수종의 생약 물 추출물의 BNL cl.2 Cells 보호효과 (Protective Effects of the Water Extracts of Herbal Medicine on BNL cl.2 Cells)

  • The protective effects of the water extracts of nine kinds of medicinal herbs, which have been reputed to having the hepatoprotective activity in Chinese herbal medicine, on BNL cl.2 cells using a MTT assay were investigated. Five extracts including Coriolus versica, Curcuma longa, Phellinus linteus, Po-lygonum aviculare, and Salvia miltiorrhiza showed the protective effects on BNL cl.2 cells damaged by $CCl_4$ with $ED_{50}$ values of less than $100\;{\mu}g/ml$. Silymarin had been used as a positive control.

Effect of Paeonia lactiflora Extracts on ${\alpha}-Glucosidase$

  • This study was carried out to investigate inhibitory effect of extracts from the root of Paeonia lactiflora on postprandial hyperglycemia. Organic solvent (hexane, ethyl acetate, butanol, aqueous) extracts from the crude drug were fractionated by high performance liquid chromatography. These fractions were examined to evaluate ${\alpha}-glucosidase$ (EC 3. 2. 1. 20) inhibition by microplate colorimetric assay. Among the fractions examined, the ethyl acetate fraction from the roots of Paeonia lactiflora showed potent inhibitory effects on ${\alpha}-glucosidase$. Therefore, further fractionation of the fraction was carried out to isolate the active principles. Finally, we isolated and Purified 1,2,3,4,6-penta-O-galloyl-beta-D-glucose (PGG) as a active principle by activity-guided fractionation. These results suggest that the extract from the root of Paeonia lactiflora can be used as a new nutraceutial for inhibition on postprandial hyperglycemia and PGG might be a candidate for developing an ${\alpha}-glucosidase$ inhibitor.

  • Studies on the stem bark extracts of Garcinia cuneifolia have furnished a new xanthone cuneifolin (1) and the triterpene stigmasterol (2). Structures for these compounds were elucidated based on NMR, 2D NMR, MS and GCMS data. Larvicidal activity screening of the crude bark extract using the larvae of Aedes aegypti indicated the larvae to be susceptible to these extracts. $LC_{50}$ values of the bioassays show the extracts to be moderately toxic to the larvae of Aedes aegypti.

  • In order to obtain the basic data for the preparation of yogurt containing ginseng component, the effect of ginseng component on cell growth of Lactobacillus casei YIT 9018 and on lactic acid production were investigated. Initial cell growth and acid production were markedly inhibited by the addition of ethanol extracts in the level of 8% into 15% skim milk. Crude saponin did not show any inhibitory effect on cell growth and acid production, but ether layer fraction showed inhibitory effect. It was thought to be more advantageous to add ginseng extracts after the fermentation of milk than before. The addition of ginseng extract at 8% level into liquid yogurt was most suitable in organoleptic test. Cell viability was not affected by the addition of ethanol extracts up to 8% level during storage of liquid yogurt.

  • To find anticancer agents from higher plants, DNA strand-scission assay method was employed for bioassay-guided fractionation as well as for screening the crude extracts. During the screening, an ethyl acetate extracts of the heartwood of Caesalpinia sappan L. (Leguminosae) exhibited potent DNA strand-scission activity. Therefore, the ethyl acetate extracts of the dried heartwood of C. sappan was subjected to the bioassay-guided fractionation, which led to the isolation of a known compound, brazilin (1) as the active constituent. In addition, caesalpine J (2) was also isolated as an inactive constituent.

  • Red ginseng tails were extracted with ethanol solutions over a range of concentrations and temperature conditions. Investigations were carried out to study the effects of treatments on yields, soluble solids, saponin and precipitate occured in red ginseng extract beverage during storage. It was found that: (1) Higher concentration of ethanol at low temperature resulted in less yield of crude extract (2) The amount of precipitate in the non-purified extract beverage were less with decrease in ethanol concentration used (3) The treatment for purification of extracts and storage of purified extract at 37$^{\circ}C$ for 6 months had no effect on HPLC chromatogram pattern of saponins (4) The amount of purified extract decreased by purification treatment and more decrease was found as the temperature and concentration of ethanol increased. For Preparation of red ginseng extract beverage, the treatment of extracts with ethanol at low temperature was found to be more effective to minimize precipitation in tile beverage.

프로폴리스 및 알콜 추출물의 화학성분 (Chemical Components of Propolis and Its Ethanolic Extracts)

  • 프로폴리스를 이용하여 새로운 기능성 식품을 개발하기 위하여 프로폴리스 원괴와 그 추출물의 화학성분에 대하여 조사하였다. 일반성분은 조지방 86.41%, 가용성 무질소물 7.32%, 조단백질 2.71%, 회분 1.05% 및 조섬유 0.20%로 나타났다. 프로폴리스에 함유되어 있는 주요 무기성분은 Na(120.40 mg%), Ca(115.40 mg%) 및 K(105.87 mg%) 순으로 나타났으며, Ca 함량은 알콜 추출물보다 물 추출물에서 높게 나타났다. 유리당은 총 3종이 분리, 동정되었으며, 그 중 sucrose가 152 mg%로 높게 나타났고, glucose 114 mg% 및 fructose 6 mg%로 함유되어 있었다. 프로폴리스에 함유되어 있는 주요 아미노산으로는 lysine, cystine 및 glutamic acid로 나타났으며, 그 함량은 각각 395.29 mg%, 267.66 mg% 및 248.14 mg%로 나타났다. 지방산은 총 8종이 분리되었으며, 주요 지방산으로는 oleic acid (51.89%), myristic acid (20.86%) 및 palmitic acid (20.28%)로 나타났다. 프로폴리스의 flavonol 화합물을 분석한 결과 myricetin, quercetin, apigenin 및 kaempferol이 주요 flavonol화합물로 나타났으며, 총 flavonoid 함량은 다른 추출물보다 50% 에탄올 추출물에서 가장 높게 나타났다. 프로폴리스에 함유되어 있는 polyphenol 성분들은 gallic acid, chlorogenic acid, catechin, epigallocatechin gallate, epicatechin 및 epicatechin gallate로 확인되었다.

매실 품종별 항균 및 항산화성 비교 (Comparison of Antimicrobial and Antioxidant Activities of Prunus mume Fruit in Different Cultivars)

  • 신약개발 이나 건강기능성 식품 개발의 기초 자료 제공을 목적으로 매실 품종별 일반성분을 분석하고 생리활성을 검정한 결과는 아래와 같다. 매실의 개체당 평균 수분 함량은 $89.94{\sim}90.62%$ 범위로서 품종간 큰 차이가 없었으며 단백질 함량은 앵숙, 백가하가 다소 높게 나타났지만 품종별로 큰 차이는 보이지 않았다. 조회분의 함량은 천매, 백가하가 앵숙, 남고에 비하여 회분함량이 높은 것을 확인하였으며 매실의 조섬유와 조지방의 함량변화는 품종간 유의적인 차이가 나타나지 않았다. Methanol, Water, Hexane, Ehylacetate 및 Ether 추출물의 항균활성 검색 결과 methanol에서 가장 크게 나타났으며 품종별 항균활성은 천매, 백가하가 앵숙, 남고에 비하여 약간 강한 활성을 보임을 확인하였다. 매실 추출물은 0.5 mg/mL 이상의 농도에서 세균에 대하여 억제효과를 보였으나 젖산균과 효모에서는 1.5mg/mL 이상에서도 억제효과를 나타내지 않았다. 항균활성을 가진 메탄올 추출물의 활성은 $100^{\circ}C$에서 30분 가열한 조건에서도 안정함을 확인하였고, pH 변화에 따른 안정성은 큰 차이를 보이지 않았다. 용매에 따른 전자공여능은 methanol > ethylacetate > water > ether > hexane 순으로 높게 나타났으며 품종간의 차이는 크지 않았다.

건조방법을 달리한 땅콩호박의 영양성분 분석 및 생리활성 평가 (Analysis of Nutritional Components and Physiological Activity of Butternut Squash (Cucurbita moschata) by Drying Methods)

  • The purpose of this study was to provide the basic data of butternut squash (Cucurbita moschata) according to the drying methods. The proximate composition and chromaticity of freeze-dried and hot air-dried Cucurbita moschata powders were evaluated, and the antioxidant effects of these ethanolic extracts were analyzed by DPPH, ABTS, FRAP, reducing power activity, total phenolic, and flavonoid content. As a result, Cucurbita moschata was revealed to have high carbohydrate and crude protein contents, while crude ash and crude fat contents were relatively low. Also, it was confirmed that the hot air-dried Cucurbita moschata had lower L, a, and b values than the freeze-dried Cucurbita moschata. Additionally, as a result of analyzing the antioxidant activity of Cucurbita moschata ethanolic extracts, hot air-dried Cucurbita moschata ethanolic extract showed significantly superior antioxidant activity than freeze-dried Cucurbita moschata ethanolic extract. Also, hot air-dried Cucurbita moschata ethanolic extract was revealed to have a higher polyphenol content, but slightly lower flavonoid content than freeze-dried Cucurbita moschata ethanolic extract. Based on the data from this study, further experiments on Cucurbita moschata material are necessary.

Characterization of Undaria pinnatifida Root Enzymatic Extracts Using Crude Enzyme from Shewanella oneidensis PKA 1008 and Its Anti-Inflammatory Effect

  • This study investigated the characterization and functionality of Undaria pinnatifida root (UPT) extracts, degraded using a crude enzyme from Shewanella oneidensis PKA1008. To obtain the optimum degrading conditions, the UPT was mixed with alginate degrading enzymes from S. oneidensis PKA 1008 and was incubated at 30℃ for 0, 3, 6, 12, 24, and 48 h. The alginate degrading ability of these enzymes was then evaluated by measuring the reducing sugar, viscosity, pH and chromaticity. Enzymatic extract at 24 h revealed the highest alginate degrading ability and the lowest pH value. As the incubation time increased, the lightness (L ) also decreased and was measured at its lowest value, 39.84, at 12 hours. The redness and yellowness increased gradually to 10.27 at 6 h and to 63.95 at 3 h, respectively. Moreover, the alginate oligosaccharides exhibited significant anti-inflammatory activity. These results indicate that a crude enzyme from S. oneidensis PKA 1008 can be used to enhance the polysaccharide degradation of UPT and the alginate oligosaccharides may also enhance the anti-inflammatory effect.
