Identification of the Capsid Protein-binding Region of the SL1(+) RNA Located at the 5' Region of the Potato virus X Genome

  • $Potato$ $virus$ $X$ (PVX) contains $cis$-acting elements including stem-loop 1 (SL1) RNA at the 5' region; SL1 is conserved among all potexviruses. The SL1 at the positive-sense RNA, SL1(+), is required for PVX RNA replication, cell-to-cell movement, and translation. Previous research demonstrated that SL1(+) RNA also serves as the origin of assembly for encapsidation of PVX RNA. To identify the essential sequences and/or regions for capsid protein (CP) subunit recognition within SL1(+) RNA, we used electrophoretic mobility shift assays (EMSA), UV cross-linking, and yeast three-hybrid analyses. The EMSA and UV cross-linking analyses with PVX CP subunits and RNA transcripts corresponding to the SL1(+) RNA showed that the SL1(+) RNA formed complexes with CP subunits. We also conducted EMSA and yeast three-hybrid analyses with RNAs containing various mutations of SL1(+) RNA elements. These analyses indicated that SL1(+) RNA is required for the interaction with PVX CP and that the RNA sequences located at the loop C and tetra loop of the SL1(+) are crucial for CP binding. These results indicate that, in addition to being important for RNA accumulation, the SL1(+) RNA from the 5' region of the PVX genome is also required for specific binding of PVX CP.

Diallel Analysis for Rice Allelopathic Potential against Barnyardgrass Assessed in Field Condition

  • A complete diallel cross was performed to determine the inheritance of allelopathic potential and combining ability of six rice germplasm that showed different level of allelopathic potential against barnyardgrass. Parents and F1 of 30 crosses were assessed for allelophatic potential in field condition. Barnyardgrass (Echinochloa crus-galli (L.) Beauv. var. frumentaceae) was used for rice allelopathic potential indicator which was observed in height and shoot dry weight, then suppression percentage were calculated by comparing to barnyardgrass grown in control plot (no rice). It was found that general combining ability showed no significant effect, whereas specific combining ability and reciprocal effect were significant. Variance analysis confirmed that genetic control of allelopathic potential in rice against barnyardgrass was mostly directed by dominance gene effect. The dominant genes were associated with high allelopathic potential and recessive genes were associated with low allelopathic potential. Heritability in broad sense for barnyardgrass dry weight suppression was around 41%; whereas for barnyardgrass height suppression was 51%. However, heritability in narrow sense was very low. The finding suggests that accumulation of genes associated with allelopathic potential would be a strategy to develop promising varieties in reference with the specific combining ability of cross combinations.

A New White Waxy Corn 'Daedukchal 1' Hybrid with High Yield and High Table Quality

  • A new waxy corn hybrid 'Daedukchal 1' was developed from single cross between CNU 504 and CNU 2396 at Chungnam National University in 2007. Inbred CNU 504 used as a seed parent and inbred CNU 2396 as a pollen parent were collected throughout domestic regions. Tasseling date of this hybrid was seven days later than that of check hybrid, 'Chalok 1'. 'Daedukchal 1' was 19.7 cm in ear length and 4.5 cm in ear diameter. On yield trial, ear length of this hybrid was increased 29.7% compared with a check hybrid, 'Chalok 1'. 'Daedukchal 1' had white kernels and good eating quality due to high sugar content and thinner pericarp. The ratio of kernel set length/ear length was similar to 'Chalok 1'. It is moderately resistant to southern leaf blight and northern leaf blight but susceptible to corn borer. The yields of 'Daedukchal 1' in fresh ear weight and in number of fresh ear were 16% and 8%, respectively, higher than those of a check hybrid in regional yield trials for two years. Seed production for this hybrid was better because of the consistency of tasseling dates.

Comparison of Lines from Anther and Maternally-derived dihaploids, Single-seed Descent and Bulk Breeding Method in Flue-cured Tobacco (Nicotiana tabacum L.) (연초(Nicotiana tabacum L.)의 반수체 육종법 1주1계통법, 집단육종에 의한 육성계통의 특성비교)

  • The present study was conducted to compare the relative efficiency of four different breeding methods in tobacco varietal development. A single Fl hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L Bright Yellow 4(BY4) and NC95, was selfed. F2population above cross was screened for resistance against bacterial wilt caused by Pseudommonas solanacearum E.F.Smith under the naturally infested field conditions, and the 30 lines were developed from F2 individual plant by anther culture (ADH), maternal method utilizing Nicotiana afpicana (MDH), single- seed descent(SSD) and Bulk breeding method, respectively. All characters except content of total alkaloids of ADH and MDH which wore bred by haploid methods reduced more than that of lines bred by conventional methods(SSD & Bulk) : however, the yields were 8% lower than other lines. The total alkaloid content of ADH was higher than that of MDH, and yield was reduced about 4n even though the number of leaf was identical with the MDH. All other characters of ADH were also reduced significantly. In the lines bred by conventional methods, population developed by SSD showed significantly wider leaf width, shorter plant height, later days to flower, and lower in percent reducing sugar than those by Bulk. The populations derived from haploid method showed greater phenotypic variance and wider range of variation than conventionally developed ones. The results obtained indicate that selection will provide a significantly greater genetic gain for leaf number and leaf length in the ADU and MDH populations, and for plant height and days to flower in the SSD and Bulk populations.

A New Self-Incompatible Interspecific Hybrid Pasqueflower Variety, 'Yeonhong' (자가불화합성 종간교잡종 할미꽃 신품종 '연홍')

  • A new hybrid variety of pasqueflower, 'Yeonhong', was derived from an interspecific cross between Pulsatilla davurica and P. koreana at the Jeollanamdo Agricultural Research and Extension Services (JARES) in Naju, Korea. The original cross was performed in 2001, and the 'Yeonhong' hybrid was established in 2003 after two years of selective breeding. 'Yeonhong' is characterized by deep pink flowers and is self-incompatible. The inherent characteristics of the variety are deep yellow anthers, a deep pink stigma, six petals, and green leaf color. 'Yeonhong' blooms twice a year. The agronomic characteristics of the variety are 26.5 flowers per plant, 58.6 cm in flower height, 46.8 cm in cut flower length, 11.5 cm in bract width, and 34.4 cm in leaf length. Flower characteristics include 8.4 days of longevity for cut flowers and 34 days of longevity for bracts. The new variety is suitable for use as a cut flower.

  • New standard carnation cultivar "Jasujung" was bred from the cross of red standard cultivar "Francesco" and purple standard cultivar "Saturn" at the National Institute of Horticultural & Herbal Science. It was crossed in 1996 and finally selected in 2000 after investigation of the characteristics for 4 years from 1997 to 2000. "Jasujung", purple standard cultivar for cut flower, has good fragrance and long stem. Especially, it is strong resistance against Fusarium wilt. This cultivar is recommended to grow over $8^{\circ}C$ at night and under $25^{\circ}C$ at days.

Mapping QTL for Grain Quality Traits Using an Introgression Line Population from a Cross between Ilpumbyeo and Moroberekan in Rice (일품벼/모로베레칸 이입계통을 이용한 미질특성 관련 QTL 분석)

  • We conducted a QTL analysis of grain quality traits using 117 $BC_3F_4$ and $BC_3F_5$ lines developed from a cross between Ilpumbyeo and Moroberekan. Genotypes of 117 $BC_3F_5$ lines were determined using 134 simple sequence repeat (SSR) markers. A linkage map constructed using 134 SSR markers was employed to characterize quantitative trait loci (QTL). The 117 $BC_3F_4$ and $BC_3F_5$ lines were evaluated for eleven grain quality traits in 2005 and 2006. A total of 18 QTLs were identified for eleven traits, and the phenotypic variance explained by each QTL ranged from 9.9% to 35.2%. Moroberekan alleles contributed positive effects in the Ilpumbyeo background at two QTL loci for 1,000 grain weight. Four QTLs, two for chalky rice and one each for 1,000 grain weight and head rice were consistently detected in two consecutive years indicating that these QTLs are stable. Clusters of QTLs were observed in three chromosome regions. One cluster harboring five QTLs including head rice and brown rice ratio near SSR markers RM190 and RM314 was detected on chromosome 6. Another cluster harboring grain weight and white belly was detected on chromosome 2. Increase in white belly at this locus might be due to the increase in grain weight due to the presence of the Moroberekan allele. The Moroberekan alleles at two QTL loci, gw3 and gw4 associated with increased grain weight might be useful in breeding programs to develop high-yielding cultivars.

Characterization of a New High-lysine Mutant in Barley (Hordeum vulgare L.)

  • A chemical, MNU-induced hulless barley mutant line designated as 'Mutant 98 (M98)' was developed from a Korean hulless waxy barley cultivar, 'Chalssalbori'. The objective of the study was to determine the genetic basis of 'M98' and the possibility of using 'M98' as breeding parent to improve lysine level. Compared to 'Chalssalbori', 'M98' had large embryo and higher lysine content in both the embryo and endosperm. Significantly different lysine content in 'M98' and the other high-lysine barley mutant stocks was observed for two years. However, the genotype by year interaction was not significant. 'M98' was higher than the other high-lysine barley mutant stocks in the percentage of lysine of total amino acid composition (0.75%). The trait of shrunken endosperm of 'M98', which was typical in the high-lysine mutants, was inherited by a single recessive gene. Based on seed morphology and lysine content of $F_1$ seeds, 'M98' had a genetically different gene from the other high-lysine mutants for shrunken endosperm. Segregation of $F_2$ for plump/shrunken endosperm did not fit the expected ratio of Mendelian inheritance except for only one cross combination (GSHO1784 (lys1)/M98). The amino acid analysis of $F_5$ and $F_6$ progenies from the cross between 'M98' and 'Chalssalbori' revealed that the attempt to increase the range of lysine content of plump lines did not go beyond the limit of the average high-lysine barley germplasm.

A New Rice Variety 'Superjami' with High Content of Cyanidin 3-glucoside (Cyanidin 3-glucoside(C3G) 함량이 높은 고기능성 벼 신품종 '슈퍼자미')

  • 'Superjami', a new blackish purple pigmented rice cultivar was derived from a cross between 'CG2-3-5-1-6-1' (developed from a cross between 'Heugjinjubyeo' and 'Suweon 425') as high amount of C3G (Cyanidin 3-glucoside) and 'Daeribbyeo 1' with large seed size in 2003, and selected by pedigree breeding method until $F_7$ generation. As a result, a promising line, 'SR28721-7-9-3-1-2-1', was developed and designated as the name of 'KNOU 5' in 2008. This variety has about 125 days to heading after sowing and has 74.9 cm culm height. The ratio of fertility of 'Superjami' was about 89.6% and 1,000-grain weight was about 26.2 g. It was 1.5 times heavier than 'Heugjinjubyeo'. 'Superjami' has 10 times higher C3G content compared with 'Heugjinjubyeo'. The yield potential of 'Superjami' in brown rice was about 6.3 MT/ha at ordinary fertilizer level in local adaptability test.

Analysis of genome variants in dwarf soybean lines obtained in F6 derived from cross of normal parents (cultivated and wild soybean)

  • Plant height is an important component of plant architecture and significantly affects crop breeding practices and yield. We studied DNA variations derived from F5 recombinant inbred lines (RILs) with 96.8% homozygous genotypes. Here, we report DNA variations between the normal and dwarf members of four lines harvested from a single seed parent in an F6 RIL population derived from a cross between Glycine max var. Peking and Glycine soja IT182936. Whole genome sequencing was carried out, and the DNA variations in the whole genome were compared between the normal and dwarf samples. We found a large number of DNA variations in both the dwarf and semi-dwarf lines, with one single nucleotide polymorphism (SNP) per at least 3.68 kb in the dwarf lines and 1 SNP per 11.13 kb of the whole genome. This value is 2.18 times higher than the expected DNA variation in the F6 population. A total of 186 SNPs and 241 SNPs were discovered in the coding regions of the dwarf lines 1282 and 1303, respectively, and we discovered 33 homogeneous nonsynonymous SNPs that occurred at the same loci in each set of dwarf and normal soybean. Of them, five SNPs were in the same positions between lines 1282 and 1303. Our results provide important information for improving our understanding of the genetics of soybean plant height and crop breeding. These polymorphisms could be useful genetic resources for plant breeders, geneticists, and biologists for future molecular biology and breeding projects.
