A Plan for Reducing SAR Call-outs for Fishing Vessels in Korean Waters

  • Korea has a systemic problem with lack of engine maintenance, especially among the Commercial Fishing Vessel fleet. This results in a inordinate percent of SAR call-outs for vessels with engine failure. These SAR call-outs lead to a free tow to shore by KCG or by one of its volunteer associates. Although these tows are not a terrible economic burden on KCG, it is a burden in terms of time and resources mis-allocated. This paper proposes a Commercial Fishing Vessel Examination (CFVE) program modeled after the program run by USCG. It is expected that adopting the CFVE program, KCG may be able to create a culture of safety among fishermen; thus, replicating the USCG's success rates.

개방형 액체대량배양 시스템을 통한 유칼리나무 펠리타 선발목의 대량증식 (Application of Open-type Liquid Culture for Large-scale Production of Mature Plus Tree of Eucalyptus pellita)

  • 유칼리나무 펠리타(Eucalyptus pellita) 선발목(5년생)의 기내 대량증식을 위해 1L 규모로 1) 고체배양(대조구), 2) 액체정치배양, 3) 개방형 액체배양 등 3가지 배양법이 시도 되었고, 그 결과를 토대로 배양규모를 10L로 확대하여 대량배양 가능성을 조사하였다. 1L 배양용기에서 4주간 배양한 결과, 공기가 공급 된 개방형 액체배양에서 가장 좋은 생장 결과를 얻었다. 이는 액체배지에 의한 원활한 염류 공급과 배양용기내 공기의 환기에 의해 얻어진 결과로 생각된다. 배양 규모를 10L로 확대하여 액체대량배양을 실시한 결과, 식물체의 생장은 대조구인 고체배양에 비교하여 마디 수에 있어서 370%, 엽 면적 3.6배, 그리고 신초 길이 3.3배 신장하는 결과를 보였다. 이는 개방형 액체대량배양 시스템이 유칼리나무 클론증식시 생산성을 향상시키는데 적합한 시스템임을 보여주는 결과라고 하겠다.

  • 본 논문은 윈치드럼과 함께 구성되어 있는 수중 케이블의 거동을 해석하기 위해 개발된 조출 모델링을 소개한다. 케이블의 수중 거동은 장력만 영향을 준다고 가정한다. 이러한 가정은 직진 거동을 하는 함정에 의해 예인되는 수중 케이블 거동을 해석하는데 적합하다. 수중 케이블은 절점 위치유한 요소법으로 차분한다. 이 수치기법은 기하학적 비선형성을 표현할 수 있기 때문에 대변형을 동반하는 수중 케이블의 거동 예측에 적합하다고 알려져 있다. 본 논문은 실제 역 실험에서 계측된 수중 예인케이블의 심도 정보를 활용하여 수치 기법의 타당성을 확보한다.

관류형 바이오리액터를 위한 박동 펌프 시스템 개발 (Development of the Pulsatile Pump System for a Perfusion Bioreactor)

  • This research is about the pulsatile pump system utilized in the perfusion bioreactor for the in vitro human tissue culture. A pulsatile pump system which can be applied to the culture of the vascular tissues including blood vessel is developed by using the idea of human heart's blood pumping into organs as followings: culture chamber, a pressurizing device which generates laminar pulsatile flow by controlling the x-sectional area of the culture media delivering tubing, a compliance chamber which supplies the pressuring device with a constant pressure, and a peristaltic pump which circulates the culture media in a circuit ranging from the culture chamber to the compliance chamber. The developed pulsatile pump system shows that a physiology of the human heart's blood pumping including pulsatile pressure waveform of systolic-diastolic pressure is well represented. Not only time domain but also frequency domain characteristics of pulsatile pump system which are necessary for the vascular tissue culture such as pulsatile pressure waveform's shape, the frequency, and the magnitude can be easily generated and manipulated by using the proposed system.

Spirulina platensis의 옥외배양 최적화 및 오염생물 구제 (Optimization of Outdoor Cultivation of Spirulina platensis and Control ofContaminant Organisms)

  • Outdoor cultivation of cyanobacterium Spirulina platensis was carried out for 40 days in a batch mode. A half concentration of the SOT based on the underground water was used as culture medium. Working volume was 5.7 tons with 0.2 m depth. During cultivation, mean water temperature, DO and light intensity were all in proper conditions for the S. platensis growth. The adjustment of pH to over 10 with Na2CO3 and addition of the 1.5% natural salt were very effective to delete contaminant organisms, Chlamydomonas moewusii and Chlorella minutissima occurred one after the other in the culture. The mean productivity of the biomass based on the dry cell weight from 14 to 25 days, after the contaminants were deleted, was 7.8 g ·m–2· d–1, which was relatively high productivity in that a half concentration of the SOT was used for the culture. Underground water used in the culture minimized contaminants invasion and addition of the 1.5% natural salt was effective to delete contaminants as well as acted as mineral supplement in outdoor cultivation of S. platensis. Harvesting using the floating activity of S. platensis was effective from mass floating in day time after overnight without agitation and illumination.

기내배양 사과 대목의 기외 삽목 시 발근과 순화에 미치는 배양조건 및 생장조절물질의 효과 (Effects of Growth Regulators and Culture Environment on ex vitro Rooting and Acclimatization of Apple Rootstock in vitro Propagated)

  • 본 연구는 사과 왜성대목 M.9와 M.26 조직 배양묘를 통기성 마개와 비통기성 마개의 MS배지에서 생장 특성을 비교하기 위해 시험되었다. 또한 배양묘의 생육 변화와 이 배양묘를 발근촉진제 침지 처리 후 기외 삽목을 하여 발근 및 순화를 동시에 시키기 위해 실시하였다. 배양병내의 $CO_2$와 에틸렌의 농도는 통기성 마개 배양병이 비통기성마개 배양병 보다 낮았다. 통기성 마개 배양병내의 온$.$습도 변화는 광조건에 따라 일정하게 반복되었다. 통기성 마개 배양묘의 기공은 외기에서 즉시 관찰 시 닫혀 있었으나 비통기성마개 배양묘의 기공은 외기에서 20분 후 관찰 시에도 열려 있었다. 통기성 마개 배양묘의 엽록체 지수와 잎 면적은 모두 비통기성 마개의 것 보다 높았다. 기외 삽목시 발근율은 M.9와 M.26 모두 300 mg/L IBA+ 3% sucrose 침지 처리 구에서 각각 86.5%와 85.4%로 가장 높았으며 순화율은 각각 71.7%와 70.0%로 가장 높았다. 통기성 마개 배양묘의 순화율은 비통기성마개 배양묘 보다 모든 침지 처리구에서 순화율이 비슷하거나 높았다.

Effects of Ovarian Morphology and Culture Vessel on In vitro Development and Cell Number in Embryos of Korean Native Cows

  • The main purpose of this study was to improve the efficiency and quality of in vitro embryo production in Korean Native Cows (KNC). We examined the effects of ovarian morphologies (Experiment 1) and the culture vessel (Experiment 2) on in vitro maturation (IVM). We measured the subsequent development rates and cell numbers of blastocysts. In Experiment 1, the ovaries of KNC were divided into six groups, based on follicle and corpus luteum (CL) morphology. The development rates to the 2- and 8-cell stages were similar among the six groups. The development rates to blastocyst stages were significantly higher in the group without a CL or follicle (WOCL/F) than in the groups with follicular cysts (FCs), regressive CLs (RCLs) or cystic CLs (CCLs) (p<0.05). The cell number of the inner cell mass (ICM) of blastocysts in the FCs and RCLs groups, and the number of cells in the trophectoderm (TE) in the WOCL/F group, FCs, growing CLs (GCLs) and RCLs were significantly higher than in other groups (p<0.05). The total cell number (TCN) in the WOCL/F, FC and RCL groups was also significantly higher than in other groups (p<0.05). The ICM cell number/TCN ratio was significantly higher in the FC and RCL groups than in the GCL and DF groups (p<0.05). In Experiment 2, oocyte IVM was carried out in culture dishes, in 0.25- or 0.5-ml straws used for freezing sperm. The development rate to the 2-cell stage was significantly higher in the 0.5-ml straw group than in the 0.25-ml straw group. The development rates to the blastocyst stage were similar in the dish and the two straw groups. There were no differences in the cell numbers of ICM, TE or TCN or ICM cell number/TCN ratios between groups.

선박사고의 인적요인에 대한 선원과 운항관리자 간의 인식 비교에 관한 연구 (A Study of the Comparison of Cognition between Seafarer and Shipping Operations Manager on Human Factors of Ship Accident)

  • 본 연구는 선박사고가 지속적으로 발생하고 있으며 점차 다양화, 복잡화됨에 따라 선원집단과 운항관리집단을 대상으로 선박사고의 인적요인을 분석하였다. 선박사고에서 인적요인은 큰 비중을 나타내고 있으며, 본 연구에서는 인적요인(human factor)에 대해 선행연구에서 제시된 인적요인을 기반으로 잠재적 요인을 관련 전문가 조사 등을 통해 확장적으로 도출하여 최종 20개 요인을 선정하였다. 이를 토대로 본 연구에서는 선정된 인적요인에 대해 선박안전 관련 전문가 및 실무자들의 인식 차이를 분석하여 선원집단과 운항관리자집단의 인식 차이를 검증하였고, 경영적 관점에 우선적으로 개선되어야 할 요인들을 추출하여 시사점을 제시하였다.

Comparative Study of Seeding and Culture Methods to Vascular Smooth Muscle Cells on Biodegradable Scaffold

  • How to improve the cell culture method on scaffolds is important in the tissue engineering fileld. In this study, we optimized seeding and culture methods to vascular smooth muscle cells (VSMCs) on biodegradable polymer scaffold. The primary culture of VSMCs obtained from canine external jugular vein was accomplished by applying the explant-derived method. The primary cultured VSMCs were seeded into scaffolds and then cultured by using various different methods; static or dynamic seeding, static or dynamic culture. The difference in proliferative response of VSMCs was analyzed with an alamar blue assay. Cell-polymer construct was examined by histochemical method and scanning electron microscopy. Mesh type scaffold ($10 \times 10 \times0.4 mm$) was made of polyglycolic acid (PGA) suture thread. The PGA mesh type scaffold was 45% in porosity, and 0.03 g in weight. The primary cultured VSMCs were confirmed with immunohistochemical staining using monoclonal anti-$\alpha$-smooth muscle actin. The density and distribution of proliferated VSMCs within the scaffold and cellular adherence on the surface of the scaffold showed better results in the static seeding condition than in the dynamic condition. Under the same condition of seeding method as the static condition, the dynamic culture condition showed enhanced proliferation rates of the VSMCs when compared to the static culture condition. In conclusion, to improve the VSMCs proliferation in vitro, static seeding is better than the dynamic condition. In the culture condition, however, culture under the dynamic status is better than the static condition. This was a pilot study to manufacture artificial vascular vessel by tissue engineering.
