Enhancement of the Cosmeceutical Activity by Nano-encapsulation of Thiamine Di-lauryl Sulfate (TDS) with antimicrobial efficacy (항균 효능이 있는 비타민 B1 유도체(Thiamine Dilauryl Sulfate:TDS)의 나노입자화를 통한 기능성 향장 활성 증진)

  • This study was to improve cosmetical activity of thiamine di-lauryl sulfate (TDS) by encapsulation of nanoparticle with lecithin. Results showed that most of the nanoparticles containing the TDS were well formed in round shape with below 150 ~ 200 nm diameter as well as they were fairly stable in various pH ranges by measuring zeta potentials. The nanoparticles of TDS resulted in 85% cell viability of human normal fibroblast cells (CCD-986sk) when added at the highest concentration (1.0 mg/mL). The nanoparticles of Acer mono sap showed highest free radical scavengering effect as 88.1% in adding sample (1.0 mg/mL), compared to TDS solution of non-encapsulation (81.6%). The nanoparticles of TDS reduced the expression of MMP-1 on UV-irradiated CCD-986sk cells down to as 41.4%. The TDS solution and nanoparticles showed significant anti-microbial activities agaionst the salmonella typhimurium and listeria monocytogenes at 5 and 6 days as compared with control. Anti-microbial activities of TDS nanoparticles were similar to positive control. These results indicated that TDS nanoparticles may be a source for functional cosmetic agents capable of improving cosmetical activity such as antioxidant, whitening, and anti-wrinkling effects and can be further developed as natural preservative in cosmetics.

Development of Coating Process of Citric Acid Using Vegetable Oil (식물성 유지를 이용한 구연산코팅 공정의 개발)

  • Citric acid is a natural preservative and is used to add a sour taste to foods and soft drinks. For the preparation of stable food additives of citric acid, a coating process using vegetable oil was developed. Coating materials used were purified soy bean oil and carnauba wax. The yield of coated citric acid was up to 95% in both cases. The contents of coating materials was 20~33% in the total composition and the coating efficiency was 95.2 ${\pm}$ 0.01%. The surface of coated citric acid was much smoother and more homogeneous than that of original citric acid according to SEM data. The coated citric acid can be used to as a stable food additive and also would be applied to nutraceuticals and cosmetic ingredients.

Vitellogenin Induction and Histo-metabolic Changes Following Exposure of Cyprinus carpio to Methyl Paraben

  • Methyl paraben (MP), which is used as a preservative in pharmaceutical and cosmetic (shampoo) products, foods and beverages, enters into the aquatic environment and can pose a potential fish health hazard. In this experiment, effects of MP were evaluated in adult male common carp (Cyprinus carpio) by exposing them to fractions (1/$143^{rd}$ to 1/29th) of the $LC_{50}$ dose with every change of water for 28 days. Vitellogenin induction, metabolic enzymes, somatic indices and bioaccumulation were studied at weekly intervals. The $96^{th}$ h $LC_{50}$ of MP in fingerlings was 120 mg/L. Compared to the control, except for increases (p<0.01) in alkaline phosphatase (EC 3.1.3.1), alanine aminotransferase (EC 2.6.1.2) and liver size, there were decreases (p<0.01) in activity of acid phosphatase (EC 3.1.3.2), aspartate aminotransferase (EC 2.6.1.1), and testiculosomatic index following exposure to any dose of MP. Vitellogenin induction was significantly higher (p<0.01) in exposed than unexposed (control) fish. The bioaccumulation of MP in testis, liver, brain, gills and muscle tissues of fish increased significantly (p<0.01) with increase of dose from 0.84 ppm to 1.68 ppm. Dose and duration of exposure (p<0.01) indicated that an exposure period of 1 to 2 weeks was sufficient to cause changes in the quantifiable parameters studied. Fish exposed to 4.2 ppm MP became lethargic after the $26^{th}$ d. Histologically, degeneration, vacuolization and focal necrotic changes in liver and fibrosis-like changes in testicular tissue were noted.

Identification of Anti-Microbial Material Originated from Opuntia ficus-indica var. saboten Makino (손바닥 선인장 분말로부터 추출된 항균물질의 특성)

  • In order to discover physiologically active substance, we investigated a powder obtained by processing of Opuntia ficus-indica var. saboten Makino trunk. The powder was treated by sulfuric acid and then extracted by several solvents such as methanol, methylene chloride, ethanol etc. Among them, the best antimicrobial activity was showed by methylene chloride extract. To identify materials exhibiting physiological activation, the acid hydrolyzed extract was separated by 7 fractions through preparative silica gel TLC. The effective fraction exhibiting the best broad antimicrobial activity was identified, named as MBT-01108. From structural analysis of the products extracted to acid hydrolysis, a compound exhibiting the antimicrobial activities is identified to levulinic acid. Levulinic acid isolated from Opuntia ficus-indica var. saboten Makino may be applicable as a natural preservative of food or cosmetic and for prevention of bacterial diseases, an ingredient of acne, ageing and whitening cosmetics and an antimicrobial agent.

Antimicrobial activity of fraction mixture of ethanol extracts from Eucalyptus globulus, Yucca recurvifolia, and Melaleuca alternifolia against several human skin microbes (유칼립투스, 유카와 차나무의 추출분획 혼합물의 여러 인간 피부 상재균에 대한 항균활성)

  • This study was carried out to evaluate antimicrobial effects of a mixture of resin fractionated ethanol extract of Eucalyptus globulus, Yucca recurvifolia, and tea tree (Melaleuca alternifolia). The plant fraction mixture showed low minimum inhibitory concentration (0.24~3.32 mg/ml) against several bacteria and yeast that usually used as the target skin microbes in a cosmetic industry, and it was more effective than antibiotics, triclosan and ampicillin. In a time-kill assay the plant fraction mixture reduced more than 92% of microbial populations during 4 h, and significantly increased leakage of nucleotides from all microorganisms tested. Antimicrobial effect of the plant fraction mixture was not affected by divalent cation ($Mg^{2+}$ and $Ca^{2+}$). These results suggest that the fraction mixture of ethanol extracts of E. globulus, Y. recurvifolia, and M. alternifolia may be utilized as an efficient preservative in cosmetics to prevent contamination by human skin microbes.

A Review on the Health Risks Associated with the Use of Products Containing Benzalkonium Chloride (BKC), Focusing on Humidifier Disinfectant Products (염화벤잘코늄(Benzalkonium chloride) 살균제의 용도 및 건강 위험에 대한 고찰 - 가습기 살균제를 중심으로)

  • Background: Benzalkonium chloride (BKC) has been extensively used as a preservative in industrial products and in hygiene, medical, and cosmetic applications. Humidifier disinfectant (HD) products containing BKC have been used in South Korea. Objectives: This study was aimed to review types of products containing BKC, to summarize the regulations in the US, EU, Japan, and South Korea, and to review the health effects associated with the use of HD. Methods: We reviewed and summarized documents which were searched through PubMed and Google Scholar with the key words: BKC and asthma/contact dermatitis, humidifier disinfectant-associated lung injury (HDLI), and more. Results: Regulations in most countries including South Korea do not allow its use as disinfectants in spray-type of products for medicine, cosmetics, and household products. Two types of HD containing BKC (800~1,270 ppm) were marketed in South Korea from 1996~2003. Health effects reported from people who used products containing BKC were allergic contact dermatitis, erythema, and respiratory disease, including asthma. Two people who responded as HD users containing BKC only were confirmed to have developed asthma. HD-associated lung injury (HDLI) was reported by consumers who used both HD containing polyhexamethylene guanidine (PHMG) and HD containing BKC. Conclusions: In conclusion, the use of BKC as a biocide has to be controlled considering the route and pattern of exposure. Products containing BKC as preservatives were reviewed with exposure routes and sites in the human body such as skin, eyes, and the respiratory tract. HD containing BKC was clinically evaluated to be associated with asthma.

  • This study aims to develop a new analytical method to detect zinc pyrithione, the ingredient of cosmetics appointed as restricted ingredients and used as preservatives.. The analytical method was based on data gathered from the relevant literature. Information about the amounts of these ingredients was researched in order to select the base-matrix materials used to validate the analytical method. After selecting and preparing the base-matrix materials, the analytical method was validated by method validation procedures. The analytical method was verified first by inter-laboratory validation and then through analyzing the cosmetics sold in the market. Based on the results of this study, guidelines are proposed for the analysis of restricted ingredients in cosmetics, which will provide a method to test the cosmetics circulating in the Korean market. The use of the proposed guidelines will increase the quality of the cosmetics as well as the safety of human health, which will enhance the competitiveness of the Korean cosmetics industry and lead to an increase in the exportation of cosmetics.

Optimal Conditions for Phenylethanol Galactoside Synthesis using Escherichia coli β-Galactosidase (대장균 베타-갈락토시데이즈를 이용한 Phenylethanol Galactoside 합성 조건의 최적화)

  • To circumvent the skin problem from phenylethanol (PhE), we have studied on the enzymatic synthesis of phenylethanol galactoside (PhE-gal) as an alternative to PhE. Base on the previous study, we optimized the reaction conditions for PhE-gal synthesis from PhE using E. coli β-galactosidase (β-gal). The optimal amount of β-gal, PhE concentration, pH, and temperature for PhE-gal synthesis were 0.45 U/ml, 1%, 8.0, 40℃, respectively. Under these conditions, about 81.9 mM PhE was converted into about 47.4 mM PhE-gal, in which the conversion yield was about 57.9%. Meanwhile, when the reaction mixture containing PhE and PhE-gal was mixed and fractionated with water-immiscible solvent (EA or MC), it was observed that PhE-gal was distributed in water phase, and PhE was distributed in solvent phase. Additionally, PhE-gal was clearly distributed into water phase when MC was used, but PE-gal was not when EA was used. In the future, we are planning to carried out the continuing study on developing an alternative cosmetic preservative using PhE-gal.

Synthesis of Linear 1,2-Alkylaminopropanediols as Preservatives and Antimicrobial Activity (보존제로서 선형 1,2-알킬아미노프로판디올의 합성과 항균활성)

  • The synthesis of 1,2-alkylaminopropanediols (1,2-AAPs) was designed to improve the hydrophilicity of linear 1,2-alkanediols having 10, 12, 14, and 16 carbon atoms in the alkyl chain. 1,2-AAPs were synthesized by reacting 3-monochloro-1,2-propanediol (3-MCPD) with linear alkylamines having 10, 12, 14 or 16 carbon atoms in an ethanol solvent at 40℃ for 2 h. The yield and purity of four types of 1,2-AAPs synthesized were found to be in the range of 51-58% and 85-99%, respectively. The amine salts of four types of 1,2-AAPs were prepared from a purified paste or solid compound by adding an acidic solution (HCl) to pH 7, and then their solubilities and antibacterial effects were tested. 1,2-decylaminopropanediol, 1,2-dodecylaminopropanediol, and 1,2-tetradecylaminopropanediol were all dissolved in water at concentrations of 100%, 50%, and 0.1%, respectively, however 1,2-hexadecylaminopropanediol was not. The antibacterial effect was improved as the length of the alkyl chain increased. As a result of confirming the preservative effect of the lotion (cosmetic formulation) applied with 1,2-AAP for application, it showed very strong antibacterial activity at low concentrations ranging from 0.005% to 0.2%.

Antimicrobial Synergistic Effects of Gallnut Extract and Natural Product Mixture against Human Skin Pathogens (피부 병원성균에 대한 오배자 천연 복합물의 시너지 항균 효과)

  • This study was attempted to investigate natural materials with antimicrobial activity and to apply as natural preservatives in cosmetics. The disc diffusion method was used to search for nine species of natural antibacterial material for three species of skin pathogenic bacteria (Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa) and Candida albicans. As a result of measuring the size of inhibition zone, Rhus Semialata gall (Gallnut) extract, Oak vinegar, and ε-polylysine were showed strongest antibacterial activities (> 10 mm). The Minimum Bactericidal Concentration (MBC) of gallnut and oak vinegar ranged from 10 to 20 mg/mL and from 20 to 40 mg/mL against five human skin pathogens. The MBC of ε-polylysine ranged from 0.5 to 2 mg/mL in fungus. The synergic effect of gallnut extract/oak vinegar mixture and gallnut extract/ε-polylysine mixture were evaluated by checkerboard test. Compared to when used alone, the MBC of gallnut extract/oak vinegar mixture were at 4 times lower concentration against E. coli, C. albicans, and A. brasiliensis. Also Furthermore, the MBC of gallnut extract/ε-polylysine mixture were at 4 times lower concentration against C. albicans and A. brasiliensis. It was confirmed that the combination of gallnut extract with oak vinegar or ε-polylysine resulted in synergistic antibacterial effect against three human skin pathogens. Thus, it is expected that gallnut extract and natural product mixture can not only demonstrate antibacterial synergies, but also be applied in cosmetics as a natural preservative system with a wide antibacterial spectrum.
