Production Of Gellan Gum by Pseudomonas elodea (I) -Estimation of Metabolic Parameters and Rheological Properties of Culture Broth- (Pseudomonas elodea에 의한 Gellan Gum 생산(I) -metabolic parsmeter의 추정및 배양액의 유변학적 특성-)

  • A quantitative physiological approach has been employed to estimate the metabolic parameters such as specific uptake rates of nutrients and specific production rate in continuous culture of Pseudomonas elodea for gellan gum production. The estimated values of metabolic parameters are used for process improvement. During the exponential growth phase, the specific growth rate was 0.16hr-1 in batch culture. The gellan gum concentration increased up to 0.7g dry weight/100g broth and the apparent viscosity of the culture broth was about 4,500 cp.(72hrs culture). The ratio of specific uptake rate of carbon to that of nitrogen were found to be optimum at about 3.0mg-carbon/mg-nitro-gen. With the improved medium, the maximum gellan production rate, 0.6g dry weight/1/hr, was obtained at D=0.14 hr-1. The shear stresses of culture broth were fairly well correlated with shear rates by using Casson equation and at highly viscous culture broth, oxygen transfer coefficient was greatly reduced.

Antibacterial Activity of Lactobacillus casei Isolated from a Fermented Milk Against Pathogenic Enteric Bacteria (발효유(醱酵乳)에서 분리된 Lactobacillus casei의 병원성 장내세균에 대한 항균작용)

  • The present study was undertaken to elucidate the antibacterial spectrum of L. casei phage type $J_1$ strain isolated from a fermented milk product against pathogenic enteric bacteria. Growth inhibitory effects and minimum inhibitory concentration(MIC) of culture supernatants of L. casei grown in MRS broth were measured by both plate culture method and microplate broth dilution technique against Salmonella typhi, Salmonella typhimurium, Shigella flexneri, Shigella dysenteriae, enterpathogenic E. coli, Klebsiella pneumoniae and Pseudomonas aeruginosa. The results are summarized as follows: 1. The MRS broth culture of L. casei gave a similar extent of growth inhibitory effects against S. typhi, S. typhimurium, S. flexneri, S. dysenteriae, E. coli, K. pneumoniae and P. aeruginosa, respectively. 2. The inhibitory effects of L. casei culture were observed either in whole broth culture or in culture supernatant, but neither the bacterial suspension nor the neutralized culture supernatant showed such as antibacterial activities. 3. The MIC titres of the culture supernatants were ${\log_2}5$ to ${\log_2}6$, whereas those of the neutralized culture supernatant dropped markdely to ${\log_2}2$ to ${\log_2}3$. These results indicated that major portion of growth inhibitory effects of MRS broth culture of L. casei against enteric bacterial pathogens was possibly due to the acids produced, and minor portion to other antibacterial substances.

  • In our ongoing search for new secondary metabolites from fungal strains, one novel compound (1) and nine known compounds (2-10) were isolated from the EtOAc-soluble layer of the culture broth of Panus rudis. The culture broth of P. rudis was extracted in acetone and fractionated by solvent partition; column chromatography using silica gel, Sephadex LH-20, and Sephadex G-10; MPLC; and HPLC. The structures of isolated compounds were elucidated by one- and two-dimensional NMR and LC-ESI-mass measurements. One new compound, panepoxydiol (1), and nine known compounds, (E)-3-(3-hydroxy-3-methylbut-1-en-1-yl)-7-oxabicyclo[4.1.0]hept-3-ene-2,5-diol (2), isopanepoxydone (3), neopanepoxydone (4), panepoxydone (5), panepophenanthrin (6), 4-hydroxy-2,2-dimethyl-6-methoxychromane (7), 6-hydroxy-2,2-dimethyl-3-chromen (8), 2,2-dimethyl-6-methoxychroman-4-one (9), 3,4-dihydroxy-2,2-dimethyl-6-methoxychromane (10), were isolated from the culture broth of P. rudis. This is the first report of isolation of a new compound panepoxydiol (1) and nine other chemical constituents (2-5, 7-10) from the culture broth of P. rudis.

  • Kombucha fermentation broth has been used as a popular health beverage and an alternative therapy with prophylactic and therapeutic benefit. We tried to establish optimal culture conditions for Kombucha fermentation in milk and to investigate cytotoxicity and antioxidative enzyme activity of Kombucha broth against normal human fibroblasts. The optimal conditions of Kombuch culture were established to 3$0^{\circ}C$, 20∼23 hours by DPPH radical scavenging test. There were positive effects on cell growth while no cytotoxicity against primary normal human diploid fibroblasts was found. The activites of glutathione peroxide and catalase in the cells treated by hydrogen peroxide (1 mM) alone and by hydrogen peroxide with Kombucha broth (1 mg/mι) were significantly different (p<0.05). These results suggest that Kombucha broth could be developed as an antioxidant agent for a new cosmetic material.

  • Fibrinolytic activities of culture concentrates of various yeasts were investigated. The concentrates of the culture broth of Saccharomyces cerevisiae Y99-7 showed the strongest fibrinolytic activity of 25 mm (clear zone). The fibrinolytic activity of Saccharomyces cerevisiae Y99-7 was more high in the culture concentrates from PD broth rather than that of yeast extract-peptone dextrose cultures (clear zone : 22.7 mm).

  • An automated flow injection analysis apparatus manufactured economically was used in the automatic measurement of E. col growth in shake-flask culture of nutrient broth. The detailed measurement of whole growth was possible at intervals of 10 minutes by the automatic measurement system which adopted the sterilized nutrient broth as a carrier solution. Using distilled water as a carrier solution showed less accurate results than nutrient broth.

  • RNA accumulating strain of Torulopsis versatilis MT-1 was cultured in molasses medium for higher contents of RNA in cell. Yeast cells were harvested at logarithmic phase on synchronous culture. Yield of cells on dry base to input sugar was 59.5%. Crude protein content was 55.1% in cell. RNA content was 13.9%. Some problems found in the process for the preparation of yeast extracts were improved by the addition of culture broth of Streptomyces faecalis MSF which secrete RNase (5' nuclease and 5' adenylic acid deaminase). When the culture broth of S. faecalis MSF was added in autolysis process 46% of RNA in cell was converted to I and G(5' inosinic acid and 5' guanylic acid) in extract. By addition of 3-7% culture broth of S.faecalis MSF in autolysis or enzymolysis process at the start or early stage, RNA in extract was converted easily to I and G and protein in cells was easily extracted and hydrolyzed to amino acid. Taste of those yeast extracts was delicious. The yeasty smell in yeast extracts was removed. And cell debris was easily removed from extract.

Whitening Effect of Mycelial Culture Broth of Paecilomyces japonica in the Mixture of Cucumber and Crape Extracts (포도와 오이즙액을 이용한 동충하초(Paecilomyces japonica) 균사체 배양액의 미백효과)

  • This study was carried out to investigate the whitening effect of mycelial culture broth of P. japonica in the mixture of cucumber and grape extracts. In the inhibition test of melanin biosynthesis of melanoma cell, B16BL6, the culture broth of P. japonica more then $50\;{\mu}l/ml$ (5%) concentration inhibited the melanin biosynthesis of the cell entirely without cytotoxicity. More then 10 days incubation of P. japonica in the mixture was required to have the inhibition activity. In vitro inhibition test of melanin biosynthesis of the culture broth of P. japonica was investigated in the concentration dependent manner of 10% to 50%. 30% concentration of the culture broth inhibited completely tyrosinase activity. In the cytotoxicity test, cucumber and grape extract itself has a strong cytotoxicity to the melanoma cell, B16Bl6. The value of $IC_{50}$ of the cucumber and grape extracts against the melanoma cells was 5% concentration. However, the culture broth of P. japonica incubated in the cucumber and grape extracts did not show the cytotoxcity up to 20% against melanoma cell, B16BL6. Therefore, we concluded that the culture broth of P. japonica in the mixture of cucumber and grape extracts can be used as a whitening cosmetic resource.

  • The development of an enrichment method for the rapid and effective identification of Salmonella spp. in sewage or food was studied. As a growth factor for Salmonella, 10 mM cyclic adenosine monophosphate (cAMP) in trypticase soy broth with 0.6% yeast extract (TSBYE) increased cell number five-folds and 0.6% yeast extract in selenite broth increased cell number ten-folds of control. Bile salts in selenite broth was tested for the selection of S. enteritidis in a mixture with Staphylococcus aureus, Pseudomonas aeruginosa, Lactobacillus plantarum and Escherichia coli. The latter four strains were effectively inhibited at 0.1% bile salt. A two-step culture method was used to enrich Salmonella spp.; a primary-enrichment and secondary- enrichment culture. At a primary-enrichment step, selenite broth with 0.6% yeast extract and 10 mM cAMP was used, and at a secondary-enrichment step, 0.1% bile salt was additionally used. Culture times of a primary- enrichment and a secondary-enrichment step were 8 hr and 6 hr, respectively. In this procedure, cell number increased from 10$^{0.3}$ to 10$^{8.5}$ with inhibition of other strains within 14 hr. In the case of an initial cell concentrarion as low as 10$^{-2}$ cfu/ml, a cell number increased to 10$^{7}$ cfu/ml by using a 10 hr primary-enrichment and 6 hr secondary-enrichment procedure.

Development of a High Efficient "Dual Bt-Plus" Insecticide Using a Primary Form of an Entomopathogenic Bacterium, Xenorhabdus nematophila

  • A phase variation has been reported in an entomopathogenic bacterium, Xenorhabdus nematophila. Compared with a wild-type primary form, a secondary form usually loses several physiological and biochemical characters. This study showed that the phase variation of X. nematophila caused a significant alteration in its immunosuppressive activity and subsequent entomopathogenicity. A secondary form of X. nematophila was detected in laboratory colonies and exhibited significant differences in dye absorption and entomopathogenicity. In addition, the secondary form was different in its production of eicosanoid-biosynthesis inhibitors (EBIs) compared with the primary form of X. nematophila. Production of oxindole and p-hydroxypropionic acid was significantly reduced in the culture broth of the secondary form of X. nematophila. The reduced EBI production resulted in significant suppression in the inhibitory effects on cellular nodule formation and phenoloxidase activity. Culture broth of the primary form of X. nematophila enhanced the pathogenicity of Bacillus thuringiensis ( Bt) significantly more than the culture broth of the secondary form. Furthermore, this study developed a highly efficient "Dual Bt-Plus: to control both lepidopteran insect pests Plutella xylostella and Spodoptera exigua, by mixing two effective Bt strains along with the addition of potent bacterial metabolites or 100-fold concentrated X. nematophila culture broth.
