First Record of Cleantioides planicauda (Crustacea: Isopoda: Holognathidae) from South Korea

  • Cleantioides planicauda (Benedict, 1899) has previously been recorded in China and Japan. In the present study, to our knowledge, we report for the first time the occurrence of this species in South Korea. Here, we provide detailed descriptions and illustrations of the diagnostic characteristics of C. planicauda. Further, for the first time, we determined the partial sequence of mitochondrial cytochrome c oxidase subunit 1 (CO1) gene of C. planicauda for molecular characteristic. Additionally, we provide a key to the Korean species and distribution of the genus Cleantioides. As a result of this study, four species of the genus Cleantioides, including C. planicauda, have now been recorded in South Korea.

Genetic Variability among Different Populations of Root Knot Nematodes Based on Their Encumbrance Response to Pasteuria Isolates Using PCR-RFLP

  • A great variable response was observed when PP-3 and PP-J encumbered with 116 populations of root knot nematode (RKN) at two different temperatures ($25{\pm}2^{\circ}C$ and $30{\pm}2^{\circ}C$) and concentrations ($10^4$ and $10^5$ spores/ml). The PCR reaction amplified intergenic region between cytochrome oxidase subunit II gene (COII) and large subunit of rRNA gene (lrRNA) of the mitochondrial genome of different RKN species. The primer C2F3 and 1108 identified M. incognita with the highest frequency (52.6%) followed by M. javanica (36.8%) and M. arenaria (10.5%). The sizes of PCR products were 1.7 kb for M. incognita and M. javanica populations while populations of M. arenaria produced 1.1 kb fragment. The digestion with Hinf I yielded three different fragment length patterns on 1.5 % agarose gel. From current research it is concluded that intra-Meloidogyne genetic variability exist in RKN populations which have better encumbrance with P. penetrans.

First record of the Cleantioides rotundata (Malacostraca: Isopoda: Holognathidae) from South Korea

  • Cleantioides rotundata (Kussakin, 1982) is reported for the first time in South Korea. Specimens of C. rotundata were collected with a small hand net from the intertidal zone of Opo-ri, Yeongdeok-gun in South Korea. This species makes tubes using seagrass debris at our sampling location and distinguished by the following characteristics: pleotelson long, length almost 1.6 times as long as width; dorsum of distal one third with deeply bilobed depression on plane. In this paper, we provide descriptions of diagnostic characteristics and illustrations of morphology of C. rotundata. Additionally, a key to the Korean species of the genus Cleantioides is provided and the partial sequences of the mitochondrial cytochrome c oxidase subunit 1 (CO1) are provided as molecular characteristics.

First Record of Callochiton foveolatus (Polyplacophora: Callochitonidae) from Korea

  • The genus Callochiton Gray, 1847 is small to medium sized chiton (up to $55{\times}36mm$ in size) and includes approximately 30 species worldwide, most of which are found on hard substrata in subtidal zone. To date, only three species of Callochiton Gray, 1847 have been reported in Japan. In this study, we found C. foveolatus(Is. Taki, 1938) as first record of the family Callochitonidae Plate, 1901 in Korea and its morphological features were described and compared with other northwestern Pacific species. In addition, the partial fragment of mitochondrial DNA cytochrome c oxidase subunit I sequences of C. foveolatus was determined as DNA barcoding record and compared with other congeneric species.

Variation of nuclear and mitochondrial DNAs in Korean and Chinese isolates of Clonorchis sinensis

  • We compared the DNA sequence difference of isolates of Clonorchis sinensis from one Korean (Kimhae) and two Chinese areas (Guangxi and Shenyang), The sequences of nuclear rDNA (18S, internal transcribed spacer 1 and 2: ITS1 and ITS2) and mitochondrial DNA (cytochrome c oxidase subunit 1: cox1) were compared. A very few intraspecific nucleotide substitution of the 18S, ITS1, ITS2 and cox1 was found among three isolates of C. sinensis and a few nucleotide insertion and deletion of ITS1 were detected. The 18S, ITS1, ITS2 and cox1 sequences were highly conserved among three isolates. These findings indicated that the Korean and two Chinese isolates are similar at the DNA sequence level.

Pythium subutonaiense, A New Aquatic Oomycete from Southern China Based on Morphological and Molecular Characters

  • A new species, Pythium subutonaiense, isolated from aquatic environments (lake) in China is being described based on morphological characters and molecular evidence. The isolates grew at temperatures between $5^{\circ}C$ and $38^{\circ}C$, and the optimum temperature was $30^{\circ}C$, with a radial growth rate of 17.6 mm at $25^{\circ}C$ per day. It is homothallic and characterized by globose to sub-globose shaped and mostly terminal or sometimes catenulate hyphal swellings, filamentous non-inflated sporangia, and smooth oogonia with hypogynous and monoclinous antheridia that contained one plerotic oospore. In phylogenetic analysis, inferred based on the internal transcribed spacer region of the ribosomal RNA gene and mitochondrial cytochrome c oxidase subunit 1 gene, the new species formed a distinct lineage in Pythium clade B. Differences between the new species and phylogenetically related and morphologically similar species are discussed.

성페로몬을 이용한 열대거세미나방 포획과 시토크롬 옥시다제 1(CO1)에서 종내 변이군 특이적 단일염기다형성 분포 (Sex Pheromone Trapping of Spodoptera frugiperda (Noctuidae: Lepidoptera) in Korea and the Distribution of Intraspecies-specific Single Nucleotide Polymorphisms in the Cytochrome c Oxidase Subunit 1 (CO1))

  • 2019년에 서남해안 지역인 고창군의 옥수수 밭 주변에서 성페로몬을 이용하여 열대거세미나방(Spodoptera frugiperda) 성충을 효과적으로 모니터링하는 방법을 조사하였다. 총 함량이 300 또는 1000 ㎍인 2종류 성분 조성의 성페로몬 미끼[(100%) (Z)-9-tetradecenyl acetate and (2%) (Z)-7-dodecenyl acetate]를 설치한 깔대기형 트랩과 델타형 트랩 중에서 열대거세미나방은 300 ㎍ 미끼의 깔대기형 트랩에서 8월 6일에 처음 잡혔고 가장 많이 포획되었다. 또한 깔대기형 트랩 모두에서 비표적 종인 뒷흰가는줄무늬밤나방(Mythimna loreyi)이 많이 포획되었다. 총 함량이 1000 ㎍인 위의 2종류 성분 조성과 4종류 성분 조성의 미끼[(100%) (Z)-9-tetradecenyl acetate, (8%) (Z)-11-hexadecenyl acetate, (2%) (Z)-7-dodecenyl acetate, and (1%) (Z)-9-dodecenyl acetate]를 설치한 날개형 트랩에서 열대거세미나방은 비슷한 수준의 낮은 포획수를 보였으나 뒷흰가는줄무늬밤나방은 4종류 성분 조성의 미끼에서 훨씬 더 많이 포획되었다. 성페로몬 트랩에 포획된 열대거세미나방 70마리의 미토콘드리아 시토크롬 옥시다제 1(CO1)의 부분염기서열(1,004 bp)을 이용하여 계통수를 분석한 결과, 두 개의 종내 변이군으로 나눠졌으며 66마리가 CO1-RS로, 나머지 4마리는 CO1-CS로 분지되었다. 또한 두 개의 CO1 변이군과 기주식물계통(벼, 옥수수)에서 일관되게 차이가 있는 총 12개의 CO1 단일염기다형성(SNP)이 확인되었으며, 전체 73마리 중 4마리만 CO1-CS 그룹(옥수수계통 포함)과 동일한 패턴을 보였으며 나머지 69마리는 CO1-RS그룹(벼계통 포함)과 같았다.

콩 포장에서 어리팥나방과 팥나방(나비목: 잎말이나방과)의 동시 발생과 시토크롬 c 산화효소 1 유전자 염기서열의 다형성 (Co-occurrence of Matsumuraeses falcana and M. phaseoli (Lepidoptera: Tortricidae) in Soybean Fields, and Polymorphism of Cytochrome c Oxidase Subunit 1 Gene Nucleotide)

  • 미토콘드리아 시토크롬 c 산화효소 1 (COX1) 유전자 염기서열(658 bp)을 사용하여, 콩 포장에서 채집된 어리팥나방(Matsumuraeses falcana)과 팥나방(Matsumuraeses phaseoli)의 종을 실험실 집단의 종들과 비교하여 동정하였다. COX1 염기서열 분석에서, 어리팥나방 47개체로부터 10개의 하플로타입이 발견되었고, 종내 유전적 거리는 0.15~0.46%이었다. 이중 하프로타입 A형이 약 70%로 우점형이었다. 팥나방의 30개체로부터는 모두 동일한 하나의 서열만이 확인되었고, 어리팥나방과의 종간 유전적 거리는 4.11~4.61%이었다. 두 종의 COX1 염기서열을 번역한 아미노산 서열은 모두 동일하여 동의적 염기서열 변이(동의치환, 同義置換, synonymous substitution)를 확인할 수 있었다. 포장 조사에서 두 종의 유충이 콩의 잎과 꼬투리를 가해하였고, 한 포장에서 동시에 발생하였다. 전체 포장에서 어리팥나방의 평균 밀도는 팥나방보다 약 1.5배 높았다. 이 결과는 콩이 두 종의 동일 기주임을 명백하게 제시하였다. 별도로 이 속의 유충 기생파리로서 Elodia flavipalpis (파리목: 기생파리과)가 발견되었고, COX1 서열로 동정되었다.

A New Species of Cerapus from Korea (Crustacea: Amphipoda: Ischyroceridae)

  • Cerapus longicervicum, a new species of ischyrocerid amphipods, was isolated from the vicinal sea of Deokjeokdo-Island, Incheon (South Korea). This species is characterized by a narrower coxal plate width of gnathopod 1 in male, which is 0.39 times as broad as pereonite 1, and antenna 2, which is longer than antenna 1. This paper also provides the partial sequence of the mitochondrial cytochrome c oxidase subunit 1 (CO1) of the new species, which was obtained from a specimen that was preserved in 4% formalin solution.

DNA Barcoding for the Hydrothermal Vent Crab Austinograea Species (Crustacea: Bythograeidae) from the North Fiji Basin, Southwestern Pacific Ocean

  • The brachyuran crab Bythograeidae Williams, 1980 is common in hydrothermal vent fields worldwide and has recorded to sixteen species of six genera. In this study, we firstly determined the cytochrome c oxidase subunit 1 (CO1) DNA barcodes for the fifth species of Austinograea, A. hourdezi, from hydrothermal vent regions of the North Fiji Basin in southwestern Pacific Ocean. All CO1 DNA barcodes of A. hourdezi were identical. The interspecies variations of three bythograeid genera were 10.9-13.3% for Austinograea, 6.6-15.7% for Bythograea, and 9.7% for Gandalfus. These results would be helpful to understand taxonomy of brachyuran crabs living in hydrothermal vent fields using CO1 DNA barcodes.
