Genetic Relationships of Rana amurensis Based on Mitochondrial Cytochrome b Gene Sequences

  • Inter- and intraspecific genetic relationships between Rana amurensis from Korea and Russia and other brown frogs were investigated by nucleotide sequence of a 504 base pair (bp) fragment of the mitochondrial cytochrome b gene. Nucleotide sequence similarities among Korean populations of R. amurensis ranged from 99.6% to 97.6% and 98.8% within Russian populations. The nucleotide sequence similarity between Korean and Russian R. amurensis ranged from 86.9% to 85.5%. Based on Kimura-2-parameter distance, the sequence divergence between R. amurensis from Korea and Russia was 16.18% and 18.04% among other related brown frogs. interspecific sequence divergences among R. amurensis and other related brown frogs diverged by 20.3%. Using an estimate of 2-4% mitochondrial DNA sequence divergence per million years, Korean and Russian R. amurensis diverged about 8 to 4 million years ago (Mya) and other brown frogs diverged about 9 to 5 Mya from ancestral frogs and distributed from North Asia to Sakhalin in a short time. In the neighbor-joining and UPGMA tree R. amurensis was clustered into two groups with Korean and Russian populations and the other brown frogs were grouped separately with diverged trichotomous clusters (R. dybowskii and R. pirica, R. okinavana and R. tsushimensis, and R. japonica and R. longicrus).

복숭아순나방(Grapholita molesta) 성페로몬 트랩에 포획된 미동정 나방의 발생패턴과 판별 분자지표 (Occurrence Pattern of an Unidentified Moth Captured by Sex Pheromone Trap of the Oriental Fruit Moth, Grapholita molesta, and Its Discriminating Molecular Markers)

  • 복숭아순나방(Grapholita molesta) 성페로몬 트랩에 미동정 나방이 포획되었다. 이들은 주로 봄에 채집되었으며 형태적으로 복숭아순나방과 유사하나 비교적 몸 크기가 커서 육안으로 쉽게 구별할 수 있다. 이들의 발생 패턴도 4-5월의 복숭아순나방 월동세대 발생과 유사하였다. 이 기간 동안 이들은 사과원 내부 보다는 주로 과수원에서 떨어진 곳에 설치된 페로몬 트랩에 집중적으로 포획되었다. 5월 이후 이들 미동정 나방은 복숭아순나방 페로몬 트랩에 더 이상 포획되지 않았다. 이 미동정 나방과 기존의 복숭아순나방과의 계통 분류학적 판별을 위해 분자마커가 개발되었다. 미토콘드리아 cytochrome b 영역을 기반으로 선발된 4종의 PCR-RFLP 분자지표는 이 미동정 나방과 복숭아순나방을 구분할 수 있었다.

생산공정 차이에 따른 죽력(竹瀝)이 apoptosis 및 신경세포 보호 효과에 미치는 영향 (Anti-apoptotic and Neuroprotective Effects of Bambusae Caulis in Liquamen Manufactured by Different Production Process)

  • Bambusae Caulis in Liquamen(BCL) has been commonly prescribed for stroke patients in the traditional Oriental medicine. So this study is aims to investigate the anti-apoptotic and neuroprotective effects of Bambusae Caulis in Liquamen(BCL) manufactured by different production process on the focal ischemia induced by intraluminal filament insertion in rats. The focal ischemia was induced by intraluminal filament insertion into middle cerebral artery. The animals were divided into four groups (n=15 in each group). The ischemia induced and not treated group : Control group, the ischemia induced and oral medication of the three kinds of BCL : BCL-A group, BCL-B group, BCL-C group. BCL-A was produced by heating at a low temperature$(250^{\circ} C)$ in electric kiln and filtering. BCL-B was produced by heating at a high temperature$(900^{\circ} C{\sim}1,000^{\circ}C)$ in yellow earth kiln and refining and filtering. BCL-C was produced by heating at a low temperature$(400^{\circ} C)$ yellow earth kiln and no refining and filtering. The anti-apoptotic and neuroprotective effects of the oral medication of BCL were observed by Bax, BCL-2, cytochrome c, mGluR5, cresyl violet and ChAT-stain. Our study suggests that BCl-A(was produced by heating at a low temperature in electric kiln and filtering) and BCL-C(was produced by heating at a low temperature in yellow earth kiln and no refining and filtering) show anti-apoptotic and neuroprotective effects on the focal ischemia induced by intraluminal filament insertion in rats and BCL-C is more effective than BCL-A.

  • Aspergillus awamori var. fumeus가 균체 밖으로 생산하느 aflatoxin 분해인자의 효소적 특성을 조사 하기위하여 그 배양 여액을 aflatoxin B1과 반응시킨 결과 1시간 동안 60의 기질을 분해하였다. 반응속도는 초기에 가장 높았으며 시간이 지날 수록 낮아졌다. 반응속도와 기질농도의 관계는 이중역수 식에서 직선을 보이며 겉보기 $K_m$$10.2{\mu}M$로 측정되었다. 최적온도 및 최적 pH는 각각 $30^{\circ}C$ 및 5로 나타났다. 반응은 분자상 산소를 요구하며 $Co^{2+}$에 의해서 촉진되나 $Fe^{2+}$, $Ca^{2+}$, $Zn^{2+}$, $Cu^{2+}$, 및 $Ba^{2+}$ 등의 이온에 의하여 저해되었다. 또한 KCN과 metyrapone에 의하여 저해되나, $NaIO_4$, cytochrome C 및 NADPH에 의한 영향은 관찰되지 않았다.

Ethanol이 Trichloroethylene 대사효소의 활성도와 유도성에 미치는 영향 (Effects of Ethanol on the Activities and Inducibility of Trichloroethylene Metabolic Enzyme System in Rat Liver)

  • This study was performed to find out the influences of ethanol on the metabolism of trichloroethylene(TRI) in rats. TRI in corn oil at the dosage of 150, 300, 600 mg/kg was injected peritoneally once a day for two days to two groups. In one group ethanol(4 g/kg) was taken orally 30 minutes before TRI injection, and the other group ethanol was not. The results of experiments are as follows: 1. The contents of cytochrome P-450 and $b_5$ had inverse relationship with in-jected TRI amounts in both groups. 2. The activity of NADPH P-450 reductase was decreased slowly in TRI injected group related with TRI amount, but decreased drastically in the group pretreated with ethanol. 3. The activity of NADH $b_5$ reductase had relationship with injected nt amount , but the statistical significance was found only in the groups of 300 and 600 mg/kg of TRI injected without relevance to ethanol when compared with the group that was not injected. 4. The activity of ADH was more decreased and ALDH activity was more increased in groups that TRI injected and ethanol was pretreated with ethanol groups than in group without any treatment. These results suggest that ethanol may inhibit epoxide formulation, the first step of TRI metabolism, and change from TCE-OH to TCA also.

제주 연안해역 어란의 분자동정과 분포양상 (Molecular Identification and Distribution Pattern of Fish Eggs Collected around Jejudo Island)

  • 이 연구는 제주도 주변 4개의 정점에서 채집된 어란의 월별 종조성 및 출현량을 알아보기 위해 실시하였다. 현장조사는 2006년 8월부터 2007년 7월까지 봉고네트를 이용하여 월별로 표층채집을 하였으며, 종동정은 채집된 어란의 미토콘드리아 cytochrome b 유전자 (cyt b) 염기서열을 성어 염기서열과 상호 비교함으로써 이루어졌다. 채집된 어란은 총 43개의 분류군으로 분류되었고, 이중 34종 (6목 24과 33속)은 종 (species)까지 동정이 이루어졌다. 나머지 9종 가운데 4종은 과 (family)까지 동정할 수 있었고, 5종은 미동정되었다. 지역별로 제주항 근해에서는 23개의 분류군으로 가장 많은 분류군이 나타났고, 성상포 근해에서 21개, 서귀포항 근해에서 19개, 차귀도 근해에서는 18개의 분류군이 출현하였다. 시기별로는 2006년 9월에 15개의 분류군으로 가장 많이 나타났고, 12월과 7월에 3개의 분류군으로 가장 적게 나타났다. 동정이 이루어진 34종 중 멸치 (E. japonicus)와 노랑벤자리 (C. japonicus)가 가장 높은 출현 빈도를 보였으며, 멸치는 표층수온 $14.0{\sim}28.6^{\circ}C$에서, 노랑벤자리는 $14.9{\sim}20.5^{\circ}C$에서 출현하였다. 이번 연구에서 사용된 cyt b 유전자의 PCR 증폭 성공률은 68.3%였으며, 종동정률은 79.1%였다.

Monitoring conservation effects on a Chinese indigenous chicken breed using major histocompatibility complex B-G gene and DNA Barcodes

  • Objective: We report monitoring conservation effect for a Chinese indigenous chicken (Langshan) breed using major histocompatibility complex (MHC) and DNA barcords. Methods: The full length of MHC B-G gene and mitochondrial cytochrome oxidase I (COI) gene in generations 0, 5, 10, 15, 16, and 17 was measured using re-sequencing and sequencing procedures, respectively. Results: There were 292 single nucleotide polymorphisms of MHC B-G gene identified in six generations. Heterozygosity (He) and polymorphic information content (PIC) of MHC B-G gene in generations 10, 15, 16, and 17 remained stable. He and PIC of MHC B-G gene were different in six generations, with G10, G15, G16, G17 >G5>G0 (p<0.05). For the COI gene, there were five haplotypes in generations 0, 5, 10, 15, 16, and 17. Where Hap2 and Hap4 were the shared haplotypes, 164 individuals shared Hap2 haplotypes, while Hap1 and Hap3 were the shared haplotypes in generations 0 and 5 and Hap5 was a shared haplotype in generations 10, 15, 16, and 17. The sequence of COI gene in 6 generations was tested by Tajima's and D value, and the results were not significant, which were consistent with neutral mutation. There were no differences in generations 10, 15, 16, and 17for measured phenotypic traits. In other generations, for annual egg production, with G5, G10, G15, G16, G17>G0 (p<0.05). For age at the first egg and age at sexual maturity, with G10, G15, G16, G17>G5>G0 (p<0.05). Conclusion: Combined with the results of COI gene DNA barcodes, MHC B-G gene, and phenotypic traits we can see that genetic diversity remained stable from generations 10 to 17 and the equimultiple random matching pedigrees conservation population conservation effect of Langshan chicken was effective as measured by these criteria.

Effect of TSHAC on Human Cytochrome P450 Activity, and Transport Mediated by P-Glycoprotein

  • TSAHC [4'-(p-toluenesulfonylamido)-4-hydroxychalcone] is a promising antitumorigenic chalcone compound, especially against TM4SF5 (four-transmembrane L6 family member 5)-mediated hepatocarcinoma. We evaluated the potential of TSAHC to inhibit the catalytic activities of nine cytochrome P450 isoforms and of P-glycoprotein (P-gp). The abilities of TSAHC to inhibit phenacetin O-deethylation (CYP1A2), coumarin 6-hydroxylation (CYP2A6), bupropion hydroxylation (CYP2B6), amodiaquine N-deethylation (CYP2C8), diclofenac 4-hydroxylation (CYP2C9), omeprazole 5-hydroxylation (CYP2C19), dextromethorphan O-demethylation (CYP2D6), chlorzoxazone 6-hydroxylation (CYP2E1), and midazolam 1'-hydroxylation (CYP3A) were tested using human liver microsomes. The P-gp inhibitory effect of TSAHC was assessed by [$^3H$]digoxin accumulation in the LLCPK1-MDR1 cell system. TSAHC strongly inhibited CYP2C8, CYP2C9, and CYP2C19 isoform activities with $K_i$ values of 0.81, 0.076, and $3.45{\mu}M$, respectively. It also enhanced digoxin accumulation in a dose-dependent manner in the LLCPK1-MDR1 cells. These findings indicate that TSAHC has the potential to inhibit CYP2C isoforms and P-gp activities in vitro. TSAHC might be used as a nonspecific inhibitor of CYP2C isoforms based on its negligible inhibitory effect on other P450 isoforms such as CYP1A2, CYP2A6, CYP2B6, CYP2D6, CYP2E1, and CYP3A.

냉이(Capsella bursa-pastoris)에탄을 추출물의 유리라디칼 소거 및 Xathine Oxidase 저해활성 (Free Radical Scavenging Activities and Inhibitory Effects on Xanthine Oxidase by Ethanol Extract from Capsella bursa-pastoris)

  • 냉이에탄올 추출물중의 항산화활성을 나타내는 물질의 특성을 조사하기 위해 각종 용매로 분획을 실시하여 헥산 분획(Fr.H), 에테르 분획(Fr.E), 에틸아세테이트 분획(Fr.EA), 부탄올 분획(Fr.B), 물 분획(Fr.D)의 5분획을 얻었다. 부탄을 분획은 가장 높은 수소공여능 및 과산화 지질 형성 억제 효과를 보인 반면, xanthine/xanthine oxidase-cytochrome c 환원 반응계에 의한 superoxide radical의 소거능에 있어서는 에테르 분획이 가장 높은 활성을 나타내었다. 또한 각 분획의 xanthine oxidase에 대한 저해효과를 측정한 결과 에테르분획이 가장 큰 저해 효과를 보였으며 $IC_{50}$값은 $5.65\;{\mu}g$이었다. 이러한 결과로 부터 에테르 분획의 높은 superoxide radical 소거능이 xanthine oxidase에 의한 라디칼 형성계에 대한 저해 활성으로 부터 기인한 것임을 알 수 있었다. 각 분획의 xanthine oxidase에 대한 저해 효과는 Fr.B

The Roles of Kupffer Cells in Hepatocellular Dysfunction after Femur Fracture Trauma in Rats

  • The aim of this study was to investigate the effects of trauma on alterations in cytochrome P450 (CYP 450)-dependent drug metabolizing function and to determine the role of Kupffer cells in hepatocellular dysfunction. Rats underwent closed femur fracture (FFx) with associated soft-tissue injury under anesthesia, while control animals received only anesthesia. To deplete Kupffer cells in vivo, gadolinium chloride (GdCl$_3$) was injected intravenously via the tail vein at 7.5 mg/kg body wt., 1 and 2 days prior to FFx surgery. At 72 h after FFx, serum alanine aminotransferase (ALT) activity was increased, and this increase was attenuated by GdCl$_3$ pretreatment. Serum aspartate aminotransferase (AST) and lipid peroxidation levels were not changed by FFx. Hepatic microsomal CYP 450 content and aniline p-hydroxylase (CYP 2E1) activity were significantly decreased; decreases that were not prevented by GdC1$_3$. The level of CYP 2B1 activity was decreased by Kupffer cell inactivation, but not by FFx. There were no significant differences in the activities of CYP 1A1, CYP 1A2 and NADPH-CYP 450 reductase among any of the experimental groups. Our findings suggest that FFx trauma causes mild alterations of hepatic CYP 450-dependent drug metabolism, and that Kupffer cells are not essential for the initiation of such injury.
