Pycnogenol, a Standardized Extract of French Maritime Pine Bark, Inhibited the Transcriptional Expression of Th2 Cytokines by Suppressing $NF-{\kappa}B$ Activation in Primary Splenocytes of C57BL/6 Mice with Murine AIDS (Murine AIDS 감염쥐 splenocytes에서 $NF-{\kappa}B$의 활성화 억제를 통한 pycnogenol의 Th2 cytokines의 mRNA 발현 조절 효과)

  • LP-BM5 murine leukemia retrovirus induces the immune dysfunction by imbalanced secretion of Th1 and Th2 cytokines in the murine AIDS model. In the present study, it was investigated whether pycnogenol (Pyc) administration could deactivate $NF-{\kappa}B$ to regulate the gene expression of Th1 and Th2 cytokines in C57BL/6 mice with murine AIDS. Treatment with Pyc for 12 weeks significantly inhibited the loss of body weight and enlargement of spleen and lymph node usually seen with AIDS. Moreover, Pyc increased the plasma level of Th1 cytokines, IL-2 and $IFN-{\gamma}$, while reducing the plasma level of Th2 cytokines, IL-6, IL-10, and $TNF-{\alpha}$. In primary culture of splenocytes, mRNA expression of Th2 cytokines was suppressed, but that of Th1 cytokines was not affected. The LP-BM5 retrovirus infection stimulated the cytoplasmic activation of $NF-{\kappa}B$ and nuclear translocation of $I-{\kappa}B$, whereas Pyc administration significantly reduced $NF-{\kappa}B$ activation and $I-{\kappa}B$ degradation. These results suggested that the inhibitory effect of Pyc on Th2 cytokines in mice with murine AIDS was dependent on suppression of the $NF-{\kappa}B$ signaling pathway and was not dependent on $INF-{\gamma}$ level, which regulates Th2 cytokines.

Studies on the Regulatory Effect of Cytokine Production in Patients with Cerebral Infarction by Yuldahansotang (열다한소탕(熱多寒少湯)이 태음인(太陰人) 뇌경색증(腦硬塞症) 환자(患者)의 세포활성물질생성조절(細胞活性物質生成調節)에 미치는 영향(影響))

  • 1. Background and Purpose According to Sasang constitutional medicine, Yuldahansotang(YHT) is a useful prescription for Teaumin patients with a variety of neurologic disorders. Then, I investigated about certain relationships between the effect of YHT and the changes of immune system, especially cytokine network. 2. Methods We studied 8 Taeumin patients with Cerbral infarction. They were treated with YHT in constitutional clinic of Wonkwang Kwagnju Oriental Hospital. We investigated the changes of cytokine network of them. We also investigated cytokine release by lipopolysaccharide- activated peripheral blood mononuclear cells from healthy Taeumin controls. 3. Results The mean interleukin (IL)-2 plasma levels were slightly lower in the plasma of patients than in normal group, whereas the mean IL-4, IL-6 and IgE levels were significantly higher. But there were no significant differences in $interferon-{\gamma}$($IFN-{\gamma}$) levels between each group. After administration of YHT for two to four weeks, plasma levels of $IFN-{\gamma}$ and IL-2 derived from T helper (Th) 1 cells were elevated significantly, whereas plasma levels of IL-4 and IL-6 derived from Th2 cells were reduced significantly. Plasma levels of IgE were reduced significantly, too. During the period of YHT administration, other adverse effects are not shown. It is increased significantly to Cytokine release by lipopolysaccharide -activated peripheral blood mononuclear cells from healthy Taeumin controls. And the release of $IFN-{\gamma}$, IL-2 and IL-6 was progressively decreased in the plasma treated with YHT. It shows regulatory effects of YHT to cytokine production.

Cytokine Expressions with Lectins from Allomyrina dichotoma and Bombyx mori (장수풍뎅이(Allomyrina dichotoma)와 누에(Bombyx mori) 렉틴의 사이토카인 생성 양상 비교)

  • Lectins from Allomyrina dichotoma (ADL) and Bombyx mori (BML) were partially purified by physiological saline extraction, ammonium sulfate fractionation, anion exchange column chromatography on DEAE Sephadex A-50 and gel filtration column chromatography on Sephadex G-200. An assay for cytokine expression was carried out by using reverse transcription polymerase chain reaction(RT-PCR). mRNA isolated from PBMC(human peripheral blood mononuclear cells) were stimulated with ADL(O.D.=0.2) and BML(O.D.=0.1) for various times(1,4,8,24,48 and 72 h) and various cytokine mRNA assessed by RT-PCR were shown as follows: The patterns of bands for IL-1 mRNA of BML were very similar with those from ADL and these bands were decreased along the increasing reaction times after showing a strong band at 1 h. However mRNA expressions for IL-2, IL-6, $IFN{\gamma}$ and $TNF{\alpha}$ showed different patterns between ADL and BML. With the effect of ADL, the expression of IL-2 and IL-6 mRNA were continuously detected until 72 h with the strongest band of IL-2 mRNA at 24 h. The strong bands of $IFN{\gamma}$ mRNA were observed from 4 to 8 h but the strongest one of $TNF{\alpha}$ was just observed at 1 h. Meanwhile with BML, the bands for IL-2 and $IFN{\gamma}$ were increased along the increasing reaction times until 72 h. The strongest bands were showed from 4 to 8 h with IL-6 and at 8 h with $TNF[\alpha}$. To verify quantitatively ELISA was used for assay of protein secretions of the cytokine gene with IL-2 and $IFN{\gamma}$ expressed markedly different in RT-PCR. The highest cytokine secretion for IL-2 was demonstrated at 48 h. The production of $IFN{\gamma}$ was markedly increased at 24 h and secreted highest at 72 h. These result suggest that ADL and BML, as inducers of cytokines, can elicit detectable cytokine mRNA from PBMC within the first few hours of stimulation and maintain the production of cytokines for a few days by the methods of RT-PCR and ELISA.

Effect of Fermented Cudrania tricuspidata Fruit Extracts on the Generation of the Cytokines in Mouse Spleen Cells (발효 꾸지뽕(Cudrania tricuspidata) 열매 추출물이 마우스 비장세포의 cytokine 생성에 미치는 영향)

  • We investigated a physiological function by fermenting a medicinal mushroom, (Cudrania tricuspidata fruit). A fermentation using lactic acid bacteria and the extracts isolated from 70% ethanol fractionation was included in cultured mouse spleen cells for cytokine secretion. As a result, total polyphenol content improved by 47% by organic acid fermentation. This was regarded as immune activity in fermented C. tricuspidata fruits, as the levels of interleukin (IL)-2 and IL-4 secretion increased. In addition, when the extracts were treated with a stimulant lipopolysaccharide, the secretion of helper T (Th) 1 cytokines IL-2, IL-12, and tumor necrosis factor-${\alpha}$ was suppressed, while the secretion of Th2 cytokines IL-4, IL-5, IL-6, and IL-10 significantly increased. Therefore, this study suggests that fermentative C. tricuspidata fruit extracts can contribute to the suppression of cellular immune reactions induced by the expression of Th1 cells and activation of the expression of Th2 cells inducing humoral immune reactions associated with the antibody generation by B lymphocytes.

Comparison Study of Immunomodulatory Activity of Polysaccharide and Ethanol Extracted from Sargassum fulvellum (참모자반 조다당 추출물과 에탄올 추출물의 대식세포 및 비장세포 활성 비교)

  • The immune system plays an important role in maintaining and protecting human health. In the present study, comparison of immuno-modulatory activities between polysaccharides (SFP) and ethanol (SFE) extracts separated from Sargassum fulvellum in macrophages and murine splenocytes were investigated. Immuno-modulatory activities of macrophages were estimated based on cell proliferation, nitric oxide (NO), inducible NO synthase (iNOS), and cytokine production in RAW 264.7 macrophage cells, and lipopolysaccharide was used as a positive control. SFP and SFE treatment did not affect cytotoxicity in RAW 264.7 macrophage cells, and SFP treatment significantly increased NO and cytokine production ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$), whereas SFE did not contribute to the increase in NO and cytokine production. In the case of splenocytes, SFP treatment increased splenocyte proliferation and also highly increased production of Th-1 type cytokines (IL-2 and $IFN-{\gamma}$) than those of SFE. Through this study, we confirmed that immuno-modulatory activities of Sargassum fulvellum may be due to polysaccharide extracts and this can be a potential nutraceutical.

Experimental Study of Lonicerae Caulis and Bee-venom Acupuncture on the Rheumatic Pathologic Model Induced by Adjuvant in Rats (인동등과 봉독약침이 Adjuvant 투여로 유발된 류마토이드 병태모델에 미치는 실험적 연구)

  • Objectives This study was carried out to investigate the experimental effects of Lonicerae Caulis and Bee-venom Acupuncture in Yanglingquan (陽陵泉, GB34) that have clinical efficacy in the Rheumatoid Arthritis. Methods Materials of present study are Lonicerae Caulis Extracts (LCE), Bee-Venom Acupuncture (BVP), Sprague-dawley rats (250 g or so, ♂), and various kinds of needing experimental studies. We measured several experimental items of the rats with the arthritis induced by Freund's complete adjuvant (0.2 ml/kg), such as body weight, rate of paw edema, analgesic effect by hot plate method, WBC, TNF-${\alpha}$ cytokine and IL-10 cytokine. Rats were divided into four groups; Normal group that was treated with normal saline 1.0 ml (o.p) and $15{\mu}l/kg$ (GB34) to normal rats, Control group that was treated with normal saline 1.0 ml (o.p) and $15{\mu}l/kg$ (GB34) to pathologic model rats induced by Freund's complete adjuvant 0.2 ml/kg, Experimental group A that was treated with LCE 1.0 ml (o.p) and normal saline $15{\mu}l/kg$ (GB34) to pathologic model rats induced by Freund's complete adjuvant 0.2 ml/kg and Experimental group B that was treated with LCE 1.0 ml (o.p) and BVP $15{\mu}l/kg$ (GB34) to pathologic model rats induced by Freund's complete adjuvant 0.2 ml/kg, and the experiment took over after 28 days. The results were analysed using SPSS for windows 12.0. Results Experimental group A showed the increase in body weight, paw licking times and IL-10 cytokine compared to Control group. Also it was decreased in rate of paw edema, WBC, and TNF-${\alpha}$ cytokine compared with Control group. Experimental group B showed the increase in body weight, paw licking times, and IL-10 cytokine compared to Control group, and showed the decrease in rate of paw edema, WBC and TNF-${\alpha}$ cytokine compared to Control group. Especially TNF-${\alpha}$ cytokine and rate of paw edema were accepted statistical significance compared with Control group. Conclusions It is suggested that Lonicerae Caulis (o.p) and Bee-venom Acupuncture (GB34) can be used in the treatment of rheumatoid arthritis.

The Effects of Coisis semen extract on blood serum cytokine of Allergic Rhinitis Mice model (의이인 추출물이 알레르기 비염 동물의 cytokine에 미치는 영향)

  • Objective : Allergic rhinitis(AR) is an inflammation or irritaion of nasal mucosa. Usually, serum cytokine is regarded as one of the most important factors in AR. Coisis semen have been used for the diseases in digestive organs in traditional chinese medicine. Nowaday, there were some studies reported about the effects of coisis semens on serum cytokine. However the results of theses studies were varies, and moreover there was no study which have used AR animal model. Therefore this study is aimed to determine therapeutic effects of coisis semen extract by observing changes of serum cytokine(IgE, TNF-${\alpha}$, IL-$1{\beta}$, IFN-${\gamma}$ and IL-13) with AR animal model. Methods : Thirty BALB/c mice were divided into six groups : Normal group, Control group, sample I, sample II, sample III, sample IV. Except for Normal group, all the mice in other 5 groups were sensitized intraperionealy by 0.1% ovalbumin solution three times at intervals of one week. Then intranasal sensitization was performed by diffusing 0.1% ovalbumin solution three times at intervals of two days. Normal group were used Normal Saline instead of ovalbumin solution. After the first day of study, sample I, sample II, sample III, and sample IV were orally administrated coisis semen extract by 100mg/kg, 200mg/kg, 500mg/kg, and 1g/kg respectively for 28days. Then, the changes of serum IgE, TNF-${\alpha}$, IL-$1{\beta}$, IFN-${\gamma}$ and IL-13 were observed in 6 groups. We used the statistical methods of ANOVA, post hoc by duncun, and Kruskal-Wallis test(p<0.05) Results : There were statistical changes in IgE and TNF-${\alpha}$. But, there were no statistical changes in IL-$1{\beta}$, IL-13 and IFN-${\gamma}$. Conclusions : According to above results, it is supposed that coisis semen extract has not some significant effects on cytokine of AR animal model. There was no evidence for using coisis semen in relieving symptoms of allergic rhinitis.

Factors Influencing the Production of Water-soluble Endopolysaccharides and Exopolysaccharides from Lentinus lepideus and their Effects on Immune Cytokine Production

  • An efficient method to produce water-soluble polysaccharides from Lentinus lepideus is described. The productivity of both endopolysaccharides (PPS) and exopolysaccharides (EPS) was compared under various culture conditions. The effect of treating their own PPS and EPS on immune cytokine production was also studied in relation to culture factors. High yield production of EPS required a moderate culture temperature $(25^{\circ}C)$ as well as long culture period (16-20 days). In contrast, PPS production required a high culture temperature $(30^{\circ}C)$ and short culture period (8 days). Most of the carbon sources did not affect polysaccharides and mycelial production except for sucrose. Immune cytokine levels in the EPS treatment varied among carbon sources or culture periods. PPS did not appear to affect much on the production of cytokines, regardless of the culturing factors, except for the culture period. These results suggest that the optimal culture conditions for L. lepideus vary according to culture purposes, and different culture conditions should be used for different targets including mycelial biomass, EPS, and PPS. Whereas the immunomodulating activitiy of EPS appeared to be affected by culture conditions in L. lepideus, that of PPS did not.
