 $A{\beta}$의 신경독성에 의해 HT-22 신경세포에서 발현되는 단백질인 BACE1과 iNOS의 발현을 저해하였다. 이는 미역쇠의 세포보호효과가 $A{\beta}$의 신경독성으로부터 일어난 ERK와 p38의 활성화에 연관된 세포신호전달 경로를 억제하는 것으로 보인다. 그러므로 특히 식용 갈조류인 미역쇠는 $A{\beta}$에 의해 유도된 신경독성에 대해서 신경세포 보호효과를 갖는 건강기능 식품 소재로서의 가치가 충분한 것으로 사료된다.

Lactobacillus spp. 이용 발효 붉나무의 기능성물질 검색에 대한 연구 (Functionality Analysis of Rhus javanica Fermented by Lactobacillus spp.)

  • 본 연구에서는 한약재를 Lactobacillus속 균주로 발효시켜 기능성물질을 확인하고자 하였다. 선행연구를 통해 선별한 발효에 사용한 10가지 한약은 100% methanol로 추출하여 Lactobacillus속 균주와 MRS배지를 첨가하여 사용하였고 발효 후 ethyl acetate로 추출하여 사용하였다. 10가지 중 붉나무를 Lb. brevis KCTC 3498로 발효하고 ethyl acetate로 추출한 추출물에서 마우스 해마 유래 세포주인 HT22 세포에서 glutamate로 유발된 산화적 손상으로부터 세포 보호효과가 우수했으며, 이는 heme oxygenase-1 단백질 발현에 의한 것 임을 밝혔다. 또한, 뇌세포 보호에 heme oxygenase-1 발현의 주요 기전인 Nrf2 핵 내의 전사와 직접적인 연관이 있음을 확인하였다. 붉나무의 발효 전과 후의 항균, 항산화 효과 비교 실험에서는 붉나무를 Lb. plantarum subsp. plantarum KCTC 3108, Lb. fermentum KCTC 3112, Lb. brevis KCTC 3498 균주로 각각 발효한 한약재는 발효 전보다 항산화 활성이 높아지거나 비슷한 수준으로 나타났다. Lb. plantarum subsp. plantarum KCTC 3108, Lb. casei KCTC 3109로 발효한 붉나무 발효액과 ethyl acetate 여액층은 Bacillus subtilis PCI 219, Escherichia coli KCTC 1682, Shigella flexneri KCTC 2517, Vibrio parahaemolyticus KCTC 7471, Pseudomonas aeruginosa KCTC 2004에 모두 항균활성을 보였다. 붉나무를 Lb. brevis KCTC 3498로 발효하여 ethyl acetate로 추출한 추출물은 B. subtilis PCI 219, E. coli KCTC 1682, S. flexneri KCTC 2517, V. parahaemolyticus KCTC 7471 균주에서 발효한 추출물이 단순 붉나무 추출물보다 높은 항균활성을 보였다.

제주 수산가공부산물 유래 기능성 소재 탐색 (Development of Bioactive Substances from Fishery Processing by-products in Jeju)

  • In this study, we investigated the bioactive substances of the Alcalase hydrolysate obtained from fishery processing by-products in Jeju by measuring bioactivities including radical scavenging acitivty, cytoprotective activity against 2,2-azobis-(2-amidino-propane) dihydrochloride (AAPH), and ACE inhibitory activity. This study is important because of utilization of unused fishery processing by-products in Jeju. The Alcalase hydrolysate was prepared through the hot water extraction and enzymatic hydrolysis, and then further separation of the Alcalase hydrolysate was performed by ultrafiltration using 10 kDa molecular weight cut-off membrane. The Alcalase hydrolysate showed the relatively higher DPPH and peroxyl radical scavenging activity ($IC_{50}$ value; 1.30 mg/ml and 0.888 mg/ml, respectively). Also, the Alcalase hydrolysate showed the ACE inhibitory activity with 1.87 mg/ml of $IC_{50}$ value. These biological activities are increased over 1.2 or 2.5 times through the ultrafiltration of the Alcalase hydrolysate. Therefore, the Alcalase hydrolysate obtained from fishery processing by-products in Jeju and the different molecular weight fractions should be given consideration for food and cosmetics ingredient. Furthermore, this research on the utility of fishery processing by-products might be a useful tool into the industry.

Protective Effect of Stilbenes on Oxidative Damage

  • Oxidative stress induced by reactive oxygen species (ROS) has been suggested to be the cause of various degenerative diseases as well as aging. To evaluate the antioxidant potential of stilbenes, we have investigated the cytoprotective effect of 10 stilbenes derived from plants on the oxidative stress induced by tertiary butyl hydroperoxide (t-BuOOH). Of the stilbenes tested, piceatannol (3) showed the most potent activity, which was further investigated using an animal model. When 3 (30 or 10 mg/kg) was topically administered prior to UVB irradiation, the amount of the thiobarbituric acid reactive substances (TBARS) was significantly reduced compared to that of the control (vehicle). Our findings suggest that piceatannol is capable of protecting cells and tissues from oxidative stress.

4'-O-β-D-Glucosyl-5-O-Methylvisamminol Attenuates Pro-Inflammatory Responses and Protects against Oxidative Damages

  • We attempted to examine anti-inflammatory and anti-oxidant effects of 4'-O-${\beta}$-D-glucosyl-5-O-methylvisamminol (GOMV), the first epigenetic inhibitor of histone phosphorylation at Ser10. While GOMV did not affect the viability of murine macrophage RAW 264.7 cells, it significantly suppressed lipopolysaccharide (LPS)-induced generation of prostaglandin $E_2$ ($PGE_2$) and nitric oxide (NO) through transcriptional inhibition of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS). GOMV also scavenged free radicals in vitro, increased NF-E2-related factor 2 (NRF2), and activated antioxidant response element (ARE), thereby resulting in the induction of phase II cytoprotective enzymes in human keratinocyte HaCaT cells. Finally, GOMV significantly protected HaCaT cells against 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced oxidative intracellular damages. Together, our results illustrate that GOMV possesses anti-inflammatory and anti-oxidant activity.

Cytoprotective Effect on Oxidative Stress and Inhibitory Effect on Cellular Aging of Terminalia chebula and Uncaria sinensis

  • The ethanol extract from the fruit of Terminalia chebula (Combretaceae) and the hook of Uncaria sinensis (Rubiaceae) exhibited significant inhibitory activity on oxidative stress and the age-dependent shortening of the telomeric DNA length. In the peroxidation model using t-BuOOH, human epidermal keratinocytes-neonatal foreskin (HEK-N/F) cells were treated with the T. chebula and U. sinensis extracts. The results showed a notable enhancing effect on the cell viability of 60.5 ${\pm}$ 3.8 and 65.0 ${\pm}$ 3.0%, respectively, by 50 $\mu\textrm{g}$/ml of the extracts. (omitted)
