  • In order to investigate experimentally the clinical effects of Soeuminsohapwon that was prescribed to cure cerebral hemorrhage, palpitation etc, the author tested various activities of extract from the Soeuminsohapwhangwon by the method prescribed in the experimental part. The results of the studies were summarized as follows: Suppressive action was not shown on the convulsion induced by strychnine, but significant effect was noted on the convulsion induced by picrotoxin and caffeine. In acetic acid method, analgesic effect was noted. A prolongation of anesthetic time by pentobarbital sodium and antipyretic effect was observed. Relaxing action was noted on the ileum of mice, also same effect was recognized on contraction of the ileum due to acetylcholine, barium chloride and histamine. The expansion of blood vessels by relaxation of smooth muscle and hypotensive action were noted. According to the above results, effects based on oriental medical references were approximate to the actual experimental results.

  • In order to investigate experimentally the clinical effects of Taeksa-Tang that was prescribed to cure the severe dizziness, the water extract of Taeksa-Tang was experimented about the effects on the diuretic action, the serum lipid, the isolated ileum and heart, the blood vessel, the blood pressure and respiration. The results of this study were obtained as follows: The diuretic action was significantly shown in rabbits. The level of total cholesterol, triglyceride and phopholipid in serum of rabbits given high cholesterol diet were significantly decreased. The contraction of the isolated ileum of mice induced by acetylcholine chloride and barium chloride was remarkably inhibited. The vasodilating and hypotensive actions were noted in rabbits. Negative inotropic action on the isolated frog heart was recognized.

Use of Gelatin Particle Agglutination Test for the Detection of Cymbidium mosaic virus in Cattleya Plants

  • Gelatin particle agglutination test (GPAT) was used to detect Cymbidum mosaic virus (CymMV) in Cattleya plants. Gelatin particles were coated with purified anti-CymMV immunoglobulin of 25-100 $\mu\textrm{g}$/ml and were subjected to several different concentrations of purified CyMfV as well as varying dilutions of orchid leaf extracts. The GPAT detected purified CymMV up to a minimum concentration of 10 $\mu\textrm{g}$/ml. CymMV was detected from crude sap extract of infected Cattleya leaves and roots up to 1:51,200 and 1:25,600 dilutions, respectively. However, the optimum range of leaf and root sap dilutions was between 50-100. Non-specific reactions were not encountered from any of the healthy orchid plants tested. The entire GPAT process was completed within 2-3 hours. This test was found to be very useful for the detection of CymMV in orchids because it is sensitive, economical, and easy to perform.

  • The paeonol content in roasted Moutan Cortex was increased about three times compared to that of untreated one. The paeonol content reached at its maximum level $(454.3\;{\mu}g/mg\;ethanol\;extract)$ after roasting at $190^{\circ}C$ for 30 minutes. Roasting processing did not affect on the DPPH radical scavenging activity of Moutan extracts.

  • The localization of carbon monoxide dehydrogenases (CO-DHs) in Pseudomonas carvoxydovorans and Acinetobacter sp.1 was examined by comparison of the distribution of CO-oxidizing activity between soluble and particulate fractions obtained after disruption of CO-grown cells by sonic oscillation and of spheroplasts by osmotic shock. When the cells were broken by sonic oscillation, most of the CO-DH activity was recovered from soluble fractions. However, disryption by osmotic lysis of spheroplasts revealed that the enzyme activity is present in the cell membrane. The results indicated the CO-DHs in these cells are loosely attached to the cytoplasmic membrane.

  • For the purpose of utilizing tannins in the functional foods and crude drugs the xanthine oxidase inhibition of tannins isolated from Korean green tea was determined. Acetone extract from Korean green tea showed inhibitory effect against the xanthine oxidase. The galloyl tannins showed higher inhibitory activity against xanthine oxidase than the nongalloyl tannins. In terms of stereo isomers, (-)-epicatechins had higher inhibitory activity than the (+)-catechins. The synergistic activity was also observed. Tannins isolated from Korean green tea appeared to be incompetitive inhibitor against the xanthine oxidase.

In vitro and in vivo antidiarrhoeal activity of epigallocatechin 3-gallate: a major catechin isolated from indian green tea

  • Epigallocatechin 3-gallate (EGCG), one of the major catechins of tea, was isolated from the decaffeinated, crude methanolic extract of Indian green tea (Camellia sinensis L. O. Kuntze) using chromatographic techniques. EGCG was then screened for antidiarrhoeal activity against 30 strains (clinical isolates) of V. cholerae, which is a well known Gram negative bacillus functioning as the pathogen of cholera. V. cholerae strains like V. cholerae 69, 71, 83, 214, 978, 1021, 1315, 1347, 1348, 569B and ATCC 14033 were inhibited by EGCG at a concentration of $25\;{\mu}g/ml$ whereas V. cholerae 10, 522, 976 were even more sensitive, being inhibited at $10\;{\mu}g/ml$ level. However, V. cholerae DN 16, DN 26, 30, 42, 56, 58, 113, 117, 564, 593, 972 and ATCC 14035 were inhibited at $50\;{\mu}g/ml$ level of EGCG. Only four strains were inhibited at $100\;{\mu}g/ml$. In this study the isolated compound was found to be bacteriostatic in its mechanism of action. In the in vivo experiment using the rabbit ileal loop model two different dosages of EGCG ($500\;{\mu}g/ml$ and $1,000\;{\mu}g/ml$) were able to protect the animals when they were challenged with V. cholerae 569B in the ileum.

Selective Stimulating Effect of Flavonoids on the Antioxidant Defense System in Normal and Transformed Hepatic Cell Lines

  • Previously, a flavonoid fraction, which consisted mainly of protocatechuic acid, fustin, fisetin, sulfuretin, and butein, here named RCMF $({\underline{R}}VS\;{\underline{c}}hloroform-{\underline{m}}ethanol\;{\underline{f}}raction)$, was prepared from a crude acetone extract of Rhus verniciflua Stokes (RVS) which is traditionally used as a food additive and as an herbal medicine. In this study, we evaluated the effects of RCMF on the antioxidant defense system using embryonic normal hepatic cell line (BNL CL.2) and its SV40-mediated transformed cell line (BNL SV A.8). This study demonstrates that RCMF selectively stimulated the antioxidant defense system of normal cells, as BNL CL.2 cells proved to be more sensitive to RCMF-mediated increases of superoxide dismutase, catalase, glutathione, and glutathione reductase than BNL SV A.8 cells. In particular, RCMF caused a significant increase in the malonaldehyde content of BNL SV A.8 cells, which is believed to be closely associated with cytotoxicity of RCMF and RCMF-mediated growth inhibition. Collectively, our findings suggest that the flavonoid fraction, RCMF, selectively stimulates the antioxidant defense system in normal rather than hepatic tumor cells.

  • These studies were conducted to investigate the effects of Tongkwan-San water extract on analgesic, sedatative, anti-inflammatory, blood pressure and vasodilating actions, the relaxing action of isolated ileums and actions on the contact dermatitis induced by picryl chloride and on the leakage of the dye into the peritoneal cavity. The results of these studies were summarized as follows: The analgesic effect of Tongkwan-San was noted. The prolongation of anesthetic time of Tongkwan-San was recognized. Spontaneous motilities of isolated ileum of mice were strongly suppressed by Tongkwan-San. It inhibited the contractions of isolated ileum of mice induced by acetylcholine and barium chloride and the contraction of isolated ileum of guinea-pig induced by histamine. Inhibition of the contact dermatitis induced by picryl chloride was recognized. Anti-inflammatory effects in the paw edema induced by histamine and dextran were significantly shown. The leakage of dye into the peritoneal cavity in mice was significantly inhibited. Hypotensive and vasodilating action due to vascular smooth muscle relaxation were noted in rats and rabbits.

Hepatoprotective and Antioxidative Effects of Alisma orientale

  • The rhizome of Alisma orientale Juzep (Alismataceae) has been used as a crude drug for diabetes, edema, inflammation and urinary disturbances in oriental medicine. Recent animal studies have shown that the extract of Alisma orientale rhizome (AOR) can potently lower high levels of serum lipids and improve insulin resistance, which are usually detected in patients and animals with non-alcoholic fatty liver disease. So, we studied the antioxidative effects of AOR extracts and fraction in vitro and their protective effects against acute hepatotoxicity induced by $CCl_4$ in vivo.. We then investigated the effects of each fraction on hepatotoxicity induced by tert-butyl hydroperoxide (t-BHP). DAOR (dichloromethane fraction of the Alisma orientale rhizome) scavenged free radicals and superoxide anions. DAOR protected against $CCl_4$ induced hepatotoxicity. DAOR had hepatoprotective and antioxidative effects against t-BHP-induced hepatotoxicity in HepG2 cells and in rats.
