A murine local lymph node assay(LLNA) has been developed as an alternative method to guinea pig maximization test for contact sensitization potential. This study was carried out to investigate the potential use of cytokine producing cells to discriminate between allergens and irritant in the LLNA.(omitted)
Abdominal skin tissues of ICR mouse painted with Dinitrochlorobenzene (DNCB) were observed to investigate the delayed type hypersensitivity of skin by chemical allergen as hapten. The abdominal skin tissues were obtained at hour 48 after secondary DNCB sensitization that were stained by Luna's method for mast cell, and immunohistochemical stain method for IL-2 receptor. The superficial perivascular lymphocytic aggregation were shown in basement membrane after DNCB secondary painting and the large size capillaries in dermis were appeared. The infiltration of lymphocyte to epithelium, the vacuolation of epithelial cell and intercellular space were increased. The number of mast cell in dermis was increased and these shape is degranulation type. The number of IL-2 receptor positive cell was increased in dermis. As results indicated that the hypersensitivy of immune system were induced by DNCB, subsequently to damage evoke inflammation in skin.
This study aims to verify for humans the suitability of the enzyme-fixed hydrogel used for the patch sensor of the blood sugar testing system without blood sampling, which utilizes reverse iontophoresis. Using acrylate monomers, hydrogel was synthesized to which a certain unit of enzyme is fixed. In order to analyze the material property of the synthesized hydrogel, a structural analysis was performed using FT-IR spectroscopy, while the DSC was used to verify the thermal stability. In addition, with the UV-Vis spectrophotometer, it was verified that the degree of active enzyme is at least 50% greater than the standard product. The SEM was used to verify secure fixation of the enzyme onto the surface. As a result, it was observed that the enzyme is successfully fixed to the surface. Since the hydrogel makes direct contact with a patient's skin, it is essential to evaluate the toxicity when making direct contact with the skin. For that purpose, various sets of tests were undertaken according to the ISO 10993-cytotoxicity, intracutaneous reactivity, skin irritation test and maximization sensitization. Consequently, it was successfully verified that the enzyme-fixed hydrogel have bioavailability.

Objectives : Horse oil (HO) has been used long time as the folk medicine of many Asian countries such as Korea, Mongol, China, India and Japan. HO has been used for anti-bacterial, anti-inflammatory, and anti-pruritic purposes in skin. However, it is still largely unknown whether HO modulates the skin condition. In this study, we attempted to evaluate the anti-inflammatory effect of HO on the 1 % of 2, 4-dinitro-1-chlorobenzene (DNCB)-induced contact hypersensitivity in Balb/c mice. Methods : To find the anti-inflammatory effect of HO, contact hypersensitivity, a local inflammatory response of skin, was induced on the back of Balb/c mice by sensitization and repeated application by 1% DNCB and HO treated 2 weeks on the 1% of DNCB-treated Balb/c mice. Excised mice skins were stained with hematoxylin and eosin and serum IgE level was measured by mouse IgE ELISA kit. Results : In this study, we found that HO reduced erythema by 1% of DNCB treated Balb/c mice. Also, HO recovered histopathological features such as the thickening of epidermis, hyperkeratosis and the infiltration of inflammatory cells in 1% of DNCB treated Balb/c mice. In addition, HO reduced IgE level on the serum obtained from blood of 1% of DNCB-treated Balb/c mice. Conclusion : Taken together, these results showed that HO could be used as a pharmaceutical material with anti-inflammatory effects by reducing of erythema, IgE level and recovering of histopathological features skin on DNCB-induced contact hypersensitivity in Balb/c mice model.
Allergic contact dermatitis may be caused by a wide variety of chemicals. A murine local lymph node assay (LLNA) has been developed as an alternative to guinea pig models for assessing the contact sensitization potential of chemical. However, there is a need to develop a nonradioisotopic endpoint for the LLNA, because of the radioisotopic method's requiring the use of special facilities. In this study, we investigated the development of a nonradioisotopic endpoint for LLNA using ELISA (enzyme-linked immunosorbent assay). Female Balb/c mice were treated by the topical application on the dorsum of both ears with four different strong sensitizers, 2,4-dinitrochlorobenzene (DNCB), oxazolone (OXZ), toluene diisocyanate (TDI), and trimellitic anhydride (TMA), and a strong irritant, sodium lauryl sulfate (SLS), once daily for three consecutive days. The proliferation of cells in the auricular Iymph node was analyzed by means of the labelling index (Ll) of bromodeoxyuridine (BrdU) incorporation into cells. The weights of the Iymph nodes in the mice treated with allergens, DNCB, OXZ, TDl and TMA were increased compared to the vehicle control. The stimulation index (Sl) of mice treated with DNCB, OXZ, TDl, and TMA was over three-fold increase compared to the vehicle control. However, the S1 of mice exposed to SLS was not significantly increased compared to the vehicle control, while the lymph node weight of SLS was significantly increased. These results suggest that the LLNA modified endpoint using ELISA based on BrdU incorporation could provide a useful method of screening for irritants and allergens.
To observe the effect of Demodex canis infection on the cellular immune response and hematological profile, 8 Doberman pinschers experimentally infected with D cains and 4 uninfected control dogs were sensitized with 2, 4-dinitro-chlorobenzene(DNCB) on the skin and were challenged with DNCB 14 days after the initial sensitization to elicit allergic contact dermatitis. Histological and hematological changes of these dogs were then observed. Macroscopic changes of skin challenged with DNCB in D canis-infected dogs included significantly reduced area of allergic reaction(p<0.05) than in uninfected control group. Infiltration of inflammatory cells in the D canis-infected group was also significantly reduced(p<0.05) than in the uninfected control group. These changes indicated that the cell-mediated immune response of the animals was suppressed by the infection with D canis. Total white blood cell count in dogs infected with D canis was increased when dogs were sensitized with DNCB (p<0.01). The result appeared to be caused by stress due to D canis infection, secondary bacterial infection and decreased efficacy of general body defense system. Blood eosionophils were increased in D canis-infected dogs which appreared to be caused by the allergic contact dermatitis. Blood chemistry analysis revealed that total protein and globulin were increased(p<0.05), while albumin level was decreased. This result appeared to be caused by secondary bacterial infection.
Self-cleaning fabric is a fabric having a function of decotamination via photodecomposition of photocatalyst or wash-off of contaminants on the superhydrophobic surface. TiO2 is the main photocatalyst for this purpose, but it only functions under UV light which is only a little portion of sunlight, compared to visible light. In this regard, this study aims to investigate Fe-doped TiO2 for improved photodecomposition from visible light sensitization to apply self-cleaning finishing of PET fabrics. Moreover, the Fe-doped TiO2 treated PET fabric was further treated with hexadecyltrimethoxysilane to provide superhydrophobicity on the PET fabrics. As a result of this dual treatment, the prepared fabric exhibited excellent photodecomposition of methylene blue with 96.96% in 12h under sunlight and superhydrophobicity with water contact angle of 166.5° and roll-off angle of 7°. This suggested that the excellent self-cleaning functions can be privided to PET fabric via Fe-doped TiO2 and hexadecyltrimethoxysilane treatment.
Allergic contact dermatitis (ACD) is an inflammatory skin disease and regarded as a prototype of T-cell mediated delayed-type hypersensitivity reactions. Aroma Oil Complex (AOC) is composed of lavender true oil, chamomile roman oil and tea tree oil. This study was performed to assess the effects of AOC in a canine model of ACD. ACD was induced on the back of dogs induced by sensitization and repeated application by 2,4-dinitro-1-chlorobenzene (DNCB). Topical treatment of AOC was applied once a day for 8 days and skin biophysical parameters including transepidermal water loss (TEWL), skin hydration, skin thickness and erythema index, were measured every two days during experimental periods. Histopathology and immunohistochemistry were performed to evaluate the anti-inflammatory effect. In skin biophysical parameters, TEWL, skin hydration, skin thickness and erythema index were significantly increased, with a maximum increase appeared on day 2 (p<0.05). After the completion of AOC treatment, skin biophysical parameters were significantly reached those of baseline in a time-dependent manner (p<0.05). In histopathology, marked increases of epidermal thicknesses were induced after DNCB challenge with numerous inflammatory cell infiltrations and edematous changes, decreases of connective tissue occupied regions in dermis. In addition, marked increases of cytokine - tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interferon-${\gamma}$ (IFN-${\gamma}$)-immunoreactivities in the dermis and of apoptotic markers - caspase-3 and PARP-immunoreactivities in the epidermis were observed in DNCB control as compared with intact control, respectively (p<0.01). The decrease of infiltrated inflammatory cells and related decreases of pro-inflammatory cytokine immunoreactivities were observed in AOC treated skin (p<0.01). Based on these findings, AOC may have anti-inflammatory and alleviatory effects in the allergic contact dermatitis.

Background: Toluene diisocyanate (TDI) can cause contact allergy and occupational asthma, but the mechanism underlying sensitization to this chemical compound remains controversal. Also the correlation of mast cell with contact hypersensitivity (CHS) and the role of mast cell in the TDI-induced CHS is unknown. This issue was investigated by administrating TDI on the skin of genetically mast cell-deficient WBB6F1/$J-Kit^{W}/Kit^{W-v}$ ($W/W^{V}$) and congenic normal WBB6F1/J-Kit+/+ (+/+) mice. Methods: To development of animal model of TDI-induced CHS and to investigate the correlation of mast cell with CHS and the role of mast cell in the TDI-induced CHS, $W/W^V$ and +/+ mice were sensitized with TDI on the back skin at day 1 and day 8, and then challenged with 1% TDI on the ear at day 15. At 1, 2, 4, 8, and 24 hours after 1% TDI challenge, the ear thicknesses were measured. It was investigated the histologic changes of dermis in the ear of $W/W^V$ and +/+ mice at 24 hours after 1% TDI challenge. Results: TDI induced a significant ear swelling response in $W/W^V$ and +/+ mice. TDI induced the significant infiltrations of polymorphonuclear leukocytes and eosinophils in $W/W^V$ and +/+ mice, but not of mast cells in normal mice. And TDI increased a characteristic extent of mast cell degranulation in normal mice. There were no significant differences in the ear swelling and the infiltrations of polymorphonuclear leukocytes and eosinophils of normal versus $W/W^V$ mice, either at baseline or after TDI-induced CHS. Conclusion: From the above results, TDI can be used as a murine CHS model, and the mast cells may not be essential in TDI-induced CHS.

The worldwide prevalence and severity of allergic diseases including atopic and contact dermatitis has increased dramatically over the past decade, especially in developed countries. Mast cells are important effector cells in allergic reactions. The purpose of this study was undertaken to investigate the anti-allergic and anti-pruritic effects of Diospyros lotus leaf extract (DLE). DLE was prepared by extracting with distilled water. In the present study, we investigated the effect of DLE on the production of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\alpha}$) and histamine in rat peritoneal mast cells (RPMCs), and on the skin lesion, leukocyte infiltration and scratching behavior in mice. Phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 significantly increased TNF-${\alpha}$ and IL-$1{\beta}$ production compared with media control. However, TNF-${\alpha}$ and IL-6 production increased by PMA plus A23187 treatment were significantly inhibited by DLE in a dose-dependent manner. DLE also inhibited the histamine release from RPMCs stimulated by compound 48/80, which promotes histamine release. Moreover, DLE administration had an inhibitory effects on the scratching behavior induced by pruritogen (compound 48/80, histamine) in ICR mice. Furthermore, DLE inhibited the skin lesions, inflammatory and mast cells in hairless mice sensitized by 2,4-dinitrofluorobenzene (DNFB). DLE administration reduced the IL-4 and IgE production induced by DNFB sensitization in hairless mice. These results suggest that DLE has a potential use as a herb medicine for treatment against allergy and pruritus-related disease.
