Luminescence properties and compositions of contaminating inorganic minerals separated from gamma-irradiated fresh and white ginsengs from different areas

  • Gamma-irradiation (0-7 kGy) of ginseng is permitted in Korea for the purpose of microbial decontamination; with strict labeling, traceability and monitoring requirements. An identification study was conducted to determine the photostimulated-luminescence (PSL) and thermoluminescence (TL) properties of gamma-irradiated fresh and white ginsengs cultivated in different areas. Dose- dependent PSL-based screening was possible for white ginseng samples; however, inappropriate results from non-irradiated fresh ginseng samples were obtained, showing intermediate (700 to 5,000) or positive ($T_2$ >5,000, irradiated) PSL counts due to the abundance of minerals on the surfaces of the samples. TL analysis of separated minerals from all non-irradiated samples gave TL glow curves of low intensity with a maximum peak after $300^{\circ}C$. However, well-defined irradiation-specific (high intensity with a maximum peak at about $200^{\circ}C$) glow curves were observed for all the irradiated samples, regardless of their type and origins. TL ratios (first glow curve /second glow curve) were also determined to confirm the irradiated (>0.1) and non-irradiated (<0.1) results. SEM-EDX (scanning electron microscope-energy dispersive X-ray) and XRD (X-ray diffraction) spectroscopic analyses showed that feldspar and quartz minerals were the main source for the typical radiation-specific luminescence properties.

인삼 논재배 연작지에서 윤작물 재배가 토양화학성, 토양 미생물상 및 2년생 인삼의 생육에 미치는 영향 (Crop Rotation in Paddy Soil Exhibiting Crop Failure Following Replanting: Effect on Soil Chemical Properties, Soil Microbial Community and Growth Characteristics of 2-Year-Old Ginseng)

  • Background: Crop rotation plays an important role in improving soil chemical properties, minimizing the presence of disease pathogens, and assists in neutralizing autotoxic effects associated with allelochemicals. Methods and Results: Five rotation crops of sudan grass, soybean, peanut, sweet potato, and perilla were cultivated for one year with an aim to reduce yield losses caused by repeated cropping of ginseng. In 2-year-old ginseng grown in the same soil as a previous ginseng crop, stem length and leaf area were reduced by 30%, and root weight per plant was reduced by 56%. Crop rotation resulted in a significant decrease in electrical conductivity, $NO_3$, and $P_2O_5$ content of the soil, whereas organic matter, Ca, Mg, Fe, Cu, and Zn content remained-unchanged. Soil K content was increased following crop rotation with sudan grass and peanut only. Rotation with all alternate crops increased subsequent ginseng aerial plant biomass, whereas root weight per plant significantly increased following crop rotation with perilla only. A significant positive correlation was observed between root rot ration and soil K content, and a significant negative correlation was observed between ginseng root yield and the abundance of actinomycetes. Crop rotation affected the soil microbial community by increasing gram negative microbes, the ratio of aerobic microbes, and total microbial biomass whereas decreases were observed in actinomycetes and the ration of saturated fatty acids. Conclusions: In soil exhibiting crop failure following replanting, crop rotation for one year promoted both soil microbial activity and subsequent ginseng aerial plant biomass, but did not ameliorate the occurrence of root rot disease.

산양삼 혼입 숙성 감식초 섭취에 의한 식이 제한 흰쥐의 지방 저장 및 에너지 대사 단백질 발현 (Mountain-cultivated Ginseng Ripened into Persimmon Vinegar Ingestion on Fat Storage and Metabolic Protein Expression in Diet-controlled Rats)

  • 본 연구는 4년근 산양삼을 4년 숙성된 감식초에 22주간 혼입숙성하여 만든 산양삼감식초(가칭: 산삼초)를 활용하여 식이제한을 하였을 때 나타나는 비만관련 요소들을 분석하였다. 웅성 흰쥐 63마리를 대상으로 고지방식을 공급하면서 식후 2회, 6주간 산삼초를 경구투여하였다. 집단은 대조군, 제한식이군, 웨이트사이클링군으로 구분하였으며, 이를 다시 대조군, 산삼초 2.5배 희석 섭취군, 산삼초 5.0배 희석 섭취군으로 각각 구분하였다. 각 집단은 7마리씩으로 하였다. 6주간의 사육이 종료된 후, 복강내 지방(복막후지방, 장간막지방, 부고환지방)과 대사관련 단백질(AMPK, PPAR-${\alpha}$, CPT-1)의 발현을 분석하였다. 결과를 볼 때, 모든 집단에서 산삼초 섭취에 의한 지방저장량의 유의한 감소 또는 감소 경향이 나타났으며, 지방저장량의 총합계 역시 산삼초 섭취에 의해 증가가 억제되었다. 이와는 반대로 대사관련 단백질 발현은 모든 항목에서 산삼초 섭취에 의해 유의한 증가 혹은 증가 경향이 나타났다. 이 결과는 산삼초 섭취에 의한 에너지 대사의 효과적인 상승이 고지방식이 또는 식이제한을 하였을 경우에도 체내 지방 저장량의 증가를 억제하였다는 것이다. 즉, 비만의 가장 큰 문제점인 웨이트사이클링의 억제를 위한 좋은 기능성을 보유하고 있다고 판단된다. 이를 통하여 임업산물인 산양삼과 감식초의 융합소재인 산삼초를 비만억제 식품으로 활용할 수 있을 것으로 기대된다.

  • In spite of their inferior quality, pale yellow flue-cured tobacco varieties (N. tabacum L.) are being cultivated as pre-crop of rice in paddy field because of its early leaf maturity. Fertile and male sterile Hicks as green variety , three pale yellow varieties and six single cross $F_1$ hybrids between green and pale yellow varieties, were evaluated. Fertile $F_1$ hybrids were similar to pale yellow varieties in early leaf maturity, curing characters and nicotine contents, where-as they were lower in total nitrogen, higher in reducing sugar comparing to those of the pale yellow varieties.

고려인삼으로 제조된 홍삼중의 화학적 암 예방성분 (Cancer Chemopreventive Compounds of Red Ginseng Produced from panax ginseng C.A. Meyer)

  • 발암물질을 투여하여 발생하는 마우스 폐선종은 홍삼추출믈의 투여에 의하여 그 발생율이 억제되나 수삼을 투여하면 발생율이 억제되지 않는다. 또한 암환자-대조군연구 결과에 있어서도 수삼즙 또는 수삼절편을 복용한 사람에서는 암의 위험비가 감소되지 않으나 수삼열탕 또는 홍삼을 복용하면 현저한 위험비의 감소를 볼 수 있었다. 이와 같은 결과는 열로 처리된 홍삼중에 암예방 유효성분이 있을 것이라고 추정되어 왔다. 저자들은 4종의 홍삼중의 진세노사이드 즉 Rh$_1$, Rh$_2$, Rg$_3$ 및 Rg$_{5}$ 를 고려홍삼으로부터 분리합성하여 윤의 9주 중기 항발암실험법에 의하여 항발암성을 관찰한바 진세노사이드 Rg$_3$와 Rg$_{5}$의 투여시에는 통계학적으루 유의한 폐선종 발생율이 감소되었으나 Rh$_2$에서는 폐선종발생율이 약간 감소되는 경향을 보였고 Rh$_1$에서는 전혀 영향을 주지 않았다. 이와 같은 소견으로 홍삼에 의한 항발암작용 또는 암예방작용은 홍삼중의 진세노사이드 Rg$_3$및 Rg$_{5}$가 유효성분임을 파악하였으며 이들 진세노사이드 Rg$_3$ Rg$_{5}$ 및 Rh$_2$가 단독 또는 복합적으로 작용할 것으로 추정된다.

홍삼 가공중 azoxystrobin, fenhexamid 및 cyprodinil농약의 감소율 (Reduction Rate of Azoxystrobin, Fenhexamid and Cyprodinil during Ginseng Processing)

  • 본 연구는 홍삼 및 농축액의 합리적인 농약잔류허용기준 개정을 위한 과학적인 자료를 얻고자 수행하였다. 인삼 재배 시에 사용 등록된 농약 3종(azoxystrobia, fenhexamid, cyprodinil)을 인삼포에 직접 살포하여 수확한 수삼을 홍삼과 농축액으로 직접 제조하였다. 수확한 수삼에 azoxystrobin 0.12 ppm, fenhexamid 0.19 ppm, cyprodinil 1.78 ppm이 잔류하였으며 농약이 잔류된 수삼을 원료로 홍삼 및 농축액으로 가공하여 각 제품의 농약 잔류량을 조사한 결과 fenhexamid의 경우 홍삼에 0.54 ppm, 홍삼농축액에서 1.93 ppm이 잔류하였다. Azoxysoobin은 홍삼에서 0.24 ppm, 홍삼농축액에서 0.81 ppm이 잔류하여 가공단계를 거치면서 증가하였다. 그러나 cyprodinil의 경우 1.78 ppm이 잔류된 수삼이 홍삼으로 가공되면서 1.49 ppm으로 감소하였으나 홍삼농축액에서는 3.66 ppm으로 증가하였다. 농약이 잔류된 수삼을 이용하여 홍삼과 농축액 제조시 azoxystrobin, fenhexamid, cyprodinil의 감소계수(dry base)는 홍삼에서 0.66, 0.94, 0.28, 홍삼농축액에서 3.25, 4.94, 1.01이였다.

인삼 추출물에 의한 Cylindrocarpon destructans의 주화성 반응 연구 (Chemotactic Response Study of Cylindrocarpon destructans towards Ginseng Root Exudates)

  • Background: Cylindrocarpon destructans (Zins) Scholten is an important pathogenic fungus that causes ginseng root rot in many ginseng growing areas in China. Although C. destructans have been studied worldwide, research on its chemotaxis towards ginseng (Panax ginseng C. A. Meyer) root exudates in the rhizosphere remains limited. Methods and Results: In this study, we collected ginseng root exudates with three different polarities from three-year-old ginseng roots, and performed chemotaxis and spore germination assays to investigate the ability of these exudates to induce the response in C. destructans. The results showed that, compared with other conditions, when C. destructans cultivated at $20^{\circ}C$ and a pH of 6 exhibited a strong positive chemotactic response toward $2mg/{\ell}$ aqueous phase, $20mg/{\ell}$ butanol phase, and $0.2mg/{\ell}$ petroleum ether from ginseng root exudates, the chemotactic moving indexes were 0.1581, 0.1638 and 0.1441, respectively. In addition, the spore germination rate with optimal chemotactic parameters were 48%, 53%, and 41% in the aqueous phase, butanol phase and petroleum ether groups, respectiviely, which were significantly higher than that in the control group (23%) (p < 0.05). The mycelial growth rate with optimal chemotactic parameters increased with culture time, and the maximum growth rates in the aqueous phase, butanol phase and petroleum ether groups were 0.425, 0.406 and 0.364 respectively, on the 4th day. The optimal chemotactic parameters were $39.73mg/50mg/{\ell}$, $48.93mg/50mg/{\ell}$, and $31.43mg/50mg/{\ell}$, in aqueous phase, butanol phase and petroleum ether respectively, from ginseng root exudates, compared with $5.5mg/50mg/{\ell}$, in the control group. Conclusions: The present study revealed that certain ginseng root exudates containing chemical attractants act as nutritional sources or signals for C. destructans and support its colonization of ginseng roots.

Effects of cultivation ages and modes on microbial diversity in the rhizosphere soil of Panax ginseng

  • Background: Panax ginseng cannot be cultivated on the same land consecutively for an extended period, and the underlying mechanism regarding microorganisms is still being explored. Methods: Polymerase chain reaction and denaturing gradient gel electrophoresis (PCR-DGGE) and BIO-LOG methods were used to evaluate the microbial genetic and functional diversity associated with the P. ginseng rhizosphere soil in various cultivation ages and modes. Results: The analysis of microbial diversity using PCR-DGGE showed that microbial communities were significantly variable in composition, of which six bacterial phyla and seven fungal classes were detected in P. ginseng soil. Among them, Proteobacteria and Hypocreales dominated. Fusarium oxysporum, a soilborne pathogen, was found in all P. ginseng soil samples except R0. The results from functional diversity suggested that the microbial metabolic diversity of fallow soil abandoned in 2003was the maximum and transplanted soil was higher than direct-seeding soil and the forest soil uncultivated P. ginseng, whereas the increase in cultivation ages in the same mode led to decreases in microbial diversity in P. ginseng soil. Carbohydrates, amino acids, and polymers were the main carbon sources utilized. Furthermore, the microbial diversity index and multivariate comparisons indicated that the augmentation of P. ginseng cultivation ages resulted in decreased bacterial diversity and increased fungal diversity, whereas microbial diversity was improved strikingly in transplanted soil and fallow soil abandoned for at least one decade. Conclusion: The key factors for discontinuous P. ginseng cultivation were the lack of balance in rhizosphere microbial communities and the outbreak of soilborne diseases caused by the accumulation of its root exudates.

연초 수확엽의 숙도가 색채 및 색소물질 함량에 미치는 영향 (Influence of Maturity on the Plastid Pigments and Chromatic Characteristics of Paddy Tobacco)

  • Field experiment was conducted to get the basic information about the difference in color and plastid pigments at the different stage of harvest of flue-cured tobacco cultivated in paddy field, and compared to upland ones. All tested crops were primed in lots of 3 to 4 at each priming, beginning at the bottom of the plant. Leaf pigments and chromatic values of tobacco leaves were determined in samples collected at weekly intervals, and according to analyses obtained from 4 stalk positions. By the degree of maturity, leaves harvested were separated with visual characters into four classes of immature, mature, ripe and mellow. regardless of stalk position, chlorophyll contents was higher in the order of immature > mature > ripe > mellow, respectively. In fresh leaves, chlorophyll levels was higher with ascending stalk position. chlorophyll a and b in cured leaves were less than 10% of the amount present at harvest, and it was estimated that maturity condition also influenced the rates of degradation of plastid pigments. Fully mature leaves of upland diverted from paddy field showed slightly higher in chlorophyll contents than in those of upland tobacco ones but somewhat lowered in $L^*$ and $b^*$ values, and there was no difference in $a^*$ value.

The Presence and Importance of VAM Spores in the Soil of Ginseng Gardens

  • Symbiotic associations between land plants and fungi have been known for more than one hundred years. Vesiculararbuscular mycorrhizas (VAM) are the most common symbiosis in flowering plants and can be recognized in almost all plant families. These fungal associations play a very important role in the growth and survival of plant species. However, with respect to the importance and intensity of the VAM, there is great variation among host species. Our knowledge of the VAM fungus-plant association in Araliaceae is very limited. After the first reports of the occurence of VAM in lateral roots of Panax species, mycorrhizal structures are now described as special structures representing the so-called Paris type. In this type, the development of new spores and vesicles is extremely low. This and the type of colonization of the fungus in Panax roots indicates on, one hand, the high intensity of the VAM and, on the other hand, a remarkable dependency for VAM in members of the Panax species. Therefore, it can be easily understood that cultivated Panax plants exhibit a significant uptake of nutrients and this leads to an extremely depleted soil at harvest. Further, the soil is nearly free of the spores of VAM fungi as they germinate each year on the newly developing Panax roots.
