Continuous Production of Citric Acid from Dairy Wastewater Using Immobilized Aspergillus niger ATCC 9142

  • The continuous production of citric acid from dairy wastewater was investigated using calcium-alginate immobilized Asrergillus niger ATCC 9142. The citric acid productivity and yield were strongly affected by the culture conditions. The optimal pH, temperature, and dilution rate were 3.0, 30$^{\circ}C$, and 0.025 h$\^$-1/, respectively. Under optimal culture conditions, the maximum productivity, concentration, and yield of citric acid produced by the calcium-alginate immobilized Aspergillus niger were 160 mg L$\^$-1/ h$\^$-1/, 4.5 g/L, and 70.3%, respectively, The culture was continuously perfored for 20 days without any apparent loss in citric acid productivity. Conversely, under the same conditions with a batch shake-flask culture, the maximum productivity, citric acid concentration, and yield were only 63.3 mg L$\^$-1/h$\^$-1/, 4.7 g/L and 51.4%, respectively, Therefore, the results suggest that the bioreactor used in this study could be potentially used for continuous citric acid production from dairy wastewater by applying calcium-alginate immobilized Aspergillus niger.

  • The utilization methods of algae biomass have been studied constantly in whole world. These are $\circled1$the wastewater treatment if waste stabilization pond and oxidation ditch etc. and $\circled2$the biosorption of heavy metals and recovery of strategic' precious metals and $\circled3$the single-celled protein production and the production of chemicals like coloring agent and $\circled4$the production of electric energy through methane gasification. The culture system also has been developed constantly in relation with such utilization method developments. In the result of experimental operation under continuous and cyclic irradiation of light, using high rate algae biomass culture pond(HRABCP), which had been made so as to be an association system with the various items which had been managed to have high efficiency for algae culture, the algae production of the 12 hours-irradiance pond was 41.48 Chlorophyll-a ${\mu}g/L$ only in spite of having the more chance of $CO_2$ synthesis to algae cell than the 24 hours-irradiance pond. This means that the energy supply required for dark-reaction of photosynthesis is very important like this. The difference of algae production between continuous and cyclc irradiation explains that the dark-reaction of photosynthesis acts on algae production as the biggest primary factor. The continuous irradiance on HRABCP made the good algae-production($1403.97{\;}{\mu}g$ Chlorophyll-a/mg) and the good oxygen-production(5.8 mg $O_2/L$) and the good solid-liquid seperation. especially, DO concentration through the oxygen-production was enough to fishes' survival.

  • A serum-free media for CRIP/MFG-LacZ retrovirus production was developed and applied to continuous packed-bed culture system. The serum-free media developed by fractional factorial design contains indulin ($10\mug/mL$), transferrin ($5\mug/mL$), BSA (4 mg/mL), EGF (25 ng/mL), and linoleic acid ($10\mug/mL$). Operation of continuous packed-bed reactor using Fibra-cel enabled the packging cell to stably maintain retrovirus titer for about 1 week. The optimal operation conditions for dilution rate and temperature were 0.67(h(sup)-1) and $32^{circ}C$, respectively. Using this media, the retrovirus titer(cfu/mL) in the packed continuous culture was about 50% that of continuous culture with serum containing DMEM media.

  • For the photoproduction of molecular hydrogen by photosynthetic bacteria in outdoor conditions, we constructed automatically controlled semi-continuous culture system. When the amount of hydrogen gas produced can be measured by a gas meter with a pulse generator, the same amount of substrate consumed for hydrogen production could be supplied by micro pump related with timers. Using the apparatus, we examined hydrogen production with immobilized cells of Rhodopseudomonas sPhaeroides B6 in outdoor conditions. In spite of severe fluctuation of weather and illumination, the culture was maintained under good control with regard to hydrogen productivity. It was possible to automate the semi-continuous outdoor culture of photosynthetic bacteria for hydrogen production.

  • This study was performed to determine optimum conditions of semi-continuous cultivation of chlorella sp. FC-21 cultivated under red light emitting diode (LED). Semi-continuous cultivation was conducted using red LED because red LED was found to be the best light source for chlorella sp. FC-21. During cultivation, phosphate and nitrogen were quickly diminished where cell concentration of chlorella was inversely proportional to the concentrations of phosphate and nitrogen in culture solution. To increase the period of dilution of culture solution, additional amounts of phosphate and nitrogen were inserted in the culture solution to increase the concentrations of phosphate and nitrogen. The cell concentrations of chlorella increased in the modified culture, but cell diameter was diminished as the dilution of culture was periodically conducted. When considered the cell concentration and cell diameter during the cultivation, amount of biomass produced was maintained constant.

Biocatalytic Production of Aldehyde by a Methanol Utilizing Yeast, Hansenula nonfermentans KYP-l Grown in Methanol-limited Continuous Culture

  • Aldehyde production by cells of a methanol utilizing yeast, Hansenula nonfermentans KYP-1 was improved when they were grown in a methanol-limited continuous culture, in comparison with cells grown in a batch culture. A higher cell yield was also obtained in continuous culture than in batch culture. This could be due to the fact that a lower methanol concentration was maintained in the jar fermentor to minimize growth inhibition by methanol. A maximum cell productivity of 0.219 g.$liter^{-1}.hr^{-l}$ and a cell yield of 47% were obtained at dilution rates of 0.1 $hr{-1}$ and 0.06 hr{-1}, respectively. The greatest amount of aldehyde was measured at a dilution rate of 0.08 $hr{-1}$. Under optimum reaction conditions, 915.7 mM of acetaldehyde was produced from 1.5 M ethanol after 21 hours reaction, with a conversion rate of 61%. Propionaldehyde and acrolein were produced with conversion rates of 32.7% and 44%, respectively.

Plasmid Stability and Cloned-Gene Expression in Continuous Culture of Recombinant Escherichia Coli Under Derepressed Condition

  • Continuous culture was carried out with a recombinant Escherichia coli W3110/pCR185, which encodes trp-operon enzymes when the temperature is shifted from $37^{circ}C\;t;42^{\circ}C$. Under derepressed condition of $42^{\circ}C$. plasmlid stability and gene expression were analysed as function of the dilution rate. The stability of plasmid increased with the dilution rate, but maximal levels of gene expression (tryptophan concentration) and plasmid DNA content were obtained at the lowest dilution rate, $0.075\;hr^{-1}$. The plasmid instability, observed at low dilution rates, could be explained by the unbalanced biosynthetic state of the recombinant cell harboring a high copy number of plasmid.

Controlling Mammalian Cell Metabolism in Bioreactors

  • Animal cells in culture typically convert most of the glucose they consume into lactate. The accumulation of lactate, however, is commonly cited as one of the factors that inhibit cell growth and limit the maximum cell concentration that can be achieved in culture. The specific production of lactate and the amount of glucose converted to lactate can be reduced when cells are grown in a fed-batch culture in which the residual glucose concentration is maintained at low levels. Such a fed-batch culture was used to grow and adapt hybridoma cells into a low-lactate-producing state before changing into continuous culture. The cells reached and maintained a high viable cell concentration at steady state. In a similar manner, cells that were initially grown in batch culture and a glucose-rich environment reached a steady state with a cell concentration that is much lower. The feed composition and dilution rates for both cultures were similar, suggesting steady state multiplicity. From a processing perspective the desired steady state among those is the one with the least metabolite production. At such seady state nutrient concentration in the feed can be further increased to increase cell and product concentrations without causing the metabolite inhibitory effect typically seen in a cell culture. Controlling cell metabolism in a continuous culture to reduce or eliminate waste metabolite production may significantly improve the productivity of mammalian cell culture processes.

The Moderated Mediating Effect of Organization Cultural unbalance on the relationship among the Protean Career Orientation, Continuous Learning Activity and Subjective Career Success (프로티언경력지향성, 지속학습활동, 주관적 경력성공의 관계에서 조직문화 불균형성의 조절된 매개효과)

  • This study was conducted to confirm whether organization culture unbalance plays a role as a moderating variable on the mediation process that protean career orientation influences subjective career success through continuous learning activity. To this end, a survey was carried out on 276 office workers with more than 5 years of work experience in large companies, and the data were analyzed using SPSS 25 and Process Macro v3.5. The results showed that continuous learning activity mediates the relationship of protean career orientation affecting subjective career success, but moderating effect of organizational culture unbalance and the moderated mediation effect were not statistically significant. However, statistical significance was found on the moderating effect of organizational culture unbalance on the mediation process, that 'self-direction', protean career orientation's sub-factor, affects subjective career success and its' sub-factor 'employability', and 'career satisfaction' through continuous learning activity. The significance and limitations of our findings are also discussed.

고정상세포분리기의 개발 및 Cyclosporin A 생산을 위한 고정화 연속배양공정에의 적용

  • We have developed an efficient immobilized cell separator for continuous operation of immobilized fungal cell cultures, and applied this separator to actual fermentation process for the production of cyclosporin A (CyA), a powerful immunosuppressant. In the experiments employing highly viscous polymer (carboxymethyl cellulose) solution, the decantor showed good separating performances at high solution viscosites and fast dilution rates. Air duct and cylindrical separator installed inside the decantor turned out to play key roles for the efficient separation of the immobilized cells. By installing the decantor in an immobilized perfusion reactor system (IPRS), continuous immobilized culture was stably carried out even at high dilution rate for a long period, leading to high productivities of free cells and CyA. Almost no immobilized biomass existed in effuluent stream of the IPRS, demonstrating the effectiveness of the decan- tor system for a long-term continuous fermentation. It was noteworthy that we could obtain these results despite of the unfavorable fermentation conditions, i.e., reduced density of the biosupports caused by overgrowth of cells inside the bead particles and existence of high density of suspended fungal cells (10g/l) in the fermentation broth.
