Antimutagenic and Cytotoxic Effects of Korean Wild Mushrooms Extracts (한국산 야생버섯 추출물의 항돌연변이원성 및 암세포 성장억제 효과)

  • Ames test revealed most methanol extracts of 13 Korean wild mushroom species have strong antimutagenic effects against N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and benzo(a)pyrene[B(a)P]. Methanol extracts of Coriolus versicolor and Phaeolepiota aurea showed 74-94 and 83-88% antimutagenic effects against MNNG and B(a)P in Salmonella typhimurium TA100 strain, while 89 and 91% inhibitions were observed against B(a)P in TA98 strain, respectively. Most water extracts of wild mushrooms did not show antimutagenic activeiy on MNNG and B(a)P. Wild mushrooms extracts inhibited human colon carcinoma cells (HT29), human hepatoma cell (HepG2), and humann histiocytic lymphoma cell (U937) dose-dependently, with most methanol extracts exhibiting stronger effect than water extracts, Highest toxicity was observed against HT-29 cells in methanol extracts of Coriolus versicolor and Phaeolepiota aurea, showing 84% inhibition at 1 mg/mL, whereas C. versicolor water extract showed 53-65% inhibition against HepG2 and U937. These extracts did not show cytotoxic effects against human lymphocyte. Results revealed wild mushrooms have strong antimutagenic and in vitro cytotoxic effects.

The Comparative Immunomodulatory Effects of β-Glucans from Yeast, Bacteria, and Mushroom on the Function of Macrophages

  • The comparative immunomodulatory effects of ${\beta}$-glucans isolated from mushroom fungi (Coriolus versicol), yeast (Saccharomyces cerevisiae) and bacteria (Agrobacterium) on the major functions of macrophages were evaluated. As parameters of macrophage functions, we examined tumoricidal activity, phagocytosis, nitric oxide (NO) production, and the induction of inducible NO synthetase (iNOS) in RAW264.7 cells, following treatments with ${\beta}$-glucans from the three different sources. The results indicated that all ${\beta}$-glucan treatments significantly induced tumoricidal activity in the RAW264.7 cells, with a remarkable effect shown by the beta-glucan from Agrobacterium at a concentration of $10{\mu}g/mL$. There was also a significant increase in iNOS-NO system activity in macrophages treated with ${\beta}$-glucans extracted from yeast; however, iNOS-NO system activity was not markedly changed by the treatment of ${\beta}$-glucans from C. versicolor mushroom fungi or Agrobacterium. Furthermore, the ${\beta}$-glucans from C. versicolor had a significant phagocytotic effect at concentrations of 1, 10, and $100{\mu}g/mL$. Taken together, the present data suggest that these ${\beta}$-glucans, isolated from three different sources, have different effects on macrophage function, and therefore, may have different clinical uses in different for various types of diseases.

Evaluation of Ten Wild Nigerian Mushrooms for Amylase and Cellulase Activities

  • Amylases and cellulases are important enzymes that can be utilized for various biological activities. Ten different wild Nigerian mushrooms (Agaricus blazei, Agaricus sp., Corilopsis occidentalis, Coriolus versicolor, Termitomyces clypeatus, Termitomyces globulus, Pleurotus tuber-regium, Podoscypha bolleana, Pogonomyces hydnoides, and Nothopanus hygrophanus) were assayed for production of these secondary metabolites. The results revealed that most of the tested wild fungi demonstrated very good amylase and cellulase activities. With the incorporation of carboxymethyl-cellulose (a carbon source) into the culture medium, Agaricus blazei had the highest amylolytic activity of 0.60 unit/mL (at $25^{\circ}C$, pH 6.8). This was followed in order by P. tuber-regium and Agaricus sp. with 0.42 and 0.39 unit/mL, respectively ($p {\leq} 0.05$). Maltose and sucrose supplementation into the submerged liquid medium made N. hygrophanus and P. hydnoides to exhibit very low amylase activities of 0.09 and 0.11 unit/mL, respectively. Introducing peptone (an organic nitrogen source) into the basal medium enhanced the ability of C. versicolor to produce a cellulase value of 0.74 unit/mL. Other organic nitrogen sources that supported good cellulase activities were yeast extract and urea. Sodium nitrate (inorganic nitrogen source) generally inhibited cellulase production in all mushrooms. The best carbon source was carboxymethyl-cellulose, which promoted very high cellulase activity of 0.67 unit/mL in C. versicolor, which was followed in order by P. tuber-regium, T. chypeatus, and C. occidentalis ($p {\leq} 0.05$). Sucrose was the poorest carbon compound, supporting the lowest values of 0.01, 0.01, and 0.14 unit/mL in P. hydnoides, A. blazei, and Agaricus sp., respectively.

Qualitative Evaluation of Ligninolytic Enzymes in Xylariaceous Fungi

  • Sixty-one strains representing the main genera of wood-decaying xylariaceous fungi (mainly in Daldinia, Hypoxylon, Kretzschmaria, Rosellinia, Penzigia, and Xylaria) were tested for their ability to produce ligninolytic enzymes. The phenol oxidase activity and fungal growth of the xylariaceous fungi on gallic aicid and tannic acid media showed a variation in their ability to degrade lignocellulose. A number of species showed equal 개 betterligninolytic enzyme activities than Coriolus versicolor, a known basidiomycete wood-degrader. A large variation of the enzyme activity was observed by individual strains as well as a substantial variation between the isolates of the same species. The most frequent ligninolytic enzymes were peroxidase and general oxidase. With 19% of the strains tested, peroxidase showed the strongest ligninolytic enzyme activity, while tyrosinase activity was detected only in 7% of the strains. All strains of Kretzschmaria and Rosellinia tested was positive for laccase. Xylariaceous fungi were able to degrade the macromolecule, lignin, using each specific ligninolytic enzyme in the specfic lignin degradation pathway.

  • Carbohydrate analysis is important in studying structure and activity of complex polysaccharides. New analytical method was applied to get an information on the composition of polysaccharides showing antitumor activity. Monosaccharides were labeled with 7-amino-1, 3-naph-thalenedisulfonic acid (7-AGA) by reductive amination and separated by HPLC. Five kinds of polysaccharides from Basidiomycetes were hydrolyzed and analyzed in combination with electrophresis and HPLC. At the same time, alditol acetate derivatives were prepared and analyzed by gas chromatography. Two different techniques using different derivatization methods showed very similar results. The monosaccharides from Coriolus versicolor and Cordyceps militaris were glucose and galactose. Phellinus linteus composed of glucose, glactose, mannose, arabinose and fucose. The HPLC method with fluorescence detector was very sensitive compared to other methods.

Water-solubility of β-Glucans in Various Edible Mushrooms - Research Note -

  • The amount of $\beta-glucans$ in 12 edible mushroom species was determined and their water-solubility was assessed. A large variability in $\beta-glucans$ content was observed in the mushroom species, ranging from 4.71 to $46.20\%$ on a dry basis. Gyrophora esculenta, Lentinus edodes, Coriolus versicolor, Ganodenna lucidum, and Flammulina velutipes had high levels of $\beta-glucans$ Soluble $\beta-glucans$ content, which plays a key role in the physiological effects of mushrooms, also varied greatly according to the mushroom species, ranging from 2.12 to $19.66\%$. Water-solubility of $\beta-glucans$ in the edible mushrooms, as a percentage of total $\beta-glucans$ content varied from 42.55 to $73.35\%$.

  • Many plant species are used in China in traditional medicine for the prevention and treatment of cancer. This paper presents some of these species with details on other pharmacologic evaluation and its traditional Chinese meditional uses. The known bioaktivities and some chemical constituents of each of the species given. Information on bioactivities of each species resulting from tests on experimental animals are given. Many of the known chemical constituents of each species are given. Various species of plants elective for various types. Biodegradation of aromatic hydrocarbons by several white-rot fungi

  • To investigate the biodegrading capability of several white-rot fungi isolated in Korea, biodegradation of BTX (benzene, toluene, xylene), phenanthrene and pyrene were tested in fungal cultures. Phanerochaete chrysosporium removed 20-30% of BTX mixture during 21 days of incubation in serum bottle. Coriolus versicolor KR-11W and Irpex lacteus mineralized 10.02 and 8.26% of totla phenanthrene, respectively, which were higher than in other studies with P. chrysosporium. These two strains also showed high mineralization rates (9.2-10.1%) for 4-ring pyrene. I. lacteus metabolized most of the added pyrene and 23.29% was incorporate dinto fungal biomass. Almost 50/5 of the pyrene was converted to polar metabolites and recovered from aqueous phase of culture. These results indicated that some white- rot fungi have higher biodegradability than P. chrysosporium and could be used in bioremediation of aromatic hydrocarbon contaminants in soil.

A New Synthetic Approach to 3-Amino-2-phenylthietanes as Potential Monoamine Oxidase Inhibitors

  • 3-Amino-2-phenylthietane derivatives were considered as a useful tool to elusidate the mechanism of inhibiton of MAO by tranylcypromine-type inhitors. The synthesis of 3-benzoylamino-2-phenylthieetane, 3-amino-2-phenylthietane, and 3-N, N-dimentylamino-2-p-nitrophenythietane was attempted using the reaction between 1, 3 dihalogeno alkanes with alkali sulfide. When 1-pheny1-1, 3-dihalo-2-benzolaminopropane was treated with sodium sulfide, 2-pheny 1-4 benzylidene-2-oxazoline was isolated, indicating the case of elimination reaction compared to ring formation. The reaction of 1-p-nitropheny1-1, 3-dichloro-2-N, N-dimethylaminopropane with sodium sulfide gave bis (1-p-nitropheny1-2-N, N-dimethylamino-3-chloropropane)sulfide. The mechanism of reaction was discussed.

Studies on the Anticomplementary Activity of Korean Higher Fungi (한국산 고등균류의 항보체활성 효과에 관한 연구)

  • Anticomplementary activity of 61 strains of Korean higher fungi was screened for immunostimulation. Extracts from 11 of 61 strains including 5 of Ganoderma lucidum, 3 of Lentinus edodes, 2 of Cordyceps sp. and 1 of Agaricus campestris, showed higher anticomplementary activity than krestin which was immunopotent extract from Japanese Coriolus versicolor. The most potent anticomplementary activity was found with extract from Lentinus edodes IY105 whose complement consumption was 31.7%.
