Nutritive Value of Urea Treated Wheat Straw Ensiled with or without Corn Steep Liquor for Lactating Nili-ravi Buffaloes

  • Influence of different levels of corn steep liquor (CSL) on chemical composition of urea treated wheat straw (UTWS) and its dietary effect on nutrient intake, digestibility, milk yield and its composition were studied. The 5% UTWS was ensiled with 0, 3, 6 and 9% CSL on dry matter (DM) basis. Total nitrogen and neutral detergent fiber (NDF) contents of UTWS ensiled with 0, 3, 6 and 9% CSL increased linearly with the increasing level of CSL. Increase in NDF content was due to increased neutral detergent insoluble nitrogen contents. Four experimental diets were formulated to contain 35% UTWS ensiled without CSL (control), 45 (WS45), 55 (WS55) and 65% (WS65) UTWS ensiled with 9% CSL, respectively. Dry matter, NDF and acid detergent fiber (ADF) intakes by lactating buffaloes fed diets containing varying levels of UTWS ensiled with or without CSL remained similar across all treatments. However, DM, NDF and ADF intakes as a percent of body weight and digestible DM, NDF and ADF intakes were higher in animals fed WS65 diet compared to those fed other diets. Apparent DM, crude protein (CP), NDF and ADF digestibilities were higher in diets containing UTWS ensiled with CSL compared to control. These differences may be attributed to higher rates of degradability of UTWS ensiled with 9% CSL than that ensiled without CSL. The 4% fat corrected milk and CP were statistically higher with WS65 diet compared to other diets. Percent milk fat, solid not fat and total solid remained unchanged across all treatments.

  • Fruit bodies of Ganoderma lucidum have been used for most pharmacological studies, but pharmacological effects are likely variable because the habitats and strains of Ganoderma lucidum are different. Therefore, their fermentation is required to produce constant and reliable pharmacolo­gical constituents from Ganoderma lucidum. During the studies of medium for industrial application. it was found that ginseng root residues, remaining after being extracted with ethanol, were a good carbon source for a fermentation of Genoderma lucidum and a corn steep liquor was also economical for the nitrogen source. Yield of the mycelial cultured in ginseng root residues and corn steep liquor was 2.5 times higher than that in glucose and peptone, known as a conventional medium of Ganoderma lucidum. The polysaccharide content of the extracts from the cultured mycelia was higher than that from fruit bodies, but protein content was vice versa. Extracts of the cultured mycelia were more effective and lasting than extracts of the fruit bodies in decreased hypertention of spontaneously hypertensive rats (SHR).

  • Cultural characteristics of a strain of Streptomyces sp. K-14 (KFCC 35051) producing glucose isomerase were demonstrated. The glucose isomerase was produced when the strain was grown in the medium containing pure xylan or xylan.containing materials such as wheat bran or com cob. The optimum condition was attained in a culture medium composed of 3 % wheat bran or com cob, 2 % com steep liquor, 0.1% $MgSO_4$$7H_2O$ and 0.012 % $CoSO_4$$7H_2O$ for the production of the glucose isomera,e. The production of the enzyme reached to a maximum level when the strain was cultured for 40 hrs $30^{\circ}C$ and pH 7.0.

Isolation of Microorganism with HIgh Productivity and Cultivation Optimization for Lactic Acid Production (고생산성 젖산생성균 분리 및 배양 최적화)

  • In order to screen microorganism producing lactic acid with high productivity from nature, we used a medium containing 100 g/l glucose and selected several microorganisms producing more than 80 g/l L-lactic acid. We investigated their physiological characteristics and compared them. The best microorganism was identified as Lactobacillus casei subsp. rhamnosus. The optimum pH for growth and production of lactic acid was 6.0 and this strain showed the highest growth rate at around 30$\circ$C , but the optimum temperature for lactic acid production was 45$\circ$C . The growth was inhibited proportionally from 50 g/l to 300 g/l of glucose and the maximal cell mass increased according to increasing the concentration of corn steep liquor (CSL) protein up to 30 g/l. In batch fermentation for lactic acid production, we produced 128 g/l L-lactic acid with 20 g/l CSL protein and 150 g/l glucose in 35 hours. In pH-stat fed-batch fermentation, we were able to produce 183 g/l L-lactic acid.

  • Whey based medium was optimized for the production of viable Lactobacillus crispatus KLB 46 isolated from the vagina of Korean women. Among the various nitrogen sources such as yeast extract, beef extract, and proteose peptone no. 3 supplemented to whey, beef extract showed the highest viable cell production. The addition of Tween 80 to the whey based medium increased viable cell concentration. As beef extract supplementation is not economically attractive, corn steep liquor was added as a supplementary nitrogen sources. When corn steep liquor was supplied with beef extract with the ratio 5 : 1, the viable cell count was $3.11{\times}10^9$ CFU/mL. Also, the addition of mineral salts containing sodium acetate (5 g/L), potassium phosphate dibasic (2 g/L), magnesium sulfate (0.1 g/L) and manganese sulfate (0.05 g/L) to the whey medium increased viable cell count further ($5.00{\times}10^9$ CFU/mL).

  • Spores of Trichoderma harzianum TCC 52445 for control of Rhlzocronia stem canker on potato were immobilized in various natural matrix, and germination rate and some rheological properties of the spore-matrix complex were Investigated. Germination rate of gelatin gel-spore complex and potato starch gel-spore complex were 2.8% and 2.9%, respectively, but hardness and cohesiveness of the gelstin gel-spoke complex were better than those of potato starch-spore complex. The hardness and cohesiveness were increased when the spores were immobilized in hybrid gelatin gel made by mixing several different types of natural matrix, but decreased their germination rate. Addition of corn steep liquor (1%) as spore nutrient in gelatin gel-spore complex was helpful for increasing the germination rate.

  • Identification of soil microorganism strain B-6. a producer of acyl CoA synthetase inhibitor, based on its morphological, physiological, biochemical and chemotaxonomical charact eristics was performed. The strain B-6 was identified as Bacillus subtilis. Ihe acyl CoA synthetase inhibitor produced by this strain was highly achieved in fermentation medium that contained glucose 1.0%, soluble starch 1.0%, NH$_4$Cl 0.3%, oatmeal 1.0%, pharmamedia 1.0%, basic magnesium carbonate 0.5%. pH 7.5 at 30$^{\circ}$C for 7 days. The optimal pH and temperature for growth were 9.0 and 30$^{\circ}$C, respectively. Butanol extract of culture filterate of strain B-6 in acyl CoA synthetase inhibitor production medium containing corn steep liquor exhibited high acyl CoA synthetase inhibitor activity and antimicrobial activity against C. albicans. But chloroform extract of culture filterate of strain B-6 in medium containing NH$_4$Cl, ($NH_4)_2SO_4$ or urea instead of corn steep liquor exhibited higher antimicrobial activity against C. albicans than that of butanol extract.

  • A highly cyclodextrin glucanotransferase producing strain of Bacillus sp. was isolated from soil, and basic studies on the characteristics of the strain and its enzyme, conditions for the enzyme production, and the enzyme utilization were carried out. The isolated strain was aerobic, motile, endospore-forming and rod-shaped bacterium. Optimum pH and temperature for the enzyme action were 6.0 and 45$^{\circ}C$, and the enzyme was stable within 5$0^{\circ}C$, and between pH 6.0 and 10.0. The highest yield of the enzyme was obtained using the medium containing 2% corn starch as a carbon source, and 5% corn steep liquor, 0.1% urea and 0.25% ammonium sulfate as nitrogen sources. The fermentation conditions for the enzyme production in a jar fermentor were cetermined to be 3$0^{\circ}C$, 200rpm, 0.6vvm and 60hr cultural period. Stevioside transglycosylation catalyzed by this enzyme was identified by high performance liquid chromatography.

Influence of Ruminally Protected Fat and Urea Treated Corncobs Ensiled with or without Corn Steep Liquor on Nutrient Intake, Digestibility, Milk Yield and Its Composition in Nili-Ravi Buffaloes

  • Sixteen early lactating Nili-Ravi buffaloes, four animals in each group, were used in a Completely Randomized Design to evaluate the effect of varying levels of both ruminally protected fat and urea treated corncobs ensiled with or without corn steep liquor (CSL) on feed intake, digestibility and milk production and its composition. Four experimental diets were formulated. The control (C) diet was balanced to contain 0% fat and 35% urea treated corncobs ensiled with 0% CSL. The low fat (LF), medium fat (MF) and high fat (HF) diets had 45, 55 and 65% urea treated corncobs ensiled with 9% CSL and 2, 4 and 6% ruminally protected fat, respectively. Dry matter, crude protein (CP) and neutral detergent fiber (NDF) intakes by buffaloes remained similar across all treatments. However, DM and NDF as a percent of body weight and digestible DM intakes were higher in HF diet when compared to C, LF and MF diets. Digestible NDF intakes were also significantly higher in HF diet as compared to all other diets. The intakes of ADF and digestible ADF were higher in MF and HF than C and LF diets. The significant variation in digestible DM, ADF and NDF intakes may be attributed to the ammoniation of corncobs along with CSL that caused significant changes in the degradability and digestibility of the diets. Ether extract and digestible EE intakes differed significantly (p<0.05) among all treatments. Intakes of EE were the highest in animals fed HF diet, which was because of added fat. Apparent DM digestibility was the highest in animals C diet and was the lowest in those fed LF diet. Neutral detergent fiber and ADF digestibilities were higher in animals fed diets containing urea treated corncobs ensiled with 9% CSL when compared to those fed diets containing urea treated corncobs ensiled without CSL. Apparent digestibility of CP was noted highest (71.47%) in animals fed HF diet when compared to those fed MF (67.75%), LF (67.04%) and C (65.39%) diets. Milk yield (4% FCM) was the higher in buffaloes fed HF, MF and LF diets than those fed C diet. These results indicated that increasing levels both of fat and urea treated corncobs ensiled with CSL elevated the negative effects of poor quality fibrous feed on milk production by buffaloes.

  • The production of fumaric acid in shaken flask cultures by Rhizopus oryzae KCTC 6946 was studied. The effects of various carbon and nitrogen sources on fumaric acid production were investigated. The strain fermented starch (or glucose) and corn steep liquor (or polypepton) better than other carbon and nitrogen sources to fumaric acid. When C/N ratio of carbon and nitrogen source was 41.7, the concentration of fumaric acid produced with 2N NaGH as a neutralizing agent was more than 10 g/L after 3 days.
