그라비올라(Annona muricata) 잎 에탄올 추출물의 항염증 효과 (Anti-Inflammatory Effects of Annona muricata Leaf Ethanol Extracts)

  • 본 연구는 그라비올라 잎 에탄올 추출물(AME)의 항염증 효과에 관하여 알아보기 위하여 LPS에 의해 유도되는 대식세포 염증모델에 AME를 병용 처리하여 다양한 염증매개인자들의 분비능 및 염증억제 기전에 관하여 알아보았다. LPS의 처리에 따라 염증반응이 유도된 RAW 264.7 대식세포에 AME를 병용 처리하였을 때, NO 및 염증성 cytokine(IL-6, $TNF-{\alpha}$$IL-1{\beta}$)의 분비능이 유의적으로(P<0.05) 감소하는 것을 확인하였다. 또한, AME의 처리에 따라 염증반응의 매개인자인 iNOS 및 COX-2 발현이 유의적으로(P<0.05) 감소하는 것으로 나타났으며, 이러한 항염증 활성의 기전에 관하여 분석한 결과 염증성 단백질 발현의 전사인자인 $NF-{\kappa}B$와 MAPKs 신호전달 경로 억제를 통하여 추출물의 항염증 활성이 나타난다는 사실을 관찰할 수 있었다. 따라서 이러한 결과를 통하여 AME의 처리는 염증반응의 전사인자인 $NF-{\kappa}B$의 발현을 조절함으로써 iNOS 및 COX-2의 발현을 억제하고, 염증성 매개인자인 NO 및 cytokine(IL-6, $TNF-{\alpha}$$IL-1{\beta}$)의 분비를 억제한다고 판단된다.

  • In the present study, we investigated the anti-inflammatory effects by kaempferol-3-O-${\beta}$-D-sophoroside (KS) isolated from Sophora japonica (Leguminosae) on the lipopolysaccharide (LPS)-induced nitric oxide (NO) and prostaglandin ($PGE_2$) production by RAW 264.7 cell line compared with kaempferol. KS significantly inhibited the LPS-induced NO and $PGE_2$ production. Consistent with these observations, KS reduced the LPS-induced expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the protein and mRNA levels in a concentration-dependent manner. In addition, the release and the mRNA expression levels of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and interleukin-6 (IL-6) were also reduced by KS. Moreover, KS attenuated the LPS-induced activation of nuclear factor-kappa B ($NF{-\kappa}B$), a transcription factor necessary for pro-inflammatory mediators, iNOS, COX-2, $TNF-{\alpha}$ and IL-6 expression. These results suggest that the down regulation of iNOS, COX-2, $TNF-{\alpha}$, and IL-6 expression by KS are achieved by the downregulation of $NF{-\kappa}B$ activity, and that is also responsible for its anti-inflammatory effects.

Anti-Inflammatory Effects of Water Chestnut Extract on Cytokine Responses via Nuclear Factor-κB-signaling Pathway

  • Water chestnut (Trapa japonica Flerov.) is an annual aquatic plant. In the present study, we showed that the treatment of water chestnut extracted with boiling water resulted in a significant increase 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity and decrease the intracellular $H_2O_2$-induced accumulation of reactive oxygen species. In addition, water chestnut extract (WCE) inhibited lipopolysaccharide (LPS)-induced nitric oxide production and suppressed mRNA and protein expression of the inducible nitric oxide synthase gene. The cytokine array results showed that WCE inhibited inflammatory cytokine secretion. Also, WCE reduced tumor necrosis factor-${\alpha}$- and interleukin-6-induced nuclear factor-${\kappa}B$ activity. Furthermore, during sodium lauryl sulfate (SLS)-induced irritation of human skin, WCE reduced SLS-induced skin erythema and improved barrier regeneration. These results indicate that WCE may be a promising topical anti-inflammatory agent.

  • 본 연구를 통해서, 한(寒) 처방의 대표 처방인 황련해독탕(黃連解毒湯)과 열(熱) 처방의 대표 처방인 건강부자탕(乾薑附子湯) 모두 항염 효능을 확인할 수 있었으나, 그 작용 기전에 있어 뚜렷한 차이를 나타내었다. 이러한 차이는 한의학의 기본 이론인 한열에 대한 개념에 대한 연구의 초석이 될 수 있기를 바라며, 각 개별 약물의 효능 및 다른 처방들과 다른 기전적 실험이 추가적으로 필요할 것을 보인다.

흑마늘 발효물의 항염증 활성 (Anti-inflammatory Activities of Fermented Black Garlic)

  • Lactobacillus rhamnosus로 발효시킨 흑마늘 발효물의 항염증 효능을 검증하기 위해 LPS로 염증 유도된 RAW 264.7 cells를 이용하여 관련 인자들을 분석하였다. 100, 200, 400 및 $800{\mu}g/mL$ 농도에서 세포독성은 유발되지 않았으며, 오히려 농도 의존적으로 세포 생존율은 증가하였다. LPS에 의해 염증 유도된 RAW 264.7 cells에서 흑마늘 발효물은 농도 의존적으로 NO와 $PGE_2$의 생성 감소와 염증성 사이토카인인 TNF-${\alpha}$, IL-$1{\beta}$ 및 IL-6의 단백질 생성을 감소시켰다. 또한 iNOS, COX-2, NF-${\kappa}B$$I{\kappa}B$ 단백질의 발현을 감소시키고 HO-1의 단백질의 발현을 증가시켰다. 이상의 연구 결과를 통해 흑마늘 발효물은 염증에 의한 NF-${\kappa}B$의 활성과 TNF-${\alpha}$, IL-$1{\beta}$와 IL-6의 생성을 억제시키고, iNOS 및 COX-2의 발현을 억제시키는 메커니즘을 통해 염증성 질환의 예방 및 개선 효능을 나타내는 것으로 판단된다.

만성폐쇄성폐질환의 급성악화와 회복기에서 유도객담 내 Nuclear Factor-κB(NF-κB)의 활성도와 IL-6, IL-8 및 TNF-α의 농도 변화 (Nuclear Factor-κB(NF-κB) Activity and Levels of IL-6, IL-8 and TNF-α in Induced Sputum in the Exacerbation and Recovery of COPD Patients)

  • 연구배경 : COPD의 급성악화는 세균에 의한 감염, 바이러스성 상기도 감염, 대기오염, 기후변화 등에 의하며, COPD의 급성악화 시에 객담 내 호중구의 증가, IL-6와 IL-8 농도의 증가, 그리고 산화질소의 증가는 $NF-{\kappa}B$의 활성화와 관련된 것으로 알려져 있다. 그러므로 COPD의 병인 및 급성악화의 기전에 $NF-{\kappa}B$ 활성도와 IL-6, IL-8 및 $TNF-{\alpha}$가 관련이 있는지 연구하고자 하였다. 방 법 : 정상대조군 및 COPD로 입원하였던 환자들의 급성악화기 및 치료 후 회복기에 유도객담에서 IL-6, IL-8, $TNF-{\alpha}$, $NF-{\kappa}B$ 활성화 정도를 측정하여 비교하였다. 결 과 : 1) 유도객담내의 IL-6, IL-8 및 $TNF-{\alpha}$는 COPD 환자에서 대조군에 비해 유의하게 증가되었다(p<0.01). IL-8은 급성악화 시에 비해 회복기에 유의하게 감소되었고(p<0.05), IL-6와 $TNF-{\alpha}$는 회복기에도 차이가 없었다. 2) 유도객담 내 대식세포에서의 $NF-{\kappa}B$의 활성도는 COPD 환자에서 대조군에 비해 유의하게 증가 되었고(p<0.05), 회복기에는 악화 시에 비해 감소하는 경향을 보였다. 결 론 : COPD 환자에서 유도객담 내 IL-6, IL-8, $TNF-{\alpha}$$NF-{\kappa}B$의 활성도 등이 정상대조군에 비해 증가되었고, 회복기에 IL-8은 감소하였고, $NF-{\kappa}B$의 활성도, IL-6 및 $TNF-{\alpha}$는 감소하는 경향을 보여서, COPD의 급성악화 및 COPD의 병인에 $NF-{\kappa}B$가 일부 관여할 것으로 생각된다.

Compound K Rich Fractions Regulate NF-κB-dependent Inflammatory Responses and Protect Mice from Endotoxin-induced Lethal Shock

  • In the previous studies, we isolated the compound K rich fractions (CKRF) and showed that CKRF inhibited Toll-like receptor (TLR) 4- or TLR9-induced inflammatory signaling. To extend our previous studies,1) we investigated the molecular mechanisms of CKRF in the TLR4-associated signaling via nuclear factor (NF)-${\kappa}B$, and in vivo role of CKRF for induction of tolerance in lipopolysaccharide (LPS)-induced septic shock. In murine bone marrow-dervied macrophages, CKRF significantly inhibited the induction of mRNA expression of proinflammatory mediators such as tumor necrosis factor-${\alpha}$, interleukin-6, cyclooxygenase-2, and inducible nitric oxide synthase. In addition, CKRF significantly attenuated the transcriptional activities of TLR4/LPS-induced NF-${\kappa}B$. Nuclear translocation of NF-${\kappa}B$ in response to LPS stimulation was significantly abrogated by pre-treatment with CKRF. Furthermore, CKRF inhibited the recruitment of p65 to the interferon-sensitive response element flanking region in response to LPS. Finally, oral administration of CKRF significantly protected mice from Gram-negative bacterial LPS-induced lethal shock and inhibited systemic inflammatory cytokine levels. Together, these results demonstrate that CKRF modulates the TLR4-dependent NF-${\kappa}B$ activation, and suggest a therapeutic role for Gram-negative septic shock.

Leaves of Raphanus sativus L. Shows Anti-Inflammatory Activity in LPS-Stimulated Macrophages via Suppression of COX-2 and iNOS Expression.

  • Raphanus sativus L. (RS) is a cruciferous vegetable that is widely consumed in Korea. The anticancer activity of leaves of RS (RSL) extract has been investigated; however, no studies focused on its anti-inflammatory effects. Therefore, the aim of the current study was to evaluate the anti-inflammatory effects of RSL extract. In brief, RSL powder was fractionated into n-hexane, chloroform, ethyl acetate, n-butanol, and water-soluble fractions. Lipopolysaccharide (LPS)-stimulated RAW264.7 cells were treated with each fraction for initial screening. It was found that the chloroform fraction significantly inhibited nitric oxide release in LPS-stimulated RAW264.7 cells with a half maximal inhibitory concentration value of $196{\mu}g/mL$. In addition, the mRNA and protein expression levels of inducible nitric oxide synthase, measured using reverse transcriptase-polymerase chain reaction and western blotting, respectively, were reduced in a concentration-dependent manner. Moreover, the inflammatory cyclooxygenase-2 enzyme expression decreased. Furthermore, the expression of nuclear factor-kappa B ($NF-{\kappa}B$), the key regulator of the transcriptional activation of the inflammatory cytokine genes, was reduced by the RSL chloroform fraction. Therefore, the results of our study suggest that RSL exhibits anti-inflammatory effects in LPS-stimulated macrophages via $NF-{\kappa}B$ inactivation.

선퇴가 인간의 THP-1 단핵구에서 사이토카인 유전자 발현에 미치는 영향 (Effects of Cryptotympana pustulata on the expression of cytokine genes in human monocytes of THP-1)

  • Objective : This study was performed to evaluate the effect of immune reaction inductive substances such as phorbol-myristate-acetate(PMA), lipopolysaccharide(LPS), dermato-phagoides pteronyssus crude extract(DPE), dinitrochloro-benzene(DNCB) and Cryptotympana pustulata(CP), the Cryptotympana pustulata extracting substance at simultaneously on the translocation of nuclear factor-kappa B(NF-${\kappa}B$) towards to the nucleus and the mRNA expression patterns of various cytokine genes in Human acute monocytic leukemia cell line(THP-1 cells), monocytes of human. Experiment : To analyze cytokine genes expression patterns, the RT-PCR method was used, measuring tumor necrosis factor(TNF)-$\alpha$ that had been secreted during cell culture in the ELISA method. The morphological change in the cell observed during THP-1 cell culture was observed using a scanning electron microscope (SEM) and the quantitative distribution in the cell NF-${\kappa}B$ was analyzed through immunocytochemistry and a confocal microscopy. Result : CP showed different influences onto the mRNA expression patterns of cytokine genes with PMA, LPS. DPE and DNCB according to the types of immune inductive substances in the THP-1 cells. The expressions of inter-leukin(IL)-10, interferon(INF)-$\gamma$, TNF-$\alpha$ and monocyte chemoattractantant protein(MCP)-1 induced by PMA were suppressed by CP while the expression of transforming growth factor(TGF)-$\beta$ was promoted. Regarding the secretion pattern of TNF-$\alpha$ according to PMA processing, its secretion amount was increased by CP concurrent processing, in case of processing CP onto PMA and LPS, We discovered that the secretion amount of TNF-$\alpha$ was increased. Upon processing PMA and LPS on the THP-1 cell strain at the same time or either additionally processing CP thereon, the movement increase towards the nucleus from the NF-${\kappa}B$ cell cytoplasm, a transcription factor was able to be observed. Conclusion : In this study, Cryptotympana pustulata extracting substance was confirmed that it had an influence on expression patterns of cytokine genes according to the actions of a variety kinds of immune reaction inductive substances processed on the monocyte THP-1 cell of humans. Therefore, additional studies as for the immune adjusting function of Cryptotympana pustulata are considered to be able to offer important materials for curing immune abnormal diseases such as atopy dermatitis afterward.
