Investigation into Disease and Pest Incidence of Panax ginseng in Jeonbuk Province (전북지방의 인삼에 발생하는 주요 병해충 조사)

  • This study was carried out to reduce the number of chemical treatment by optimal apply for the disease and pest and obtain the basal data of environmental-friendly cultivation in Panax ginseng in 2006. The result by checking disease Incidence and pests fried in ginseng field of jeollabuk-Do was as follows. The kind of disease occurred in Jinan was 8 including Rhizoctonia solani, 7 with Botrytis cinerea in Jeongeup, and also 6 with Botrytis cinerea in Kochang within Jeollabuk-Do. It was required thorough disease control before the rainy season because the occurrence time was peaked around July. Also, the most serious disease in Jeollabuk-Do was Alternaria alternata, Botrytis cinerera, and Colletotrichum gloeosporioides. The actual harmful pests in ginseng field were Asusta despecta steboldiana and Holotrichia sp. and in other method using black light trap, Maladera orientalis, Ostrinia furnacalis, and Holotrichia morosa were mainly trapped.

Rapid Identification of Potato Scab Causing Streptomyces spp. from Soil Using Pathogenicity Specific Primers

  • The plant-pathogenic species S. scabies, S. acidiscabies, and S. turgidiscabies cause the scab disease of potato and produce the phytotoxins, thaxtomins. necl, a gene conferring a necrogenic phenotype, is involved in pathogenicity and physically linked to the thaxtomin A biosynthetic genes. Identification of the pathogenic strains of Streptomyces from soil was performed through the polymerase chain reaction by using specific pathogenicity primer sets derived from the necl gene sequences of Streptomyces smbies. The DNA was extracted from soil using a bead-beating machine and modifications of the FastPrep system. The DNA was suitable for direct use in the PCR. The PCR products showed the bands of approximately 460 bp. This methods can be very usuful in identifying species responsible for scab diseases and studying on the ecology of plant-pathogenic Streptomyces spp.

Development of Resistance Evaluation Method for Powdery Mildew (Leveillula taurica) in Capsicum spp. (고추 흰가루병 저항성 평가방법 개발)

  • Pepper powdery mildew causes increasingly economical damage due to increased cultivation of pepper in greenhouses. To assess resistance of pepper resources against pepper powdery mildew, we developed a novel evaluation formula for pepper resistance against powdery mildew. The evaluation formula named S index is as follows; (number of the highest disease leaf/top leaf number)-(number of the lowest disease leaf/top leaf number). Positive correlation (81%, P = 0.01) between S index and authentic disease leaf rate was observed from the pepper plants infected by powdery mildew. Various pepper species from our genetic resources were evaluated to identify pepper varieties conferring resistance against powdery mild using S-index. Capsicum frutescens accessions 3CA131 and C. baccatum accessions 3CA0162, 3CA174, 3CA176 showed high resistance to powdery mildew, but none of C. annuum was resistant. Results suggest that S-index proposed in this study is useful to assess resistance evaluation of powdery mildew in chili pepper breeding.

  • Kokang is a high-quality, disease resistant and high-yielding Rehmannia glutinosa cultivar developed by the medicinal crop breeding team of National Institute of Crop Science, RDA, during the period from 1997 to 2005. The reproduction of Rehmania glutinosa has been accomplished mainly by vegetative propagation with its seedlings have many variants. The cultivar was selected from seedling of Jihwang 1. The plant type of Kokang is some rising from ground. It has higher disease resistance, catalpol content and extract content compared with Jihwang 1. Regional yield trials conducted at three site from 2003 to 2005. The root yield of Kokang was 11.8ton per hectare, which was increased 13% compared with a check variety, Jihwang 1. This cultivar is adaptable to the whole of Korea except for mountain areas.

The Disease Severity and Related Pathogens Caused by Root Rot on 6 Years Old Ginseng Cultivation Fields (인삼 6 년근 수확지의 뿌리썩음병 발생현황 및 관련 병원균 동정)

  • Ginseng (Pnanx ginseng C. A. Meyer) is famous worldwide, and is very important cash crop and medicinal herb in Korea. It takes four to five years to produce harvestable ginseng roots, and ginseng is attacked by several pathogens during cultivation. We investigated the disease rate caused by ginseng root rot from 6 years old ginseng cultivation fields (Chungnam; 9 fields, Chungbuk; 11 fields, Gangwon 5 fields). The highest disease severity was Dangjin D (2.9) and the lowest one was Gaesan C (0.6). Of the 625 isolations, 340 isolations were classified as Ilyonectria radicicola and Fusarium solani. Finally, genetic diversity of I. radicicola and F. solani was confirmed by sequence analysis. Among the I. radicicola group, I. mors-panacis, which is known as highly virulent pathogen, and I. liriodendri, I. robusta and I. cyclamicicola, which are weakly virulent pathogens, were identified. In the case of F. solani, it is divided into two groups, but it is necessary to conduct diversity research through genetic analysis and pathogenetic studies using various markers. Based on these results, it could be used as a basic data for control of ginseng root rot pathogens.

Outbreak of Fire Blight of Apple and Pear and Its Characteristics in Korea in 2019 (2019년 국내 사과와 배 화상병 대발생과 그 특징)

  • To find out the cause of the fire blight outbreak in apples and pears of Korea in 2019, we investigated disease appearing situation of thirty fire blight infected orchards, and interviewed farmers to determine the cultivation characteristics. Fire blight occurred mostly in orchards that had infected more than 2 years before. The cause of this were as follows: farmers did not know the symptoms of the disease properly. It is presumed that it has spread from the first occurrence to the surrounding orchards by flower-visiting insects or farmers and to a short distance or a long distance by the same cultivator or co-farmer. These series of processes repeated in the newly spreading area, and then disease reports increased as farmers became aware of fire blight. To minimize the spread of fire blight in Korea, it suggested that thorough education of farmers for early diagnosis and quantitative detection technology that can diagnose even in no symptom showing plants. And chemical or biological spraying systems suitable for domestic cultivation methods, which are producing large fruits, and molecular epidemiological studies of pathogens.

Improving Efficiency and Effectiveness of Disinfection by Soaking Seeds before Rice Seed Disinfection (벼 종자소독 전 침종에 따른 소독효과 및 효율성 향상)

  • Seed disinfection is a necessary process to decrease Bakanae disease in rice and is carried out at 30℃ to increase disinfection efficiency. However, the length of sprout after seed disinfection is not suitable for sowing, so additional processes such as seed soaking is needed. For this reason, this study was conducted to develop the efficient and effective seed disinfection method. When the seed disinfection finished after soaking seeds in cold water for 2 days, length of sprout was about 1.0 mm and sowing can be done without additional processes. In addition, the incidence of Bakanae disease was greatly reduced by 64% compared to conventional method. In order to find out why the newly developed method (disinfection after soaking) is more efficient than conventional method, soaked and not soaked seeds were stained using methylene blue staining. It was found that soaked seeds were more easily stained with about 50% of internal and external endosperm stained. Therefore, pesticide can be absorbed easily into internal endosperm of soaked seeds and efficiently destroy the spore of pathogen. As a results of this study, newly developed method will be useful as it can improve efficiency of agricultural work and also effectively decrease Bakanae disease compared to conventional method.

Artificial Screening for Black Rot Resistance Based on Different Disease Parameter in Early Cauliflower

  • India has maximum genetic materials in early cauliflower, which grow in subtropical conditions. Different disease parameters like linear growth, maximum growth rate per day, AUDPC, apparent infection rate and percent diseased area were calculated in artificially inoculated plants. Apparent infection rate is not co-related with the black rot disease incidence and should never be considered during characterization of disease resistance and varietal screening. Based on the above disease parameters Kunwari-18, Phool Gobhi Kunwari, Kataki-7 and BT-10-2 were selected as moderately resistance to black rot in early cauliflower. These lines can be used for black rot prone area and also for black rot disease improvement programme. Considering the qualitative and quantitative parameters, slow rotting resistance cauliflower lines are selected as such for cultivation and would be best suited in integrated disease programme.

Antifungal Activity of Impatiens balsamina against Ginseng Pathogen Alternaria panax (봉선화(Impatiens balsamina)의 인삼점무늬병균에 대한 항균활성)

  • This study was conducted to find out environment-friendly disease control method to Alternaria blight caused by Alternaria panax of ginseng. For this study, 150 methanol extracts from medicinal plants were evaluated and the extract of Impatiens balsamina showed most strong antifungal activity against A. panax. The methanol extract of I. balsamina showed also stable antifungal activity against other ginseng pathogens such as Botrytis cinerea and Fusarium sp., when treated by heat or pH. In vivo, Alternaria blight incidence rate was low of 13% with the treatment of I. balsamina methanol extract compared to 35% of the non-treatment. The antifungal compound of I. balsamina was purified and identified by using a silica gel column chromatography, TLC and ESI-LC/MS/MS analysis. The compound which showed strong antifungal activity was identified as 2-methoxy-1,4-naphtoquinone ($C_{11}H_8O_3$).

Development of Early Maturing Rice Stripe Virus Disease-Resistant 'Haedamssal' through Marker-Assisted Selection (MAS를 이용한 줄무늬잎마름병 저항성 조생종 벼 '해담쌀' 개발)

  • 'Haedamssal' is an early maturing and rice stripe virus disease-resistant cultivar adaptable for early-transplanting cultivation that was developed by the rice breeding team of the Department of Southern Crop, NICS, RDA, in 2014. This cultivar was derived from the cross YR25869 (YR21247-B-B-B-49-1/Sasanishiki BL4//Koshihikari) and YR25868 (Unkwang//YR21247-B-B-B-49-1/Sasanishiki BL4) made in the 2005/2006 winter season and was advanced to the F5 generation by a bulk breeding method using rapid generation advance. To incorporate rice stripe virus resistance, marker-assisted selection on the RSV gene was conducted in 3-way and 6-way cross F1 generation using the tightly linked marker RM6897. From testing in the replicated yield trial in 2011, a promising line YR26258-B-B-B-33-3 was selected and it was designated as 'Milyang276'. A local adaptability test of 'Milyang276' was performed at three locations from 2012 to 2014 and it was named as 'Haedamssal', which was a good eating quality variety. The culm length was 67 cm in yield trials, which was 4 cm shorter than 'Jopyeong'. The number of spikelets per panicle was lower than 'Jopyeong', whereas the number of tillers per hill was higher. This variety was resistant to RSV disease, bacterial blight, and leaf blast disease. The milled rice yield of 'Haedamssal' was 5.48 MT per ha at the early transplanting in the local adaptability test. 'Haedamssal' is well adapted to early transplanting cultivation in the southern plain area (Registration No. 6811).
