A new aromatic amide, methyl-N-(3-carbamoylpropionyl) anthranilate was isolated for the first time as a natural compound and one known $C_{19}-diterpene\;alkaloid$, avadharidine was also obtained from the root of Aconitum pseudolaeve var. erectum. The\;LD_{50}$ values of water extract and MeOH extract of the root of Aconitum pseudolaeve var. erectum in mice were 1. 23 g (13. 6 g crude drug) and 0. 77 g(5. 13 g crude drug)/kg, p.o., respectively.
We report here the use of high-speed countercurrent chromatography (HSCCC) in the preparative isolation and purification of the bioactive component, fucosterol, from Pelvetia siliquosa. A crude extract was obtained by ultrasonic extraction of powdered P. siliquosa using methylene chloride and was then subjected to separation and purification by HSCCC, coupled with evaporative light-scattering detection. Preparative HSCCC was performed successfully using a two-phase solvent system, n-heptane:methanol (3:2, v/v), to obtain 10.96 mg fucosterol with 96.8% purity from 50 mg of crude extract; the recovery rate was approximately 90.5%.

Soybean lipoxyeenase isozymes were isolated from acetone-defatted soybean seeds(Glycine max [L.] Merr. variety AmSoy) by ammonium sulfate fractionation, eel filtration, and ion exchange chromatoeraphy. The final preparation of lipoxygenase-1 and -2 obtained was 19- and 32-fold purified, respectively, to the crude extract. But a considerable loss of total enzyme activity occurred during purification. On 7% polyacrylamide gel electrophosis at pH 9.0, employing lipoxigenase specific staining technique, lipoxyeenase-1, -2, and -3 showed distinctive Rf values of 0.38, 0.29, and 0.33, respectively.
This study investigated the physiological and antioxidant activities of green, oolong, and black tea extracts. The crude catechin extract yields of green, oolong, and black tea were 4.9%, 3.4%, and 2.5%, respectively. Total phenol contents of green, oolong, and black tea were 40.9%, 43.0%, and 38.5%, respectively. The order of the electron donating abilities of green, oolong and black tea were green tea>oolong tea>black tea extracts. The SOD-like activities of green, oolong and black tea extracts at 5,000 ppm were 21.2%, 17.5% and 13.9%, respectively. The nitrite-scavenging abilities of green, oolong and black tea extracts were higher than that of ascorbic acid (p<0.05). Antioxidant activities in soybean oil substrates at 500 ppm were in the order of green tea>oolong tea>black tea${\geq}$BHT (200 ppm). Therefore these results showed that the physiological and antioxidant activities of green tea were better than those of oolong and black tea.
This study was carried out in order to utilize ginseng marc (GM), a byproduct produced during the preparation of ginseng extract. For the utilization of GM as coffee-replacing beverage, GM was roasted at 140-23$0^{\circ}C$, for 10-30 min to generate coffee-like flavor and taste. The physicochemical changes in GM induced by roasting process was measured. Weight and moisture contents of GM abruptly decreased while roasting at various temperature and period of time, and the amount of water-extracted solid substances decorated beyond 23$0^{\circ}C$. Among the proximate components, total and free sugar decreased remarkably, while crude fat content increased. Crude protein content did not change by the roasting process. pH derides to pH 4.9 after 30 min at 20$0^{\circ}C$, but increased at higher temperatures. Hunter color L value of the GM powder decreased, whereas a and b values, increased up to 10 times as compared to that of the control. The brown color(O, D 490 nm) in 50% ethanol extract of the roasted GM was 46 times higher than that of the control.
Extract of drugs in commerce was administered to mice for 3 days and its fatigue effects were compared with those of P. ginseng extract and A. sessiliflorum extract. Mice were used for these experiments divided into ethyl alcohol and water extract of P. cocos, S. japonica, P. multiflorum, L. chinense, A. cochinchinensis, S. indicum, N. nucifera were given ordinary dose orally. For antifatigue test, swimming performance were determined after administration of the extract The group of P. cocos gave high improvement of the swimming performance in ethyl alcohol extract group. And the group of S. japonica an improvement was also observed in water extract group.
This study was performed to investigate the inhibitory activity of Sargassum thunbergii (ST) against ${\alpha}$-amylase and elucidate the availability of ST extract as a functional food agent. To test the inhibitory activity of ST against ${\alpha}$-amylase, porcine pancreatic ${\alpha}$-amylase and potato starch were used as substrates. It was revealed that ST crude ethanol extracts have high ${\alpha}$-amylase inhibitory activity. Subsequently, ST crude ethanol extract was separated into five partition layers by solvent extraction: n-hexane, chloroform, ethyl acetate, butanol, and water. Chloroform and n-hexane fractions showed higher inhibitory activities than did acarbose (positive control). To confirm the changes in enzyme inhibitory activity by physical treatments, ST crude ethanol extract was subjected to heat, pH, and ${\gamma}$-irradiation treatments. In all heat treatments with the exception of one ($121^{\circ}C$, 15 min), the inhibitory activity was increased compared with the untreated group. With regard to pH stability, ST extract showed no significant changes at pH 4.6, but somewhat decreased inhibitory activity was revealed at pH 2, 8, and 10. On the other hand, ST ethanol extract was stable under ${\gamma}$-irradiation under all conditions (3.20 kGy). In summary, ST ethanol extract can be used in the food industry as a natural ${\alpha}$-amylase inhibitor.
Anti-oxidative activities of Heracleum moellendorffii Hance extracts were measured after extraction with 50% (v/v) ethanol or water (at $80^{\circ}C$ or $20^{\circ}C$). The total polyphenol content was highest (64.73 mg GAE/g) in extract from 50% ethanol extraction at $80^{\circ}C$. In extracts obtained by water extraction at $80^{\circ}C$ and $20^{\circ}C$, the total polyphenol contents were 39.78 mg GAE/g and 23.17 mg GAE/g, respectively. The antioxidant activities of the ethanol extract at $80^{\circ}C$ were highest, as assessed by DPPH radical scavenging activity (69.14% at 50 ppm), reducing power (OD 0.93), and $ABTS^+$ radical-scavenging activity (85.62% at 1,000 ppm). At $80^{\circ}C$, the antioxidant activity of the 50% (v/v) ethanol extract was 8-20% higher than that of the water extract. Also, the chemical composition of extract from ethanol extraction at $80^{\circ}C$ was analyzed. The levels of moisture, crude protein, crude fat, and crude ash were 6.38%, 4.35%, 0.67%, and 1.96%, (all w/w), respectively. The Na, K, Mg, Zn, Fe, Ca, and P contents of the extract were 53.41, 398.26, 5.80, 0.40, 5.27, 3.56, and 47.17 mg/100 g, respectively. The total levels of polyphenolic compounds and flavonoids in the ethanol extract were 64.73 mg GAE/g and 49.54 mg RE/g.
The effect of temperature and ethanol concentration on lipids yield and free fatty acids were investigated during 1-5 times of 8 hours extraction. The crude lipids recovered was generally increased as the ethanol concentration increased while it was decreased at higher extraction temperature. Fatty acids composition in free fatty acids showed linoleum and plasmatic acids to be the major fatty acids, accounting 60% of 16 fatty acids identified. Fatty acids pattern was little affected by the changes in ethanol concentration while increase in temperature resulted significant changes in fatty acids composition.
