To protect the plant against several soil-borne pathogens, we are currently constructing disease-resistant transgenic root stock for the growth of cucurbitaceae vegetable plants, watermelon and gourd. We made a watermelon cDNA library from Cladosporium cucumerinum-Infected leaves for substractive hybriazation and differential screening. We isolated the several pathogen inducible cDNA clones, such as caffeoyl-CoA-methyltransferase, LAA induced protein, receptor-like kinase homolog, hydroxyproline-rich glycoprotein, catalase, calmodulin binding protein, mitochondrial ATPase beta subunit, methyl tRNA synthetase and WRKY transcription factors. We previously obtained CaMADS in pepper and galactinol synthase ( CsGolS) in cucumber that were confirmed to be related with disease-resistance. CaMADS and CsGolS2 were transformed into the inbred line 'GO701-2' gourd, the inbred line '6-2-2' watermelon and the Kong-dye watermelon by Agrobacterium tumerfaciens LBA4404. Plant growth regulators (zeatin, BAP and IAA) were used for shoot regeneration and root induction for optimal condition. Putative transgenic plants were selected in medium containing 100mg/L kanamycin and integration of the CaMADS and CsGO/S2 into the genomic DNA were demonstrated by the PCR analysis. We isolated major soil-borne pathogens, such as Monosporascus cannonballus, Didymella bryoniae, Cladosporium cuvumerinum from the cultivation area of watermelon or root stock, and successfully established artificial inoculation method for each pathogen. The effect of the electrolyzed oxidizing water on Sphaerotheca fuliginea was investigated with germination and sporulation of the fungal conidia. The sporulation was inhibited by the electrolyzed oxidizing water of pH 2.5, 3.5, and 4.5, but was not inhibited by the distilled water adjusted pH with 1N-HCL solution. However, the electrolyzed oxidizing water did not affect conidial germination. The oxidation-reduction potential at pH 2.5 and pH 3.5 of electrolyzed oxidizing water were 1130 mV and 1060 mV, respectively, but those of distilled water adjusted with HCL solution were 550 mV and 490 mV, respectively. When the electrolyzed oxidizing water of ORP over 1100 mV was sprayed on cucumplanting, the disease severities of powdery mildew were about 8.5% and 19.2%, respectively. Disease severity of a standard control (triflumizole 30% WP, $500\textrm{mg}\textrm{/L}$) was about 3.0%, while that of plants without electrolyzed oxidizing water was to 45.8%.
An efficient plant regeneration system has been developed for figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), which is exclusively used as a rootstock for cucumber. The protocol is based on results obtained from a series of culture experiments involving different parts of the cotyledons and various media. The culture of cotyledon explants was critical for the enhancement of shoot regeneration frequency. The lower parts of the cotyledon excised at the plumule base were found to display a markedly enhanced production of adventitious shoots compared to other cotyledon regions. Culture in silver nitrate-supplemented Murashige and Skoog (MS) medium was not beneficial for shoot regeneration and suppressed root regeneration. Efficient shoot regeneration was obtained on MS medium containing 1.0 $mg\;l^{-1}$ zeatin and 0.1 $mg\;l^{-1}$ indole-3-acetic acid. Regenerated shoots successfully elongated and rooted in medium containing 0.1 $mg\;l^{-1}$ 1-naphthalene-acetic acid after 10-15 days of subculturing. The plantlets were satisfactorily acclimatized in a greenhouse and grew into normal plants without any morphological alterations.
Powdery mildew is one of the most devastating diseases in cucurbits. Crop yield can decline as the disease severity increases. In this study, we evaluated the effect of silver nanoparticles against powdery mildew under different cultivation conditions in vitro and in vivo. Silver nanoparticles (WA-CV-WA13B) at various concentrations were applied before and after disease outbreak in plants to determine antifungal activities. In the field tests, the application of 100 ppm silver nanoparticles showed the highest inhibition rate for both before and after the outbreak of disease on cucumbers and pumpkins. Also, the application of 100 ppm silver nanoparticles showed maximum inhibition for the growth of fungal hyphae and conidial germination in in vivo tests. Scanning electron microscope results indicated that the silver nanoparticles caused detrimental effects on both mycelial growth and conidial germination.
Three three hundred and ninety seven isolates of Botrytis cinerea were isolated from infected plants of strawberry, tomato and cucumber from several areas in Korea during 1994∼1996 and the resistance of these isolates against some fungicides were examined. The isolation frequency of phenotypes resistant to carbendazim, procymidone, and diethofencarb were found to be 69.9 43.7, and 31.8%, respectively. The isolates were divided into six phenotypic groups; SSR, SRR, RSS, RRS, RSR and RRR, representing sensitive (S) or resistant (R) to benzimidazole, dicarboximide, and N-phenylcarbamate fungicides in order. The percentage of six phenotypes were 28.2, 2.0, 27.2, 41.0, 0.9 and 0.8%, respectively. On the basis of the mycelial growth inhibition (%) B. cinerea isolates were divided into three classes (class 1; 0∼50%, class 2; 51-99%, class 3; 100% inhibition) on carbendazim and three classes (class 1; 0∼75%, class 2; 76∼99%, class 3; 100% inhibition) on procymidone and the mixture of carbendazim+diethofencarb, respectively. Changes in sensitivity levles to carbendazim and carbendazim+diethofencarb were affected by introduction and increasing ratio of the use of diethofencarb.
The Gustatory system enables animals to detect toxic bitter chemicals, which is critical for insects to survive food induced toxicity. Cucurbitacin is widely present in plants such as cucumber and gourds that acts as an anti-herbivore chemical and an insecticide. Cucurbitacin has a harmful effect on insect larvae as well. Although various beneficial effects of cucurbitacin such as alleviating hyperglycemia have also been documented, it is not clear what kinds of molecular sensors are required to detect cucurbitacin in nature. Cucurbitacin B, a major bitter component of bitter melon, was applied to induce action potentials from sensilla of a mouth part of the fly, labellum. Here we identify that only Gr33a is required for activating bitter-sensing gustatory receptor neurons by cucurbitacin B among available 26 Grs, 23 Irs, 11 Trp mutants, and 26 Gr-RNAi lines. We further investigated the difference between control and Gr33a mutant by analyzing binary food choice assay. We also measured toxic effect of Cucurbitacin B over 0.01 mM range. Our findings uncover the molecular sensor of cucurbitacin B in Drosophila melanogaster. We propose that the discarded shell of Cucurbitaceae can be developed to make a new insecticide.
An antiviral protein (CAP30) with ribosome-inactivating activity was purified from the leaves of Chenopodium album L. through ammonium sulfate precipitation and column chromatography using S-Sepharose, Blue-Sepharose, FPLC Suprose12 HR, and FPLC Mono-S. The molecular wight of CAP30 was estimated to be 30kD. CAP30 was thermostable, maintaing its activity even after incubation at $70^{\circ}C$ for 30 min, and was stable in the pH range of 6 to 9. In a cell-free in vitro translation system using rabbit reticulocyte lysate, protein synthesis was inhibited by the addition of CAP30 with an $IC_{50}$ of 2.26pM. The comparison of N-terminal amino acid sequences of this protein with known ribosome-inactivating proteins (RIPs) revealed that it had some sequence homology with PAP-S and PAP-R from pokeweed (Phytolacca americana)and dodecandrin from P. dodecandra, but had no sequence homology with RIPs from other plants belonging to different orders. The mosaic symptoms on tobacco leaves caused by cucumber mosaic virus infection was completely inhibited by 100 ng/ml of the pure CAP30 protein.
The family Peronosporaceae, an obligate biotrophic group of Oomycota, causes downy mildew disease on many cultivated and ornamental plants such as beet, cucumber, grape, onion, rose, spinach, and sunflower. To investigate the diversity of Peronosporaceae species in Korea, we performed morphological analysis for dried plant herbariums with downy mildew infections by two largest genera, Peronospora and Plasmopara. As a result, it was confirmed that there are five species of Peronospora and two species of Plasmopara, which have been so far unrecorded in Korea, as well as rarely known in the world; Pl. angustiterminalis (ex Xanthium strumarium), Pl. siegesbeckiae (ex Siegesbeckia glabrescens), P. chenopodii-ambrosioidis (ex Chenopodium ambrosioides), P. chenopodii-ficifolii (ex Chenopodium ficifolium), P. clinopodii (ex Clinopodium cf. vulgare), P. elsholtziae (ex Elsholtzia ciliata), and P. lathyrina (ex Lathyrus japonicus). In addition, their phylogenetic relationship was inferred by molecular sequence analysis of ITS, LSU rDNA, and cox2 mtDNA. By rediscovering the seven missing species and barcoding their DNA sequences, this study provides valuable insights into the diversity and evolutionary studies of downy mildew pathogens.

Biological control of important fungal diseases such as Phytophthora blight of red pepper, gary mold rot of vegetables, and powdery mildew of many crops was attempted using an antagonistic bacterium, Bacillus sp. AC-1 in greenhouses and fields. The antagonistic bacterium isolated from the rhizosphere soils of healthy red pepper plant was very effective in the inhibition of mycelial growth of plant pathogenic fungi in vitro including Phytophthora capsici, Rhizoctonia solani, Pyricularia oryzae, Botrytis cinerea, Valsa mali, Fusarium oxysporum, Pythium ultimum, Alternari mali, Helminthosporium oryzae, and Colletotrichum gloeosporioides. Culture filtrate of antagonistic Bacillus sp. AC-1 applied to pot soils infested with Phytophthora capsici suppressed the disease occurrence better than metalaxyl application did until 37 days after treatment in greenhouse tests. Treatments of the bacterial suspension on red pepper plants also reduced the incidence of Phytophthora blight in greenhouse tests. In farmers' commercial production fields, however, the controlling efficacy of the antagonistic bacteria was variable depending on field locations. Gray mold rot of chinese chives and lettuce caused by Botrytis cinerea was also controlled effectively in field tests by the application of Bacillus sp. AC-1 with control values of 79.7% and 72.8%, respectively. Spraying of the bacterial suspension inhibited development of powdery mildew of many crops such as cucumber, tobacco, melon, and rose effectively in greenhouse and field tests. The control efficacy of the bacterial suspension was almost same as that of Fenarimol used as a chemical standard. Further experiments for developing a commercial product from the antagonistic bacteria and for elucidating antagonistic mechanism against plant pathogenic fungi are in progress.

Chemical properties of soils and mineral contents in plants grown at seventy one regions located in southern part of Korea were surveyed to obtain the basic information for sustainable management of plastic film house soils. The averaged soil chemical properties showed optimum levels in pH, organic matter, available boron, manganese, zinc contents, whereas available phosphate and exchangeable rations were highly accumulated. Available phosphate showed highly positive correlation with continuous cropping period. The contents of phosphate, potassium, calcium, and magnesium in the top soils were ranged from 912(Pusan)~1.796mg (Masan) ; $1.52(Pusan){\sim}2.93cmol^+$(Sunchon) ; $7.44(Masan){\sim}18.58cmol^+$(Changweon) ; $2.82(Changnyong){\sim}6.24cmol^+/kg$(Changweon), respectively. There were high levels of phosphorus and potassium in the leaves of plastic film house plants of pepper, cucumber, pimento, chrysanthemum, and carnation. And calcium and magnesium were more than two fold high in fruiting vegetables than flowering plants showing antagonistic utilization against potassium.
