Two Cyanidin compound from the Fruits of Acanthopanax divaricatus var. albeofructus

  • Acanthopanax divaricatus var. albeofructus is one of the indigenous medicinal plant and the fruits of Acanthopanax spp. used as a remedial for "wipe out evil wind". Two anthocyanin were isolated from the fruits of Acanthopanax divaricatus var. albeofructus. Their structures were elucidated as cyanidin 3-lathyroside (1) and cyanidin 3-galactoside (2) by chemicophysical and spectroscopic analysis. And also, four chemical, syringin, chlorogenic acid, caffeic acid and acanthoside D were identified. Both anthocyanide were isolated for the first time from Acanthopanax species. cyanidin 3-lathyroside is one of the rare anthocyanin in natural resources.

물리적 처리조건 변화에 따른 아로니아(Aronia melancocarpa) 유래 안토시아닌 함량변화 특성 (Changes in Anthocyanin Content of Aronia (Aronia melancocarpa) by Processing Conditions)

  • 식품의 가공 시 농축, 건조, 살균 등을 목적으로 가열공정이 필수적으로 요구된다. 또한 다양한 원료가 혼합될때 원료 본연의 pH는 타 혼합원료의 영향으로 변화되게 된다. 본 연구는 아로니아의 가공조건에 따른 안토시아닌의 함량감소에 영향에 미치는 인자인 온도, pH, 가온시간, 수분함량 및 citric acid 첨가 등에 따른 안토시아닌 함량감소 억제효과를 검토하였다. 가공공정에서 안토시아닌 함량을 감소시키는 요인으로는 pH, 온도, 가온시간, 수분함량이 가장 영향을 주었다. 아로니아에 함유된 안토시아닌은 산성에서 안정하다고 보고된 바가 있으나, 수분함량 9%이하의 분말의 경우 pH 6이하에서는, 가열온도와 시간에 따른 안토시아닌 함량감소가 고수분함량 시료에 비하여 적었다. 가열온도에 따른 총 안토시아닌 함량변화는 $40^{\circ}C$이하에서는 안정한 편이나, $40^{\circ}C$ 이상 $60^{\circ}C$는 감소속도가 서서히 증가하고, $60^{\circ}C$ 이상에서 급격하게 감소속도가 증가되었다. 일반적으로 사용하는 열풍순환건조기를 이용한 건조에서 건조효율을 참작한 적정최저온도라 할 수 있는 $65^{\circ}C$에서 수분함량 9%의 아로니아 분말은 40시간 가열처리로 50%가 소실되었다. 이러한 결과는 아로니아를 건조할 때 유효성분인 안토시아닌의 감소를 억제하기 위하여 가열온도를 $40^{\circ}C$ 이상에서 장시간 건조하지 않아야 함을 나타낸다. Juice 상태의 $65^{\circ}C$ 수용액 중에서는 10시간 만에 50%가 감소되었으며, juice 상태의 $85^{\circ}C$ 수용액 중에서는 4시간만에 50%가 감소되었다. 저수분 함유 조건에서 아로니아 분말에 함유된 안토시아닌의 안정성에 영향을 미치는 것은 온도 및 시간 변수가 가장 큰요인이며, 이러한 요인은 아로니아 뿐만 아니라 안토시아닌을 함유하는 과일류에 공통으로 적용될 것으로 생각된다. 본 연구는 아로니아에 함유된 안토시아닌의 함량을 다양한 방법을 이용하여 분석하였다. 총 안토시아닌 분석은 이전부터 사용하던 UV-VIS 분광광도계를 이용하는 방법을 사용하였으며, 이 방법은 최근 방법인 LC-MS/MS 분석법에 비하여 신속하다는 장점은 있지만 정확도가 떨어지는 것으로 사료된다. 특히 안토시아닌 종류(Cyanidin-3-Galactoside, Cyanidin-3-Glucoside, C3-Arabinoside)별 함량의 구분도 불가능하다. 이러한 분석의 어려움으로 2013년, 2015년 특허(Jang and Park et al., 2013; Lee and Jung et al., 2015)에서도 아로니아에 함유된 주종 안토시아닌으로 Cyanidin-3-glucoside를제시하고 있으며, 본 연구에서는 LC-MS/MS 분석결과 아로니아에 함유된 주종 안토시아닌은 Cyanidin-3-galactoside인 것으로 확인되었다.

Simultaneous analysis of anthocyanins and flavonols in various flower colors of Rhododendron schlippenbachii (royal azalea)

  • Anthocyanins contained in Rhododendron schlippenbachii (royal azalea) are expressed in a variety of colors and affect flower colors. R. schlippenbachii flowers of seven colors (white, red group: pink, deep pink, red, purple group: light purple, purple, deep purple) were collected from the garden around KT&G building in the college of agriculture and life science. Seven types of anthocyanins [cyanidin 3-O-diglucoside, cyanidin 3-O-arabinoside-5-O-glucoside, cyanidin 3-O-galactoside, peonidin 3-O-arabinoside-5-O-glucoside, cyanidin 3-O-glucoside, cyanidin 3-O-(6"-O-malonyl) arabinoside, cyanidin 3-O-(6"-O-coumaroyl) glucoside] turned out to be from the cyanidin and peonidin series in R. schlippenbachii flowers. Also, seven types of flavonols [azaleatin 3-O-glucoside, azaleatin 3-O-arabinoside, azaleatin 3-O-rhamnoside, quercetin 3-O-galacatoside, quercetin 3-O-glucoside, quercetin 3-O-arabinoside, quercetin 3-O-rhamnoside] were identified in R. schlippenbachii flowers. Total anthocyanin amounts decreased in R. schlippenbachii flowers in the following order: 'deep pink' (8.07) > 'red' (6.37) > 'pink' (5.35) > 'deep purple' (0.78) > 'purple' (0.43) > 'light purple' ($0.22mg{\cdot}g^{-1}$ dry weight, DW) > 'white' (not detected). Total flavonol amounts decreased in the following order: 'pink' (97.78) > 'deep pink' (63.79) > 'deep purple' (61.98) > 'white' (57.58) > 'light purple' (47.06) > 'purple' (46.76) > 'red' ($7.60mg{\cdot}g^{-1}$ dry weight, DW). This study provided the quantitative and qualitative information for the variation of anthocyanin and flavonol compounds in R. schlippenbachii flowers. Furthermore, this information can contribute to the identification of anthocyanin and flavonol compounds in other Rhododendron flowers.

Enhancing the Effect of Aronia Extract on Hyaluronic Acid Synthesis through Liposome Formation

  • Background and objective: Aronia melanocarpa, called black chokeberry, is a natural product belonging to the family rosaceae, and is known to contain polyphenolic antioxidants including cyanidin-3-galactoside, cyanidin-3-arabinoside, cyanidin-3-xyloside, and cyanidin-3-glucoside Because of the abundance of anthocyanins, Aronia has been studied to be used in various industries. Methods: Aronia melanocarpa extract was treated 24 hours a day to RAW 264.7 cells with inflammations induced by LPS. After extracting total RNA, the amount of inflammatory cytokine expression was measured using RT-PCR. After processing the Aronia liposome using Aronia extract and the layer-by-layer electrostatic deposition method in keratinocyte cells at the same time, we checked the synthesis of Hyaluronic acid enhanced through the formation of Aronia liposome using ELISA. Results: The treatment of Aronia extract in inflammation-induced RAW 264.7 cells conducted to check the anti-inflammatory efficacy of Aronia extract inhibited inflammatory cytokines including TLR4, TNF-α, IL-1β, COX-2, and iNOS and increased the mRNA expression of HAS2 genes related to moisturizing. Based on the anti-inflammatory and moisturizing effect of Aronia extract, the Aronia liposome technology was introduced to Aronia extract to produce Aronia liposome. Conclusion: The liposome formation of Aronia extract is expected to be used as a functional material in treating various inflammatory skin diseases by controlling the moisture content of the corneocytes by increasing the expression rate of genes associated with the synthesis of hyaluronic acid, while retaining the efficacy of its components.

Anthocyanin Profiling and Radical Scavenging Activity of Selected Pigmented Rice Varieties

  • BACKGROUND: Anthocyanins have been recognized as health-enhancing substances due to their antioxidant activity, anti-inflammatory, anticancer, and hypoglycemic effects. The objective was to identify anthocyanins-rich rice grains for the development of functional foods and/or functional food colorants METHODS AND RESULTS: Rice grains of one black and three red-hulled rice varieties were extracted with acidified 80% aqueous methanol. The antioxidant activity of the methanolic extracts was screened on TLC plates and in an in vitro assay using DPPH (1, 1-diphenyl-2-picrylhydrazyl) as a free radical source. Red-hulled rice varieties exhibited higher antioxidant activity (88%, 1 mg/mL) than black rice (67%, 1 mg/mL). Among the red-hulled varieties tested, rice variety SSALBYEO54 (901452) was the most active (72%, 0.5 mg/mL). Rice extracted anthocyanin compounds were analyzed by HPLC-DAD-FLD and LC-MS/MS. Red-hulled varieties comprised cyanidin-3-glucoside in addition to ferulic acid esters, apigenin and kaempferol glycosides. CONCLUSION(s): Anthocyanins identified in the black rice variety were cyanidin-7-O-galactoside, cyanidin-3-Oglucoside, cyanidin-3'-O-glucoside, cyanidin-5-O-glucoside, cyanidin-3, 7-O-diglucoside, cyanidin-3, 5-O-diglucoside and peonidin-4'-O-glucoside. The results of this study show that the black rice (IT212512) and red-hulled rice variety SSALBYEO54 (901452) contain notable antioxidant activity for potential use in nutraceutical or functional food formulations.

Sustainable water extraction of anthocyanins in aronia (Aronia melanocarpa L.) using conventional and ultrasonic-assisted method

  • The demand for sustainable extraction of bioactive compounds from food matrices has been increasing. Water extraction of anthocyanins in aronia was investigated using conventional and ultrasonic-assisted methods. The optimum extraction conditions for the conventional method included a sample-to-water ratio of 1:40 g/mL, extraction temperature 71℃, and extraction time of 39 min. The optimized conditions for ultrasonic-assisted extraction were a sample-to-water ratio of 1:40 g/mL, extraction temperature 80℃, extraction time of 20 min, and an amplitude of 87.2 ㎛. The anthocyanin contents of the two extracts were 155.32 and 158.02 mg/100 g fresh weight, respectively. The major anthocyanins were cyanidin 3-galactoside (65% of the total) and cyanidin 3-arabinoside (30% of the total). The contents of individual anthocyanins and phenolic acids were not significantly different between the two optimized extracts.

유색 농산물 중 안토시아닌과 폴리페놀 함량 비교 연구 (A Comparative Study on Anthocyanin and Polyphenol Contents in Colored Agricultural Products)

  • 본 연구에서는 인터넷 쇼핑몰에서 유통되는 컬러푸드 중 블랙푸드 76건을 대상으로 폴리페놀 17종 및 안토시아닌 5종을 동일한 전처리 방법으로 HPLC, HPLC-MS/MS 분석하여 정량 및 정성시험을 실시하였다. 분석대상 폴리페놀은 gallic acid, protocatechuic acid, chlorogenic acid, vanillic acid, caffeic acid, syringic acid, p-coumaric acid, t-ferulic acid, t-cinnamic acid 등 9종의 phenolic acid와 (+)catechin, syringic aldehyde, rutin, epicatechin gallate, naringin, luteolin, naringenin, kaempferol 등 8종의 flavonoid로 총 17종의 폴리페놀을 동시분석 하였고, 안토시아닌은 delphinidin-3-glucoside, delphinidin-3-rutinoside, cyanidin-3-galactoside, cyanidin-3-glucoside, cyanidin-3-arabinoside 등 5종을 동시분석 하였다. 폴리페놀 17종의 총 함량은 서리태 $255.1{\pm}7.5{\mu}g/g$, 서목태 $275.8{\pm}5.3{\mu}g/g$, 흑미 $78.5{\pm}4.6{\mu}g/g$, 흑임자 $75.8{\pm}3.2{\mu}g/g$, 블루베리 $143.3{\pm}5.5{\mu}g/g$, 아로니아 $195.2{\pm}4.9{\mu}g/g$, 블랙커런트 $131.6{\pm}3.2{\mu}g/g$로 나타나 서목태 > 서리태 > 아로니아 > 블루베리 > 블랙커런트 > 흑미 > 흑임자 순으로 높은 함량을 나타냈다. 안토시아닌 5종의 총 함량은 서리태 $82.4{\pm}17.2{\mu}g/g$, 서목태 $95.2{\pm}6.1{\mu}g/g$, 흑미 $74.1{\pm}9.7{\mu}g/g$, 흑임자 불검출, 블루베리 $110.8{\pm}1.9{\mu}g/g$, 아로니아 $218.9{\pm}6.1{\mu}g/g$, 블랙커런트 $209.7{\pm}4.0{\mu}g/g$로 나타나 아로니아 > 블랙커런트 > 블루베리 > 서목태 > 서리태 > 흑미 순으로 높은 함량을 나타났다. 본 실험결과 폴리페놀은 서목태, 안토시아닌은 아로니아가 가장 높은 함량을 나타내었으며 향후 유색 농산물을 활용한 기능식품 개발이 필요한 것으로 판단된다.

Photoprotective Potential of Anthocyanins Isolated from Acanthopanax divaricatus Var. albeofructus Fruits against UV Irradiation in Human Dermal Fibroblast Cells

  • Ultraviolet (UV) A penetrates deeply into the skin and induces the generation of reactive oxygen species (ROS) causing damage to fibroblasts, which leads to aging of the skin. However, the body has developed an antioxidant defence system against the harmful effects of ROS. Enzymes such as superoxide dismutase (SOD) and catalase (CAT) play critical roles on the removal of excess ROS in living organisms. In this study, the antioxidant activities of anthocyanins (cyanidin 3-galactoside and cyanidin 3-lathyroside) from Acanthopanax divaricatus var. albeofructus (ADA) fruits were investigated by xylenol orange, thiobarbituric acid reactive substances (TBARS), and antioxidant enzyme assay. As a result, generation of $H_2O_2$ and lipid peroxide induced by UVA-irradiation in human dermal fibroblast (HDF-N) cells was reduced by treatment of anthocyanins. Also, augmented enzyme (SOD and CAT) activities were observed in UVA-irradiated cells when treated with anthocyanin. In conclusion, the results obtained show that anthocyanins from ADA fruits are potential candidates for the protection of fibroblast against the damaging effects of UVA irradiation. Furthermore, anthocyanin may be a good candidate for antioxidant agent development.

추출용매에 따른 아로니아 추출물의 생리 활성 (Physiological activities of Aronia melanocarpa extracts on extraction solvents)

  • 본 연구에서는 페놀성물질 및 안토시아닌을 함유하고 있어 다양한 생리활성을 가지는 아로니아의 산업적 이용 증대 및 기능성식품 소재 개발을 목적으로 열수추출, 50% 에탄올 추출 및 50% 메탄올을 사용하여 추출물을 제조하였으며 추출용매에 따른 생리 활성을 조사하였다. 추출수율은 50% 에탄올, 열수 및 50% 메탄올 추출물 순으로 나타났으며, 총 당 함량은 35.56~37.68 g/100 g으로 유의적인 차이는 나타나지 않았다. 총 안토시아닌 함량은 50% 메탄올 추출물이 395.10 mg/100 g으로 가장 높은 함량을 나타내었으며, 50% 에탄올 및 열수추출물이 각각 318.61 mg/100 g과 252.82 mg/100 g이었다. 50% 메탄올 추출물의 안토시아닌조성은cyanidin-3-O-galactoside, cyanidin-3-O-arabinoside 및 cyanidin-3-O-glucoside순이었으며, 100 g당 각각 364.65 mg, 163.06 mg 및 35.69 mg으로 가장 높은 함량을 나타내었다. 총 페놀 함량은 50% 에탄올 및 50% 메탄올 추출물에서 각각 121.38 mg/g 및 122.43 mg/g으로 열수추출물 80.14 mg/g보다 유의적으로 높은 함량을 나타내었으며, 총 플라보노이드 함량은 총 페놀 함량과 유사한 경향을 나타내었다. ORAC은 열수 추출물, 50% 에탄올 및 50% 메탄올 추출물에서 각각 $715.66{\mu}M/g$, $768.15{\mu}M/g$$780.77{\mu}M/g$로 나타나 50% 에탄올 및 50% 메탄올 추출조건의 항산화 활성이 높게 나타났다. DPPH radical 소거활성은 50% 에탄올 및 50% 메탄올 추출물이 $100{\sim}1,000{\mu}g/mL$ 농도에서 각각 7.96~70.01% 및 8.90~69.21%로 높은 활성을 나타내었으며, superoxide radical 소거활성은 모든 추출물에서 농도가 증가함에 따라 소거활성이 증가하였다. FRAP는 $100{\sim}1,000{\mu}g/mL$ 농도에서 50% 에탄올 추출물이 $57.14{\sim}817.87{\mu}M$이었고 50% 메탄올 추출물이 $67.32{\sim}812.78{\mu}M$로 나타나 높은 함량을 보여주었다. Tyrosinase 저해활성은 50% 에탄올 추출물이 $100{\sim}1,000{\mu}g/mL$의 농도에서 23.03~33.82%로 가장 우수한 활성을 나타내었다. 인간 자궁경부암세포주인 HeLa에 암세포생육 저해활성을 분석한 결과 50% 에탄올 추출물이 $100{\sim}1,000{\mu}g/mL$ 농도에서 6.58~76.86%로 유의적으로 높은 저해활성을 나타내었다. 따라서, 50% 에탄올 추출방법이 아로니아의 생리 활성이 우수한 추출물 제조방법으로 적합하였으며 기능성 식품 소재 개발에 있어 산업적으로 적용 가능할 것으로 사료된다.

블루베리(Vaccinium corymbosum L.) 품종별 안토시아닌 조성 및 함량 (Compositions and Contents Anthocyanins in Blueberry (Vaccinium corymbosum L.) Varieties)

  • BACKGROUND: Anthocyanins, potential health-promoting compounds, were major natural pigment in the blueberry (Vaccinium corymbosum L.). The objectives of this study was to investigate anthocyanin glycosides in the blueberry varieties.METHODS AND RESULTS: A total of seventeen anthocyanins were identified from highbush blueberry using HPLC (representatives, 530 nm) and ESI-MS in positive ion mode. The individual anthocyanins are containing cyanidin, delphinidin, malvidin, peonidin, and petunidin moieties which are acylated with aliphatic acid (acetic acid) and conjugated with sugar moieties (arabinose, galactose, and glucose). Among them, delphinidin 3-O-galactoside (D3Ga), peonidin 3-O-glucoside (Pn3G) + malvidin 3-O-galactoside (M3Ga) were major compounds in varieties. Total anthocyanins were found the highest level in 'Elizabeth' (1,406.3 mg/100 g dry weight) which was 3-fold higher than 'Darrow' (465.7). Especially, D3Ga was presented the 32% of total anthocyanins followed by Pn3G + M3Ga (20%) in 'Elizabeth'.CONCLUSION: This result was showed as valuable information regarding nutritional properties of the different varieties of the highbush blueberry.
