Production of high-quality horses is important to make the horse industry grow. Grazing during the growing period can be an important factor affecting the production of high-quality horses. The objective of this study was to determine the effects of grazing on growing horses by analyzing their blood components and intestinal microbiota. Twelve growing horses for evaluating blood components and ten growing horses for evaluating intestinal microbiota were raised for about seven months and separated by two treatments: grazing vs. stable. Complete blood count, blood chemistry, and creatine kinase levels were analyzed as blood components and a 16s rRNA gene sequence analysis was performed to analyze intestinal microbiota. Calcium ions tended to be lower in the group with grazing treatment. Alkaline phosphatase and creatine kinase tended to be higher in the group with grazing treatment. These results indicate that grazing can provide horses with more exercise than staying in stables. At the phylum level, Firmicutes/Bacteroidetes ratios in grazing and stable groups were 4.2 and 6.5, respectively. Because various studies have reported that a. high Firmicutes/Bacteroidetes ratio indicates obesity, the method of raising horses might affect their physical ability. At the species level, rates of Clostridium butyricum in grazing and stable groups were 3.2% and 13.1%, respectively. Some strains of C. butyricum can cause several diseases such as botulism. These results indicate that grazing can positively affect growing horses in terms of blood components and intestinal microbiota. Moreover, grazing can be helpful to make growing horses healthy through proper exercise.
본 연구는 매실식초 및 홍삼농축액 혼합 음료의 증진된 피로회복 효과를 확인하고자 유기산 함량과 고강도 운동 쥐모델에서 항피로 관련 인자를 측정하였다. 홍삼농축액 및 매실식초 혼합 음료 내 유기산 함량은 acetic acid가 1,654.17 ppm으로 가장 높은 함량을 보였고, malic acid와 citric acid가 각각 617.28, 600.12 ppm으로 확인되었다. 항피로 관련 인자 중 지구력을 측정한 결과, 홍삼매실식초 혼합음료(RPV)를 섭취한 군에서 138.83 min으로 가장 높은 지구력을 보였으며, 혈중 무기인산과 암모니아 및 젖산 함량은 0.67 mM, 24.28 mmol/L 및 1.75 mM으로 유의적으로 가장 낮은 수치를 보였다. 또한, 간 및 근육 내 글리코겐 함량에서 RPV는 각각 288.4, 22.84 ㎍/g으로 가장 높은 함량을 나타냈다. RPV의 혈청 및 근육조직 내 creatine kinase 함량은 465.42와 33.05 U/L로 유의적으로 가장 낮은 값으로 확인되었으며, 근육 내 lactate dehydrogenase 함량은 RPV에서 가장 높은 값인 140.13 U/L로 나타났다. 본 연구결과를 통해 풍부한 유기산을 가진 매실식초 및 홍삼농축액 혼합 음료의 섭취는 매실식초 또는 홍삼농축액의 단일 섭취에 비해 증진된 항피로 효과를 나타낸다는 것을 확인하였으며, 이는 피로회복 기능성식품 소재로 이용할 수 있을 것으로 기대한다.
Development of mouse fetus brains can be defined morphologically and functionally by three developmental stages, embryo day (ED) 16, postnatal stage one week and eight weeks. These defined stages of brain development may be closely associated with differential gene expression rates due to limited cellular resources such as energy, space, and free water. Complex patterns of expressed genes and proteins during brain development suggests the changes in relative concentrations of proteins rather than the increase in numbers of new gene products. This study was designed to evaluate early protein expression pattern in mouse fetus brain. The mouse brain proteome of fetus at ED 15.5, and 19.5 was obtained using 2-dimensional gel electrophoresis (DE). Analysis of the 2-DE gels in pH 3-10 range revealed the presence of 15 differentially expressed spots, of which 11 spots were identified to be known proteins following MALDI-TOF analysis; 3 spots were up-regulated and 8 spots were down-regulated in the mouse fetus brain at ED 15.5. UP-regulated proteins were identified as MCG18238, isoform M2 of pyruvate kinase isozymes M1/M2, isoform 2 of heterogeneous nuclear ribonucleoprotein K, heterogeneous nuclear ribonucleoprotein H2, creatine kinase B-type, 40S ribosomal protein SA and hemoglobin subunit beta-H1. Down-regulated proteins were putative uncharacterized protein, lactoylglutathione lyase and secreted acidic cysteine rich glycoprotein. Our results revealed composite profiles of mouse fetus brain proteins related to mouse fetus development by 2-DE analysis implying possible roles of these proteins in neural differentiation.
The water extract of Sophorae radix was tested for its preventive effects against cardiovascular anaphylaxis elicited in experimental animals. H₂O and ethyl acetate fractionation of Sophorae radix water extract improved anaphylactic cardiac dysfunction in passively sensitized isolated guinea hearts: improvement was noted in the maximal contractile force, post-challenge contractile force, post-challenge coronary flow and creatine kinase change elevation. These results suggest that H₂O and ethyl acute fractionation of Sophorae radix water extract possesses anti-anaphylactic effect in cardiovascular system.
High-intensive endurance exercises induce cell changes in body, changes in structures and functions of the heart, the muscles, the cartilages, and the liver, as well as increase of inflammatory cytokine. The purpose of this study was to estimate the biochemical changes in the liver and muscles during ultra-marathon race (100 km) by sections. The blood of the subjects was collected before the marathon as a control in order to analyze serum creatine kinase (CK), lactic dehydrogenase (LDH), asprtate aminotransferase (AST), alanine aminotransferase (ALT), total(T)-bilirubin, direct(D)-bilirubin, total protein, albumin, uric acid, gamma-glutamyltranspeptidase (${\gamma}$-GTP), alkaline phosphatase (ALP), creatinine, blood urea nitrogen (BUN), and high sensitive C-reactive protein (hs-CRP) concentrations. The CK, LDH, D-bilirubin, AST and ALT concentrations at 50 km and 100 km were significantly increased compared to the control (P<0.05). The markers at 100 km were higher than those at 50 km (P<0.05). The T-bilirubin and hs-CRP concentrations showed no difference among the groups, whereas the markers at 100 km were higher than those of the control and at 50 km (P<0.05). In conclusion, this study shows that the ultra-marathon race (100 km) may induce the damage of the skeletal muscle, liver and kidney, intravascular hemolysis and inflammatory responses.
An experiment was conducted to study the effects of high dietary copper supplementation on the gastrointestinal tract morphology of broiler chickens. Eighty 3-week-old broiler chicks were divided randomly into eight groups of four dietary treatments and over three week were fed isoenergetic and isonitrogenous diets that contained 0, 100, 250, or 500 mg/kg of supplemental copper from cupric sulfate. The copper supplementation in the broiler diet up to 250 mg/kg did not significantly influence broilers' performance. A high dietary copper supplementation of 500 mg/kg did significantly depress growth and feed conversion in the broilers (p<0.05). Copper supplementation more than 250 mg/kg in the broiler diet significantly influenced the morphology of the GI tract, as shown by severe oral lesions and gizzard erosion. It also significantly depressed the villi height and significantly thickened the muscular layer in the duodenum (p<0.05). The severely damaged villi were observed by scanning electronic microscope from the duodenum samples of broilers fed a 500 mg/kg copper supplemented diet. The 500 mg/kg copper supplemented diet also significantly influenced the plasma constituents. Plasma glucose concentration was significantly depressed (p<0.05).
In recent, many cardiac centers have preferred off-pump coronary artery bypass grafting (CABG) to on-pump CABG to prevent the adverse effects of cardiopulmonary bypass. The present study was performed to prove beneficial effects of off-pump CABG. Sixty adult patients scheduled for elective CABG were randomly assigned to On-pump group (n=30) or Off-pump group (n=30). Arterial blood samples were drawn before and after the operation (Pre-OP and Post-OP, respectively) for measuring CBC, prothrombin time, activated thromboplastine time, blood gas analysis, creatine kinase-MB (CK-MB) level, and lactate dehydrogenase (LDH) level. Perioperative parameters including heparin and protamine usages, complications, blood components usages, blood loss, ventilation and ICU-staying time, and hospitalization were also evaluated. Platelet count at Post-OP was high in Off-pump group whereas CK-MB and LDH levels were low compared with On-pump group. Off-pump group had significantly lower heparin and protamine usages, lower total leukocyte count, higher hematocrit and hemoglobin levels, less blood loss, lower usages of blood components, shorter ventilation and ICU-staying time, and lower incidence of pleural effusion than On-pump group. Other variables did not significantly differ between two groups. These results showed that Off-pump CABG was a satisfactory technique with less inflammatory reaction, less cardiac damage, less postoperative complications, and less cost.
An 8-year-old female, German Shepherd dog was presented with history of dyspnea and seizure. One Year Previously a lumpectomy had been performed for surgical resection of mammary mass. In serum chemistry, severe hypoglycemia and elevation of aspartate aminitransferase and creatine kinase were shown. In thoracic radiography, there were variable sized nodules in the overall lung field. On computed tomography examination, the mass was shown in left longissimus lumborum muscle. Histopathological examination revealed adenocarcinoma derived from the mammary gland. Muscular metastasis of mammary gland tumor is uncommon. This is a rare observation and could easily be overlooked or misinterpreted.
Calcium channel blockers may prevent myocardial injury during cardioplegia and reperfusion. This study was done to evaluate the effects of diltiazem cardioplegia on myocardial protection during ischemic arrest and recovery of myocardial function after reperfusion. Four formulations of crystalloid cardioplegic solutions, GIK solution[group I, n=12], diltiazem[lug/ml GIK] in GIK solution[group II, n=7], ],diltiazem[2ug/ml GIK] in GIK solution[group III, n=6] and diltiazem[4ug/ml GIK] in GIK solution[group IV, n=6] were compared in isolated working rat heart subjected to a long period [2 hours] of hypothermic arrest with multi-dose infusion. Diltiazem cardioplegia[group II, III and IV]was found to be superior in nearly all aspects. Diltiazem cardioplegia showed faster recovery of regular rhythm and lower incidence of ventricular fibrillation than group I did. In comparing mechanical function in all experimental hearts, the mean postischemic recoveries of aortic flow, cardiac output, peak aortic pressure, stroke volume and stroke work[expressed as a percentage of its preischemic control] were significantly greater in group II, III and IV[diltiazem cardioplegia] than in group I. The infused amount of cardioplegic solution was more increased by the addition of diltiazem to GI K solution. [p < 0.01] Creatine kinase leakage tended to be lower in hearts receiving diltiazem cardioplegia, especially in group III and IV[p<0.05] than in those receiving GIK solution only[group I]. Diltiazem cardioplegia results in the increased flow of cardioplegic solution and the decreased ischemic injury of myocardium during ischemic arrest and the improved recovery of myocardial function after reperfusion, and a dose-response relation must be established before clinical use.
In order to evaluate the myocardial injury in cardiac valvular and coronary surgery, variables of creatine kinase[CK], myocardial band of CK[CK-MB], lactate dehydrogenase[LDH], aspartate aminotrasferase[AST] were measured in the preoperative[Preop], the operation day[POD0], and the first[POD1], third[POD3], fifth[POD5], seventh[POD7], ninth[POD9] day after operation in 29 patients. The subjects were divided into two groups according to the diseases: group V [valvular disease, n=16] and group C[coronary artery disease, n=13]. Each group was subdivided into two subgroups according to the duration of aortic crossclamping time[ACT]; group VI[ACT 120min, n=7] and group VII[ACT>120min, n=9]; group CI[ACT 120min, n=6] and group CII[ACT>120min, n=7]. The results were as followed 1. The values of CK between group V and group C had no significant difference. The values of CK in group CII were significantly greater than those in group CI and the values of CK in group VII were significantly greater than those in group VI. 2. Percentages of CK-MB between groups had no significant difference. 3. The serum levels of LDH in group V were significantly greater than those in group C. The serum levels of LDH in group VII were significantly greater than those in group VI. 4. The serum levels of AST in group VII were significantly greater than that in group VI. We were concluded that myocardial injury was more related with the duration of aortic cross clamping time rather than the type of diseases.
