Corm Steep Liquor를 이용한 젖산균이 생산배지에 관한 연구 (A Study on the Production Medium of Lactic Acid Bacterial Cells by Using Corn Steep Liquor)

  • 젖산균의 생장을 위한 질소, 탄소 공급원으로서 corn steep liquor의 이용 가능성을 시험하고 반응 표면 분석(Response surface metho-dology)을 이용하여 젖산균의 최적 생상 배지 조성을 연구하였다. 반응 표면 분석에서 L.fermentum의 생장배지에 첨가되 corn steep liquor와 yeast extract의 농도(p<0.01) 그리고 corn steep liquor와 yeast extract의 교호작용(P<0.05)이 L.ferm-entum의 생장에 큰 영향을 미치는 것으로 나타났으며, 이때 생균수가 최대인 corn steep liquor의 함량은 10.77%, yeast extract는 3.39%.Tween 80은 1.69%으로 예측되었다. 한편, Lc, lactis ssp. lactis 의 생장배지는 corn steep liquor 의 농도(P<0.01) 그리고 corn steep li-quor와 $\beta$-glycerophosphate disodium salt의 교호작용(P<0.05)인 Lc. lactis ssp. lactis의 생장에 큰 영향을 미치는 것으로 나타났으며 이때 생균수가 최대인 corn steep liquor의 함량은 3.5%, $\beta$-glycerophosphate disodium salt는 4.38%로 예측되었다. MRS broth와 예측된 최적 배지에서 L.fermentum의 젖산과 초산의 생성량은 각각 0.166, 0.114과 0.273, 0.081 M이고 M 17glc broth와 최적배지에서 Lc. lacti ssp. lactis의 젖산과 초산의 생성량은 각각 0.089, 0.003과 0.189, 0.003M이었다. 따라서 corn steep liquor는 L. fermentum와 Lc. lactis ssp, lactis 의 생장을 위해 질소 또는 탄소 공급원으로서 배지에 첨가 될 수 있는 우수한 농업 부산물로 판단되었다.

  • Lactobacillus cristatus KLB 46 isolated from Korean woman was grown on supplemented whey medium and medium compositions were optimized for maximum viable cell count. Among the nitrogen sources tested, beef extract yielded the highest viable cell number. When corn steep liquor was applied as an additional nitrogen source, the viable cell number was highest $(3.11{\times}10^9\;CFU/ml)$ in the medium containing 50g/ l corn steep liquor and 10g/ l beef extract. The highest viable cell $count(5.00{\times}10^9\;CFU/ml)$ was obtained from the supplemented whey medium that contains beef extract(10g/ l ), corn steep liquor(50g/ l ), tween 80(0.1%, v/v) and trace amounts of sodium acetate(5g/ l ), dipotassium phosphate(2g/ l ), magnesium sulfate(0.1g/ l ), and manganese sulfate (0.05g/ l ).

  • The production of fumaric acid in a novel fixed-layer fermenter using Rhizopus oryzae KCTC 6946 was studied. When yeast extract and corn steep liquor were used as nitrogen sources, fumaric acid produced was 6.4 g/L and 6.8 g/L, respectively. Compared with the flask cultivation, fumaric acid concentration was about 200% when yeast extract was used as a nitrogen source, but reached to 93% when corn steep liquor was used.

  • 강력(强力)한 inulin 분해력(分解力)을 가진 Streptomyces chibaensis균주(菌株)를 이용하여, 효소생산(酵素生産)에 효과적인 배양조건 및 배지조성을 검토(檢討)하여 다음과 같은 결과를 얻었다. 1. 효소생산배지(酵素生産培地)의 최적 초기 pH는 7.5였으며, 이 조건하에서 $30^{\circ}C$에서 84시간 배양했을 때 가장 높은 효소역가를 나타냈다. 2. Inulase를 다량생산(多量生産)하기 위한 배지의 탄소원(炭素源)으로서는 다른 탄소원(炭素源)에 비하여 inulin이 가장 효과적이었다. 3. 무기질소원(無機質素源)으로는 $(NH_4)_2HPO_4$가 inulase생산(生産)에 가장 효과적(效果的)이었고, 유기질소원(有機窒素源)으로는 corn steep liquor가 효과적(效果的)이었다. 4. 무기염(無機鹽)으로 KCI 0.01%, $MgSO_4$ 0.05%, 그리고 $FeSO_4$ 0.0001% 첨가하였을 때 가장 효과가 컸다. 5. 탄소원(炭素源)(inulin) 1%와 질소원(窒素源)(corn steep liquor) 2%의 비율(比率)로 혼합(混合)하여 만든 배지(培地)에서 가장 효소생산(酵素生産)이 높았다.

High-Cell-Density Fed-Batch Culture of Saccharomyces cerevisiae KV-25 Using Molasses and Corn Steep Liquor

  • High-cell-density cultivation of yeast was investigated using the agricultural waste products corn steep liquor (CSL) and molasses. The Saccharomyces cerevisiae KV-25 cell mass was significantly dependent on the ratio between C and N sources. The concentrations of molasses and CSL in the culture medium were statistically optimized at 10.25% (v/v) and 16.87% (v/v), respectively, by response surface methodology (RSM). Batch culture in a 5-l stirred tank reactor using the optimized medium resulted in a cell mass production of 36.5 g/l. In the fed-batch culture, the feed phase was preceded by a batch phase using the optimized medium, and a very high dried-cell-mass yield of 187.63 g/l was successfully attained by feeding a mixture of 20% (v/v) molasses and 80% (v/v) CSL at a rate of 22 ml/h. In this system, the production of cell mass depended mainly on the agitation speed, the composition of the feed medium, and the glucose level in the medium, but only slightly on the aeration rate.

Fermentative Production of Succinic Acid from Glucose and Corn Steep Liquor by Anaerobiospirillum succiniciproducens

  • Anaerobiospirillum succiniciproducens requires expensive complex nitrogen sources such as yeast extract and polypeptone for its growth and succinic acid production. It was found that A. succiniciproducens was able to grow in a minimal medium containing glucose when supplemented with corn steep liquor (CSL) as the sole complex nitrogen source. The concentration of CSL had a significant effect on the glucose consumption by A. succiniciproducent. When 10-15 g/L of CSL was supplemented, cells were grown to an OD(sub)600 of 3.5 and produced 17.8 g/L succinic acid with 20 g/L glucose. These results are similar to those obtained by supplementing yeast extract and polypeptone, thereby suggesting that succinic acid can be produced more economically using glucose and CSL.

Candida magnoliae에 의한 에리스리톨 생산을 위한 최적 배양환경과 질소원 선별 (Optmization of Culture Conditions and Nitrogen Sources for Production of Erythritol by Candida magnoliae.)

  • Culture conditions and nitrogen sources were optimized for production of erythritol, a natural sweetener, by Candida magnoliae M26. The optimal culture conditions were found to be culture temperature of $28^{\circ}C$, initial pH of 7, aeration of 1 vvm and agitation speed of 500 rpm in a 2.5 1 jar-fermentor. Glucose was chosen as the best carbon cource bsed on cell growth and erythritol productivity. Kight steep water(LSW) and corn steep liquor (CSL) which are by-products in starch processing from corn were tested as a nitrogen source substitute for yeast extract. The use of either LSW or CSL did not change the fermentation performance. The experimental results using LSW and CSL showed 1.5 times higher in cell growth and almost the same value in erythritol productivity com-pared with the control fermentation using yeast extract as a nitrogen source. These results suggested that either LSW of CSL could be used as a nitrogen source in a large-scale fermentation for erythritol production.

Effect of Lactate and Corn Steep Liquor on the Production of Bacterial Cellulose by Gluconacetobacter persimmonis $KJ145^T$

  • In this study, we attempted to assess the effects of lactate and com steep liquor (CSL) on the production of bacterial cellulose (BC) by Gluconacetobacter persimmonis $KJ145^T$. The optimal condition for the production of BC was a lactate concentration of 1% (w/v) and a CSL concentration of 10% (w/v). Under these optimal conditions, 6 days of fermentation produced 6.90 g/L of BC. Both the BC production yield and cell growth increased continuously until the 20th day of fermentation, by which time 17.0 g/L had been produced. In a static culture trial, in which plastic containers were used as fermentation chambers for 6 days of fermentation, the BC production yield in the group initially cultured with 500 mL medium was higher than that of the 750 and 1000 mL media. In addition, the texture of the BC was examined according to its post-treatment in order to determine conditions for optimal textural characteristics. The strength, hardness, and other characteristics of the BC were negatively correlated with sucrose concentration, but were largely positively correlated with NaCl concentration. With regards to the effect of pH on textural change, BC strength and hardness were elevated at pH 2 and 8 but reduced at pH 4 and 6, indicating that the texture of the BC is extremely sensitive to treatment conditions.

  • We investigated the fermentative production of lactic acid from cheese whey and corn steep liquor as cheap raw materials using Lactobacillus sp. RKY2 to reduce the manufacturing cost of lactic acid. Lactic acid yields were obtained at more than 0.98 g/g from medium containing whey lactose. Lactic aid productivities and yields obtained from whey lactose were slightly higher than those obtained from pure lactose. The final concentration of lactic acid increased with increase, in whey lactose concentration, whereas the lactic acid productivity decreased probably due to substrate inhibition. The fermentation efficiencies were improved by addition of more corn steep liquor to the medium.

Influence of Corn Steep Liquor on Feeding Value of Urea Treated Wheat Straw in Buffaloes Fed at Restricted Diets

  • Influence of different levels of corn steep liquor (CSL) on chemical composition of urea treated wheat straw (UTWS), ruminal characteristics, digestion kinetics, nitrogen (N) utilization, and nutrient digestibility by ruminally-cannulated buffalo bulls was studied in a $4{\times}4$ Latin Square Design. The CSL was used to ensile 5% UTWS at the rate of 0, 3, 6, and 9% on a dry matter (DM) basis. Total N and neutral detergent fibre (NDF) were increased with increasing level of CSL. Increased NDF content was attributable to increased neutral detergent insoluble nitrogen. Four diets were formulated to contain 20% concentrate and 80% UTWS ensiled with 0, (control), 3 (CSL3), 6 (CSL6), and 9% CSL (CSL9). All diets were mixed daily and fed at 1.5% of body weight twice daily. Ruminal NH3 concentration decreased with level of CSL used to ensile UTWS at 3 and 6 h post prandial, however, at 9 h post parandial it was similar across all diets and at 12 h post prandial was higher with diets containing UTWS ensiled with CSL. Concentrations of total ruminal volatile fatty acid and acetate were increased with the CSL level used to ensile UTWS. Increased rate of disappearance and reduction in lag time of DM and NDF was recorded with diets containing UTWS ensiled with CSL. Dry matter and NDF digestibilities were higher with CSL diets than on the control diet. Buffalo bulls retained more N with diets containing UTWS ensiled with CSL. The present results indicated that UTWS could be ensiled with CSL to improve its nutritive value and N utilization by ruminants.
