Effects of Roscovitine on Nuclear Maturation, Spindle Configuration, and Chromosome Alignment in Porcine Oocytes

  • In the present study, effects of concentration and time of culture in presence of roscovitine on nuclear maturation and meiotic spindle configuration, chromosomal alignment were examined in porcine oocytes. In experiment 1, porcine cumulus oocyte complexes (COCs) were cultured at $39^{\circ}C$ in a 5% $CO_2$ atmosphere in North Carolina State University 23 (NCSU-23) supplemented with 25, 50, 75 or $100\;{\mu}M$ roscovitine for 22 h and then were cultured for additional 22 h after removal of roscovitine. Nuclear maturation and morphology of the meiotic spindle and chromosomal alignment were examined to determine the optimal concentration of roscovitine in oocyte maturation. In experiment 2, COCs were cultured in NCSU-23 supplemented with $50\;{\mu}M$ roscovitine for 17, 20, 27 or 42 h and then an additional 22 h without roscovitine was followed to determine the optimal time of culture. The optimal concentration of roscovitine to arrest and resume meiosis of porcine oocyte was $50\;{\mu}M$ by examining nuclear status (p<0.05) and normal spindle and chromosome configuration. The optimal time of culture in presence of roscovitine to arrest meiosis of porcine oocyte was 17 h (p<0.05), although MII rates and normal morphology of the meiotic spindle and chromosomal alignment were not significantly different among various times of culture. In conclusion, the optimal concentration and time of culture in presence of roscovitine to arrest porcine oocytes are $50\;{\mu}M$ and 17 h, respectively.

  • This article analyzes statistical indicators on the infrastructure of S&T culture in Korea. In the case of visible infrastructure, science museums, S&T programs in TV, and S&T books are not sufficiently secured except for internet. The utilization of visible infrastructure tends to be increased but in rudimentary stage. In relation to invisible infrastructure, public interest and understanding of &T is underdeveloped, social status of S&T personnel is low compared to social importance, and participation in S&T policy of S&T personnel and citizens organization is limited. For the development of statistical indicators in S&T culture, personal, institutional, and social contexts should be much considered, surveys on the particular social groups are systematically pursued, and a white paper on the S&T culture in Korea needs to be published.

  • Reading and writing in the past make noble significance in 'narration', in other words, in 'the creation of narration', through the process of 'recording'; but in the modern times, it engenders controversies over narration and linearity. In other words, reading/writing in digital era, is devalued as the simple arrangement of test or the connection of vast information without narration nor linearity. However, the reading/writing through text and hypertext reading is not the phenomenon which should be criticized because of the lack of narration or linearity-not only the lack of narration and linearity -, a process of social and cultural transition; it should be revalued as a result. The change of reading and writing methods will inevitably accompany the layers, status, significance and value of the contents; thus it makes more sense, when the reading and writing methods in digital contents are approached as new pop culture phenomenon. This is the "The prose of everyday life", based on pop culture and "The society of Conversation", based on communication; proposed 'Digitelling' ; this is the reason why we should pay attention to the digital contents, created infinitely by the citizens of the world, as new mass-culture phenomenon.

Study on the Consumption Status of Beans and the Soybean Food Culture in the Mid-Joseon Period According to Shamirok (조선 중기 두류 수급 현황과 콩 음식 문화 고찰 - 오희문(吳希文)의 『Shaemirok (쇄미록(瑣尾錄))』을 중심으로 -)

  • This study researched the food culture and bean economy of the Joseon dynasty during the 16th century and according to the primary lifestyle reference "Shaemirok (?尾錄)". The research analyzed the textual contents of the "Shaemirok (?尾錄)". It is clear that the people of the Joseon dynasty produced more beans than grain, at a ratio of 41 to 50, respectively. The soy bean sauce consumption was split into family consumption and non-family consumption. It was evident that there was more family consumption compared to that of non-family consumption at a ratio of 7 to 3, respectively. People of the Joseon dynasty annually recorded their way of making soy sauce from 1595 to 1600. The Joseon writers edited the record six times for making meju and four times for making soy sauce. The recorded ratio displays the ingredients of soy sauce, which were: 6 Du of Mal Jang and 2 Du of salt. Mal Jang and salt had a three to one ratio, respectively. The most mentioned food was Tofu during the mid-Joseon period with fifty six mentions. The Joseon people regarded making Tofu in a Buddhist temple as a family-bonding experience. Porridge was the second most prominent food next to Tofu, among the bean-related food. Porridge appears thirty five times. There were 3 types of porridge named: bean porridge, bean powder porridge and mung bean porridge.

A program for enhancing the South Koreans' and North Korean defectors' interpersonal abilities and accultural abilities (남한주민과 북한이탈주민의 대인관계와 문화적응 향상을 위한 프로그램)

  • This study was performed to develop a group counseling program for the South Koreans and the North Korean defectors to identify mutual cultural characteristics and personal traits, and to acquire adaptation capacity for the other's culture and interpersonal relationship. And then we validated the effect of this group program. This program was based on the existing program even if that was not validated through research work. Actually this program was for encouraging the South Koreans and the North Korean defectors to acquire interpersonal ability through mutually dependent and cooperative work based on the equal status naturally and to adjust themselves to corresponding culture. Each session continued for 1.5 hour per week. And the sessions were done for 4 weeks. This research was analysed, and the results were compared with control group's results. The results showed that treatment group revealed significantly positive outcomes than control group. But, five participants among the North Korean defectors were dropped out in the course. So North Korean defectors' credibility got lowered.

Present Status and Prospects of Oyster Industry in Korea

  • Oyster is the most important item next to tuna based on the market value in Korea's fisheries exports and it shares more than $90\%$ of the world oyster market. Oyster industry is a vital component of the Korea's aquaculture industry. However, it has faced many problems such as (1) a lower productivity of the culture system, (2) an environmental deterioration of the farming area, (3) a higher labor cost, (4) a shortage of of oysters to export, and (5) a poor seedling in nature. Therefore, including it's business forecast, the above problems and some countermeasures are discussed in this paper.

A Study on the Level of Stress Recognition of Urban Housewife and the Method of Coping to Stress (도시 주부의 스트레스 인지수준 및 적응 방법에 관한 연구)

  • The purpose of this study is to investigate the relationship between the level of stress recognition of urban housewife and the method of coping, and to explore bow these factors are influenced by socio-demographic variables such as the age of housewife, level of education, status of employment, number of children, durations of marriage, types of family, religion and socio-economic status. The research was conducted on 431 housewives in Seoul in August, 1985. As for the measurement of the instrument, 48 item questionnaire made by investigator was used. The questionnaire was based upon modified and upplemented Holme & Rahe's SRRS and Bell's 18-item Questionnaire to be appropriate to Korean culture. Data were analyzed by percentage, frequency and mean, and verified significant difference by ANOVA and performed Spearman's correlation coefficient. The results of this study are as follows; 1) There is some similarity in distribution of the level of stress recognition of urban housewife. 2) the level of education and the durations of marriage have influence upon the level of stress recognition of urban housewife. In each area, there are differences among groups : age, level of education, durations of marriage, number of children and types of family in the area of education ; age, status of employment, and durations of marriage in the area of health; level of education, durations of marriage, number of children and socio-economic status in the area of finance; status of employment in the area of household work. 3) There are several methods in the method of coping to stress of housewife and the score of long-term coping method appears higher than that of short-term. 4) The level of education, number of children, religion and socio-economic status were variables to have influence on the method level of education, religion and socio- economic status were variables to have influence and in the long-term coping method level of education, number of children, religion, and socio-economic status were to have influence. 5) There is very low positive correlation between the level of stress recognition of urban housewife and the method of coping to stress( ρ=.10, P<.05). 6)In the relation between several variables in socio-demographic variables and the method to coping to stress, the lower the level of stress recognition there are negative correlation (ρ=-.28, P<.01) between religion and the method of coping and also negative correlation (ρ=-.16, P<.05) between number of children and the method of coping. There are positive correlation between socio-economic status and the method of coping.
