Process Development for Concentration and Stabilization of Recombinant Endoxylanase Expressed in Bacillus subtilis

  • A strong constitutive $P_{JH}$ promoter from Bacillus sp. was applied to overexpress the endoxylanase gene in Bacillus subtilis. The expression plasmid, pJHKJ4, was designed to contain the $P_{JH}$ promoter and open reading frame of endoxylanase including its own promoter. The plasmid was introduced into B. subtilis DB431 and the resulting transformant was grown on LB glucose medium. The endoxylanase activity in the culture supernatant reached about 140 unit/ml. The enzyme in the supernatant was efficiently concentrated to 70% by two-step treatments of ammonium sulfate saturation and ultrafiltration. The stabilization of concentrated enzyme solution at different storage temperatures was examined with various stabilizers such as NaCl, $CaCl_2$, sucrose, sorbitol, polyethylene glycol, and Tween-80.

Optimal Conditions for the Production of Antioxidant by Nocardiopsis sp. S-1

  • This study investigated the production of antioxidant from Actinomyces culture supernatant. For the research of the natural marine antioxidant, several bacteria were isolated from the coast of Je-ju in Korea. An actinomycetes strains, S-1 was identified to a genus level 16S ribosomal DNA sequence and fatty acid analysis. From these results and other characteristics described in the Bergey's Manual, this strain was identificated as a Nocardiopsis dassonvillei. Strain S-1 showed high activity of 1,1-diphenyl-2-prcrylhydrazyl radical scavenging. The hydroxyl radical scavenging ability of Nocardiopsis sp. S-1 supernatant was 53%. Nutritional and cultural conditions for the production of antioxidant by this organism under shake-flask conditions have optimized. Similary initial medium pH 7.6, incubation temperature of $25^{\cicr}C$, sodium chloride concentration 2.5 and incubation time of 8 day were found to be optimal.

  • The ginseng growth-promoting bacterium Pseudomonas fluorescens KGPP 207 synthesized indole-3- acetic acid (IAA) from L-tryptophan, indole-3-pyruvic acid (IPyA), and indole-3-acetaldehyde (IAAld), but not from indole-3-acetamide (lAM) and other intermediates of various IAA biosynthetic pathways in the experiment with indole compound supplemented cell suspensions. TLC, HPLC, and GC-MS analyses revealed the presence of IPyA, indole-3-ethanol, indole-3-lactic acid and its methyl ester, IAA and its methyl, and ethyl esters in the culture supernatant of the bacterium. IAAld was detected in the supernatant using sodium bisulfite and TLC. The results indicate that unlike gall-forming bacteria which can synthesize IAA by lAM, the indole-3-pyruvic acid pathway is the route for IAA biosynthesis in this beneficial strain of P. fluorescens.

Co-cultured methanogen improved the metabolism in the hydrogenosome of anaerobic fungus as revealed by gas chromatography-mass spectrometry analysis

  • Objective: The purpose of this study was to reveal the metabolic shift in the fungus cocultured with the methanogen (Methanobrevibacter thaueri). Methods: Gas chromatography-mass spectrometry was used to investigate the metabolites in anaerobic fungal (Pecoramyces sp. F1) cells and the supernatant. Results: A total of 104 and 102 metabolites were detected in the fungal cells and the supernatant, respectively. The partial least squares-discriminant analysis showed that the metabolite profiles in both the fungal cell and the supernatant were distinctly shifted when co-cultured with methanogen. Statistically, 16 and 30 metabolites were significantly (p<0.05) affected in the fungal cell and the supernatant, respectively by the co-cultured methanogen. Metabolic pathway analysis showed that co-culturing with methanogen reduced the production of lactate from pyruvate in the cytosol and increased metabolism in the hydrogenosomes of the anaerobic fungus. Citrate was accumulated in the cytosol of the fungus co-cultured with the methanogen. Conclusion: The co-culture of the anaerobic fungus and the methanogen is a good model for studying the microbial interaction between H2-producing and H2-utilizing microorganisms. However, metabolism in hydrogenosome needs to be further studied to gain better insight in the hydrogen transfer among microorganisms.

Fucoidan Upregulates Chemotactic Activity of Canine Peripheral Blood Polymorphonuclear Cells Through Interleukin-8 from Peripheral Blood Mononuclear Cells in vitro (개 말초혈액 다형핵백혈구의 유주활성에 있어 fucoidan의 효과)

  • Fucoidan has been shown to enhance immune function. The objective of this study was to examine the in vitro effect of fucoidan on the chamotactic activity of canine peripheral blood polymorphonuclear cells (PMNs). The chemotactic activity of PMNs was evaluated by method of a modified Boyden chamber assay. The amount of interleukin (IL)-8 in the culture supernatants from peripheral blood mononuclear cells (PBMCs) treated with fucoidan was determined by means of ELISA. Fucoidan itself could not have chemoattract effects for PMNs. However, the chemotaxis of PMNs was remarkably enhanced by culture supernatant from PBMCs treated with fucoidan. Similarly, it was also increased by recombinant canine (rc) IL-8. These chemotactic activities of PMNs were inhibited by addition of anti-rcIL-8 polyclonal antibody (pAb). The amount of IL-8 in the culture supernatant from PBMCs was shown to increase upon treatment of fucoidan as compared with that of untreated PBMCs culture supernatant. These results suggest that fucoidan upregulates the chemotaxis of PMNs, which is mainly mediated by IL-8 released from fucoidanstimulated PBMCs.

Mechanism of Heat-Libile E. coli Enterotoxin Production (대장균의 이열성장독소 생산기전)

  • Enterotoxigenk E. coli is one of the major causative agents of the infantile diarrhea and traveler's diarrhea. The heat-labile enterotoxin is thought to be a virulence factor in the pathogenesis of the diarrhea and to be a marker for identification of the enterotoxigenic E. coli from non pathogenic E. coli. Therefore knowledge about the heat-labile enterotoxin is essential not only for understanding the pathogenesis but also for the diagnosis of the diarrhea. However the in-vitro heat-labile enterotoxin production is reported to be greatly affected by the cultural condition. In this regards, this study was designed to know the optimal conditions for the production of the heat-labile enterotoxin by assaying the permeability factor in the 18 hours culture supernatant of E. coli 08K25(B2) H9 and of E. coli 015 H11. Results obtained were summerized as follows: 1. Amounts of heat-labile enterotoxin produced were greater at initial pH 8.5 than at 7.0 of CYES-2 broth culture. However, the bacterial growth itself was more abundant at 7.0 than at 8.5. 2. Heat-labile enterotoxin per unit volume of culture supernatant was greater at shaking culture than at standing culture condition, but ratio of the enterotoxin produced over the unit mass of E. coli calculated was greater at standing culture than shaking culture condition, indicating that the greater yields of the toxin produced at shaking culture was due to increase in E. coli cell mass compared to the standing culture condition: 3. The enterotoxin produced in the lincomycin(128 microgram/ml) supplemented media was 5 or 11 times greater on the basis of enterotoxin per unit mass of E. coli, compared to the lincomycin-non-supplemented media, indicating that lincomycin itself increases the enterotoxin production. 4. Treatment of 18 hours culture of E. coli with polymyxin B(0.2 mg/ml) for 1 hour increased the yields of enterotoxin amounting to 2 or 5 times of the non-treated control cultures.

Isolation and Cultural Conditions of an with Antifungal Activity Bacillus sp. (항진균성 활성 Bacillus sp.의 분리 및 생산 조건)

  • An antifungal antibiotic-producing bacterium was isolated from soil and identified as Bacillus sp. CJ-1. The culture supernatant was found to have a strong and stable antifungal activity against Colletotrichum gloeosporioides. Culture conditions for the maximum antifungal activity were examined. Glucose and yeast extract were selected as the best carbon and nitrogen sources. The optimum C/N ratio was 3. The optimum temperature and initial pH were determined as $35^{\circ}C$ and 6.0, respectively. Under these conditions, the production for the antibiotic was maximized at 72 hr at $35^{\circ}C$ after cultivation. Microscopic observation showed that the culture supernatant of Bacillus sp. CJ-1 had a strong inhibitory activity on the mycelial growth of the test strain at above $12.5\;{\mu}L\;mL^{-1}$ of concentration.

Production of IL-6 and IL-8 in Human Fibroblasts Stimulated with Mycoplasma Lysates and Bacterial Toxins (세균독소와 Mycoplasma 항원으로 자극시킨 사람 섬유아세포의 Interleukin-6와 Interleukin-8 생성의 변화)

  • Cytokines are hormone-like proteins which mediate and regulate inflammatory and immune responses. The purpose of this study was to investigate the effect of lipopolysaccharide (LPS), Staphylococcus enterotoxin B (SEB), and Mycoplasma lysates on regulation of IL-6 and IL-8 production by human nasal fibroblasts. Primary cultured cells were incubated with LPS ($1.0\;{\mu}g/ml$) from E.coli, SEB ($1.0\;{\mu}g/ml$) from S.aureus, or Mycoplasma lysates (M.pneumoniae, Mp; M. fermentans, Mf; M. hominis, Mh, each $1.0\;{\mu}g/ml$). The culture supernatants were collected at 2, 6, and 24 hr and assessed for IL-6 and IL-8 production by enzyme-linked immunosorbent assay. The production of IL-6 in the culture supernatant was downregulated by LPS, SEB, or Mycoplasma lysates. But IL-6 was upregulated by mixed exposure with Mp+LPS (2 hr), Mp+LPS+SEB (24 hr), Mf+LPS (24 hr), Mf+LPS+SEB (2 hr), Mh+LPS (24 hr), Mh+SEB (24 hr), or Mh+LPS+SEB (24 hr). The production of IL-8 in the culture supernatant was similar to that of IL-6 by same stimulants. But IL-8 was upregulated by mixed exposure with Mf+LPS+SEB (2 hr), Mh+LPS (24 hr), Mh+ SEB (24 hr), or Mh+LPS+SEB (24 hr). These studies show that costimulation of LPS or SEB with Mycoplasma whole cell lysates upregulates the production of IL-6 and IL-8.

Production and Properties of Hemicellulases by an Isolate of Microbacterium sp. (Microbacterium sp. 분리균의 Hemicellulases 생산성과 효소특성)

  • A bacterium producing the extracellular mannanase and xylanase was isolated from Korean farm soil by successive subcultures in a minimal medium supplemented with palm kernel meal (PKM) and rice bran. The isolate YB-1106 showed 98% similarity with Microbacterium arabinogalactanolyticum on the basis of 16S rRNA gene sequences. The additional carbohydrates including locust bean gum (LBG) and PKM increased the mannanase productivity of the YB-1106, while the xylanase productivity of the isolate was increased by wheat bran, oat spelt xylan, rice bran and xylose. Particularly, maximum mannanase and xylanase activities were obtained in the culture filtrate of tryptic soy broth supplemented with 1% LBG or 2% wheat bran, respectively. Both enzyme activities were produced at stationary growth phase. The mannanase of culture supernatant was the most active at $50^{\circ}C$ and pH 6.0, while xylanase of culture supernatant was the most active at $55^{\circ}C$ and pH 6.5. The predominant products resulting from the mannanase or xylanase hydrolysis were oligosaccharides for LBG or xylan, respectively.

Anti-Immunomodulating Activities in Mycelial Filtrates and Culture Broth of Cordyceps ochraceostromat (동충하초의 균사 및 배양액에 의한 면역 활성 비교)

  • This study investigated the effects of mycelium and culture supernatant of Cordyceps ochraceostromat(Co) on air way hyper-responsiveness, pulmonary immune cell infiltration, and Th2 cytokine expression in animal models of atopy and asthma. After ConA(+/-) activation of mouse primary spleen cells, decreased IL-4 and IL-13 cytokine production were seen in the presence of Co mycelium extracts and culture supernatant. The asthma model involved mice sensitized to ovalbumin by i.p. injection treatment; Co mycelium extract was also injected. The atopy model was the dinitrofenylbenzene-treated mouse ear. Ear thicken ing induced by DNFB was decreased by Co mycelium extract, and the extract also inhibited lung cell infiltration in ovalbumin-induced asthmatic mice. The results thus indicated that the Co mycelial extract reduced the undesirable immune responses seen in asthma and atopy.
