Comparison of the Antioxidative Effects and Content of Anthocyanin and Phenolic Compounds in Different Varieties of Vitis vinifera Ethanol Extract

  • This study was a quantitative HPLC analysis of four anthocyanins and five phenolic compounds contained in the skins, vines and seeds of the Campbell Early, Muscat Bailey A and Neo Muscat grape varieties. In the phenolic analysis, the seeds of the Campbell Early were found to contain 1.9, 1.8 and 1.6 times higher quantities of gallic acid, catechin and epicatechin relative to other grape seeds. Three anthocyanins, cyanidin, peonidin and pelargonidin, were also found to be higher in the skins of the Campbell Early relative to other grape skins. Therefore, the Campbell Early is the most useful grape variety with regard to the extraction of these six compounds from these grape seeds and skins. The free radical scavenging effects of grape seeds were also compared, and the results indicated that the Campbell Early seeds were most effective among them.

Determination of C3G Content in Blackish Purple Rice using HPLC and UV-Vis Spectrophotometer

  • Cyanidin 3-glucoside (C3G) content contained in the grains of blackish purple rice varieties, Heugjinjubyeo, Kilimheugmi, Heugnambyeo, Sanghaehy-anghyeolla, and the progenies derived from their crosses was evaluated by HPLC and UV-Vis spectroscopy. C3G content was higher in the range of 10-30% by using UV-Vis method compared to HPLC method. A significant linear relationship was, however, observed between two analytical methods. The correlation coefficient was 0.98. Thus, this results suggested that it would be able to use UV-Vis spectroscopy to determine C3G content which does not demanded precise value like selection.

The Anthocyanin Components and Cytotoxic Activity of Syzygium cumini (L.) Fruits Growing in Egypt

  • Four anthocyanins were isolated from the acidic alcoholic extract of Syzygium cumini fruits growing in Egypt: Pelargonidin-3-O-glucoside, pelargonidin-3,5-O-diglucoside, cyanidin-3-O-malonyl glucoside, and delphenidin-3-O-glucoside. They were identified by the chromatographic, TLC and PC, and spectral analyses, UV, $^1$H-NMR and FAB/MS. The fruits were found to contain 0.03 gm % anthocyanins calculated on fresh weight basis calculated by spectrophotometric assay. Cytotoxic activity of total alcoholic extract of the fruits was performed against several types of tumor cell lines using the SRB assay. The tested extract exhibited significant cytotoxic activity for MCF7 (breast carcinoma cell line) (IC$_{50}$= 5.9 ${\mu}$g/mL), while the IC$_{50}$ was > 10 ${\mu}$g/mL for both Hela (Cervix carcinoma cell line), HEPG2 (liver carcinoma cell line), H460 (Lung carcinoma cell line) and U251 (Brain carcinoma cell line).

Effects of Different Transplanting Dates and Agroclimatic Zones on Quality of Brown Rice and Yield of a Pigmented Rice Variety 'Josaengheugchal' (이앙시기와 농업기후지대의 차이가 조생흑찰의 현미 품질과 수량에 미치는 영향)

  • This experiment was carried out to clarify the effect of various transplanting dates and agroclimatic zones on quality of brown rice and yield of a pigmented rice variety ‘Josaengheugchal’. The black density of brown rice in Jecheon (central inland region) showed a slight difference among the transplanting dates, but in Cheongwon (western sobaek inland region) that had increased at the transplanting dates of June 10th to 20th. cyanidin 3-glucoside (C3G) content at the same transplanting date in Jecheon was higher than Cheongwon, and the overall C3G content was increased at the later transplanting dates. The optimum transplanting date estimated by brown rice yields and C3G content was May 11th (491 kg/10a) in Jecheon and June 16th (468 kg/10a) in Cheongwon.

Grain Quality of New Large-Grain Pigmented Rice Variety, "Daeripjamibyeo" (대립자미벼의 품질특성)

  • Daeripjami, a novel black rice variety developed by conventional breeding has high contents of Cyanidin-3-glucoside(C3G) and a more strong antioxidant than normal black rice. This study aimed at informing ordinary people of the superiority of Daeripjami, a variety of rice proven to be superb physiologically and valuable as natural coloring, and making it popular by examing the physicochemical features of the rice. Recently, an increasing number of people in modern society are suffering from various adult diseases and atopic diseases. Therefore, in order to resolve the problem by making people consume more C3G, the main coloring contained in functional rice Daeripjami. The general components of Daeripjami, Superjami, Heugjinju and Ilpum were compared, As for water content, Ilpum took first place, followed by Heugjinju, Daeripjami and Superjami, As for crude protein and fat content, Heugjinju was highest, followed by Daeripjami, Superjami and Ilpum. This indicated that Daeripjami likely has better cooking qualities than Heugjinju. Amylose content turned out to be related with the volume, stickness and retrogradation of cook rice. As Heugjinju contained more amylose than Daeripjami, the latter was expected to have better eating quality than the former. The 1000 grain weight of daeripjami was 1.67 times heavier than that of Heugjinju. The C3G content of Daeripjami is 3.49 times higher compared with that of Heugjinju. As for total polyphenol and electron donating ability, Daeripjami was high in physiological functionality.

Antioxidant Activities of Rubus coreanus Miquel and Morus alba L. Fruits (복분자와 오디의 항산화 특성)

  • The antioxidant activities of extracts from Rubus coreanus Miquel (black raspberry) and Morus alba L. (mulberry) fruits were investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, 2,2-azino-bis-(3-ethylbenzo-thiazoline-6-sulfonic acid) (ABTS) assay, and reducing power assay. Aqueous mixtures of ethanol, methanol, and acetone were analyzed in order to determine the most effective extraction solvent for the two fruits. Black raspberry and mulberry extracts with the 60:40 acetone-water mixtures (v/v) showed the highest DPPH radical scavenging activities (56.2 and 85.2%, respectively) compared to the other extraction solvents. The 60% acetone extract was finally selected as an extraction solvent and then sequentially fractionated according to solvent polarity. Among the fractions of the two fruits, the ethyl acetate fraction showed the highest antioxidant activity as well as total phenolic and total flavonoid contents. In addition, there were high correlation coefficients between antioxidant activities and their contents. The $EC_{50}$ value of the ethyl acetate fraction from mulberry fruit was 2.2 times lower than that of butylated hydroxytoluene (BHT) in DPPH assay. The major phenolic acid and anthocyanin of the two fruits were protocatechuic acid and cyanidin-3-glucoside, respectively.

Physicochemical properties and storage stability of blueberry fermented by lactic acid bacteria (블루베리 유산균 발효물의 이화학적 특성 및 저장안정성)

  • The physicochemical properties and storage stability of blueberries fermented by lactic acid bacteria were investigated. The viable cell count of lactic acid bacteria slowly increased to 5.96 log CFU/mL after 72 hr of fermentation. The pH decreased whereas titratable acidity increased after fermentation. The contents of total anthocyanin (31.52 mg/100 g) and cyanidin-3-glucoside (C3G) (5.41 mg/100 g) after 72 hr of fermentation were higher than those of non-fermented blueberries (16.10 mg/100 g and 2.21 mg/100 g, respectively). The L and a value decreased, and the b and ${\Delta}$E value increased. The total polyphenol and flavonoid contents of fermented blueberries (2.21 g/100 g and 0.91 g/100 g, respectively) were higher than those of non-fermented blueberries (1.13 g/100 g and 0.49 g/100g, respectively). The DPPH radical scavenging activity and superoxide radical scavenging activity of the fermented blueberries were 30.74%, and 52.76%, respectively. The ferric reducing antioxidant power of the fermented blueberries ($256.42{\mu}M/g$) was higher than that of non-fermented blueberries ($191.52{\mu}M/g$). Anthocyanin and C3G content was stable in fermented blueberries after 42 days of storage. The results suggest that blueberries fermented by lactic acid bacteria have the potential to be functional materials in the food industry.

Variation of Functional Compounds in Leafy Chinese Cabbage Grown Under Different Light Conditions in a Plant Factory (광 조건에 따른 식물공장 재배 쌈 배추의 기능성 성분 변화)

  • A recently developed Chinese cabbage cultivar, Shinhong Ssam, was cultured under different light conditions at a plant factory to evaluate effects of light on the production of functional compounds. The effects of the following combinations of red (R), blue (B), and white (W) LED light were compared to that of fluorescent light: R+B, R+W, and R+B+W. Under R+B light, the Chinese cabbage failed to grow well. In contrast, under R+W, R+B+W, and fluorescent light conditions, cabbages had similar growth rates. Cyanidin, the only anthocyanidin detected, accumulated in cabbages under LED conditions to levels more than 2-fold of that in cabbages grown under fluorescent light. Phenolic acid and flavonol levels varied subtly, whereas caffeic acid was found to be between 70- and 160-fold greater in LED-illuminated cabbages than in those exposed to fluorescent light. The amounts of free amino acids and sugars that affect the taste of vegetables were greater in the cabbages exposed to LED light than in those grown under fluorescent light conditions. In conclusion, R+W LED light produced Shinhong Ssam with greater nutritional value than those grown under R+B+W LED and fluorescent lights.

Anthocyanin and Polyphenol Analysis and Antioxidant Activity of Small Fruit and Berries in Korea (국내 재배 소과류 및 베리류의 안토시아닌 및 폴리페놀 분석)

  • Anthocyanin, polyphenol contents, and antioxidant activities of introduced small fruits and berries varieties were investigated. The anthocyanin and polyphenol contents were different according to the varieties. The highest total anthocyanin was 927.9~1,149.1 mg, $100g^{-1}FW$, and the highest total polyphenol content was 502.2 to 530.5 mg, $100g^{-1}FW$ in black chokeberry. A linear correlation ($r^2$) was shown between DPPH free-radical scavenging activity and total anthocyanin ($r^2=0.7669^{**}$) and phenolic content ($r^2=0.8956^{***}$). The content of quercetin, catechin and epicatechin was higher among the all types of polyphenols in small fruits and berries varieties. The main anthocyanin of blueberry and black currant was delphinidin-3-glucoside, and the major anthocyanin of saskatoon berry and black chokeberry was cyanidin-3-galactoside.

Antioxidant and Anti-inflammatory Effects of Fermented Blackcurrant Fruit Extracts with Saccharomycopsis fibuligera (Saccharomycopsis fibuligera로 발효된 블랙커런트추출물의 항산화 및 항염증 효과)

  • While searching for useful microorganisms, Saccharomycopsis fibuligera which can be used as cosmetic materials were divided from Jeju island's traditional fermented foods. In this study, blackcurrant extract which contains a large amount of anthocyanin glycosides was fermented with S. fibuligera. HPLC analysis was performed to analyze the components of blackcurrant extract (BE) and fermented blackcurrant fruit extract (FBE). As a result, bio-conversion of delphinidin and cyanidin were able to be identified. In order to verify the anti-oxidant effect of BE and FBE, we investigated radical scavenging ability with DPPH and ABTS. In addition, to confirm anti-inflammatory effect, we investigated inhibition effect of nitric oxide (NO) production on lipopolysaccharide (LPS) - stimulated RAW264.7 macrophages, and inhibition effect of the expression of inflammatory-related proteins (iNOS, COX-2) by western blot analysis. As a result, as FBE has anti-oxidant and anti-inflammatory effects, we suggest that it might be used as an active ingredient for cosmetics.
